Effects of Residual Hypochlorite Ion on Methane Production during the Initial Anaerobic Digestion Stage of Pig Slurry

  • The hypochlorite ion ($OCl^-$) is a widely used disinfecting agent in pig rearing in Korea, but its residual effect on $CH_4$ production from pig slurry is unclear. The objective of this study was to investigate the inhibition effects of residual $OCl^-$ on $CH_4$ production during the initial anaerobic digestion stage of pig slurry. Three organic concentrations (9.9, 26.2 and 43.7 g/L) of volatile solids (VS) were tested with the addition of 52.3 mg/L $OCl^-$, ten times of the typical concentration used in Korea, or without $OCl^-$ (Control) in anaerobic batch culture. The culture was run under mesophilic ($38^{\circ}C$) conditions for 20 d. At the lowest organic concentration with $OCl^-$, the VS degradation was 10.3% lower (p<0.05) than Control, while at the higher organic concentration with $OCl^-$, it did not differ from Control. $CH_4$ yields were higher in the control treatments than their $OCl^-$ counterpart cultures, and $CH_4$ yields of Control and $OCl^-$ treatments at the organic concentrations of 9.9, 26.2 and 43.7 g/L differed in the probability level (p) of 0.31, 0.04, and 0.06, respectively. Additionally, $CH_4$ concentration increased steeply and reached 70.0% within 4 d in the absence $OCl^-$, but a gradual increase up to 60.0% was observed in 6 d in the $OCl^-$ treated cultures. The $R_m$ (the maximum specific $CH_4$ production rate) and ${\lambda}$ (lag phase time) of 9.9 g/L with $OCl^-$ were 8.1 ml/d and 25.6 d, while the $R_m$ was increased to 15.1 ml/d, and ${\lambda}$ was reduced to 11.4 d in PS-III (higher organic concentration) with $OCl^-$. The results suggest that a prolonged fermentation time was necessary for the methanogens to overcome the initial $OCl^-$ inhibitory effect, and an anaerobic reactor operated with high organic loadings was more advantageous to mitigate the inhibitory effect of residual hypochlorite ion.

Integrated Wet Oxidation and Aerobic Biological Treatment of the Quinoline Wastewater (퀴놀린 폐수의 습식산화와 호기성 생물학적 통합처리)

  • The treatment of a model wastewater containing quinoline in an integrated wet oxidation-aerobic biological treatment was investigated. Partial wet oxidation under mild operating conditions was capable of converting the original quinoline to biodegradable organic acids such as nicotinic, formic and acetic acid, the solution of which was subjected to the subsequent aerobic biological treatment. The wet oxidation was carried out at 250$^{\circ}C$ and the initial pH of 7.0, and led to effluents of which nicotinic acid was oxidized through 6-hydroxynicotinic acid by a Bacillus species in the subsequent aerobic biological treatment. Either homogeneous catalyst of $CuSO_4$ or phenol, which is more degradable in the wet oxidation compared to quinoline, was also used for increasing the oxidation rate in the wet oxidation of quinoline at 200$^{\circ}C$. The oxidation of quinoline in the catalytic wet oxidation and the wet co-oxidation with phenol resulted in effluents of which nicotinic acid was biodegradable earlier in the aerobic biological treatment compared to those out of the non-catalytic wet oxidation at 250$^{\circ}C$. However, the lag phase in the biodegradation of nicotinic acid formed out of the wet oxidation at 250$^{\circ}C$ was considerably shortened after the adaptation of Bacillus species used in the aerobic biological treatment with the effluents of the quinoline wet oxidation.

Lactic Acid Bacteria Isolated from Fermenting Kimchi and Their Fermentation of Chinese Cabbage Juice (김치에서 젖산균의 분리 및 이 세균들의 배추즙액 발효)

  • Lactic acid bacteria(LAB) were isolated from fermenting Kimchi and were cultivated in filter-sterilized Chinese cabbage juice individually or in combination. LAB isolated were Lactobacillus leichimannii, Lac. fermentum, in addition to the already known Leuconostoc mesenteroides, Lac. plantarum, lac. brevis and pediococcus pentosaceus. Lac. leichimannii, Lac. fermentum and Lac. sake, the early lactobacilli, were high in number exceeding $10^4cells/ml$ at 0 time and multiplied up to $10^9cells/ml$ altogether at the 3rd day of kimchi fermentation. When the representative LAB were cultivated singly in Chinese cabbage juice with or without 3.0% NaCl, one strain of Leu. mesenteroides and La. leichmannii were not different in acid producing ability while the other strain of Leu. mesenteroides and Lac. fermentum Lac. plantarum, produced less acid when NaCl was present. When the bacteria in combination were cultivated in Chinese cabbage juice with 3.0% NaCl, the presence of Leu. mesenteroides was essential to eliminate the lag phase in acid production with higher amounts of acid produced than without. The total number of lactobacilli in the mixture of kimchi ingredients was about $2.9{\times}10^4 cells/ml$ while the number of Lac. plantarum was 7.3 cells/ml. The number of Lac. plantarum in individual ingredients were normally in the range between $0.0{\sim}240cells/g$ except garlic sold in ready-to-use form with $9.0{\times}10^3 cells/g$.

Reduction of Hydraulic Conductivity in the Subsurface by the Formation of Aerobic Biobarrier (토양 내 호기성 생물벽체(Biobarrier)의 형성에 의한 투수계수의 제어)

  • A series of batch and column experiments were conducted for the development of biobarrier technology which can be applied to containment and reduction of contaminants in soil and ground waters. The growth kinetic constants of Pseudomonas fluorescens on glucose or molasses were determined using batch experiments. The maximum specific growth rate (Vmax) of P. fluorescens at $23^{\circ}C$ on glucose or molasses were $0.246\;hr^{-1}$ and $0.073\;hr^{-1}$, respectively. However, molasses was selected as carbon source due largely to the absence of lag phase of P. fluorescens growth on molasses and economic reason. In constant head column experiments, the hydraulic conductivity of the column soil reduced by $6.8{\times}10^{-3}$ times from $4.1{\times}10^{-2}cm/sec$ to $2.8{\times}10^{-4}cm/sec$ after the inoculation of P. fluorescens and administration of carbon source and nutrients. The biomass concentration was observed highest in the column inlet. Measurements of carbon source and electron accepter (dissolved oxygen) concentration showed that the growth of P. fluorescence, which is the main reason for hydraulic conductivity reduction, was limited not by the concentration of carbon source but by the concentration of electron acceptor.

Drag Coefficient Variations of an Oscillating NACA 0012 Airfoil (진동하는 NACA 0012 에어포일에서의 항력계수 변화)

  • An experimental study was performed in order to investigate the influence of Reynolds number on the drag coefficient variations of an oscillating airfoil. A NACA 0012 airfoil was sinusoidally pitched at the quarter chord point with an oscillating amplitude of ${\pm}6^{\circ}$. The free-stream velocities were 1.98, 2.83 and 4.03 m/s and the corresponding chord Reynolds numbers were $2.3{\times}10^4$, $3.3{\times}10^4$ and $4.8{\times}10^4$, respectively. The drag coefficient was calculated from the ensemble average velocity measured by an X-type hot-wire probe(X-type, 55R51) in the near-wakes region. In the case of Re=$2.3{\times}10^4$, variation of drag coefficient shows a negative damping (counter-clockwise variation), which implies an unstable state which could be excited by aerodynamic force, whereas the drag coefficient represents the positive damping (clockwise variation) as the Reynolds number increases from Re=$3.3{\times}10^4$ to $4.8{\times}10^4$. Hence, the drag coefficient variations show significant differences between Re=$2.3{\times}10^4$ and $4.8{\times}10^4$이다.

Improvement of Anodic Performance by Using CTP Binder Containg Nickel (니켈을 함유한 콜타르 피치 결합제를 이용한 미생물연료전지 산화전극 성능개선)

  • The composite anodes of expanded graphite (EG) and multiwall carbon nanotube (MWCNT) for microbial fuel cells were fabricated by using coal tar pitch (CTP) binder containing nickel (Ni), and the effect of the anodes with the binders on the performance of the MFCs were examined in a batch reactor. During the start-up of the MFCs, quick increase in voltage was observed after a short lag phase time, indicating that the CTP binder is biocompatible. The biomass attatched on the anode surface was more at higher Ni content in the binder, as well as at smaller amount of CTP binder for the fabrication of the anode. The internal resistance of the MFC was smaller for the anode with more biomass. Based on the results, the ideal combination of CTP and Ni for the CTP binder for anode was 2 g and 0.2 g, respectively. The maximum power density was $731.8mW/m^2$, which was higher 23.7% than the anode with Nafion binder as control. The CTP binder containing Ni for the fabrication of anode is a good alternative in terms of performance and economics of MFCs.

Effects of Light Wavelengths on the Growth and Paralytic Shellfish Toxin Production of Alexandrium catenella and A. pacificum (유독 와편모조류 Alexandrium catenella와 A. pacifcium의 생장과 마비성 독소 생산에 미치는 빛의 파장의 영향)

  • To supply a stable amount of standard material to detect paralytic shellfish toxin (PST), we examined possible increases in toxic content in Alexandrium catenella and A. pacificum using a light emitting diode (LED), which is one of the most eco-friendly and economical lighting method. When comparing the growth rates of organisms and wavelengths of light used, the half saturation constants (Ks) of red wavelength were higher than those of other wavelengths. In contrast, the Ks of blue wavelength were lower than those of other wavelengths. Moreover, when comparing the toxic contents and wavelengths of light used, red wavelength produced approximately 8 times more toxic content in A. catenella and approximately 3.2 times more toxic content in A. pacificum than other wavelengths. Thus, the toxic content present in the organism might be closely related to the Ks of light. The optimum light source to be used to ensure economically ef ective and productive growth in an Alexandrium culture system (photo-bioreactor) would likely consist of a two-phase culture, wherein a blue LED is used during the lag and exponential phases to increase growth rates, followed by the use of a red LED during late exponential and stationary phases to achieve increased PST yields.

Nitrite Removal by Autotrophic Denitrification Using Sulfur Particles (황입자를 이용한 독립영양탈질에서의 아질산성질소 탈질 조건 탐색)

  • Swine wastewater contains high amounts of organic matter and nutrients (nitrogen and phosphorus). The biological nitrogen removal can be achieved by nitrification and denitrification processes. Nitrification-denitrification can be performed via nitrite which is called as the short-cut process. This Short-cut process saves up to 25% of oxygen and 40% of external carbon during nitrification and denitrification. In this study, the batch tests were conducted to assess the different parameters for the nitrite sulfur utilizing denitrification, such as alkalinity, temperature, initial nitrite concentration, and dissolved oxygen. The experimental results showed that the nitrite removal efficiency of the reactor was found to be over 95% under the optimum condition ($30^{\circ}C$ and sufficient alkalinity). Autotrophic nitrate denitrification was inhibited at low alkalinity condition showing only 10% removal efficiency, while nitrite denitrification was achieved over 95%. The nitrite removal rates were found similar at both $20^{\circ}C$ and $30^{\circ}C$. In addition, nitrite removal efficiencies were inhibited by increasing oxygen concentration, but sulfate concentration increased due to sulfur oxidation under an aerobic condition. Sulfate production and alkalinity consumption were decreased with nitrite compared those with nitrate.

Quality Characteristics of High Acidity Apple Vinegar Manufactured Using Two Stage Fermentation (2단계 발효에 의한 고산도 사과식초의 품질특성)

  • This study investigated the manufacturing conditions of apple vinegar with high acetic acid content following fermentation according to alcohol concentration without any nutrients. We compared and analyzed the quality characteristics of high acetic acid fermentation by varying the initial alcohol content (6%, 7%, 8%, and 9%). In the results, it was possible to manufacture high acetic acid vinegar with 12% titratable acidity and an alcohol content of 6% and 7%. Lower initial alcohol content was associated with higher yield due to a shorter lag phase. For quality characteristics of the high acetic acid apple vinegar, pH was 2.91~3.20, titratable acidity was 12.0%, and organic acid consisted of acetic acid, malic acid, critic acid and oxalic acids. Based on the results, high acetic acid apple vinegar was produced using a two stage fermentation process after alcohol fermentation but the further research is needed to reduce the time of fermentation in fed-batch culture for industrial use.

Characteristics for Co-digestion of Food Waste and Night Soil using BMP Test (BMP실험을 이용한 음식물폐기물 및 분뇨의 병합소화 특성)

  • BMP test was carried out to evaluate the characteristics for co-digestion of night soil and food waste. 6 types of sludge were tested in 30 days which were raw, excess, digested, night soil/septic tank (1:1), food waste (food : dilution water = 1:1), and mixed sludge. Bio gas was produced actively after 2 days, and continued in 2 weeks. Gas generation amount was decreased rapidly after considerable space of time. Especially maximum productivity of gas was shown in 7~8 days. The ultimate methane yields of raw, excess, digested, night soil/septic tank, food waste, and mixed sludge were 64.63, 67.49, 66.45, 72.44, 107.85, and 46.71 mL $CH_4/g$ VS respectively from Modified Gompertz model. The lag growth phase time and maximum specific methane production rate of mixed sludge were 1.88 day and 80.4 mL/day respectively. The methane potential of mixed sludge was higher than individual sludge. So high methane potential was expected by controlling mixing ratio of food waste. Besides stable operation of digestion tank and the solution of oligotrophic problem were possible.
