For more convenient and rapidly isolation of Bacillus thuringiensis subsp. israelensis(Bti), 1) heat treatment spore forming bacteria, 2) growth in enrichment culture media for Bacillus sp. and 3) selection of bacteria producing a lecithinase for Bacillus thuringiensis subsp. israelensis, were performed. Spore forming bacteria were counted 4.8 $\times $ 10$^{8}$cells/g soil on NAPGCY media, 9.2 $\times $ 10$^{7}$cells/g soil on NA media, and 3.6 $\times $ 10$^{8}$cells/g soil on NAAC media, respectively. Bacteria producing only a lecithinase were reached at 25.2% on medium contained egg york, bacteria only producing a delta-endotoxin were reached at 23.2% by phase contrast microscope, and bacteria producing a lecithinase & a delta-endotoxin simultaneously were reached at 13.7%. Bacillus thuringiensis which producing a lecithinase and a delta-endotoxin simultaneously among bacteria producing a lecithinase, were reached at 56.5%; A half of Bacillus thuringiensis was produced a delta-endotoxin, but not produced a lecithinase. Among 8 isolates of Bacillus thuringiensis, two strain of Bti which has a mosquito-cidal toxin, were detected by PCR using a specific primer of $\delta $-endotoxin gene from Bti.
현재 Bacillus cereus의 정량 및 정성 분석을 위해서 MYP 배지가 주로 사용되고 있으나 식품공전과 축산물의 가공기준 및 성분 규격 내에서 MYP의 주요 첨가물인 egg-yolk의 농도는 각각 2.5%와 5%로 서로 상이하여 대상이 되는 시료의 유래에 따라 각기 다른 MYP 배지를 제조해야 하는 불편함이 있다. 따라서 본 연구는 MYP 배지에 1%에서 10%로 10개 구간으로 egg-yolk의 농도를 첨가하여 이에 따른 B.cereus의 정량적 특성, lecithinase 활성대의 크기 및 육안 확인이 가장 좋은 구간대에 대한 관능평가를 실시하였다. 결과적으로 정량적 특성과 lecithinase 활성대의 크기는 농도가 증가할수록 비례하지 않음을 알 수 있었으며, lecithinase 활성대에 대한 관능평가는 2%의 egg yolk가 가장 확인이 용이한 것으로 평가되었다. 본 연구를 통해서 MYP에 첨가되는 egg-yolk의 최종 농도가 현재의 2.5%(식품), 5%(축산물)로 다르게 기준 되어 있는 것을 2% 또는 식품공전의 2.5%로 통합하여 개정하는 것이 가능하다고 판단되며 이러한 개정을 통하면 MYP 조성물 중 고가인 egg-yolk 제조 비용을 상당히 줄여 줄 것으로 기대된다.
Seventy-two strains of Pseudomonas aeruginosa isolated from the patients were tested for pigment production, exoenzyme production and antimicrobial susceptibility. In the pigment production test, 23.6% of total 72 strains produced both pyocyanine and pyoverdin. Pyoverdin and pyomelanin producing strains were in 9.7%, and 5.5% produced pyoverdin and pyorubin. Strains producing of all of exoenzyme, protease, elastase and lecithinase were in 5.6%. The most common type of exoenzyme production was both protease and elastase producing. Protease producing strain were 23.6%, Among the 72 strains, 50% produced protease. As the result of antimicrobial susceptibility in the isolated 20 strain, most strain were resistant to sulfamethoxazole(90%), but sensitive to other tested antibiotics more than 60%. The MIC50 and MIC90 level of tested antibiotics to 70 strains were 128 $\mu\textrm{g}$/$m\ell$, 512 $\mu\textrm{g}$/$m\ell$ for KM, 8,256 $\mu\textrm{g}$/$m\ell$ for GM, 8, 128 $\mu\textrm{g}$/$m\ell$ for CPZ, and 8, 64 $\mu\textrm{g}$/$m\ell$ for PIPC respectively.
The 23 serovars of Bacillus thuringiensis strain were commonly gram-positive and motile, formed endotoxin crystals, produced acid and alkali in the KIA media, and acid from glucose, hydrolyzed starch, and reduced nitrate but did not produce H$_2$S, oxidase and indole, did not decompose lysine, ornithine, phenylalanine, malonate, lactose, dulcitol, adonitol, inositol, sorbitol, arabinose, raffinose, rhamnose, maltose, and xylose. Eighteen serovars were positive in the MR tests and 15 in the VP tests. Four serovars used citrate. Five serovars produced urease, 5 $CO_2$ from glucose, 2 DNase, and 15 lecithinase. Twelve serovars decomposed arginine, 11 did sucrose, 2 manitol, and 9 salicin Serovar tohokuensis did not hemolyze, but the others did.

Raw and washed spinaches were tested to evaluate the incidences of Aeromonas hydrophila, Escherichia coli O157:H7, Plesiomonas shigelloides, Pseudomonas aeruginosa, Salmonella spp., Shigella spp., Yersinia enterocolitica, Bacillus cereus, Campylobacter jejuni, Clostridium perfringens, Listeria monocytogenes, and Staphylococcus aureus. Four pathogenic bacteria were isolated from spinach samples, and identified by morphological and biochemical methods, including API and ATB identification systems. Isolates from MacConkey, Cereus Selective, Clostridium Perfringens, and Baird-Parker agar media were in 99.9, 99.8, 99.9, and 97.8% agreements with A. hydrophila, B. cereus, C. perfringens, and S. aureus at the species level, respectively. SET-RPLA revealed, among the five strains of S. aureus isolates, two produced type A enterotoxin. All five strains of B. cereus isolates produced enterotoxin as revealed with CRET-RPLA.
This study was performed to assess the presence of contaminated microorganisms of Escherichia coli, Clostridium perfringens, and Bacillus cereus in the 112 commercial Saengshik products. E. coli was not detected in all the samples, but C. perfringens was detected in 11 products (9.8%). The number of the bacteria was less than 100 CFU/g, which was satisfactory to KFDA microbiological requirement. B. cereus was detected less than $10^2{\sim}10^3$ CFU/g in 7 products and $10^3{\sim}10^4$CFU/g in 13 products out of 25 products. Those detected bacteria from tryptose sulphite cycloserine agar and mannitol egg yolk polymyxin agar showed the typical characteristics of Gram positive and contained lecithinase, which can decompose egg-yolks layers in the biochemical test. Therefore, much more attention must be applied to satisfy the B. cereus requirement for Saengshik products sold in the market.
Bacillus thringiensis strain 108 was isolated from soil and had nematicidal activity against second stage juvenile of plant root-knot nematode, Meloidogyne incognita. The strain 108, a rod shape, spore forming and Gram positive bacterium, produced lecithinase, catalase, and ${\delta}$-endotoxin. The strain 108 belongs to H serotype 3, Bacillus thuringiensis var. kurstaki. A nematicidal substance of the strain 108 was partially purified on Sephadex G-25 gel filtration, activated carbon adsorption, silica gel adsorption, and Sephadex G-10 gel filtration. $LC_{90}$ of the partially purified substance against M incognita was $1.2\;{\mu}g/ml$. The nematicidal substance was stable by heat treatment at $100^{\circ}C$ for 1hr, but was perfectly lost nematicidal activity after autoclave ($110^{\circ}C$, 30 min).
The aim of the present research program was to develop a strain of actinomycetes producing antifungal substance. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying selective agar for actinomycetes. Among over 440 isolates, a strain (NA-4803) producing antifungal substance against Trichophyton spp. Nannizzia otae and Pyricularia oryzae was selected. The strain NA-4803 was identified as strain similar to Streptoverticillium blastmyceticum with respect to morphological and physiological characteristics, lecithinase and lipolytic activity, degradation of organic compounds, resistance to antibiotics and utilization of carbon and nitrogen sources. But it showed some differences such as positive reaction of nitrate reduction, negative reaction of L-tyrosine degradation, resistance to cephaloridine, and utilization of I -rhamnose and inulin. The strain NA-4803 was named as Streptoverticillium sp. NA-4803.
To investigate the effects of lactic acid bacteria administration on fecal microflora and putrefactive metabolites in human being, Lactobacillus acidophilus and Bifidobacterium longum powder (1.5 $\times$ 10$^{9}$ cells, respectively) was administrated to six healthy volunteers (average 28 years old) twice a day for 2 weeks. During the administration of lactic acid bacteria, the numbers of bifidobacteria, lactobacilli, and enterococci in feces were increased significantly, whereas those of Staphylococcus and lecithinase-negative Clostridium were decreased considerably. In addition, a number of anaerobic Bacteroides were increased. However, the contents of fecal ammonia and putrefactive metabolites (indole, skatole, p-cresole) were not changed during the administration.
The present study was conducted to investigate the prevalence, the biochemical properties and the enterotoxin types of Staphylococcus aureus in healthy human and patient. A total of 61 S. aureus strains were isolated from 142 samples. The prevalence of S. aureus isolated from healthy human and patient were 17.7% and 14.0%, respectively. All the isolates showed the production of coagulase, lecithinase and hemolysis (${\alpha}-hemolysis;\;32.8%,\;{\beta}-hemolysis;\;67.2%$) on sheep blood agar. Coagulase type VII (38.4%) and type 111 (26.0%) were dominant among coagulase types I through VIII. Twenty-four (52.2%) of 46 strains tested produced one or more enterotoxin; enterotoxin A, Band C were produced by 3, 9 and 12 strains, respectively. Enterotoxins were produced by 100% of type 11 strains, 75% of type 111 and 39.1 % of type VII. Commonly, coagulase type II produced enterotoxin B or C, and type VII produce enterotoxin C.
