Quantitative Mass Spectrometric Analysis of Mixed Self-Assembled Monolayers for Biochips

  • Formation and characterization of self-assembled monolayers (SAMs) on various surfaces are the essential basis for many other applications, including molecular switches, biosensors, microfluidics, and fundamental studies in surfaces and interfaces. To improve the performance at these applications, it is a key to control the quantity of each molecule in various mixed SAMs on the surface. In this study, using mixed SAM of carbamate-based hydroquinone (HQ)-PhBr and11-mercaptoundecanol, the quantitative mass spectrometric method of mixed SAM was developed based on comparison study with XPS and FT-IR methods. In addition, our method was applied to another mixed SAM of biotinylated PEG alkane thiol and 11-mercaptoundecanol for verification purpose. Time-of-flight secondary mass spectrometry (ToF-SIMS) analysis was performed to identify and quantify each molecule of mixed SAM along with principal component analysis (PCA). Since there is no matrix effect in the X-ray photoelectron spectroscopy (XPS) and Fourier transform-infrared (FT-IR) techniques, we compared ToF-SIMS results with XPS and FT-IR results. Because PCA results from ToF-SIMS analysis are well matched with XPS and FT-IR results from both mixed SAMs, we are expecting that our method will be useful to identify and quantify each molecule in various mixed SAMs.

Detection of deoxynivalenol using a MOSFET-based biosensor (MOSFET형 바이오 센서를 이용한 디옥시 니발레놀의 검출)

  • We have detected deoxynivalenol(DON) using a metal-oxide-semiconductor field-effect-transistor(MOSFET)-based biosensor. The MOSFET-based biosensor is fabricated by a standard complementary metal-oxide-semiconductor(CMOS) process, and the biosensor's electrical characteristics were investigated. The output of the sensor was stabilized by employing a reference electrode that applies a fixed bias to the gate. Au which has a chemical affinity for thiol was used as the gate metal to immobilize a self-assembled monolayer(SAM) made of 16-mercaptohexadecanoic acid(MHDA). The SAM was used to immobilize anti-deoxynivalenol antibody. The carboxyl group of the SAM was bound to the anti- deoxynivalenol antibody. Anti-deoxynivalenol antibody and deoxynivalenol were bound by an antigen-antibody reaction. In this study, it is confirmed that the MOSFET-based biosensor can detect deoxynivalenol at concentrations as low as 0.1 ${\mu}g$/ml. The measurements were performed in phosphate buffered saline(PBS; pH 7.4) solution. To verify the interaction among the SAM, antibody, and antigen, surface plasmon resonance(SPR) measurements were performed.

Regulation of Sulfur Metabolism in Cephalosporium acremonium (Cephalosporium acremonium에서 황화합물 대사의 조절)

  • A DL-seleno-methionine resistant mutant, Cephalosporium acremonium MS-92 showed increased activities of sulfate and L-methionine uptake than the parent strain, and accumulated excess methionine and S-adenosylmethionine (SAM) intracellularly. And the sulfate uptake system was severely inhibited by L-cysteine. In crude enzyme extracts, the mutant MS-92 showed lower L-serine sulfhydrylase (identical with cystathionine $\beta$-synthase) activity than the parent. Also, cysteine desulfhydrylase activity, an index of intracellular L-cysteine concentration, of the mutant MS-92 was decreased by about 50% as com-pared with that of the parent. Thus, it was supposed that the mutant MS-92 should have n lower level of L-cysteine than the parent. In C. acremonium like A. nidulans, the enzymes related to the biosynthesis of methionine might be regulated by L-cysteine, but not by methionine or SAM.

고 진공 (UHV) 조건을 이용하여 구리 나노 분말에 도포한 1-octanethiol 기상 자기조립박막(SAMs)의 두께 조절에 관한 연구

  • Alkanethiol (CH3(CH2)nSH) 자기 조립 박막은 금, 은, 팔라듐 그리고 구리와 같은 금속 물질과 결합하여 산화 방지 보호막, 생화학적 멤브레인 그리고 케미컬 센서로 널리 이용되었다. 전도성을 가진 많은 금속 분말 중에서, 구리는 뛰어난 열, 전기 전도성과 풍부한 양으로 다른 귀금속에 비교하여 경제성까지 갖춘 물질이다. 그러나 이러한 구리 나노 분말은 대기에 노출된 구리 분말이 쉽게 산화된다는 결정적인 단점 때문에 그동안 널리 이용되지 못하였다. 이러한 구리의 단점을 극복하고 뛰어난 전도성의 특징을 이용하고자, Langmuir-Blodgett (LB), layer by layer (LbL), electrophoretic deposition (EPD), self-assembled monolayer (SAM)과 같은 구리 나노 분말 위에 유기 박막을 형성하고자 하는 많은 방법이 시도되어왔다. 이러한 방법들 대부분은 습식 방법으로 진행되었으며, 약 2-nm 두께의 SAM 구조를 형성할 수 있음이 많은 연구를 통하여 확인되었다. 그러나 습식 기반의 SAM 구조는 단지 수일 동안만 유효하며, 이는 코팅을 수행하면서 점차 떨어지는 source solvent의 순도와 적합하지 않은 코팅 조건, 그리고 이러한 원인으로 형성된 부실한 막질 구조 때문으로 추측된다. 게다가 이러한 습식 기반 공정은 코팅 막의 두께 조절과 코팅 시 solvent의 순도를 일정하게 유지하는 것이 매우 복잡하고 어려운 작업으로 알려져 왔다. 본 실험에서는 고 진공 챔버 (< $4.0{\times}10-6$ torr) 시스템을 이용하여 습식 기반 공정의 문제점을 극복하고 구리 나노 분말의 산화를 막기 위한 실험을 진행하였다. 1-octanethiol (CH3(CH2)7SH)은 중간 길이의 hydrocarbon (n=7) 구조를 가진 특징 때문에 코팅 물질로 사용되었다. 게다가, alkanethiol 족 특유의 물질인 황(sulfur)은 구리와 결합하여 산화방지 보호막의 역할을 수행할 수 있다. 저 진공 조건에서는 10-nm의 multilayer가 일괄적으로 코팅됨을 확인할 수 있었다. 본 실험에서는 약 10-nm 두께의 자기 조립 박막(self assembled monolayers: SAMs)이 고 진공 조건에서 구리 나노 분말 표면 위에 코팅 조건의 변경을 통해서 5-nm에서 10-nm 두께의 1-octanethiol SAMs 구조를 얻어낼 수 있었다. 이는 고 진공 조건에서 1-octanethiol SAMs의 코팅 두께를 조절함으로 다양한 크기의 분말에 코팅 물질로 쓰일 수 있음을 알 수 있다.

Localization Development of Axial Fan for KM-SAM Multi-function radar (KM-SAM 다기능레이더용 축류형 송풍기 국산화 개발)

  • This paper describes the localization development of an axial fan for KM-SAM multi-function radar. The multi-function radar, which is constantly affected by the external environment, is a key instrument for detecting and tracking low and medium altitude threat targets. Operating this equipment smoothly requires a fan for controlling the internal temperature and humidity. Presently, all such fans are imported. To solve these problems, localization development research was proposed. The development of localization includes analysis of requirements through review of related technical reports such as original equipment and system equipment specification, prototype design, and verification of design requirement through performance test and environmental test. The study results are described. The blower consisted of an axial fan with guide vanes and the motor was designed to generate a maximum airflow of 970 CFM and a wind pressure of 4.8 IWG. Six prototypes were manufactured for performance evaluation. In addition, for reliable data acquisition, AC power supply, fan performance tester and data acquisition equipment were designed and tested. All prototypes were verified as having design requirements equal to or better than those of imports.

Clinical Characteristics of Terminal Lung Cancer Patients Who Died in Hospice Unit (일개 호스피스 병동에서 임종한 말기 폐암 환자의 임상적 고찰)

  • Purpose: The prevalence of lung cancer is increasing continuously these days. We studied clinical characters of the terminal lung cancer patients who had died in hospice units and our study is the basic report for efficient hospice and palliative care to the lung cancer patients. Methods: We retrospectively reviewed the medical records of 129 terminal lung cancer patients who had died in Sam Anyang Hospice Unit from March 2003 to December 2006. The survival days during the hospice and palliative care were analyzed using Kaplan-Meier method of SPSS 13.0. Results: There were 93 males (72%) and 36 females (28%), and median age of patients was 68 years (range $37{\sim}93$). Eighty two patients (64%) took analgesics, the others 47 (36%) not. The most prevalent reason for admission was dyspnea (47 patients, 36%) and it was different from the terminally ill cancer patients being hospitalized because of pain. And the most common symptom was general weakness (103 patients, 80%). One hundred twenty of the paitents (93%) were administered opioid analgesics, and IV morphine shots were mostly used (103 patients, 80%). Sedation was used in 87 patients (67%), and midazolam was mostly used (68 patients, 53%). The median survival in hospice and palliative care was 35 days and the median hospitalization was 24 days. Conclusion: It is very important to manage dyspnea in terminal lung cancer patients. The length of hospice and palliative care for the terminal lung cancer patients is still short. Therefore continuous education and promotion of hospice and palliative care is needed for an effective care for the patients, their families and doctors.

The Effects of Astragali Radix Pharmacopuncture at $CV_{12}$ on Osteoporosis of Senescence Accelerated Mice (SAM) P6 (중완(中脘)($CV_{12}$) 황기약침이 노화촉진 생쥐의 골다공증 개선에 미치는 영향)

  • Objectives : The purpose of this study was to examine the effects of the Astragali radix Pharmacopuncture on improvement of senile osteoporosis (Type 2) in SAM P6. Methods : At 10 weeks after birth, Astragali radix Pharmacopunctures were given 100mL, 200mL/kg/day, i.p. 3times a week for 4 weeks at $CV_{12}$ in SAM P6 mice. We measured complete blood cells (CBC) such as RBC, HGB, Hct, PLT, MPB and MCHC. And we analyzed the plasma concentrations of blood urea nitrogen, creatinine, inorganic phosphate and total iron. In addition, we tested bone mineral density (BMD) using the soft X-ray. Results : The results were as follows. 1. At $CV_{12}$ in SAM P6, treatments of Astragali radix Pharmacopuncture were showed a trend of increase in bone mineral density (BMD) of the vertebrae lumbales, femurand tibia in P6 mice. 2. At $CV_{12}$ in SAM P6, treatment of Astragali radix Pharmacopuncture increased in RBC, HB, HCT and PLT, in comparison with control group. It was also found that the inorganic phosphate levels increased in the treatment on groups of the Astragali radix Pharmacopuncture from that of the control group, but blood urea nitrogen was no significant. Conclusions : These results are suggested that at $CV_{12}$ the Astragali radix Pharmacopuncture help on improvement of osteoporosis in SAMs.

Performance Measurement and Analysis of RTI in the HLA-based Real-time Distributed M-SAM Simulation (HLA 기반 실시간 분산 M-SAM 시뮬레이션에서 RTI성능 측정 및 분석)

  • The HLA is the simulation architecture standard that the civilian and military simulation communities are deeply interested in. We can find various successful practices applying HLA to constructive simulations such as war games in domestics and overseas. However, any case of real-time distributed simulations has not been reported. The reason is that a message transmission period via RTI in a network layer varies according to computing power, simulation nodes, transmission types, and packet size; further a message processing time in an application layer depends on its processing methods, thus too difficult to set up real-time constraints for the enhancement of a real-time resolution. Hence, in this paper we have studied the real-time constraints of RTI for the development of the M-SAM simulator. Thus we have developed a HLA based pilot simulator using 6 PC's in LAN and then measured and analysed the performance of the RTI. As the results of our work, we could obtain the quantitative values for message delay, RTI overhead and RTI packet transmission ratio by a real operation scenario and loads, which are not shown in the previous works. We also expect that the results can be used as a guideline to set up the number of targets, transmission frequency and message processing method in the development of the M-SAM simulator and similar applications.
