Structural Characteristics of Low Molecular Weight Laminarin Prepared by Ionizing Irradiation (이온화 방사선 조사에 의해 얻어진 저분자 laminarin의 분자구조 특성)

  • Recently, it has been reported that low molecular weight laminarin had the enhanced biological activities. In this study, molecular structure of low molecular weight laminarin prepared by ionizing irradiation was studied. Low molecular weight laminarin samples of 13.5, 8.5, 7, and 6 kDa were obtained from 15 kDa laminarin by irradiation. From gel permeation chromatography data, low molecular weight laminarin was shown to have low polydispersity. To define the changes of functional groups in laminarin with different molecular weights, Fourier-transform infrared analysis was carried out. There was found no significant changes of functional groups in low molecular weight laminarin, except the increase of carbonyl group. The granular fissures from scanning electron microscopy showed the breakage of glycosidic bond in low molecular weight laminarin. These results could be utilized for the investigation of the enhanced biological activities of low molecular weight polysaccharides including laminarin.

Antioxidant Activity of Low Molecular Weight Laminarin Prepared with Gamma Irradiation (감마선 조사된 저분자 laminarin의 항산화 활성 연구)

  • In this study, it was investigated the antioxidant activity of laminarin degraded by gamma irradiation. Because the activities of antioxidants have been attributed to various mechanisms, different assay methods have been conducted and compared. 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power, and lipid peroxidation inhibitory activity of degraded laminarin were measured and compared with non-degraded. All of these results showed that the antioxidant activity of laminarin degraded by irradiation was increased depending on the absorbed dose. Therefore, gamma irradiation could be an alternative method for the preparation of degraded laminarin with higher antioxidant activity.

Application of β-1,3-Glucanase from Pyrococcus furiosus for Ethanol Production using Laminarin (Pyrococcus furiosus의 β-1,3-glucanase를 처리한 laminarin 분해 산물을 이용한 바이오 에탄올의 생산)

  • $\beta$-1,3-glucanase from Pyrococcus furiosus was applied for the saccharification of laminarin, which is a major oligo-saccharide component of brown algae, and the reaction mixture produced from laminarin was utilized as a substrate for alcohol fermentation using yeast. To prepare the recombinant $\beta$-1,3-glucanase, a $\beta$-1,3-glucanase gene was overexpressed in Escherichia coli and purified. Laminarin was degraded to an oligo- and mono-saccharide, such as glucose, after reaction with the purified recombinant $\beta$-1,3-glucanase, and the products after enzymatic treatment were confirmed by TLC and HPLC analysis. Decomposed laminarin after enzyme reaction was only added to the medium as a C-source for yeast alcohol production reaction. 0.3% alcohol production was detected from the cultured broth by gas chromatography after 48 hr of incubation. Further evaluation for optimal conditions of saccharification and alcohol fermentation can be suggested, as well as the possibility of using this enzymatic method to produce ethanol using laminarin.

  • The objective of this study was investigated to apply the fucoidan and laminarin extract from low-grade Undaria pinnatifida to real food product, a pork patty. Pork patties added with fucoidan and laminarin showed lower lipid oxidation and the inhibition against lipid oxidation was shown to be dependent on the irradiation doses. The Hunter color values of pork patty added with fucoidan increased significantly with an increase of irradiation dose. The hardness profiles of the patties with fucoidan and laminarin was decreased, but the amount of water in the patties was increased. Also, the combination of gamma irradiation and addition of fucoidan and laminarin was shown to be effective for the microbiological control. These results suggested that gamma irradiation and fucoidan and laminarin treatment showed the positive effect on microbial stability and quality of the pork patty.

SEASONAL VARIATION IN THE CHEMICAL COMPOSITION OF BROWN ALGAE WITH SPECIAL REFERENCE TO ALGINIC ACID (갈조류의 알긴산 함량의 계절적인 변화에 관하여)

  • The present investigations were carried out with the purpose of making clear the fundamental features of the seasonal variations in chemical composition of the brown algae. Three species of samples, Ecklonia cava, Sargassum sagamianum and Hizikia fusiforme have been analyzed monthly for their contents of total ash, crude protein, alginic acid, mannitol, and laminarin over a year period. Three kinds of samples were collected from the same locality, situated on the southern coast of the Che-ju Island, from September 1966 to August 1967. In addition, the comparative analysis was made on fronds and stipes of the plant for their chemical composition. The results obtained are summarized as follows: 1, In general, the three species examined underwent a similar mode of seasonal variation, and no essential difference was detected among them. 2. The chemical composition of the plant exhibited a considerable difference between the species. The content of total ash in H. fusiforme was remarkably higher than those in the two other species respectively. The alginic acid content was relatively high in S. sagamianum and low in H. fusiforme. The contents of crude protein, mannitol and laminarin were appreciably high in E. cava and low in H. fusiforme in general. 3. The most outstanding feature in the seasonal variation was that, in general, the total ash, crude protein, and alginic acid contents were at a maximum in the winter months while laminarin and mannitol contents were at a minimum. The converse was true in summer. Total ash-Maximum values were observed from December to February and minimum from August till October. Crude Protein-All species exhibited maxima in January and February, and minima from August to October. Alginic acid-Maximum contents occured from January to March and minimum from September to November. Mannitol-The maximum content of mannitol was In May and lune and minimum in January, February and March. Leminerin-Maximum content was in September and October, and minimum in January and February. 4. The wide seasonal variation in chemical constitution occured in the fronds, but the stipes showed a slight seasonal variation. In the chemical composition, the stipe was high in alginic acid, low in mannitol and laminarin. The reverse was in the frond.

Novel substrate specificity of a thermostable β-glucosidase from the hyperthermophilic archaeon, Thermococcus pacificus P-4 (초고온 고세균 Thermococcus pacificus P-4로부터 내열성 β-glucosidase의 새로운 기질 특이성)

  • Based on the genomic analysis of Thermococcus pacificus P-4, we identified a putative GH1 ${\beta}$-glucosidase-encoding gene (Tpa-glu). The gene revealed a 1,464 bp encoding 487 amino acid residues, and the deduced amino acid residues exhibited 77% identity with Pyrococcus furiosus ${\beta}$-glucosidase (accession no. NP_577802). The gene was cloned and expressed in Escherichia coli system. The recombinant protein was purified by metal affinity chromatography and characterized. Tpa-Glu showed optimum activity at pH 7.5 and $75^{\circ}C$, and thermostability with a half life of 6 h at $90^{\circ}C$. Tpa-Glu exhibited hydrolyzing activity against various pNP-glycopyranosides, with kcat/Km values in the order of pNP-${\beta}$-glucopyranoside, pNP-${\beta}$-galactopyranoside, pNP-${\beta}$-mannopyranoside, and pNP-${\beta}$-xylopyranoside. In addition, the enzyme exhibited exo-hydrolyzing activity toward ${\beta}$-1,3-linked polysaccharide (laminarin) and ${\beta}$-1,3- and ${\beta}$-1,4-linked oligosaccharides. This is the first description of an enzyme from hyperthermophilic archaea that displays exo-hydrolyzing activity toward ${\beta}$-1,3-linked polysaccharides and could be applied in combination with ${\beta}$-1,3-endoglucanase for saccharification of laminarin.

Characterization of yeast cell wall lytic enzyme from Fusarium moniliforme (Fusarium moniliforme이 생산하는 효모세포벽 분해효소의 특성)

  • Yeast cell wall lytic enzyme was purified from Fusarium moniliforme by ammonium sulfate fractionation and gel column chromatography. The lytic activity was found to consist of three enzyme activities which were resolved on Sephadex G-100. The first peak on chromatogram exhibited proteolytic, lytic and laminarinase activities, and the second had both lytic and laminarinase activities, whereas the third peak was shown to contain lytic activity only. Three enzyme activities showed the synergistic effect and reducing agents accelerated the yeast roil wall lysis. This indicates that lytic, proteolytic and laminarinase activity acted cooperatively in the lysis of intact cells. Tannic acid precipitate of crude enzyme constituted of three enzyme activities had a high lytic activity on viable yeast cell and has proved useful in yeast protoplast formation.

Production of the Convertible Enzyme of Ginsenoside $Rb_1$ by Rhizopus japonicus (Rhizopus japonicus 에 의한 Ginsenoside $Rb_1$ 전환효소의 생산조건)

  • In ginseng saponin, $ginsenoside-Rb_1$ was contained the most abundantly. But ginsenoside-Rd which is similar to ginsenoside $Rb_1$ in structure was known to be superior to $ginsenoside-Rb_1$ pharmaceutically. A strain of Rhizopus japonicus is able to produce the convertible enzyme which can convert selectively $ginsenoside-Rb_1$ to ginsenoside-Rd without the change of any other ginsenoside. The strain can produce the most enzyme after 5 day-culture on wheat bran medium. The enzyme production was promoted best efficiently by addition of red ginseng powder in ginseng products, xylose in sugars, laminarin in polysaccharides, naringin in flavonoids, and potassium nitrate in nitrogen substrates.

Identification, sequence characterization and expression analysis of the arginine kinase gene in response to laminarin challenge from the Oriental land snail, Nesiohelix samarangae (동양달팽이(Nesiohelix samarangae)의 arginine kinase 유전자 분석 및 발현 패턴에 관한 연구)

  • Arginine kinase (ArK) is known to play an important role in most invertebrates the level of ATP by phosphorylation of phosphagens in cell and immuninty in living organisms. ArK has been identified in many kinds of organisms ranging from invertebrate to vertebrate. However, no ArK gene has been cloned and investigated from N. samarangae. This leads us to identify ArK cDNA (NsArK) from the expressed sequence tag (EST) sequencing of N. samarangae. Sequence analysis indicated that the coding region of 1,065 bp contains 355 amino acid residues. Molecular phylogenetic analysis shows that NsArK had very high similarities with mollusca and arthropoda. In an attempt to investigate a potential role of NsArK in the digestive gland of N. samarangae, expression patterns were analyzed. RT-PCR analsysis shows that NsArK mRNA is induced in the rane of 1.2 fold at 6 hr by laminarin when compared with the control. The immunnologial and physiological role of NsArK remains to be further investigated in N. samarangae.
