Inhibition of flowering in Lemna perpusilla 6746 by 30 mM sucrose was reversed by the addition of acetylcholine (>$10^{-4}M) supplemented with 10^{-4}M$ ascorbic acid to 1/10-strength Hunter's growth medium. The reversible effect of acetylcholine was found to be greater at early stages of flowering than in the later period. Promotive effects of both acetylcholine ($10^{-3}M) and eserine(10^{-5}M$) on flowering in the short-day plant under various photoperiodic conditions were studied. It was indicated that the application decreased length of the critical dark period for the floral induction, and it was also shown that the endogenous status of acetylcholine was involved in the floral response which had a correlation with phytochrome. Interruption of inductive dark periods by red irradiation (1min) immediately followed by far-red light (1 min) completely inhibited flowering, while the addition of acetylcholine and eserine to the medium under the same condition slightly promoted flowering, indicating possible involvement of phytochrome system in acetylcholine activity for photoperiodic sensitivity of floral response in Lemna perpusilla 6746.
In this study, water hyacinth plants(Eichhornia crassipes), water parsley(Oenanthe javanica) and Lemna paucicostate were used to treat contaminants such as COD, T-N and T-P in far m·fishing village wastewater. The results were as follows : In the batch system experiment, water hyacinth was showed at the high removal efficiency in the 173∼412kgCOD/ha·day concentration that is rather than 260mg/1 of the high concentration. The next is Oenanthe javanica, Lemna. Oenanthe javanica was showed the high removal efficiency in the 96∼173kgCOD/ha·day concentration that is less than 260mg/1 of the low concentration.
Lemnaceae의 반공생적 질소고정(窒素固定)을 확인(確認)하기 위(爲)하여 SEM으로 관찰(觀察), 조사(調査)한 바 다음과 같은 결과(結果)를 얻었다. 1) 반공생적 질소고정(窒素固定)을 하고 있는 암조(藍藻)는 Calthrix sp.와 Microcrate sp.가 많았으나 L. gibba에서는 Anabaena sp.도 보였다. 2) L. oligorhiza는 거의 공생적(共生的) 질소고정(窒素固定)을 하고 있는 것으로 보였으나, 타(他)의 Lemnaceae는 이와같이 보이지 않았다.
좀개구리밥(Lemna perpusilla J. Torrey)은 오염물질의 독성시험에 사용되어 온 대표적인 부유성 수생식물이다. 본 연구에서는 오염된 공장지대나 농경지에서 식물의 생육에 악영향을 미치는 독성물질인 PAHs(polycyclic aromatic hydrocarbons; 다환방향족탄화수소)와 암모늄($NH_4{^+}$)의 식물 독성 평가를 위해 좀개구리밥을 이용하였다. 독성물질인 PAHs와 암모늄이 들어있는 배양액에 좀개구리밥 엽상체를 배양하면서 반응을 조사하였다. 조사결과, 좀개구리밥의 생장은 PAHs의 종류와 농도에 따라 다르게 반응하였다. PAHs중 fluorene의 처리 때 좀개구밥의 생장이 가장 저해되었으며, fluoranthene과 pyrene은 생장 저해 효과가 적었다. fluoranthene과 pyrene의 경우 30ppm정도의 낮은 농도에서는 좀개구리밥의 생장 저해는 거의 일어나지 않았다. 식물색소 함량도 또한 처리한 독성물질의 종류에 따라 다르게 나타났다. Phenanthrene과 fluorene 처리 시 fluoranthene과 pyrene 처리 때보다 chlorophyll의 함량이 많이 감소되었다. 그러나, 항산화 화합물로 잘 알려진 carotenoids의 경우 fluoranthene과 pyrene을 30ppm 이하의 낮은 농도로 처리 시 오히려 함량이 약간 증가하는 것으로 나타났다. 이는 저농도의 fluoranthene 또는 pyrene 처리 시 이들 물질이 좀개구리밥의 황산화 작용을 자극하여 황산화 물질인 carotenoids를 많이 만들게 하는 작용을 하는 것으로 추측된다. 암모늄은 3mM의 낮은 농도에서도 좀개구리밥 엽상체의 생장에 강한 저해효과를 갖는 것으로 나타났다.
A study was conducted to evaluate the nutritive potential value of different aquatic plants: duckweed (Lemna trisulaca), duckweed (Lemna perpusila), azolla (Azolla pinnata) and water-hyacinth (Eichhornia crassipes) from Bangladesh. A wide variability in protein, mineral composition, gas production, microbial protein synthesis, rumen degradable nitrogen and in situ dry matter and crude protein degradability were recorded among species. Crude protein content ranged from 139 to 330 g/kg dry matter (DM). All species were relatively high in Ca, P, Na, content and very rich in K, Fe, Mg, Mn, Cu and Zn concentration. The rate of gas production was highest in azolla and lowest in water-hyacinth. A similar trend was observed with in situ DM degradability. Crude protein degradability was highest in duckweed. Microbial protein formation at 24 h incubation ranged from 38.6-47.2 mg and in vitro rumen degradable nitrogen between 31.5 and 48.4%. Based on the present findings it is concluded that aquatic species have potential as supplementary diet to livestock.
The inhibiotory effect of ethphon on the flowering in Lemna perpusilla 6746 was shown to be related to sucrose concentrations and dilution factors of Hutner's medium. When grown in 1/10-strength Hutner's medium under 10(14) cycles, the plants have been completely inhibited in the floral induction by ethephon (>5ppm) in the presence of sucrose (>20 mM) in the meduim. However, in a less diluted Hutner's medium (1/2-strength), the inhibition of flowering by ethephon was observed to be partially diminished by sucrose at a high concentration (30mM), while a low concentration of sucrose enhanced the inhibitory effect of ethephon in flowering. As inductive dark periodswere extended, the effects of both compounds were partially nullified. Since no significant amount of ethylene possibly released in ethephon decomposition in the medium was detected, the inhibitory effect of ethephon in flowering was postulated to be exerted only through ethylene production within the plants. Plants were incubated in 10 ppm ethephon-containing medium during either dark or light periods, singly or periodically. The most effective single treatment with ethephon was observed during the 4th dark period, when formation of floral stimulus was assumed to be completed beyond a critical level. This postulation can be partially supported by a fact that the plants should be exposed to at least more than four consecutive 10(14) cycles for flowering.
High frequency plant proliferation system via direct frond regeneration of endemic duckweed plants Lemna paucicostata and Spirodela polyrhiza was established. Fronds of L. paucicostata and S. polyrhiza were able to multiply half-strength MS basal medium without plant growth regulators. However, addition of BA at a range of 1 to 3 mg/L was more effective than high concentration of BA treatments for fronds proliferation. Also half-strength MS salts was suitable for the fronds proliferation. Increase of salts concentration had inhibitory effect on fronds proliferation. Also the frequency of callus formation from fronds of L. paucicostata was 3.3%, when they cultured onto 1/2 MS medium supplemented with 1 mg/L of BA. Similarly the frequency of callus formation from S. polyrhiza was very low. After subculture of white globular structures derived from fronds of L. paucicostata, numerous globular somatic embryos and calluses were developed onto the surface of fronds. However these somatic embryos did not fully develop into normal plants when transferred to 1/2 MS basal medium. Therefore direct frond regeneration system was more efficient for mass proliferation of L. paucicostata and S. polyrhiza. The plant regeneration system of L. paucicostata and S. polyrhiza established in this study, might be applied to mass proliferation and genetic transformation for molecular breeding.
The treatment of high-strength swine wastewater by anaerobic digestion combined with an aquatic plant system was investigated. Anaerobic digestion of swine wastewater gave volatile solids (VS) removal efficiencies of 43.3%, 52.1% and 54.5% for hydraulic retention times (HRTs) of 20, 30, 40 days, respectively. The removal efficiencies of VS, total chemical oxygen demand (TCOD) and soluble chemical oxygen demand (SCOD) decreased with increasing VS volumetric loading rate (VLR). Higher organic removal efficiency was observed at longer HRTs for the same VS volumetric loading rate. As VS volumetric loading rate increased, biogas production increased and the methane content of the biogas decreased. Experiments using duckweed (Lemna species) as an aquatic macrophyte gave the following results. In the case of nitrogen, removal efficiency was above 60% and effluent concentration was below 10.0 mg/L when the influent ammonia-N loading was about $1.0\;g/m^2/day$. In the case of phosphorus, removal efficiency was above 55% and effluent concentration was below 2.0 mg/L when the influent $PO_4$-P loading was about $0.15\;g/m^2/day$. In addition, crude protein and phosphorus content of duckweed biomass increased from 15.6% to 41.6% and from 0.8% to 1.6%, respectively, as the influent nutrient concentration increased. The treatment of high-strength swine wastewater by anaerobic digestion combined with an aquatic plant system offers good performance in terms of organics and nutrient removal for relatively low operation and maintenance costs. The results indicate that under appropriate operational conditions, the effluent quality is within the limits set by Korean discharge criteria.
