Isolation and Identification of Lactic Bacteria Containing Superior Activity of the Bile Salts Deconjugation (담즙산 분해능이 뛰어난 젖산균의 분리 및 동정)

  • The purpose of this study is to isolate probiotic lactic acid bacteria (LAB) that produced bile salts hydrolase. One hundred twenty strains were initially isolated from human feces. Based on their resistance of acid, tolerances of bile salts, and inhibitory activity against Escherichia coli, five strains were selected. A strain producing highest activity of bile salts hydrolase was identified as Lactoacillus plantarum using API carbohydrate fermentation pattern and 16S rRNA sequences, and named CK102. Lactobacillus plantarum CK102 survived at a level of 1.36${\times}$10$\^$8/ CFU/$m\ell$ in pH 2 buffer for 6 h and showed exhibited excellent bile tolerance. When L plantarum CK102 was cultured with E. coli in MRS broth, no viable cells of E. coli was detected after 18 h fermentation. These results suggest that Lactobacillus plantarum CK 102 may be commercially used for the probiotic culture.

Microbiological Characteristics of Gamma Irradiated and Low-Salted Fermented Squid (감마선 조사된 저염 오징어젓갈 발효의 미생물균총 특성)

  • Microbiological characteristics of gamma irradiated low salt squid Jeot-gal were examined. Following the fermentation periods, total bacterial cell, Lactobacillus spp., Staphylococcus spp., Streptococcus spp., Pseudomonas spp. and yeast cell number were counted on their selective media and some acid forming bacteria and Pseudomonas spp. were identified. As the gamma irradiation dose increased, the microbial density of early fermentation phase was reduced and the growth rate was delayed. The repression effects on microbiological growth by gamma irradiation were to be higher as salt concentration increased. Adequate conditions of salt concentration and gamma irradiation for low-salt squid Jeot-gal preparation were 10% and 10 kGy, respectively. Lactobacillus sp. 2, Micrococcus varians and Streptococcus sp. I were isolated from 5% salt containing squid Jeot-gal, and Micrococcus morrhuae was from 20% only while Lactobacillus plantarum and Lactobacillus brevis were widespread. Lactobacillus brevis, Pediococcus halophilus and Pseudomonas diminuta were sensitive and Lactobacillus plantarum, Micrococcus morrhuae and Pseudomonas sp. 3 were resistant to gamma irradiation. The diversity of microflora decreased as salt concentration decreased and gamma irradiation dose increased.

Effects of Fermentation to Improve Hygienic Quality of Powdered Raw Grains and Vegetables Raw Grains and Vegetables Using Lactobacillus sp. Isolated from Kimchi (김치분리균주 Lactobacillus sp.를 Starter로 한 발효생식 제조에서의 위생미생물 살균효과)

  • Improvement of hygienic quality of powdered raw grains and vegetables by fermentation was investigated. Luc-tobacillus sp. isolated from kimchi was used as a starter. The cell counts of coliform group and SS enteric bacteria on the SS agar plate in raw grains and vegetables were 2.3$\times$103 cfu/mL and 8.6$\times$10$^3$ cfu/mL, respectively. The starlet, Lactobacillu sp., reached 10$^{7}$ cfu/mL after 48 hr in fermentation. At that time, the coliform group and enteric bacteria on the SS agar plate were gradually inactivated and eliminated after 60 hr of fermentation. During the fermentation process, pH of the suspension was lowered and acidity increased. Antimicrobial activity of the acidic supernatant of fermented raw grains and vegetal]les against the E. coli sp. and Salmonella sp. was higher than that of lactic acid solution or neutralized supernatant. Therefore, it was considered that antimicrobial effect of the fermented raw grains and vegetal]les might be accelerated by tile synergic effect of acid and bacteriocin, and liquid fermentation of powdered raw grains and vegetables will be effective for inactivation of hygienic microorganisms.

Characteristics of Lactate Dehydrogenase Produced from Lactobacillus sp. FFy111-1 as a Ruminant Probiotic (반추동물용 활성제로서 Lactobacillus sp. FFy111-1이 생산한 Lactate Dehydrogenase의 특성에 관한 연구)

  • The objective of this experiment is to study the possibility of lactate dehydrogenase(LDH) enzyme to prevent lactate accumulation in the rumen, For understanding capacity of bacterial LDH in rumen environments, this study was conducted to explore the effects of temperature, pH, VFAs and metal ions on Lactobacillus sp. FFy111-1's LDH activity, and the LDH activation in rumen fluid accumulated lactate. The optimum pH and temperature of LDH were pH 7.5 and 40$^{\circ}C$, respectively. The LDH activity had a good thennostability at range from 30 to 50$^{\circ}C$. The highest pH stability of the enzyme was at ranges from pH 7.0 to 8.0 and the enzyme activities showed above 64% level of non-treated one at pH 6.0 and 6.5. The LDH was inactivated by VFAs treatments but was enhanced by metal ion treatments without NaCl and $CuSO_4$ Especially, the LDH activity was increased to 127% and 124% of its original activity by 2 mM of $BaCl_2$ and $MnSO_4$, addition, respectively. When the acidic rumen fluid was treated by LDH enzyme of Lactobacillus sp. FFy111-1, the lactate concentration in the rumen fluid was lower compared with non-treated rumen fluid(P<0.05). This lactate reduction was resulted from an action of LDH. It was proved by result of purified D,L-LDH addition that showed the lowest lactate concentration among the treatments(P<0.05). Although further investigation of microbial LDH and ruminal lactate is needed, these findings suggest that the bacterial LDH has the potential capability to decrease the lactate accumulated in an acidic rumen fluid. Also, screening of super LDH producing bacteria and technical development for improving enzyme activity in rumen environment are essential keys for practical application.

Characteristics of Fermented Omija (Schizandra chinensis Baillon) Sugar Treatment Extracts by Lactobacillus sp. (Lactobacillus sp. 균주를 이용한 오미자 당침액의 발효특성)

  • This study was carried out to investigate the characteristics of FOST (fermented omija sugar treatment extracts) using Lactobacillus brevis HLJ59. Antioxidant activities of FOST were evaluated through viable cell number of L. brevis HLJ59, DPPH radical scavenging activity, reducing power and SOD-like activity, compared to non-FOST(non-fermented omija sugar treatment extracts). Also it was to evaluate Angiotensin Converting Enzyme (ACE) and Urease inhibitory activity of FOST. The viable cell number of L. brevis was about $2.05{\pm}0.21{\times}10^8$, $6.31{\pm}0.56{\times}10^{11}$, and $8.14{\pm}0.14{\times}10^9$ at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. DPPH radical scavenging activity of FOST was about 60.3%, 71.8%, and 44.5% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. The reducing power of FOST was about 0.92, 1.19, and 0.73 (OD at 700 nm) at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, and SOD-like activity of FOST was about 50.4%, 53.7%, and 33.4% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. ACE and Urease inhibitory activity by FOST was about 47.4%, 78.2%, 56.4% and 58.1%, 83.4%, 63.2% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. The results indicated that the fermentation of omija sugar treatment extracts using Lactobacillus brevis HLJ59 increased the antioxidant activities campared to the non-fermented omija sugar treatment extracts.

Purification and Enzyme Property of a Cell-Wall Lytic Enzyme Produced by Bacillus sp. LM-8 against Lactobacillus plantarum. (Bacillus sp. LM-8이 생산하는 Lactobacillus plantarum 용균 효소의 정제 및 효소 특성)

  • Purification and characterization of enzyme property of a cell-wall lytic enzyme against Lactobacillus plantarum were carried out. Final specific activity of purified enzyme was 5.8 units/mg and purity of the enzyme was increased 8.3 fold compared with the enzyme activity in culture broth. The molecular weight of purified enzyme was estimated to be 60,000 kDa by gel filtration and SDS-polyacrylamide gel electrophoresis. Optimal pH and temperature for the activity of this enzyme were 3.0 and 4$0^{\circ}C$, respectively. The cell-wall lytic enzyme activity was maintained at 3$0^{\circ}C$ when treating the enzyme for 30 mins, whereas the activity was decreased to 80% of the maximum level at 4$0^{\circ}C$ The enzyme activity exhibited good stability at the range of pH 4~7.

Characterization of Antimicrobial Substance Produced by Lactobacillus sp. HN 235 Isolated from Pig Intestine (돼지 장관으로부터 분리한 Lactobacillus sp. HN 235 균주가 생산하는 항균물질의 특성)

  • In order to develop probiotics which may be a viable alternative of antibiotic use in pig industry, five bacterial strains (Lactobacillus sp. HN 52, 92, 98, 235 and AP 116) possessing antimicrobial properties were selected from 500 strains isolates of pig intestines. The bacteriocin produced by Lactobacillus sp. HN 235 displayed a relative broad spectrum of inhibitory activity against all Enterococcus strains, Pseudomonas aeruginosa, Listeria monocytogenes and Clostridium perfringens using the spot-on-lawn method. The production of antimicrobial substance started in the middle of the exponential growth phase, reached maximum levels (6,400 AU/mL) in the stationary phase, and then declined. Bacteriocin activity remained unchanged after 30 min of heat treatment at $95^{\circ}C$ and stable from pH 2.0 to 10 for 1 h, or exposure to organic solvents; however, it diminished after treatment with proteolytic enzymes. The molecular weight of the bacteriocin was about 5 kDa according to a tricine SDS-PAGE analysis.

Isolation and Partial Characterization of Isoflavone Transforming Lactobacillus plantarum YS712 for Potential Probiotic Use (Isoflavone 비배당화 및 항산화 활성을 지닌 Lactobacillus plantarum YS712의 선발)

  • Lactic acid bacteria (LAB) are typical probiotic microbes that are used in various industries including fermented foods, feed additives, and pharmaceuticals. The purpose of this study was to compare the ability of isoflavone biotransformation and antioxidative activity of 17 LAB. Six strains including the Lactobacillus species exhibited a 100% hydrolysis rate for daidzein during fermentation. The content of total genistein in soymilk fermented with these strains was $872-943\;{\mu}g/g$. The DPPH (1, 1-diphenyl-2-picrylhydrazyl) radical scavenging ability of the LAB was widely variable and ranged from 23-78%. A selected strain was isolated from kimchi and the strain was identified as Lactobacillus plantarum ssp. through the API carbohydrate fermentation pattern and 16S rDNA profile. The strain exhibited excellent acid tolerance in an artificial gastric solution. L. plantarum YS712 showed high $\beta$-glucosidase activity in fermentation. The concentration of genistein and daidzein in soymilk fermented with L. plantarum YS712 increased from 3.64 to $917.3\;{\mu}g/g$ and from 58.18 to $1062.17\;{\mu}g/g$, respectively. These results demonstrate the potential of L. plantarum YS712 as a probiotic culture that can be utilized in the manufacturing of fermentation foods and dietary supplements.

Physiological Characterization of Lactobacillus sp. JK-8 Isolated from Shrimp Aquaculture Pond (새우양식장에서 분리한 Lactobacillus sp. JK-8의 생리적 특성)

  • The purpose of this work was to investigate the physiological characteristics of Lactobacillus sp. JK-8 isolated from a shrimp aquaculture pond. The strain JK-8 was grown on MRS media, and morphological and physiological characteristics of the strain were examined. The bacterium was identified as a strain of the genus Lactobacillus on the basis of BIOLOG test. Strain JK-8 produced both lactic acid and acetic acid, which were responsible for the pH decrease during growth. Concentrations of lactic acid and acetic acid increased to 192.8 mM and 43.6 mM, respectively, and the initial pH 7.0 of the cultures decreased to 3.8 at the end of incubaction. The bacteriocidal effect against eight target bacteria was examined with 5-fold concentrated culture supernatants. All bacteria tested in this work were completely killed within 3 hrs after treatment with the culture supernatant. The bacteriocidal effects were clearly observed, only when the pH of the culture supernatants were not adjusted. HPLC was used to reslove lactic acid and acetic acid in the culture solution, and GC-MS was used to verify the metabolites.

Evaluation of Functional Properties of the Tissue Cultured Wild Ginseng Fermented by Lactobacillus sp. (Lactobacillus sp.균주를 이용한 산삼 배양근 발효물의 기능성 평가)

  • A tissue cultured wild ginseng (TCWG) suspension was inoculated with lactic acid bacteria and fermented to improve the functionality of TCWG. The utilization of TCWG was increased directly using the freeze-dried powder. The optimal ratio of TCWG powder and water for fermentation was 1:19 (5%), which was selected by measuring the fluidity and viable cell count according to concentration. The effects on ADH activation and immune cell activation by each ferments with 10 kinds of Lactobacillus sp. strains were examined. The ferments with the Lactobacillus casei KFRI 692 strain showed 5.4 times higher ADH activity and 1.3 times higher ALDH activity than the non-fermented TCWG powder (control). The level of NO production and cytotoxicity was also measured by Raw 264.7 cells. The ferment with the Lac. casei KFRI 692 strain showed the highest level of NO production and lower cytotoxicity than the others. Therefore, the Lac. casei KFRI 692 strain was selected as a strain for fermentation of a TCWG suspension to maximize its functionality. To identify the optimal fermentation time of the selected Lac. casei KFRI 692 strain on the 5% TCWG suspension, the viable cell count of lactic acid bacterial and the changes in pH were observed for 72 hours. 24-hrs was found to be the optimal fermentation time. In this way, fermented TCWG with lactic acid bacteria showed higher ADH activation efficacy and immune cell activation than non-fermented TCWG.
