Isolation and Identification of Burkholderia pyrrocinia CH-67 to Control Tomato Leaf Mold and Damping-off on Crisphead Lettuce and Tomato

  • A bacterial strain CH-67 which exhibits antagonism towards several plant pathogenic fungi such as Botrytis cinerea, Fulvia fulva, Rhizoctonia solani, Sclerotinia sclerotiorum, Colletotrichum sp. and Phytophthora sp. was isolated from forest soil by a chitin-baiting method. This strain was identified as Burkholderia cepacia complex (Bcc) and belonging to genomovar IX (Burkholderia pyrrocinia) by colony morphology, biochemical traits and molecular method like 16S rRNA and recA gene analysis. This strain was used to develop a bio-fungicide for the control of tomato leaf mold caused by Fulvia fulva. Various formulations of B. pyrrocinia CH-67 were prepared using fermentation cultures of the bacterium in rice oil medium. The result of pot experiments led to selection of the wettable powder formulation CH67-C containing modified starch as the best formulation for the control of tomato leaf mold. CH67-C, at 100-fold dilution, showed a control value of 85% against tomato leaf mold. Its disease control efficacy was not significantly different from that of the chemical fungicide triflumidazole. B. pyrrocinia CH-67 was also effective in controlling damping-off caused by Rhizoctonia solani PY-1 in crisphead lettuce and tomato plants. CH67-C formulation was recognized as a cell-free formulation since B. pyrrocinia CH-67 was all lethal during formulation process. This study provides an effective biocontrol formulation of biofungicide using B. pyrrocinia CH-67 to control tomato leaf mold and damping-off crisphead lettuce and tomato.

Studies on Differentiation of a Paddy Weed, Bur Beggarticks(Bidens tripartita L.) (논 잡초(雜草) 가막사리(Bidens tripartita L.) 생태종(生態種)의 분화(分化)에 관(關)한 연구(硏究))

  • Variation of morphological and physiological traits of 50 Bidens tripartita accessions were studied and the accessions were grouped through cluster analysis based on four major characters; plant type, leaf partition, achene length, days to flowering. Bidens tripartite accessions have shown significant variations in plant type, stem length, days to flowering, leaf shape, leaf partition, chlorophyll content, leaf color, stem color, achene color, achene length and achene shape. Most of Bidens tripartite accessions appeared to have strong dormancy and also photodormancy with some exceptions. Plants could be classified into 5 types from straight(I) to triangle(V), and intermediate diamond type(III) was prevalent. The plant type score has negative correlation with the stem length. None, three, and five part leaved plants were observed and most of them were three or five parted. Leaf partition had negative correlation with achene length and chlorophyll content. Average days to flowering was 108 days in the range of 94~141 days. It had positive correlation with achene length and leaf shape and negative correlation with achene color. Average achene length was 10.0mm and it had positive correlation with achene shape, stem length, days to flowering and leaf shape. It also had negative correlation with leaf color, stem color, achene color, leaf partition. Bidens tripartite accessions could be divided into identifiable six groups from the cluster analysis at the distance 0.06 using Ward's minimum-variance method.

Growth and heteromorphism of Leaves along Leaf Rank (Plastochron) of Woody Plants (목본식물의 엽순에 따른 잎의 생장과 이형성)

  • The growth, size and morphology of leaves, which foliated on the same shoot and time in the early growth season, were studied in 12 woody species in temperate deciduous forests of Korea, and the equation for the index of heteromorphism of leaves was fonnulated. The numbers of leaves per shoot were from 3 to 7 in early growth season and the numbers of synchronous leafing were from 2 to 6. All the leaves on a shoot were reached maximum values of specific leaf arells at the SIlme time nearly. The differences of the length (L) and breadth (B) of leaves along leaf rank (plastochron) were conspicuous, except for Quercus variabilis. Especially, in 10 species, the ~fferences of the neighbouring leaves were significant at 0.001 or 0.05 level. The index of leaf heteromorphism was calculated from the data based on $L\;{\times}\;B$ and LIB, and similar to differences of actual leaves along leaf rank. The values of index of heteromorphism were high in Carpinus tschonosldi, Styrax obassia, Ulmus davidiana, Rhamnus yoshinoi and Symplocos chinensis for. pilosa, and low in Sorbus alnifolia and Tilia amurensis. The index of heteromorphism had no relation to other characters of the leaf or life fonn of the species. It was thought that the differences along leaf rank were related to the degree of adaptation to environment, especially to changing air temperature in early growth season.season.

Agronomic Characteristics and Chloroplast Morphology of a Pale-green Leaf Line in Rice (벼 연녹색잎 유전자계통의 농업형질 및 엽록체 구조)

  • The leaf color varies with the contents of pigments, especially chlorophylls and carotenoids. Teichung 65 (T.65), a japonica rice, with pgl(pale-green leaf) gene exhibits pale green color on the whole plant from seedling to harvest. This study conducted to evaluate the agronomic characters and examine the chloroplasts of 'pgl' plants in parents and BC$_1$F$_2$ of T.65(pgl) xSuweon ${345}^2$. The average grain yield of pale-green-leaf individuals in F$_2$ was the same as T.65(pgl) but that of green-leaf individuals was much higher than that of Suweon 345. The contents of chlorophyll a(Ca), chlorophyll b(Cb) and total chlorophyll content(Ct) of T.65(pgl) in flag leaf were lower than those of Suweon 345, but the Ca/Cb ratio of T.65(pgl) was higher than that of Suweon 345 during from 15 days after heading (DAH) to 60 DAH. The SPAD value of T.65(pgl) in flag leaf was lower than that of Suweon 345, but that in the second and the third leaves was similar to that of Suweon 345. The SPAD value of pale-green-leaf individual group was lower than that of green individual group in upper three leaves. The structural difference of chloroplasts in flag leaf between T.65(pgl) and Suweon 345 through TEM at 20 DAH was not detected, but the number of osmium granules in chloroplast of T.65(pgl) were higher than that of Suweon 345.

  • Ditylenchus acris was isolated from diseased strawberry plants. Frequently, D. acris and Aphelenchoides fragariae occur together in a strawberry plant. Both species appeared very similar in the shape, length, swimming behavior and causing symptoms, and difficult to distinguish them by a stereomicroscope. But they were easily distinguished under a compound microscope especially by their tail shape, median bulb, vulva position, and bursa.

  • This study was made on three taxa of Chrysanthemum that grew on the Ewha Womans University campus on the basis of gross morphology, pollen epidermal patterns and chromosomes. The three taxa were recognized as Chrysanthemum zawadskii subsp. zawadskii var. acutilobum(2n=54); Chrysanthemum zawadskii subsp. latilobum (2n=36), and possible hybrid(2n=45) between the two taxa in view of leaf morphorlogy and chromosome number counts.

Arcuphantes catillus n. sp., a new spider species (Araneae: Linyphiidae) from Korea

  • Arcuphantes catillus n. sp., a new species of the genus Arcuphantes Chamberlin and Ivie, 1943 is described from Korea. The present species is distinguishable from its known similar congeners by a conspicuous proximal process and a blunt lateral process of the paracymbium, feathery tipped lamellar extension of the pseudolamella, and bowl-like posterior part of radix. The present new species was collected from the leaf litter layer of mixed forest on a hillock around rice fields with a pitfall trap.

Morphological Characteristics and Growth Rate of Medium-Leaf Type Zoysiagrasses Collected at Major Sod Production Area in S. Korea (국내 잔디 주 생산지역에서 수집된 한국잔디류의 형태적 특성 및 생육속도)

  • Field experiments were conducted to evaluate the morphological characteristics and growth rates of 101 medium-leaf type zoysiagrasses (Zoysia spp.) collected at the major sod production area (Jang Seong Gun) in South Korea. Collected lines with distinctive morphology and visual growth rate were planted in plastic pots and measured morphological characteristics under the plastic house conditions. Variation of leaf width, plant height, leaf angle, length of leaf sheath, trichome, stolon length, and color were measured. Six lines were selected by evaluating growth rates from one hundred one collected lines. Eight standard cultivars and three other superior lines previously collected were compared to 7 selected lines form Jang seong area by checking growth rates and morphological characteristics. Average leaf blade width was 3.4 mm, leaf angle was 45.8 degree, plant height was 21.6 cm, height of lowest leaf was 5.0 cm, and length of leaf blade was 14.1 cm. Ground cover rates of selected lines 'CY6097' and 'CY6069' were 70% and 68.3%, respectively. These are believed to be faster than 60% ground cover rate of zoysiagrass 'Anyang', and also, twice as faster than the 31.7% ground cover rate of Z. matrella. Selected line 'CY6069' showed fast growth rate with shorter internode length (5.1 cm) compared to zoysiagrass 'Anyang'. Based on the results of this study, we could select useful fast growing zoysiagrass breeding lines from the major sod production area (Jang Seong Gun) in Korea.

Leaf Morphological Characteristics and Variation of Sorbus alnifolia (Sieb. et Zucc.) K. Koch in 11 Natural Habitats (국내자생 팥배나무 11개 천연집단의 잎 형태적 특성과 변이)

  • This study was conducted to provide a basic data such as leaf morphological characteristics, total 110 individual trees selected from 11 wild population, for selective breeding. As a result of investigation of the twenty morphological characteristics of the leaf, there were statistically significant differences in all leaf characteristics among the populations. Especially, Mt. Mani population had larger leaf length (LL), width (LW) and area (LAR) than other populations. On the other hand, Mt. Beakwoon and Mt. Duryun had smaller leaf size (LL, LW and LAR) among the populations. Its principal component analysis (PCA) results showed that it represented 72.9% accumulated explanation from three principal component. The characteristics such as leaf area, leaf length and leaf width were highly contributed for classification among populations. According to the cluster analysis, the natural S. alnifolia populations were classified into five groups and Mt. Mani population was different from the other population.

Characteristics of Leaf Morphology, Vegetation and Genetic Variation in the Endemic Populations of a Rare Tree Species, Koelreuteria paniculata Laxm (희귀수종(稀貴樹種) 모감주나무 자생집단(自生集團)의 잎의 형태적(形態的) 특성(特性), 식생특성(植生特性) 및 유전변이(遺傳變異))

  • Characteristics of leaf morphology and vegetation were examined in 6 populations of rare endemic tree species, Koelreuteria paniculata Laxm. Additionally we studied genetic variation employing isozyme marker. Nested anova showed that there were statistically significant differences among populations as well as among individuals within populations in all 8 leaf morphological traits. In 7 of 8 traits, variance components among populations were larger than those among individuals within populations. Only a few tree species were found within each population and all the populations seemed to have been disturbed by man. Level of genetic diversity for this rare endemic species was very low, especially compared with those of geographically widespread tree species(A/L=1.1, P=9.5%, $H_o=0.021$, $H_e=0.035$) and degree of genetic differentiation was high($F_{ST}=0.114$). Factors contributing to the low level of genetic diversity and high level of genetic differentiation might include genetic drift, inbreeding and limited gene flow. Finally, implications for the conservation of genetic diversity of Koelreuteria paniculata were discussed.
