온도감수성 대장균의 형질전환조건 및 Ampicillin 내성의 표현 (Transformation Conditions and Ampicillin-resistant Expression of E. coli Ts-mutant)

  • 온도감수성 대장균 M5248 균주에 있어서의 plasmid $pPL-\lambda$와 pAS1을 $30^{\circ}C$에서 형질전환 시킬 때의 조건을 조사한 바 배양시간에 있어서는 균주의 대수 증식기 중반인 2시간 30분 배양시, 즉 cell농도는 $4.5\times10^7\;cells/ml$이며 파장 590nm에서의 흡광도가 0.45 일 때 plasmid $pPL-\lambda$와 pAS1 모두 형질전환율은 $2.4\times10^6$$1.5\times10^{-6}$로서 가장 높았다. 그리 고 competent cell $200{\mu}l(9\times10^{-6}cell)$에 대하여 plasmid 농도가 $6.4{\mu}g/ml$ 형질전환율이 $4.4\times10^{-6}$로서 높게 나타났으며 이때 calcium chloride만으로 처리한 것 보다 calcium chloride와 thymidine을 혼합하여 처리했을 때 2배 정도 더 많은 transformant를 유도하였다. Ampicillin 내성유전자는 LB배지를 첨가하여 $30^{\circ}C$에서 2시간 동안 배양했을 때 그 형질이 표현되었다.

B형 간염 바이러스의 돌연변이 내면항원의 발현 및 분비 (Expression and Secretion of Hepatitis B Viral Mutant Core Antigen)

  • 포유동물 세포내에서 간염 바이러스의 내면항원의 발현과 전위내면 항원(precore) 부위의 역할을 규명하기 위하여 고등동물세포 발현용 벡터에 전위내면항원 부위를 갖거나 또는 갖지 않는 내면항원 유전자를 클로닝 하여 COS 세포내에서의 발현을 조사하였다. 전위내면항원 부위를 포함한 내면항원 유전자를 갖는 플라스미드로감염시킨 COS 세포는 항원들이 세포추출물과 배양액에서 검출되었다. 분비된 항원의 증가율은 감염후 2일과 3일 사이가 가장 높았고, 부분결실된 제조합 플라스미드 중 내면항원의 ATG codon에서 180bp 떨어진 것이 가장 발현이 잘 되었다. 전위내면항원을 갖지 않거나 하나의 염기가 첨가되어 변형된 전위내면항원을 갖는 제조합 플라스미드의 경우 항원들이 세포 추출물에서만 검출되었다. 이러한 사실은 전이내면항원 부위가 HBe 항원의 분비에 관여 한다는 경우 항원들이 세포 그러나 대장균이나 효모 세포의 경우는 전위내면항원의 존재와 상관없이 항상 세포추출물에서만 존재하는 것으로 보아 이들 세포의 경우에서는 전위내면항원 부위가 HBe 항원의 분비에 영향을 줄 수 없음을 의미한다.

Periodic Change in DO Concentration for Efficient Poly-${\beta}$-hydroxy-butyrate Production Using Temperature-inducible Recombinant Escherichia coli with Proteome Analysis

  • Recombinant Escherichia coli strain harboring the ${\lambda}$pR-pL promotor and heterologus poly-${\beta}$-hydroxybutyrate (PHB) biosynthesis genes was used to investigate the effect of culture conditions on the efficient PHB production. The expression of phb genes was induced by a temperature upshift from $33^{\circ}C\;to\;38^{\circ}C$. The protein expression levels were measured by using two-dimensional electrophoresis, and the enzyme activities were also measured to understand the effect of culture temperature, carbon sources, and the dissolved oxygen (DO) concentration on the metabolic regulations. AcetylCoA is an important branch point for PHB production. The decrease in DO concentration lowers the citrate synthase activity, thus limit the flux toward the TCA cycle, and increase the flux for PHB production. Since NADPH is required for PHB production, the PHB production does not continue leading the overproduction of acetate and lac-tate. Based on these observations, a new operation was considered where DO concentration was changed periodically, and it was verified its usefulness for the efficient PHB production by experiments.

인간 인터루킨-2를 코드하는 유전자의 합성과 대장균에서의 발현 (Total Synthesis and Expression in E, coli of a Gene Coding for Human Interleukin-2)

  • DNA 자동합성기를 이용하여 오릴고뉴클레오타이드들을 합성하였으며 이로부터 인간 인터루킨-2를 코드하는 유전자를 제조하였다. 합성 유전자의 염기서열은 대장균에서 사용빈도가 높은 코돈을 고려하여 결정하였으며, 이때 인터루킨-2의 아미노산 서열은 변화시키지 않았다. 합성된 유전자는 람다 피엘 프로모터를 사용하여 대장균에서 발현시켰다. 인터루킨-2는 대장균에서 불용성의 침전물로 생성되었다. 생성된 인터루킨-2는 인터루킨-2 요구 배양세포주의 성장을 촉진하였다.

Mycobacterium tuberculosis-induced Expression of Interleukin-1 Beta is Mediated Via Protein Kinase C Signaling Pathway

  • Interleukin-1${\beta}$ $(IL-1{\beta})$ is one of the key proinflammatory cytokines and it plays an important role for the antimycobacterial host defense mechanisms. In this study, we examined Mycobacterium tuberculosis (MTB)-stimulated induction of IL-1${\beta}$ and evaluated the associated signal transduction pathways. In PMA-differentiated THP-1 cells, MTB infection increased mRNA expression of IL-$1{\beta}$ in a dose-dependent manner. The expression of IL-1${\beta}$ mRNA began to be induced at 1.5 h after infection, and induced expression of IL-1${\beta}$ was retained for 48 h after MTB infection. The increase in expression of IL-1${\beta}$ caused by MTB was reduced in cells treated with Ro-31-8425 (an inhibitor of PK$C{\alpha}$, ${\beta}I$, ${\beta}II$, ${\gamma}$, ${\varepsilon}$) or PD98059 (an inhibitor of MEK1), meanwhile, pre-treatment with $G\ddot{o}6976$ (an inhibitor of $Ca^{2+}$ dependent PK$C{\alpha}$ and PK$C{\beta}I$) or Rottlerin (an inhibitor of PK$C{\delta}$) has no effect on MTB-induced expression of $IL-1{\beta}$ mRNA. These results show that the expression of $IL-1{\beta}$ mRNA caused by MTB may be mediated via MEK1 and PKC isoforms including PK$C{\beta}II$, $PKC{\gamma}$, or $PKC{\varepsilon}$. Further studies are required to determine whether other PKC isoforms $(PKC {\eta},\;{\theta},\;{\varepsilon},\;and\;{\lambda}/{\iota})$, except $PKC{\delta}$, $PKC{\alpha}$, and $PKC{\beta}I$, are also involved in $IL-1{\beta}$ mRNA expression after mycobacterial infection.

  • To clone the gene coding for steroid $\Delta^1$-dehydrogenase of Arthrobacter simplex, its genomic library was constructed with a , $\lambda$gt11 expression vector and immunoscreened with antiserum against the enzyme. One positive clone was found to carry a 1.6-kb EcoR I restriction endonuclease fragment of A. simplex DNA. The restriction map of the 1.6-kb EcoR I fragment was determined after cloning of the DNA into pBS vector.

Little's 법칙의 미시적 활용 사례 (A MIrcroscopic Application of the Little's Formula)

  • The Little's formula, $L={\lambda}W$, expresses a fundamental principle of queueing theory: Under very general conditions, the average queue length is equal to the product of the arrival rate and the average waiting time. This useful formula is now well known and frequently applied. In this paper, we demonstrate that the Little's formula has much more power than was previously realized when it is properly decomposed into what we call the microscopic Little's formula. We use the M/G/1 queue with server vacations as an example model to which we apply the microscopic Little's formula. As a result, we obtain a transform-free expression for the queue length distribution. Also, we briefly summarize some previous efforts in the literature to increase the power of the Little's formula.

  • A stochastic model for an inventory system in which depletion of stock takes place due to random demand as well as random loss of items is studied under the assumption that the intervals between cussessive unit demands as well as those between cussessive unit losses, are independently and identically distributed random variables having negative exponential distributions with respective parameters .mu. and .lambda. It is further assumed that leadtime for each order is an outstanding-order-dependent random variable. The steady state probability distribution of the net inventory level is derived under the continuous review (S -1, S) inventory policy, from which the total expected coast expression is formulated.

여자의 부모선호태도와 그 요인 분석 (The analysis of the child's preference for one parent and its factors)

  • The purpose of this study is to investigate the child's preference attitude for one parent and to analyze the difference in the four variables(sex, age, birth order and maternal employment) and the factors influencing it. To meet this purpose, three kinds of questionnaire for child (school-age/late adolescence) and his parents were used. The data were obtained through 136 pairs(parents and child) and analyzed by chisquare test and Guttman's lambda. The results of this test are summerized as follows. 1. The majority of children prefer mother to father and especially adolescent daughters prefer mother more than sons. 2. The majority of parents answered that their child would prefer mother to father and the correlation coefficient between mother's answer and fathers is .53. But child's actual preference is no related with parent's awareness of being preferred. 3. The majority of parent's answer were consistent with child's preference but the correlation coefficients are very low from the analysis of factors influencing the child's preference. Therefore parents and child must have more communication, concern and expression of love for mutual consistence.

  • In this paper, we consider a model problem arising from a classical planar Heisenberg ferromagnetic spin chain $-{\Delta}u+V(x)u-{\frac{u}{\sqrt{1-u^2}}}{\Delta}{\sqrt{1-u^2}}={\lambda}{\mid}u{\mid}^{p-2}u$, x ∈ ℝN, where 2 ≤ p < 2*, N ≥ 3. By the Ekeland variational principle, the cut off technique, the change of variables and the L estimate, we study the existence of positive solutions. Here, we construct the L estimate of the solution in an entirely different way. Particularly, all the constants in the expression of this estimate are so well known.
