Food Organisms of juveniles of Chaenogobius mororanus Inhabited at Intertidal Zone of the Western Coast of Korea

  • To investigate the food organisms of the Chaenogobius mororanus during the juveniles stage, the stomach contents of fish, captured in the intertidal zone of Chungchong- nam-do Sochon-gun Su-myon Dodun-ri (Fig. 1) between on early June from end of May 2003, were observed. Total length of the juveniles of C. mororanus was 5.0 mm ${\sim}$ 8.4 mm size, and the participation rate of feeding was 71.4%. Main food organisms were such as copepods, decapods, polychaete larvae, amphipods and these occupied more than dry weight 3.9%. Copepods among them dominated the most quantitys by average 62.0%, and next, food organisms appeared much by order of decapods, polychaete larvae and amphipods etc.. Therefore, most important food organisms of juvenile stage of C. mororanus were copepods, decapods, polychaete larvae, amphipods etc.

Gonadal Sex Differentiation of Hatchery-Reared Longtooth Grouper (Epinephelus bruneus)

  • For the gonadal sex management of younger longtooth grouper (Epinephelus bruneus), this work investigated the timing and histological process of ovary differentiation and oocyte development of longtooth grouper larvae and juvenile. Specimens (from 1 to 365 DAH) were collected for gonadal histological study from June 2008 to August 2009. Rearing water temperature was ranged from 20 to $24^{\circ}C$. The primordial germ cells could be observed from 10 to 15 DAH, while undifferentiated gonad occurs from 20 to 50 DAH in longtooth grouper. The initial ovarian phase was 60 to 110 DAH with the formation of ovarian cavity and the increased in size of gonad. The ovarian phase started at 140 DAH with appearance of oogonia. The gonad at 365 DAH appeared to have full of oogonia and primary growth stage oocyte. Formation of ovarian cavity indicates that the ovarian differentiation beginning at 60 DAH in longtooth grouper. The gonads in longtooth grouper differentiated directly into ovaries in all fish examined.

사할린가자미(Limanda sakhalinensis) 자치어의 골격발달 (Osteological Development of the Larvae and Juvenile in Sakhalin Sole Limanda sakhalinensis)

  • This study is designed to observe the development of skeleton of Sakhalin sole Limanda sakhalinensis in order to use the findings as the basic data for the taxonomic research. As for the development of skeleton, on 20 days of the hatch when its average total length was 5.86 mm, the clavicle and the paraspenoid in the cranium were ossified. As for the jaw bone, the premaxillary and the dentary were ossified. On 28 days of hatch when its total length was 7.05 mm, 25 neural spines and 22 hemal spines were ossified and 5 fin rays appeared. On 34 days of hatch when its average total length was 8.40 mm, the end of tail was twisted by $45^{\circ}$ and 3 hypural bones were ossified. On 48 days of hatch when its average total length was 10.1 mm, 2 actinosts below the postcleithrum were ossified. On 54 days of hatch, when the average total length was 10.4 mm, the ethmoid, prootic and exoccipital were ossified, thus completing the ossification of skeleton.

다슬기 (Semisulcospira libertina libertina)와 곳체다슬기 (Semisulcospira gottschei)의 출산 유도 (Parturition Induction on Melania Snails, Semisulcospira libertina libertina and Semisulcospira gottschei)

  • 난태생종인 다슬기(Semisulcospira libertina libertina)와 곳체다슬기(Semisulcospira gottchei) 어미에게 여러 가지 물리 화학적 자극을 주어 출산 개시 소요 시간, 출산 유생 치패수 및 치패 출산율을 조사하였다. 곳체다슬기는 온도, 공기 노출, serotonin 및 acetylcholine 자극에 대하여 다슬기보다 더욱 민감하게 반응하였으나, $H_2O_2$$NH_4OH$ 자극에 대하여는 두 종 모두 반응하지 않았다. 다슬기는 acetylcholine $10^{-9}M$ 자극에서 모패당 68개체로 가장 많은 유생과 치패를 출산하였으며, 치패 출산율도 57.5%로 높았다. 곳체다슬기는 수온을 $9^{\circ}C$ 상승시켰을 때, 모패당 113개체의 유생과 치패를 출산하였고 치패 출산율은 56.3%였으며, acetylcholine $10^{-12}M$ 첨가했을 때는 각각 83개체, 61.7%였다. 결론적으로, 다슬기와 곳체다슬기의 인공 종묘 생산 시 치패의 대량 출산 유도에는 신경전달물질인 acetylcholine 자극이 효과적인 것으로 나타났다.

볼락, Sebastes inermis 자.치어의 성장에 따른 소화효소 활성 변화 (Developmental Changes in Digestive Enzymes Activity of Black Rockfish Sebastes inermis)

  • 본 실험은 볼락 종묘를 안정적으로 대량 생산하는 데 필 수적인 자치어기의 성장과 소화효소인 trypsin, pepsin-like enzyme, amylase 그리고 lipase의 activity를 측정하여 소화생리에 대한 기초 자료를 제공하고자 수행하였다. 산출 직후 자어의 체장은 $5.01{\pm}0.22 mm$이었던 것이 산출 후 100일에는 $36.01{\pm}1.22 mm$ 까지 성장하였고, 습중량은 산출 후 70일 전후로 급격히 증가하였다. trysin 활성은 산출후 2일째 처음으로 확인되어 $7.00{\pm}1.48$ unit을 나타내고 산출후 66일 $1425{\pm}230$ unit으로 peak를 나타내었다. pepsin-like enzyme 활성은 산출 후 11일째 처음으로 확인되었고 산출후 66일째 $902{\pm}160$ unit까지 급격히 증가하였다. Lipase 활성은 산출후 2일째 $4.5{\pm}1.4$ unit으로 확인되었고 산출후 47일계에는 $38.3{\pm}0.4$unit로 peak를 나타내었다. Amylase 활성은 산출후 11일째 $0.18{\pm}0.11$ unit으로 처음 확인되었고 산출후 50일째 $23.48{\pm}5.11$ unit까지 급격히 증가하였다. specific activity는 trypsin이 산출후 14일째, pepsin-like enzyme이 61일째 peak를 나타냈다. 볼락의 체중이 현저하게 증가하는 시기에 맞춰서 각 효소와 전활성이 급격히 상승하는 것을 확인 할 수 있었다. 이것은 이시기가 위선이 기능적으로 역할을 하는 시기와 일치하므로 단백질 소화능력의 비약적인 증가가 체중 증가에 크게 관여하였음을 나타낸다. 그러므로 본 실험에서 확인 된 체중이 급격히 증가하는 시기는 볼락 양식에 있어서 사료 전환기 즉, 생물사료에서 배합사료로 전환하기에 적당하다고 할 수 있겠다.

여수 연안에서 채집한 양태(Platycephalus indicus)의 난발생 및 자치어 형태발달 (Morphological Development of Eggs, Larvae and Juveniles of Platycephalus indicus Caught from Yeosu)

  • 여수 연안에서 채집한 양태(Platycephalus indicus)의 난발생과 자치어 형태발달을 연구하였다. 건식법을 통해 인공수정한 수정란은 구형 형태의 분리부성란이었으며, 직경 1.03~1.12 mm (mean =1.08 mm, n =50)였다. 수온 20.0℃에서 50시간 30분 만에 부화하였으며, 전기 자어의 전장은 2.72~3.04 mm (mean =2.93±0.21 mm, n =50)였다. 부화 5일 후 전장 3.88~4.42 mm (mean =4.11±0.31 mm, n =15)였으며, 난황을 모두 흡수하였고, 이빨이 발달하였다. 부화 39일 후 치어기 도달하였으며, 전장은 10.23~11.95 mm (mean =11.09±0.86 mm, n=5)였다. 부화 45일 후 전장은 12.01~13.25 mm (mean=12.63±0.62 mm, n=5)였으며, 체형, 체색이 모두 미성어 시기와 유사하였다.

Early Life History of the Sevenband Grouper, Epinephelus septemfasciatus from Korea

  • This study is conducted to monitor the morphological developmental features of the egg development, larvae and juvenile of Epinephelus septemfasciatus, the fertilized eggs were gotton using artificial insemination. Matured parents are collected from marine caged fish farms in Geomun-ri, Samsan-myeon, Yeosu-si, Jeollanamdo Korea in June 2012. The fertilized eggs were pelagic eggs containing one oil globule, and measured 0.81~0.89 mm ($0.85{\pm}0.04mm$, n=50) in diameter. In regard to rearing environment, the water temperature is $21.0{\sim}23.0^{\circ}C$ and the salinity is 32.0~33.2‰. Hatching was observed from 48 hours after fertilization, the mouth and anus of prelarvae was not opened but had egg yolk at newly hatched. 4 days after hatching, the mouth and anus of postlarvae was opened and began to eat Rotifer and was measured 2.40~2.49 mm ($2.45{\pm}0.03mm$ n=10) in total length. 12 days after hatching, postlarvae was measured 3.77~4.67 mm ($4.27{\pm}0.33mm$) in total length, its the second pole tide of dorsal fin and the first pole tide of pelvic fin was extended longitudinally. 71 days after hatching, juvenile was measured 40.5~45.4 mm ($42.6{\pm}2.04mm$) in total length. Seven bands were observed in body, and pole tides of dorsal and pelvic fins were shortened.

한국산 남방돛양태[Bathycallionymus kaianus (돛양태과)] 자치어의 분자 동정 및 형태 기재 (Molecular Identification and Morphological Description for Larvae and Juveniles of Deepwater Dragonet Bathycallionymus kaianus (Callionymidae, PISCES) from Korea)

  • Dragonet fish (Callionymidae), living in benthic upper 900 m of all subtropical, tropical and temperate oceans, comprises 200 species in 20 genera worldwide, of which 18 species in 6 genera occur in Korea. Although dragonet fish plays an important role in linking between top predators and lower trophic levels, there are only few studies about their early life history. Herein, we present molecular and morphological data on larvae and juveniles of Bathycallionymus kaianus (Günther, 1880) collected from the Korean waters. During 2016 to 2018, one preflexion larvae [2.69 in total length (TL)], three flexion larvae (3.65 and 4.77 mm TL), six postflexion larvae (6.07 and 7.94 mm TL), and three juveniles (10.81 and 12.26 mm TL) were collected in the East Sea, Korea Strait, and Jeju Island using Bongo net. Of them, 13 individuals were identified through molecular markers (COI or 16S rRNA) and morphologically described. The larvae of B. kaianus are well distinguished from other species of Callionymidae in melanophore distribution, body shape and development of preopercular spine. It was very similar to larvae of two Repomucenus species, R. valenciennei and R. virgis, but was clearly distinguished in melanophore distribution, preopercular spine development, and head shape.

Study on the Seed Production of Olive Flounder Paralichthys olivaceus in a Pilot Scale Recirculating Aquaculture System

  • Seed production of olive flounder Paralichthys olivaceus was performed in a pilot RAS. The growth of juvenile olive flounder and changes in water quality were monitored for the entire production period. The pilot RAS consisted of 8 circular culture tanks($4.0mD{\times}1.0mH$), 2 trickling biofilters($1.7mD{\times}2.0mH$), 2 protein skimmers ($0.8mD{\times}2.5mH$), and 4 sedimentation chambers($0.7mD{\times}1.5mH$). The culture surface area was about $100.5m^2$ and the actual working volume was about $106.9m^3$. As many as 300,000 fertilized olive flounder eggs were initially distributed into 2 culture tanks with the water temperature at $19.0^{\circ}C$. Live feeds such as rotifers and Artemia nauplii were fed until the 32nd day after hatching, and a commercial diet was fed from the 19th day to the end of the experiment. After 70 days, 150,256 juveniles with a body length of $65.8{\pm}3.9mm$ were produced in the RAS, with a daily growth rate for body length of 4.7%/day. At this time, the final culture density was 1,495 individuals $m^{-2}$, and 13.6 L of makeup water, 0.071 kW of electricity and 0.025 L of diesel fuel were used to produce a juvenile olive flounder. During metamorphosis of the larvae, the TAN concentration increased to 0.99 mg/L, which made the larvae sensitive to result in some mortality. However no more massive mortality occurred at the juvenile stage after metamorphosis even at a TAN concentration of 4.25 mg/L and a ${NO_2}^{-}-N$ concentration of 2.45 mg/L.

수온과 염분이 양태 자치어의 성장과 생존에 미치는 영향 (Effects of Water Temperature and Salinity on the Growth and Survival of Larvae and Juvenile of Platycephalus indicus)

  • 실험구의 수온은 13, 16, 19, 22, 25℃로 설정하였고, 염분농도는 각각 7, 14, 21, 28, 32 psu로 설정하였으며, 200 L 플라스틱 원형수조에 부화 자어를 각각 500마리씩 수용하였다. 성장은 수온 25℃(21.62±0.14 mm TL)와 염분 28 psu (15.02±0.05 mm TL)에서 가장 높았고, 수온 13℃ (7.04±0.05 mm TL)와 염분 7 psu에서 가장 낮았다. 생존율은 수온 22℃(69.2%)와 염분 32 psu (84.1%)에서 가장 높았으며, 수온 13℃(15.1%)와 염분 7 psu에서 가장 낮았다. 양태는 강어귀 또는 연안 해역에 서식하여 채집이 가능한 어종으로 다른 해수 어종에 비해 염분 내성이 강하였다.
