For determining the insecticidal effect of Carbofuran on the Brown planthopper, Nilaparvata lugens, sucking toxicity by drenching application, sucking and contact toxicity by leaf dipping application, and contact toxicity by topical application were examined. Drenching caused two types of mortality patterns. One was logarithmic curve at a relatively high concentration (8~30 ppm) with over 40% mortality in 24 h, and the other was an S-shaped curve at low concentrations (1~4 ppm) with over 60% mortality on the fifth day after Carbofuran treatment. Leaf dipping application caused a rapid increase in mortality in a day, and this effect decreased steadily with time. Topical application showed steep increase in mortality in a day, and hardly increased thereafter. The best mortality evaluation time for the drenching application was the second day (42 h), and that for the leaf dipping and topical applications was the first or second day after Carbofuran application. When the insecticide has systemic effects, drench application provides the best efficacy and its insecticidal effects persist for a longer time than any other application method.
Mixture study of bifenthrin and prothiofos was conducted to control insecticide resistant diamondback moth (DBM), Plutella xylostella, which had shown 581, 18, 19, 11 fold resistance to fenvalerate, cypermethrin, furathiocarb, and prothiofos in Chinju strain, respectively and 38 and 9 fold resistance to fenvalerate and furathiocarb in Seosang strain, respectively. Optimal mixture ratios of bifenthrin and prothiofos was selected against Seosang strain of DBM by leaf dipping method in laboratory and by field test. Maximum co-toxicity coefficient by dipping method was shown at a ratio of 1:50 mixture of bifenthrin 1EC and prothiofos 50EC in active ingredient(a.i.) by 243.2 and then suddenly decreased. The mixed formulation from the dipping test was examined at the cabbage field of Seosang, Hamyang. Although insecticides were reduced to half, control efficacy was similar to farmer's conventional method. In addition, the mixture combination was also more effective against Chinju DBM population which had very high level of resistance to several representative insecticides. Mixture methods of insecticides will be effective countermeasures to the resistance problem of pests.
Effects of BA treatment and cut planting methods on shooting and seedling characteristics were investigated to increase seedling products by cuttings of lacquer tree. Dipping on 1,000mg/L BA solution was better than dripping with same solution at 5 weeks after cut planting. As shooting rate was increased suddenly, it was 85% at 10 weeks after cut planting. Among 3 dripping treatments, interval was not effective on shooting rate. Dripping per 2 days with 100 ㎕ of 1,000 mg/L BA solution was most effective on seedling height and leaf number and than dipping was more effective than dripping per 5 and 7 days. Branch developed on near part from dripped scion. Shooting of horizontal planting was earlier about a week and rate was increased suddenly from 3 to 6 weeks, it was highest with 96% than others at 10 weeks after planting. Seedling of horizontal cut planting was tallest about 40cm and branch developed lower than vertical and oblique cut planting. In case of horizontal cut planting, 2-3 shoots were formed on different part of scion and than those were could divided to seedling, so horizontal cut planting was most effective to propagation of seedling than any others.
In order to examine the factors affecting herbicidal injury of pepper, crop injury and growth response of 2 red pepper cultivars and 2 sweet pepper cultivars to napropamide and pendimethalin were evaluated under defferent conditions of soil texture, organic matter, soil temperature and seeding depth in the greenhouse. Growth response of 4 pepper cultivars was also examined by roos dipping to napropamide and pendimethalin. More inhibition of top leaf growth by root dipping to napropamide was occurred in red pepper cultivars than in sweet pepper cultivars. However, sweet pepper cultivars showed more severe inhibition of top leaf growth by root dipping to pendimethalin compared to red pepper cultivars. Crop injury due to napropamide and pendimethalin was more severe in sandy soil than in loam soil and this trend was more remarkably shown in sweet pepper cultivars. Crop injury due to napropamide and pendimethalin was reduced with the increase in organic matter especially in cv. Walgeykwan and cv. Orient al Pimento. As seeding depth of pepper cultivars became deeper, crop injury due to napropamide and pendimethalin was reduced in cv. Walgyekwan and cv. Oriental Pimento.

After treatment with imidacloprid, there were clear differences in the time to the first reaction of Myzus persicae among the concentrations treated. The time taken for the proboscis of the aphids to penetrate, during the recording plants increased as the imidacloprid concentration increased. Imidacloprid concentration inflow into a leaf was investigated using high-performance liquid chromatography, and the residues of the imidacloprid varied slightly with the different concentrations treated. However, the inflow rates of this insecticide into a leaf increased as the dipping times increased. Furthermore, it was shown that there was no relationship in inflow concentration between the concentrations and times of treatment. However, the concentration in the leaf differed according to the dipping time. Judging from the fact that the first reaction behavior against imidacloprid displayed at an inflow concentration of 0.32-0.35 mg/L, we concluded that inflow concentrations causing the first reaction of the aphids to the insecticide were much lower than the concentration treated. The general feeding characteristics of the aphids indicated that xylem and/or phloem feeding behavior continued after a series of probing behaviors and stylet activity during the first 3 h from the start of EPG recording. After 90 min treatment with imidacloprid, feeding behavior over the next 30 min indicated a significant increase in the withdrawal of the stylet from the plant at all treated concentrations. Xylem and/or phloem feeding patterns were significantly decreased during this time. In particular, the proportion of xylem feeding differed according to the concentration of imidacloprid.
Background: Glycosylation of natural compounds increases the diversity of secondary metabolites. Glycosylation steps are implicated not only in plant growth and development, but also in plant defense responses. Although the activities of uridine-dependent glycosyltransferases (UGTs) have long been recognized, and genes encoding them in several higher plants have been identified, the specific functions of UGTs in planta remain largely unknown. Methods: Spatial and temporal patterns of gene expression were analyzed by quantitative reverse transcription (qRT)-polymerase chain reaction (PCR) and GUS histochemical assay. In planta transformation in heterologous Arabidopsis was generated by floral dipping using Agrobacterium tumefaciens (C58C1). Protein localization was analyzed by confocal microscopy via fluorescent protein tagging. Results: PgUGT72AL1 was highly expressed in the rhizome, upper root, and youngest leaf compared with the other organs. GUS staining of the promoter: GUS fusion revealed high expression in different organs, including axillary leaf branch. Overexpression of PgUGT72AL1 resulted in a fused organ in the axillary leaf branch. Conclusion: PgUGT72AL1, which is phylogenetically close to PgUGT71A27, is involved in the production of ginsenoside compound K. Considering that compound K is not reported in raw ginseng material, further characterization of this gene may shed light on the biological function of ginsenosides in ginseng plant growth and development. The organ fusion phenotype could be caused by the defective growth of cells in the boundary region, commonly regulated by phytohormones such as auxins or brassinosteroids, and requires further analysis.
Methanol and hexane extracts from 35 species in 27 families of herbal plants were evaluated for their acaricidal activities against two-spotted spider mite, Tetranychus urticae by leaf-dipping and spraying methods in laboratory, pot and field, respectively. Acaricidal activities were different depending on herbal plants. When T. urticae was released on leaves of Phaseolus vulgaris var. humilis that were dipped in the 1,000 ppm hexane extracts from fruits of Torreya nucifer and Daphan genkwa, seeds of Xanthium strumarium and Pharbitis nil at one minute they experienced 56.8, 47.8, 47.7, and 47.7% mortalities, respectively. Dipping time influenced acaricidal activity, that is, one minute dipping was more effective than 30 second dipping of Phaseolus vulgaris var. humilis leaves. However, extracts of X. strumarium and P. nil seeds had comparatively high activities. Concentration of hexane extract of X. strumarium and P. nil seeds revealing LC50 were 1,824 and 1,899 ppm, respectively. Extracts of both plants from hot and cold water were not effective representing <20% mortality at 1,000 ppm. However, hexane extracts of X. strumarium and P. nil seeds were effective against T. urticae on P. vulgaris var. humilis representing 76.3 and 71.3% mortalities in pot, respectively. Control effects of hexane extract of P. nil seed were 50.8 and 35.1% at 2,000 and 1,000 ppm against T. urticae on Chrysanthemum morifolium in greenhouse, respectively.
Turnip mosaic potyviruses (TuMV) were isolated from Rorippa indica and Armoracia lapathifolia showing mosaic symptoms in field. Identification of the TuMVs were carried out by host reactions of indicator plants, electron micrograph, serological properties and reverse transcription-poly-merase chain reaction (RT-PCR). Both viruses systemically infected Chenopodium quinoa, Nicotiana clevelandii, Brassica rapa, B. campestris subsp. pekinensis, B. juncea and Raphanus sativus, and developed local infection on inoculated leaves of C. quinoa, C. amaranticola, C. album, N. tabacum cv. Xanthi nc and Gomphrena grobosa. However, the viruses did not infect on N. glutinosa, Cucumis sativus and Vigna unguiculata. The filamentous particles, about 720 nm in length, and inclusion bodies were observed from the infected leaf tissues by dipping on electron microscopy. Crude sap of leaf infected with the viruses was reacted positively with an antiserum of TuMV in agar gel double diffusion. For detection of the viruses, RT-PCR was carried out with TuMV--specfic oligonucleotide primer. The RT-PCR products, a 1,092 bp DNA fragment, were obtained from naturally infected leaves of R. indica and A. lapathifolia. In inoculation test to seven cruciferous weeds with TuMV, infection occurred in Arabis glabra, Barbarea orthoceras, Capsella bursa-pastoris, Draba nomorosa var. hebecarpa, Rorippa cantoniensis and Thlaspi arvense.
The root-out of mulberry saplings has been studied by pour into soil and spray on leaves of the plant growth substances. In addition, the effect of "Rutin", a kind of plant growth hormones has been also studied on promoting the rootability of mulberry scions by dipping the scions at the various concentrations. The results are summarized as follows: (1) It is likely to be effective on acceleration of the rootability resulting in increase of rooting ratio by pour into soil at the concentration of 1 ppm and 10 ppm NAA, 10 ppm and 100 ppm Rutin, and 5000 fold solution of Atonic, respectively (2) Growth of branch is only promoted by leaf spray of 0.5% urea, 0.005% and 0.01% Rutin, and 5000 fold solution of Atonic without increasing the root weight. (3) It seems that 0.05% of Rutin is of practical use, and 0.05% to 0.4% of "Rutin" accelerates the root-out of mulberry scions as well as NAA does.
Tolclofos-methyl applied as seed dipping at 1,000 ppm for 3 hrs before sowing and soil drenching at the rate of 300 g ai./10 a in the middle of April protected emerging seedlings of Panax ginseng from damping-off caused by Rhiiutonia solani(AG2-1) in Yangjik Soil artificially infested with the pathogen. Germination rates with tolclofos-methyl, pencycuron, and control were 53.7%, 45.8%, and 7.5%, respectively, while the rate of the seeds at non-infested soil was 62.6%. The effectiveness of Tolclofos-methyl against the pathogen in the soil lasted upto 32 days in vitro. However, the transpiratio of ginseng seedlings increased greatly with chemical treatment, showing 0.02, 0.12, and 0.24 m1/cm2 leaf area/day at 0, 1,2, and 4 ppm a.i. of the fungicide, respectively.
