Preliminary Screening of Some Jeju Island Native Plants for Whitening and Antioxidant Activity (제주도 자생식물들에 대한 미백 및 항산화 효능 탐색)

  • In this study, we investigated the whitening and anti-oxidant activity of 37 Jeju island native plants. The active ingredients of the plants were prepared by methanol extraction. Whitening activity of plant extracts was examined from the inhibitory effect of tyrosinase and the inhibition of melanin synthesis of the B16-F1 cell line. Their anti-oxidant activity was measured by electron donating ability of DPPH (1,1-diphenyl-2-picrylhydrazyl) and ROS (reactive oxygen species) scavenging activity in V79-4 lung fibroblast cells using DCF-DA (dichlorofluorescin diacetate). Cytotoxicity of the extracts on cell s based experiments was investigated by MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) assay. Also, the toxicity test using a rabbit and the human skin patch test were carried out for examining the safety of the extracts which showed the high whitening activity. It is interesting that the extracts of Lespedeza cuneata, Ligustrum lucidum (stem), Morus bombycis (stem) and Prunella vulgaris var. lilacina showed both potent whitening and anti-oxidant activities.

Influence of the Cr-Carbides on the Mechanical Characteristics during Isothermal Heat-Treatment of the Mod.9Cr-1Mo Steel (Mod.9Cr-1Mo강의 항온변태시 기계적 특성변화에 미치는 Cr탄화물의 영향)

  • In this study, mechanical tests and microstructural analyses including TEM analyses with EDX of precipitates in modified 9Cr-1Mo steel were carried out to determine the cause of embrittlement observed after heat-treatment, which limits the usage of the alloy for power plants. Mod. 9Cr-1Mo steel specimens at austenite temperature were quenched to the molten salt baths at $760^{\circ}C$ and $700^{\circ}C$, in which the specimens were kept for 10 min ~ 10 hr with subsequent air-cooling. Impact tests showed that the impact value dropped abruptly when the specimens were kept longer than 30 min at $\sim760^{\circ}C$ reaching to minima in about 1 hr, and then increasing at further retention. The tensile strength of the specimens reached the minimum value without much change afterward, whereas the values of elongation showed the same trend as that of the impact value. The isothermally heat-treated steel at $700^{\circ}C$ also showed a minimum impact value in about 1 hr. These results suggest that the isothermal heattreatment at 760 and $700^{\circ}C$ for about 1 hr induces temporal embrittlement in Mod. 9Cr-1Mo steel. The microstructural examination of all the specimens with extraction replica of the carbides revealed that the specimens with temporal embrittlement had $Cr_2C$, indicating that the cause of the embrittlement was the precipitation of the $Cr_2C$. In addition, TEM/EDX results showed that the Fe/Cr ratio was 0.033 to 0.055 for $Cr_2C$, whereas it was 0.48 to 0.75 for $Cr_{23}C_6$, making the distinction of the $Cr_2C$ and $Cr_{23}C_6$ possible even without direct electron diffraction analyses.

  • In hydrology, it is a difficult task to deal with multivariate time series such as modeling streamflows of an entire complex river system. Normal distribution based model such as MARMA (Multivariate Autorgressive Moving average) has been a major approach for modeling the multivariate time series. There are some limitations for the normal based models. One of them might be the unfavorable data-transformation forcing that the data follow the normal distribution. Furthermore, the high dimension multivariate model requires the very large parameter matrix. As an alternative, one might be decomposing the multivariate data into independent components and modeling it individually. In 1985, Lins used Principal Component Analysis (PCA). The five scores, the decomposed data from the original data, were taken and were formulated individually. The one of the five scores were modeled with AR-2 while the others are modeled with AR-1 model. From the time series analysis using the scores of the five components, he noted "principal component time series might provide a relatively simple and meaningful alternative to conventional large MARMA models". This study is inspired from the researcher's quote to develop a multivariate simulation model. The multivariate simulation model is suggested here using Principal Component Analysis (PCA) and Independent Component Analysis (ICA). Three modeling step is applied for simulation. (1) PCA is used to decompose the correlated multivariate data into the uncorrelated data while ICA decomposes the data into independent components. Here, the autocorrelation structure of the decomposed data is still dominant, which is inherited from the data of the original domain. (2) Each component is resampled by block bootstrapping or K-nearest neighbor. (3) The resampled components bring back to original domain. From using the suggested approach one might expect that a) the simulated data are different with the historical data, b) no data transformation is required (in case of ICA), c) a complex system can be decomposed into independent component and modeled individually. The model with PCA and ICA are compared with the various statistics such as the basic statistics (mean, standard deviation, skewness, autocorrelation), and reservoir-related statistics, kernel density estimate.

Study on Characteristics for Local Deposit of Sediment by Surveying River Bed's Layer History in High Berm of River Channel (하도 층구조 이력조사를 통한 하도내 국지퇴적 특성 분석)

  • More recently, there have been significant changes in the forms of channels due to runoff characteristics driven by climate changes and other alterations in basin/channel environments. Particularly, increasing local deposition in major channels is being observed nationwide. Of such phenomena, it is noteworthy that flood-plains show unidirectional growth and lowering of channels within compound channels in the form of a high-flow plain. These changes are supposed to affect management of the river ecology as well as flood control. In this study, the research on channels in Korea confirmed that the phenomenon of local deposition in those channels is actually taking place, rendering a problem to be urgently addressed. Previous studies on bed changes have been focused on low channels based on bed materials distributed over the channels. However, this research has proved that surface-layer deposition of a high-flow plain is closely related with changes in the conditions of ground surfaces and, ultimately, affects the bed of the entire channel as well. According to the intensive research on the condition of the high-flow plain of the mouth of the Han River, the silt deposited in the high-flow plain was the main cause of settlement/growth of vegetation. And this leads to landforming along with woods-forming, disturbing flood control as well as the normal river ecology.

  • In this study, nucleotide sequence structures of intergenic transcribed spacer (ITS) 1, complete 5.8S ribosomal RNA gene and ITS 2 region were analyzed to identify three Peruvian entomopathogenic fungal isolates. The isolates had highly conserved sequence region in 5.8S rRNA gene and unique sequences in ITS 1 and 2 region among them. 5.8S rRNA gene regions were highly conserved and showed high homoloies among tested isolates. In contrast, ITS region showed species-specific sequence region, resulting in inter-genus differencies. Scanning electron microscopic images of these isolates supported the result of ITS-based identification. From these result, Peruvian entomopathogenic fungal isolate J270, J278, were identified as Beauveria bassiana and J271 was identified as Lecanicillium attenuatum.

Clinical Implications of the Epidermal Growth Factor Receptor Overexpression in the High-grade Astrocytomas (악성 성상세포종에서 표피성장인자 수용체 과발현의 임상적 의의)

  • Purpose : To determine the incidence and prognostic effects of EGFR over-expression in the high-grade astrocytomas. Materials and Methods : With 23 paraffin blocks of the high-grade astrocytomas (7 anaplastic astrocytomas and 16 glioblastoma multiforme), expression of EGFR were evaluated by immunohistochemical staining employing polyclonal antibody raised to short cytoplasmic domain of the molecule. Result : Two out of 7 anaplastic astrocytomas and 9 out of 16 glioblastoma multiforme patients showed overexpression of EGFR(p=0.44). Three out of 11 patients of age below 55 and 8 out of 12 patients of age over 54 showed EGFR overexpression(p=0.141). Median survival of the EGFR negative anaplastic astrocytoma patient was 37 months. Median survival of the glioblastoma multiforme Patients were 11 months in EGFR negative group and 7 months in EGFR positive group. But survival difference was not significant (p=0.17). Conclusion There was a marked trend of increasing overexpression of EGFR in older patients. But survival of the glioblastoma multiforme decreased by the overexpression of the EGFR without significant.

High Throughput-compatible Screening of Anti-oxidative Substances by Insect Extract Library (약용곤충추출물 라이브러리를 이용한 항산화 활성의 초고속 검색)

  • Oxidant stress is well-known for a pivotal parameter related to neuro-inflammatory diseases including Alzheimer's disease, Parkinson's disease, and ALS (Lou Gehrig's disease). In order to effectively screen for anti-inflammatory agents, we first established the infrastructure of high throughput screening for anti-oxidant agents from medicinal insect library extracted with water, methanol, ethanol, and dimethyl sulfoxide. By the screening system, we found that Tenodera angustipennis Saussure, Pyrocoela rupa Olivier and Papilio maackii Mntris had strong anti-oxidant activity. Moreover, Tenodera angustipennis Saussure and Tenodera aridifolia (Stoll) exhibited protection effects of cellular damage by treatment of an oxidant hydrogen peroxide. Together, the results suggest that some selected hits could be a potential agent against neuro-inflammation, although the in vivo studies should be clearly tested.

Changes of Major Constituents by Soaking of Kumquats and Citrus platymama with Ethanol Solution (금귤과 병귤의 침출 중 유용성분의 변화)

  • In order to prepare liqueur of citrus fruit, changes of major constituents, flavonoid pH, color changes, and extract, by soaking 3 kg/6 L kumquats for $1{\sim}70$ days and 1 kg/3 L Citrus platymama for $1{\sim}50$ days in $30{\sim}95%$ ethanol solution were investigated 1.5kg of kumquats, and 1kg of citrus platymama were soaked in 3 L of $30{\sim}95%$ ethanol solution for $50{\sim}70$ days. pH and color changed largely by ethanol concentration. Glucose and fructose were more extracted in $60{\sim}95%$ ethanol concentration. Citric acid and malic acid were extracted $10{\sim}15$ times with kumquats than with Citrus platymama in 30% ethanol solution. Ascorbic acid was more extracted in 60% ethanol solution for kumquats, and in 95% ethanol for Citrus platymamma. The content of ascorbic acid was $3.19{\sim}41.91{\mu}g/mL$ in kumquats, and $21.90{\sim}30.12{\mu}g/mL$ in Citrus platymamma. $312.82{\sim}688.12{\mu}g/mL$ of rutin were extracted in 95% ethanol solution, $9.32{\sim}74.49{\mu}g/mL$ of neohesperidin were extracted in 60% ethanol as for kumquats. Rutin and neohesperidin were more extracted in 30% ethanol concentration contrary to hesperidin. Hesperidin was extracted $38.93{\sim}136.86{\mu}g/mL$ in 95% ethanol solution.

Changes in Fruit Quality of Hallabong Tangor (Citrus kiyomi ${\times}$ ponkan) by Film Packaging during Storage (포장재 처리에 따른 한라봉 감귤의 저장 중 품질변화)

  • We investigated changes in fruit quality of Hallabong tangor (Citrus Kiyomi ${\times}$ ponkan) that was packaged with Si+CaO and LDPE film. The flesh ratio during storage was 74.33% (${\pm}3.66$) to 81.56% (${\pm}1.38$). Firmness of M16A, a variant of Hallabong tangor, was higher 100g-force than that of Hallabong tangor, changes of firmness was not shown among film packages. A fruit juice was $12-14^{\circ}$Brix, and this increased somewhat at the end of storage without film packaging. The titratable acidity of the M16A variety was 0.2% lower than that of the Hallabong tangor. Hallabong and M16A maintained freshness and taste for 120 and 60 days, respectively. The level of reducing sugars in the Hallabong tangor was 1% higher than that of the M16A variety. Reducing sugars increased at room temperature storage without film packaging. Total sugar content was 9.19% (${\pm}2.03$) to 12.78% (${\pm}0.75$). The content of vitamin C declined slowly after 105 days of storage. In conclusion, storage of Hallabong tangor with film packaging coated with Si+CaO was effective for maintaining freshness and quality.

Wahsing Effect of Micor-Bubbles and Changes in Quality of Lettuce (Lacutuca sativa L.) during Storage (마이크로버블에 의한 상추의 세척효과 및 저장 중 품질변화)

  • We assessed quality changes in and washing effects (time and method) on lettuce (Lactuca sativa L.) treated with micro-bubbles. Samples were treated with micro-bubbling for 1, 3, or 5 min, and the 5-min treatment yielded the best results in terms of reduced total microorganism counts, sensory aspects, and degree of washing. Total microorganism counts were 4.30 log colony-forming units (CFU)/g in unwashed lettuce(CT), 4.10 log CFU/g in hand-washed lettuce (HW), 3.98 log CFU/g in conventional, bubble-washed lettuce (BW) and 3.25 log CFU/g in micro-bubble-washed lettuce (MW). In comparison, total counts of samples examined after 10 days of storage were 7.00 log CFU/g for CT, 6.19 log CFU/g for HW, 6.02 log CFU/g for BW, and 5.89 log CFU/g for MW. The lowest counts were seen after micro-bubble treatment. BW and MW samples showed significantly higher counts than did CT and HW samples. In general, BW and MW samples did not vary significantly in count numbers. MW showed a 2.3-fold lower residual pesticide level compared with CT, and also had the lowest level of impurities. HW and BW samples were not well washed.
