본 연구는 포도가공부산물로부터 유용성분을 추출하여 자원의 활용과 환경오염을 줄이고, 공정을 간결하게 하여 실제산업현장에서 이용할 수 있는 공정의 개발을 목적으로 하였다. 포도가공 부산물을 포도과피와 포도씨로 나누어 과피에서는 anthocyanin을 씨에서는 oligomeric proanthocyanidin (OPC)를 주 대상물질로 하여 추출하기 위한 공정을 개발하였다. 추출된 항산화물질은 HPLC를 이용하여 정량하였으며, 이들의 효능은 DPPH법, SOD법을 이용하여 대조군들과의 비교를 통하여 확인하였다. 두 방법 모두 포도씨 추출물의 효능이 포도과피 추출물보다 뛰어난 항산화도를 보였다. 이렇게 생산된 추출물을 이용하여 multiple lamella emulsion의 화장품 제형을 개발하였고 이 제형의 안전성과 안정성을 실험적으로 확인하였다.
This study was intended to observe the changes of pectic substances and enzyme activities, texture, microstructure of anchovy added kimchi during fermentation for 4 weeks at 4$^{\circ}C$. Content of alcohol insoluble solid(AIS) and HCl soluble pectin(HClSP) were decreased, whereas content of hot water soluble pectin(HWSP) was increased during fermentation. Content of HClSP was higher and HWSP was lower in anchovy added kimchi than control. Activity of pectinesterase(PE) was decreased, whereas activity of polygalacturonase(PG) was increased during fermentation. In anchovy added kimchi, PG activity was lower than control. Changes in microstructure of Chinese cabbage and kimchi during fermentation was lower than control. Changes in microstructure of Chinese cabbage and kimchi during fermentation was observed ; in the raw cabbage, parenchyma cells, intercellular space and middle lamella were clearly shown. But in salted cabbage, middle lamella became separated. In the late stage of fermentation, parenchyma cell walls were wrinkled and collapsed. Puncture forces of kimchi were decreased, whereas cutting forces of kimchi were increased as fermentation proceeded. The firmness was slightly higher in anchovy added kimchi than control at the late stage fermentation, which may be explained by the PG activity.
오래전부터, 많은 여성들은 자신들의 젊음을 연장하고, 외모를 더 아름답게 가꾸고, 이를 죽을 때까지 유지하는 것을 바래왔다. 이에 의사와 약사들뿐만 아니라, 많은 화장품 연구자들도 노화와 관련된 기술개발에 총력을 기울여 왔다. 따라서, 이들 연구자들은 노화방지를 위한 새로운 원료를 찾고, 이를 안정화하고, 피부로 전달하는 기술개발에 항상 관심을 쏟아왔다. 뛰어난 노화방지 화장품 개발을 위해서, Ubiquinone의 일종인 ldebenone에 대해 연구하였고, 이를 비수계 피부유사막 기술을 가지고 캡슐화하고 약물전달하는 연구를 진행하였다. 먼저, 편광현미경(PM, Polarized Microscope), X-선 회절분석(XRD, X-ray Diffractions) 및 시차주사열량계 (DSC, Differential Scanning Calorimetry)를 이용하여 Idebenone을 담지한 피부유사막 액정을 비수계 조건에서 구조 및 열적특성을 조사하였다. 그 결과 비수계 조건에서도 규칙적으로 패킹(Packing)된 지질이중층(Lipid bilayer)과 용매의 연속층으로 이루어진 고밀집된 라멜라(Lamella) 구조의 형성유무와 이때의 상거동을 확인할 수 있었다. 결론적으로 높은 극성도로인해 물분자와 접촉하면 불안정해지는 경향이 있는 Idebenone을 비수계 조건에서 각질층(SC, Stratum Corneum)과 구조 및 조성이 유사한 피부유사막을 디자인하여 안정하게 캡슐화 하였다. 이를 적용한 화장품은 모든 보관조건에서 유화입자의 안정성을 유지함을 확인하였고, Idebenone의 활성역가 또한 $40^{\circ}C$에서 6개월 동안 약 90%이상을 유지하는 우수한 결과를 나타냈다.
Cellular compatibility in the self-parasitism of Cuscuta australis R. Brown was studied at the ultrastructural level. The front cells of the haustorium penetrated the host stems independently grew within the host tissues and transformed into elongate, filamentous hyphae. Each hyphal cells contained a large nucleus and dense cytoplasm with abundant cell organelles. Multilamellar structures were contained in the cytoplasm and cell walls of the penetrating hyphal cells. When the hyphal cells did not yet invade the host cells, the middle lamella and the fused cellulosic cell walls of the two partners at the host-parasite interface were preserved well. As the invasion of the parasitic hyphal cells progressed, however, the middle lamella was not found at the interface and the host cell walls and plasma membranes were partially broken down. A hyphal cell penetrated deeply into the host cell had a more darkly stained cytoplasm with numerous of cell organelles. In the host cells attacked by the hyphal cells the limiting membranes of plastids were broken down and several vesicles were arrayed near the cell walls. No plasmodesmatal connections between the host and parasite cell walls were found; however, half-plasmodesmata were observed frequently on the side of the hyphal cell walls. These results suggested that the compatibility response in the self-parasitism of Cuscuta was expressed by cell walls, not by plasmodesmata, between the host and the parasite cells.
The cell wall components of fruit include cellulose. hemicellulose, pectin, glycoprotein etc., and the cell wall composition differs according to the kind of fruit. Fruit softening occurs as a result of a change in the cell wall polysaccharides : the middle lamella which links primary cell walls is composed of pectin. and primary cell walls are decomposed by a solution of middle lamella caused due to a result of pectin degradation by pectin degrading enzymes during ripening and softening, During fruit ripening and softening, contents of arabinose and galactose among non-cellulosic neutral sugars are notably decreased, and this occurs as a result of the degradation of pectin during fruit repening and softening since they are side-chained with pectin in the form of arabinogalactan and galactan Enzymes involved in the degradation of the cell wall include polygalacturonase, cellulose, pectinmethylesterase, glycosidase, etc., and various studies have been done on the change in enzyme activities during the ripening and softning of fruit. Among cell wall-degrading enzymes, polygalacturonase has the greatest effect on fruit softening, and its activity Increases during the maturating and softening of fruit. This softening leads to the textural change of fruit as a result of the degradation of cell wall polysaccharides by a cell wall degrading enzyme which exists in fruit.
Micromorphological changes in waterlogged archaeological woods excavated from Sweden and Germany were investigated. Especially bacterial attacks on those wood samples under near anaerobic conditions were examined by transmission electron microscopy(TEM). The major feature of micromorphological alterations in those wood samples was the preferential destruction of secondary wood cell wall. In contrast, the middle lamella was not extensively degraded. Three distinct degradation patterns by bacteria were observed : erosion, cavitation and tunnelling bacteria. Erosion and cavitation bacteria attacked primarily $S_2$ layer, whereas tunnelling bacteria made the tunnel-like degradation along the $S_1$ layer. Tunnelling bacteria, in some samples, were able to degrade tunnel in the lignin-rich areas, such as middle lamella, suggesting that these bacteria had the capacity to degrade the lignin. IR spectra indicate that hemicellulose and cellulose in the waterlogged woods were preferentially decomposed. Breakdown of the lignin, on the other hand, was much slower.
The spermatogenesis of Korean slug, Incilaria fruhstorferi are observed by electron microscope. The results are as follows: The spermatogenesis of Korean slug, Incilaria fruhstorferi, is processed through the five stages; Spermatogonia, primary spermatocyte, secondary spematocyte, spermatid and spermatozoon. The spermiogenesis, the differentiation of the spermatid, is also processed through the five stages. In stage 1, the numerous and round mitochomdria in the cytoplasm are moved around the nucleus of spermatid. In stage 2, the nucleus of spermatid transformed into the oval shape, and the oval nucleus is surrounded by many rough endoplasmic reticulum. In stage 3, the oval nucleus of spermatid is changed to be curved as an arrow, and then two centrioles appeared behind nucleus. The centriole is sucked into the cytoplasm. and almost all the chromatins are changed into heterochromatins. In stage 4, the nucleus of spermatid are transformed into the oval shape, when the lamella plate chromatin of spermatid form in the nucleoplasm. In stage 5, the oval nucleus is then transformed into the stream-line shape when the lamella plate chromatin of spematid gradually concentrated in the nucleus, and long axoneme ($65{\mu}m$ in length) form from the distal centriole. Two long mitochondria in the middle piece and the main piece of spermatozoon array spirally along a long axoneme, and the mitochondria matrix is gradually filled with electron-dense glycogen particles ($0.1{\mu}m$ in size). The axoneme of spermatozoon consists of typical 9+2 microtubular pattern.
Chemical and micro morphological changes of archaeological waterlogged woods from shipwrecked materials in marine situations were investigated which were submerged in seabed for over 900 years. Tested Wood species were Pinusdensiflora, Zelkova serrata, Quercus acutissima and Camellia japonica. The obtained results were summarized as follows; Chemical analysis showed that lignin content was increased, whereas the amout of holocellulose was heavily decreased in the degraded archaeological lwoods(DAW), when compared to the recent woods. The amount of alkalineextractives in the DAW was extremley high. IR spectra showed that disappearance of absortion band at $1,730㎝^-1$ intensity increase at 1,600, 1,500 and $1,270㎝^-1$ and the emergence of single band around $1,050㎝^-1$.Microscopic investigation showed that cell wall of latewood tracheids and fiber in the DAW were severely degraded while, early wood tracheids less degraded. Degradation in the cell wall was mainley occurred in $S_2$layer, while the middle lamella was the least degraded. The micro morphological characteristics of DAW were separation of secondary wall from middle lamella, cavities aligned with micro fibril angle in $S_2$layer and granular appearance of secondary wall by the bacterial attack.
본 연구는 유사세라마이드(N-Ethanol-2-myristyl/palmityl-3-oxostearamide/arachidamide (PC-9))를 함유한 다중층상 유화물의 조제와 이의 효능 평가에 대한 연구이다. 다중층상 유화물의 제조를 위하여, 일차적으로 지질유사성분 혼합조성(유사세라마이드(PC-9), 지방산, 콜레스테롤)의 phase 연구를 통하여 액정형성이 용이한 조성비를 결정하고, 액정형성이 용이하다고 알려진 유화제인 Glyceryl monostearate와 polyoxyethylene glyceryl monostearate를 사용하여 유화물을 제조하였다. 유성성분으로서는 올리브 오일과 같은 천연오일이 다중층상 유화물이 잘 형성되는 경향을 보였으며, 전체적으로 유성성분의 양이 많아질수록 제조된 유화물의 편광하에서 광학 이방성을 나타내는 정도는 감소하였다. 지질유사성분과 유화제 성분의 양이 유성성분에 비해 높은 경우 건조과정에서도 높은 유화입자의 안정성을 보여주었으며, 유성성분을 증가하여 다중층상 유화물을 제조할 경우 건조시 액정상(liquid crystal)으로의 전이과정을 보여주었으며, 완만한 수분 건조속도와 실제 피부 적용시 뛰어난 경피수분상실 억제효과를 보여 주었다.
사과과실(Malus Pumila Mill var. Fugi)을 preclimacteric단계에 수확하여 380mmHg, $20^{\circ}C$의 조건에서 감압저장하고 climacteric rise의 각 단계별로 전자현미경적으로 관찰한 결과, 사과저장 중 감압처리가 엽록체의 초미세구조변화에 미치는 영향과 이로 인한 저장기간 연장효과를 규명하였다. 760mmHg, $20^{\circ}C$의 조건에서 저장한 상압저장 과실은 25일간의 저장으로 이미 막투과성의 변화로 인한 공포화현상으로 인하여 엽록체의 초미세구조에 상당한 변화가 일어나 stroma부위의 전자밀도가 낮아지고, lamellar system이 와해되었으며, 50일간의 저장에서는 광범위한 breakdown현상으로 인하여 stroma부위의 전자밀도상실, lamella system상실 등의 초미세구조변화가 매우 심하게 나타나 저장성이 상실되었다. 한편 380mmHg, $20^{\circ}C$의 조건에서 저장한 감압저장과실은 상압저장과실에 비하여 엽록체의 초미세구조변화가 크게 억제되었다. 특히 thlakoid막은 climacteric rise가 진전되어 공포화현상이 심하게 나타나서 lamella구조가 완전히 와해되었을 경우에도 비교적 건전한 상태로 유지되어 저장기간이 2개월정도 연장되었는데, 저장 중의 감압처리로 인하여 막투과성변화가 억제되어 사과과실의 숙성 및 연화가 지연됨으로써 저장기간이 연장된 것으로 판단되었다.
