  • This experiment was conducted to investigate the effects of starch addition on the fermentative quality and dry matter digestibility(DMD) of Kudzu(Puerari~ thzmbergii Bentham) silage. The herbages was ensiled by the conventional methods in small plastic silo of 7.5 liters with addition of starch of 0 %, 2 %, 4 74, 6 % and 8 %, respectivery. The samples of kudzu silage were separated into leaf and stem, and was determined the pH, organic acid and characteristics of fiber such as neutral detergent fiber(NDF), acid detergent liber(ADF) and acid detergent lignin(ADL). The DMD of leaf and stem silage were evaluated by pepsin-cellulase technique method. The energy values(tota1 digestible nutrients, TDN; digestible energy, DE; metaboliz;~bie energy.   • These studies were conducted to investigate the changes of starch, sugar and nicotine content in relation to maturity and cultural conditions and correlation between growth characteristics and chemical components in order to improve the leaf quality. The results were summarized as follows; 1. Nicotine content, total nitrogen content and the ratio of total nitrogen to nicotine of the green leaf were increased with heavier fertilizer, wider planting space and lower topping, whereas starch, total sugar and the ratio of sugar to nicotine were decreased. 2. Starch content, sugar content and the ratio of total sugar to nicotine(T-S/Nic) in the cured leaf were decreased with milder planting space, less application of MH and delaying harvesting date. 3. It was estimated that 58.1% of starch of the harvested leaf was converted into sugar of the cured leaf, and also they were significantly correlated each other.

  • According to the content of sugar and starch of each positional leaf sheath and internode at heading and 4 weeks after it using IR667-Suwon 214 (high yielding var. having tropical Indica parantage) and Jinheung (local leading var. temperate Japonica) rice grown in various cultivation seasons the suitability of grouping into the high sugar type (sugar>starch), sugar, tendency (increasing tendency in sugar content), high starh type (starch>sugar) and starch tendency (increasing tendency in starch) in carbohydrate metabolism was reexamined as follows. 1. Sugar tendency appeared strongly in IR667 than Jinheung, internode than leaf sheath, late season cultivation than early season, 4 weeks after than heading and high temperature than low temperature. Thus at heading, leaf sheath and internode of Jinheung in early and late season cultivation were high starch type, and lower internode in early season cultiattion and leaf sheath and internode in late season for IR667 were high sugar type. In very late season all internodes of both varieties except 1st internode of Jinheung at heading were high starch type. At four week after heading all leaf sheaths except 1st and 4th one of Jinheung and all internodes were high sugar type. High sugar type was intensified 4 weeks after heading in leaf sheaths than in internodes of IR667 in early season and of both varieties in late season. 2. The upper three leaf sheaths and internodes seem to work in the same way for carbohydrate translocation. Among them upper ones showed sugar tendency at heading and starch tendency 4 weeks after heading and it was clear in Jinheung. 3. The later the cultivation season, the higher the carbohydrate content (sugar+starch), and such tendency was clear 4 weeks after heading and in IR667, suggesting teanslocation inhibition by low temperature. 4. Grain filling rate (weight increase per day) was more rapid in early season cultivation and IR667 took shorter days to reach maximum rate. 5. The later the cultivation season, the greater the percent contribution of carbohydrate before heading to yield and it was always greater in IR667, a leaf sheath type. 6. Sugar and starch ratio appears to be determined principally by metabolic characteristics of variety according to growth process and secondly but considerably by environmental factors.

  • This experiment was conducted to clarify the effect of pyroligneous liquor on endogenous substances that were GA (gibberelic acid)-like substances, starch, and protein of Neofinetia falcata cultured in vitro. When seedlings of Neofinetia falcata were treated with several concentrations (0, 0.5, 1.0, 2.0, 3.0 ml/L) of pyroligneous liquor, the growth of seedlings was enhanced in 1.0ml/L pyroligneous liquor added media. The activity of GA-like substances was lower in the leaf, but higher in the and root than that of the control. The content of starch in 1.0ml/L pyroligneous liquor was lower in the leaf, but higher in the root than that of the control. And the content of protein in 1.0ml/L pyroligneous liquor was also lower in the leaf, but higher in the root than that of the control.

Effects of Leaf and Pod Removal on Assimilate Translocation in Soybean Plants (적엽 및 제협처리가 콩의 동화물질 전류에 미치는 영향)

  • Effects of leaf and pod removal on changes in dry weight and on the contents of soluble sugar, starch, protein and oil in leaves and seeds of soybean [Glycine max(L.) Merr.] cultivar ‘Hwangkeumkong’ were measured at the research farm of Korea University in 1992. The upper 40% and lower 60% of leaves and pods were subjected to treatments at the growth stage of beginning pod(R3). Upper leaf-lower pod removal showed the highest leaf and the lowest seed dry weights. Soluble sugar content was no different among treatments in leaves and seeds. The highest starch content was found in leaves of upper leaf-lower pod removal. Protein content was higher in lower leaves than upper leaves and the lowest in seeds of lower leaf-upper pod removal which had the highest oil content in leaves and seeds. These results apparently indicated that photoassimilates were mobilized from upper leaves to lower seeds, and protein sources were moved from lower to upper parts but weak in remobilization from leaves for the long distance translocation during the reproductive growth period.

  • Starch contents in healthy and powdery mildew-infected spring barley leaves at first and fifth leaf stages were measured at different intervals after inoculation. Different patterns of starch accumulation in the susceptible cultivar Peruvian and the adult plant resistant cultivar Asse were found in infected first and fifth leaves. At the early phase of infection, the amount of starch in infected first leaves was slightly changed in both cultivars. During colony development and sporulation, the decrease in starch content was .more marked in the susceptible Peruvian, whereas the amounts of starch in the adult plant resistant Asse increased during colony development and then decreased sharply before and after sporulation. In heavily infected first leaves, the susceptible cultivar Peruvian showed a drastic decrease in starch content. In the adult plant resistant cultivar Asse, the higher amount of starch retained in these infected leaves, regardless of infection intensity. The regulation of starch accumulation in mildewed barley plants at different leaf stages was discussed in relation to adult plant resistance of spring barley.

Changes in the Expression of ADP-Glucose Pyrophosphorylase Genes During Fruit Ripening in Strawberry

  • Starch contents play important roles in determining the fruit quality. Stawberry accumulates starch in the early stages and then mobilized into soluble sugars during fruit ripening. To date the molecular studies on the ADP-glucose pyrophosphorylase (AGPase), a key enzyme of starch biosynthesis, were not reported. cDNAs encoding small (FagpS) and large (FagpL1 and FaspL2) AGPase subunits were isolated from strawberry (Fragaria ${\times}$ ananassa Duch. cv. Niyobou). Both FagpS and FagpL1 cDNAs have open reading frames deriving 55-58 kDa polypeptides, where FagpL2 contains a partial fragment. Sequence analyses showed that FagpS has a glutamate-threonine-cysteine-leucine (ETCL) instead of a glutamine-threonine-cysteine-leucine (QTCL) motif found in all the dicot plants except for Citrus. In fruits, FagpS and FagpL1 were expressed in all stages with a little change in the amounts of transcripts. In the case of FagpL2, we were not able to detect any signal from all stages of fruit development and all tissues except for very a weak signal from the leaf. The results indicate that FagpL1 and FagpL2 show ubiquitous and leaf-specific expression patterns, respectively. The studies suggest that the starch contents in strawberry might be controlled by the expression of AGPase gene at both the transcriptional and post-transcriptional levels during fruit development.

Characterization of Carbohydrate Metabolism during Dark-Induced Senescence

  • To investigate the changes of carbohydrate metabolism in the senescing leaves of Zea mays during dark-induced senescence, the changes in the contents of reducing sugar, sucrose and starch as well as the activities of sucrose synthase, three isozymes of invertase, and ${\alpha}$-amylase were measured. In the senescing leaves, the content of reducing sugars temporarily increased at 4 d and rapidly decreased thereafter, whereas sucrose contents gradually decreased thereafter, whereas sucrose contents gradually decreased until 3 d of senscence and significantly decreased thereafter. The activities of intracellular invertases such as soluble acid and alkaline formed gradually enhanced until 4 d of leaf senescence and significantly declined thereafter. The extracellular invertase activity showed no significant changes during leaf senescence. The deactivation of sucrose synthase was observed within 3 d of leaf senscence. On the other hand, the starch contents gradually declined during 2 d of leaf senescence, and showed a temporary increase at 3 d, which is similar to the pattern of sucrose synthase activity., These results imply that sucrose in the senescing leaves. The major enzymes which correlated to the breakdown of sucrose during dark-induced senescence were soluble acid and alkaline invertases, not sucrose and ABA accelerated leaf senescence by inducing the accumulation of reducing sugar. These result, therefore, that leaf senescence may be mediated by the temporary quantitative changes of reducing sugar induced by the activation of intracellualr inveertases.

Carbon-Nitrogen Transport in Response to Control of Leaf-Pod Ratio in Soybean (콩의 엽-협 비율 조절에 따른 탄소와 질소의 전류)

  • Effects of translocation on seed yield and carbon-nitrogen compounds at five leaf-pod ratios of soybean [Glycine max. (L.)Merr. ] culti bars, 'Paldalkong', 'Baekunkong', and 'Danyeobkong' were measured. The upper 50 and lower 50% of leaves and pods were subjected to treatments at growth stage R3. Three soybean cultivars showed the similar trends on changes in dry matter accumulation and on the contents of soluble sugar, starch and protein in seeds among the treatments. Mean stem dry weight was increased with upper leaf-lower pod and lower leaf-upper pod removals, and decreased with upper leaf and lower leaf removals. Leaf dry weight was appeared higher at the upper leaves among the treatments. Seed numbers and dry weights were decreased with leaf and leaf-pod removals, and were higher in lower part of the plants. Soluble sugar and starch contents in seeds were also showed slightly higher in lower part. Protein content of seeds was decreased in upper part with upper leaf removal and in lower part with lower leaf removal, however, that of the upper seeds was the highest with lower leaf removal. The results of this study are assumed that carbon and nitrogen compounds were translocated opposite directions and protein source was weak in remobilization for the long distance transport during the reproductive growth period of soybean plants.

Identification of the quantitative trait loci for breaking and bending types lodging resistance in rice, using recombinant inbred lines derived from Koshihikari and a strong culm variety, leaf star

  • To develop rice cultivars with increased biomass and grain yield, superior lodging resistance is an essential trait. The new breeding approach can be adopted for the improvement of stem lodging resistance by enhancing culm strength. The resistance to breaking type lodging is attributed to bending moment of basal culm (M), which is composed of the section modulus (SM) and bending stress (BS). The resistance to the bending type lodging is attributed to flexural rigidity (FR) of stem, which is composed of the secondary moment of inertia (SMI) and Young's modulus (YM). Starch and cell wall components such as cellulose, hemicellulose and lignin also play a significant role in physical strength of culm, and thus affect lodging. Leaf Star has a superior lodging resistance due to its thick and stiff culm because of its high M and FR compared with Koshihikari. Furthermore, Leaf Star contains high densities of hemicellulose, cellulose and low lignin density in culm compared with Koshihikari. In this study, we performed QTL analysis for these traits associated with culm strength, using 94 recombinant inbred lines (RILs, $F_8$), derived from a cross between Leaf Star and Koshihikari. The SM in the RILs showed a continuous distribution. QTLs for SM were detected on chrs.2, 3 and 10. Leaf Star alleles increased SM on chrs. 2 and 3, but Koshihikari allele increased on chr.10. These QTLs overlapped with those QTLs identified using backcrossed inbred line derived from a cross between Chugoku 117 and Koshihikari, the parents of Leaf Star. The FR in Leaf Star was higher than that in Koshihikari due to the larger SMI and YM. 3 QTLs for SMI were detected on chrs.2, 3 and 10. Leaf Star alleles increased SMI on chrs.2 and 3, and Koshihikari alleles increased on chr.10. One QTL on chr.3 and two QTLs on chr.5 for hollocelulose content were detected with Leaf Star alleles contribution. Moreover, two QTLs were detected for hemicellulose density on chrs.3 and 5. Leaf Star allele increased hemicellulose density on chr.5, and Koshihikari allele increased on chr.3. Furthermore, two QTLs for cellulose density were detected on chr.5, and one QTL on chr.2. For starch content, one QTL on chr.3 and two QTLs on chr.5 with Leaf Star alleles contribution were detected. TULK-6 carrying a chromosome segment of Leaf Star on chr.5 in the Koshihikari genetic background showed higher densities of starch and hemicellulose than those in Koshihikari. These results suggest that the detected QTLs for culm strength could be utilized for the improvement of lodging resistance in rice by marker-assisted selection.
