Biochemical Properties of Acetylcholinesterase from the Larval Head of Bombyx mori

  • We investigated some biochemical properties of acetylcholinesterase (AChE) in the Bombyx mori larval head. 1% Triton X-100 (v/v) was suitable for extracting AChE from the silkworm larval head but 1 M NaCl was not suitable. PAGE analysis showed a single band of AChE that was detected by histochemical staining using acetylthiocholine as a substrate. AChE was also partially purified with Sepharose 6B and DEAE-cellulose column. Finally, the specific activity of partially purified enzyme solution was 7.6. The study on inhibitor specificity indicated that the enzyme under study was a true cholinesterase (ChE) or AChE. AChE activity was maximum at the substrate concentration of $5{\times}10^{-4}$ M and the excess substrate inhibited the AChE activity. The optimal pH and temperature were pH 7.0-9.0 and 30-35$^{\circ}C$.

Quantitative Analysis of Lignans in the Fruits of Acanthopanax Species by HPLC

  • High performance liquid chromatography (HPLC) was used for the analysis of the lignans eleutheroside B and eleutheroside E in the fruits of Acanthopanax species. A reverse-phase system using a gradient of $H_2O$ and acetonitrile as the mobile phase was developed and detection was at 210 nm. The analysis was successfully carried out within 20 min. The content of eleutheroside B and eleutheroside E in Acanthopanax species was measured in the fruits of A. senticosus (0.58 and $1.66\;{\mu}g/mg$, respectively), A. sessiliflorus (1.15 and $8.49\;{\mu}g/mg$, respectively), A. koreanum (2.16 and $1.80\;{\mu}g/mg$, respectively), and A. divaricatus (1.06 and $7.08\;{\mu}g/mg$, respectively).

The model development and verification for surface branch wood fuels moisture prediction after precipitation during spring period at the east coast region (영동지역 봄철 소나무림에서 강우후 지표연료 직경별 연료습도변화 예측모델 개발 및 검증)

  • In this study, we developed a fuel moisture variation prediction model on each day after precipitation during a spring forest fire exhibition period. For this research, we selected plots in pine forest on Sam-Chuck si and Dong-hae si in Kangwon do according to a forest density(low, mediate, high) and classified a surface woody fuel by a diameter.(below 0.6cm, $0.6{\sim}3cm$, $3{\sim}6cm$, and above 6cm). A validity of this model was verified by applying a fuel moisture variation after precipitation in this spring. In the result, $R^2$ was $0.76{\sim}0.92$. This model will be a useful for improvement of a forest fire danger rate forcast through a prediction a fule moisture in forest.

  • Poly (vinyl acetate) (PVAc) was used as a precursor of PVA/PVAc (skin/core) bicomponent. In order to investigate the possibility of PVA particles for electrical applications, PVA/PVAc particles were produced with an emulsifier, SDS (Sodium Dodecyl Sulfate) and an initiator, V-50 (2,2'-azobis(2-amidinopropane)digydrochloride). In this study, we investigated the electrical property of PVA/PVAc (skin/core) particles. The hydroxyl group of the PVA/PVAc (skin./core) was confirmed by the analysis of PVAc and PVA/PVAc (skin/core) using Fourier Transform Infrared Spectroscopy (FT-IR). The zeta-potential of the PVA/PVAc (skin/core) and PVAc has similarity; however, charge control agent (CCA) treated PVA/PVAc (skin/core) particles has lower zeta-potential than untreated PVA/PVAc particles. The zeta-potential (negative values) of the PVA/PVAc (skin/core) were enhanced in proportion to the increased concentration of CCA.

  • Development and life history of the Korean native firefly, Pyrocoelia rufa, were investigated throughout the indoor rearing. Average size of egg with an oval shape was 1.7 m and the hatchability of egg of P. rufa was approximately 88.5%. The larvae were pupated at the ${5^th}$ instar and the body length of the matured larvae was 27.1 mm. The total periods of larval stage to the end of the 5$^{th}$ instar were approximately 104.7 days. Average pupal period was 10.3 days and average number of eggs oviposited by a female was 87.3 eggs. The body size of female in the pupa and adult was larger than that of male. The wings of female adult were deteriorated.

  • 문산천 하구와 임진강 장단반도 부근은 일반 하천과는 달리 특이한 하도특성을 지니는 구간이다. 문산천의 종단특성은 전형적인 상시 퇴적구간이다. 유사레짐에서도 실제 문산천 하구와 임진강 반구정 구간의 흐름특성 또한 감조구간으로서 밀물, 썰물에 민감하게 작용하여 거시적으로 유사이동이 정체성을 띠는 구간인 것이 확인되었다. 평면특성으로서는 임진강 반구정 구간에서 만곡하도 구간으로서 그 만곡 정도가 매우 크며(곡률 반경 709 m), 이에 따른 양안의 수위차가 실제 약 30 cm 이상 발생하는 것으로 평가되었다. 그리고 문산천 하구역의 하상경사가 1/5,400으로 임진강의 1/3,000보다 완만하다. 문산천 합류부 부근의 하상재료를 살펴 볼때도 임진강보다 문산천에서 더 작은 입경이 비교적 널리 분포하고 있다. 따라서, 홍수시 특히 조석이 창조기나 정체기에 임진강으로 직각 유입하는 형태를 취하고 있는 문산천 하구에서 문산천 홍수량이 유하하지 못하여 배수영향이 가중되고 임진강의 우안이 상대적으로 높은 홍수위가 역류하고 있다고 추정되었다. 이와 같은 현상들은 평균하상고의 변화에서도 나타나듯이 문산천 하구에서 외견상 하구막힘과 같은 사주 형태가 발달하는 양상에서도 찾을 수 있었다. 결국 문산천 하구에서 수위 및 하상변동량 저감을 위한 궁극적인 대책으로서 새로운 문산천 방수로 건설 및 현 문산천 하구에서의 배수기장 도입이 필요한 것으로 분석 되었다.

  • High Performance Concrete(HPC) has been widely used in high-rise building. The HPC has several benefits including high strength, high fluidity and high durability. However. spatting is susceptible to occur in HPC and HPC also tends to be deteriorated in the side of fire resistance performance at fire. This paper focuses on the analysis of the temperature history and residual compressive strength with finishing material, in order to protect HPC from sudden-high-temperature, which is one of the main reason spatting occurs. Test results show that spalling occurs in all specimens. The most serious spalling took placed in HPC covering fire enduring spray-on material, whose covering thickness is 20mm but temperature history indicates that fire enduring spray effectively protected HPC from fire for more than 2hours. In addition, residual compressive strength ratio of HPC using fire enduring paint was more than $90\%$ of original strength, thus minimizing spatting and indicating significant fire resistance performance.

Zonation and soil factors of salt marsh halophyte communities

  • Background: The structures and soil factors of Suaeda glauca-Suaeda japonica zonal communities and Phragmites australis-S. japonica zonal communities were studied in salt marshes of west and south coasts of South Korea to provide basic data for coastal wetland conservation and restoration. Results: S. glauca community mean length was 67 m and S. japonica community mean length was 567 m in zonal communities, and P. australis and S. japonica community mean length were 57 m and 191 m in zonal communities. Regarding the electrical conductivity, sodium content, and clay contents in Upnae-ri, Shinan-gun, there were significant differences among zonal communities at significance level of 0.05 for two-sided t test. However, other factors were not significantly different. Conclusions: The results indicate that multiple factors such as electronic conductivity, total nitrogen level, clay, and sodium might play important roles in the formation of zonal plant communities of salt marshes.

Isolation of a Cerebroside from Panax notoginseng (삼칠근(Panax notoginseng)으로부터 Cerebroside의 분리)

  • A mixture of cerebrosid was purified from the roots of Panax notogiseng (Araliaceae) and characterized as 1-O-{\beta}-D-glucopyranosyl-(2S,3S,4R,8E)-2-[(2'R)-2‘-hydroxypalmitoylamino]-8-octadecene-1,3,4-triol (Aralia cerebroside) and its 8Z-isomer (1-O-{\beta}-D-glucopyranosyl-(2S,3S,4R,8E)-2-[(2'R)-2’-hydroxypalmitoylamino]-8-octadecene-1,3,4-triol, a major component of poke-weed cerebroside) by means of spectroscopic methods.
