The present study was planned to examine the effect of different feeding regimens on milk urea concentration and milk protein concentration. The objectives are to describe the diurnal variations of milk urea (MU) concentration and to predict plasma urea (PU) concentration from MU concentration. Six lactating Murrah buffaloes were distributed in two groups and were fed two different diets in a crossover design. The diets consisted of leguminous crops as diet 1 (berseem (Trifolium alexandrinum)+concentrate mixture 1+wheat straw)) and non-leguminous crops as diet 2 (oats (Avana sativa)+concentrate mixture 2+wheat straw). All the diets were isocaloric and isonitrogenous. Each diet was fed to the animals for a period of 28 days, followed by a 10 day gap to obviate the carry over effect of the previous diet and then a switch over to the other diet. Digestibility trials were conducted on the last 7 days of each feeding period. Milk samples were collected on day 3, 7, 10, 14, 17, 21, 24 and 28 of the feeding period and blood samples were collected on the same day at morning within 30 minutes after morning milking. The average milk urea (MU) values (mg/dl) differed significantly (p<0.01) and were 44.83${\pm}$0.62 and 42.53${\pm}$0.73, respectively, for diets 1 and 2. Milk urea concentrations (mg/dl) also varied (p 0.01) among the days of feeding period, but were stabilized after 10th day of feeding period. In contrast, diets and days of feeding period had no significant effect on percent milk protein. Plasma urea concentration showed a significant (p<0.01) positive correlation (r = 0.93) with MU concentration. To predict the PU from MU the following equation was developed 'PU = 10.67${\pm}$0.76${\times}$MU (mg/dl) with $R^2$ = 0.87'. A clear diurnal variation of MU was found with lowered morning value (42.04${\pm}$0.68 mg/dl) than the evening value (45.32${\pm}$0.66 mg/dl). Present findings suggested that MU or PU concentration could be used as an indicator to monitor the feeding strategy. Plasma urea can be predicted from MU, whenever interpretation of milk urea data required consideration of diurnal variation.
In order to clarify the morphological changes in the mammary glands of cows affected with coliform (Escherichia coli) mastitis, histopathological observations were undertaken on the mastitis of the lactating rabbits which was experimentally induced with E. coli or its endotoxin isolated from cases of acute and chronic matitis in dairy cattle. In the bacterial suspension-infused groups the affected quarters of udder showed cloudy swelling, hyperemia and hemorrhage to local necrosis and firmness. The microscopic findings of early stage of the mastitis were appearance of large numbers of heterophils in the glandular lumina and ducts accompanied by degeneration, necrosis and desquamation of epithelial cells, and also infiltration of heterophils, hemorrhage and edema in the interstitial tissue, and destruction of alveoli. Later, proliferation of firoblasts, plasma cells, lymphocytes, eosinophils and histiocytes appeared in the glandular tissue and necrotic foci of glandular tissue were surrounded by highly proliferated connective tissue. Granuloma-like inflammatory changes could be observed in the glandular tissue on the 7th days after infusion. The inflammatory response in the group infused with E. coli strain isolated from the natural case of acute mastitis was rapid and severe as compared with that of chronic mastitis. In the endotoxin-infused group the morphological changes were similiar to those of the bacterial suspension-infused groups.
Objective: The objectives of the study were to assess the effect of cacao bean husk as bedding material in free-stall barn on the behavior, productivity, and udder health of dairy cattle, and on the ammonia concentrations in the barn. Methods: Four different stall surfaces (no bedding, cacao bean husk, sawdust, and chopped wheat straw) were each continuously tested for a period of 1 week to determine their effects on nine lactating Holstein cows housed in the free-stall barn with rubber matting. The lying time and the milk yield were measured between d 4 and d 7. Blood samples for plasma cortisol concentration and teat swabs for bacterial counts were obtained prior to morning milking on d 7. The time-averaged gas-phase ammonia concentrations in the barn were measured between d 2 and d 7. Results: The cows spent approximately 2 h more per day lying in the stalls when bedding was available than without bedding. The milk yield increased in the experimental periods when cows had access to bedding materials as compared to the period without bedding. The lying time was positively correlated with the milk yield. Bacterial counts on the teat ends recorded for cows housed on cacao bean husk were significantly lower than those recorded for cows housed without bedding. Ammonia concentration under cacao bean husk bedding decreased by 6%, 15%, and 21% as compared to no bedding, sawdust, and chopped wheat straw, respectively. The cortisol concentration was lowest in the period when cacao bean husk bedding was used. We observed a positive correlation between the ammonia concentrations in the barn and the plasma cortisol concentrations. Conclusion: Cacao bean husk is a potential alternative of conventional bedding material, such as sawdust or chopped wheat straw, with beneficial effects on udder health and ammonia concentrations in the barns.
Recently, Jersey cattle was introduced and produced by embryo transfer to Korea. This study was conducted to investigate the differences of milk compositions between Jersey and Holstein cows and the relationship between days in milk (DIM) and milk compositions during early lactation. Data were collected from twelve lactating cows from Department of Animal Resources Development at National Institute of Animal Science. Cows in parity 1 were used, and calved at spring from April to March of 2017. All cows were housed in two sections within a free-stall barn, which divided into six from each breed, and received a basal total mixed ration. Milk samples of each cow were collected at 3 DIM and 30 DIM for analyzing the milk compositions, including fatty acids (FA), amino acids and minerals. Total solids, citrate, and milk urea nitrogen level were differed between the breeds (p < 0.05). As DIM went from 3 to 30, milk protein, total solids, and somatic cell count decreased (p < 0.05), but lactose increased in all breed milk (p < 0.05). Citrate and free fatty acid (FFA) elevated in Jersey milk (p < 0.05), whereas reduced in Holstein milk (p < 0.05). Proportions of some individual FA varied from the breeds. Myristic (C14:0), palmitic (C16:0), and arachidonic acid (C20:4) in milk from all cows were higher at 3 DIM than at 30 DIM (p < 0.05). Also, stearic (C18:0) and oleic acid (C18:1) were lower at 3 DIM than at 30 DIM (p < 0.05), and the C18:1 to C18:0 ratio was significantly differed in DIM × breed interactions (p < 0.05). The contents of the individual amino acids did not differ from the breeds. Calcium, phosphorous, magnesium, and zinc (Zn) contents was significantly increased in Holstein milk than Jersey milk at 3 DIM. Also, K and Zn concentrations were higher in Holstein milk than in Jersey milk at 30 DIM (p < 0.05). It was concluded that Jersey cows would produce more effective milk in processing dairy products and more proper energy status compared with Holstein cows in early lactation under the same environmental and nutritional conditions.

Objective: Feed additives that modify rumen fermentation can be used to prevent metabolic disturbances such as acidosis and optimize beef cattle production. The study evaluated the effects of liquid and powdered forms of polyclonal antibody preparation (PAP) against Streptococcus bovis and Fusobacterium necrophorum on rumen fermentation parameters in ruminally cannulated non-lactating dairy cows that were adapted or unadapted to a high concentrate diet. Methods: A double 3×3 Latin square design was used with three PAP treatments (control, powdered, and liquid PAP) and two adaptation protocols (adapted, unadapted; applied to the square). Adapted animals were transitioned for 2 weeks from an all-forage to an 80% concentrate diet, while unadapted animals were switched abruptly. Results: Interactions between sampling time and adaptation were observed; 12 h after feeding, the adapted group had lower ruminal pH and greater total short chain fatty acid concentrations than the unadapted group, while the opposite was observed after 24 h. Acetate:propionate ratio, molar proportion of butyrate and ammonia nitrogen concentration were generally greater in adapted than unadapted cattle up to 36 h after feeding. Adaptation promoted 3.5 times the number of Entodinium protozoa but copy numbers of Streptococcus bovis and Fibrobacter succinogens genes in rumen fluid were not affected. However, neither liquid nor powdered forms of PAP altered rumen acidosis variables in adapted or unadapted animals. Conclusion: Adaptation of cattle to highly fermentable carbohydrate diets promoted a more stable ruminal environment, but PAP was not effective in this study in which no animal experienced acute or sub-acute rumen acidosis.
Appropriation of partial substitution of concentrate mixture by urea-molasses-mineral block (UMMB) lick supplements for 20 lactating crossbred cows in 2nd and 3nd lactation was studied. Animals fed on wheat straw ad lib. and Berseem (Trifolium alaxandrium) fodder @ 1.5 kg/d on dry matter basis. Animals of control group were given concentrate supplement, while in treatment groups 10% of the concentrate requirement was substituted with 3 different types of UMMB lick type A ($T_1$), type B ($T_2$) and type C ($T_3$). CP content of the ration was 15%. Total dry matter intake (DMI) was about 1.0 kg/kg of fat corrected milk (FCM) yield and was not significantly different between control and treatment groups. Digestibility of neither proximate principles nor cell wall constituents were deviated on UMMB licks partial supplementation. FCM yield was increased by 140, 410 and 460 g/d, in $T_1$, $T_2$ and $T_3$, respectively, in comparison to control group but differences were statistically invalid. Though fat per cent was reduced, fat yields were remain constant among treatments. Milk composition was unaltered except significant difference (p < 0.01) in non-protein nitrogen (NPN) content. Gross-N and digestible-N conversion was significantly higher (p < 0.01) with $T_1$, $T_2$ and $T_3$ than control group. Energy utilization efficiency for milk production was only 36%. Result demonstrated that UMMB licks could be partial supplemented up to 10% of the concentrate requirement of crossbred cows yielding on an average 14kg/d without any adverse effect on feed intake, nutrient utilization and mild production. Comparatively, UMMB lick type B and C was proved better than type A and also economically viable.
Fifteen lactating crossbred cows were randomly allotted to three groups of 5 each, and fed three isoproteinous concentrate mixture varying in RDP and UDP ratios, viz. 71: 29 (T1) and 58: 42 (T2) and 44: 56 (T3), along with green maize and wheat straw given as 2/3 and 1/3 of total roughage respectively, for a period of 120 days. The DM intake (kg/d) differed significantly (p<0.01) among the treatments as well as among the fortnights over a period of 120 days. DMI (kg/d) progressively increased from first to eighth fortnight in all the treatments. The daily DMI (% BW) was significantly (p<0.01) lower in T1 (2.37) than those of T2 (2.82) and T3 (2.67). The body weights of cows decreased up to 4th fortnight in T1 and up to 3rd fortnight in T2 and T3, then it showed an increasing trend till the end of the experiment in all the treatments. Cows in T1 lost 10 kg body weight but cows in T2 and T3 gained 23 and 12 kg the body weight, respectively. Both the milk and FCM yield differed significantly (p<0.01) among the fortnights. The FCM yield increased up to 2nd fortnight in all the treatments and thereafter, the FCM yield declined gradually as the lactation advanced. The FCM yield (kg/d) was significantly (p<0.05) higher in T3 (10.47) than in T2 (9.81) and T1 (9.68), however, milk yield, SCM yield and milk energy yield did not differ among the treatments as well as among the fortnights. Fat and protein % in milk increased as the lactation advanced. However, fortnightly SNF % in milk showed an irregular trend. The % fat, protein, SNF and total solids in milk differed significantly (p<0.01) among the fortnights. The % fat and protein in milk varied significantly (p<0.01) among the treatments, being lowest in T1 and highest in T3. The feed efficiency for milk production showed a non-significant variation among the treatments as well as among the fortnights, but increased with the increase in UDP level. It is concluded that by increasing the UDP level from 29 to 56 per cent of CP in the diet of medium producing cows, the milk production increases and cost of milk production reduces.
This study was to evaluate the usefulness of a programmed reproductive management including Presynch-Ovsynch-Resynch protocol in lactating dairy cows. Nine hundred and thirty four cows calved during February to May 2008 were used for this study. Cows received im injections of 25 mg $PGF_{2{\alpha}}$ at $45{\pm}3$ and $59{\pm}3$ days in milk (DIM). Fourteen days later, Ovsynch was initiated with an im injection of $100\;{\mu}g$ GnRH ($73{\pm}3$ DIM), 25 mg $PGF_{2{\alpha}}$ 7 days later ($80{\pm}3$ DIM), $100\;{\mu}g$ GnRH 56 h later and timed artificial insemination (TAI) 16 h after the GnRH injection. Cows showed estrus during the Presynch-Ovsynch, were inseminated artificially according to am-pm rule. Pregnancy was determined on 32 days after AI using ultrasonography. Cows diagnosed pregnant were re-examined for pregnancy by rectal palpation at 60 days after AI. Non-pregnant cows were resynchronized by receiving $100\;{\mu}g$ GnRH, 25 mg $PGF_{2{\alpha}}$ 7 days later, $100\;{\mu}g$ GnRH 56 h later and TAI 16 h after. Cows with estrus since the first AI before the completion of Resynch protocol were also inseminated artificially according to am-pm rule. Pregnancy was determined by the same manner as following the first AI. Fifty five percents of the cows treated showed estrus and received AI before completion of the Presynch-Ovsynch protocol, while 45% received TAI, regardless of estrus exhibition following the completion of the protocol. The pregnancy rate following the first AI was higher in cows that showed estrus and received AI (43.3 and 38.5%) during the Presynch-Ovsynch than cows with TAI (34.6 and 29.6%) on 32 and 60 days after AI (P < 0.01). Sixty six percents of cows diagnosed non-pregnant following the first AI showed estrus before the completion of Resynch protocol and received AI, while 34% received TAI, regardless of estrus exhibition following the completion of the protocol. The pregnancy rate following the second AI was higher in cows that showed estrus before the completion of Resynch protocol and received AI (40.2 and 36.8%) than cows with TAI (21.2 and 18.2%) on 32 and 60 days after AI (P < 0.01). The cumulative pregnancy rates following the first and second AIs were 60.8 and 53.9% on 32 and 60 days after AI, respectively. These data suggest that inclusion of Presynch-Ovsynch-Resynch protocol and estrus detection into the programmed reproductive management might be an alternative option to control dairy cattle breeding.

Objective: The study aimed to evaluate sugarcane bagasse as roughage in lactating cow on feed intake, digestibility, ingestive behavior, milk production and composition, and microbial protein synthesis. Methods: Ten Girolando cows at initial body weight of $450{\pm}25.6kg$ and at $143.7{\pm}30.7days$ in milk were assigned in two $5{\times}5$ Latin square designs. Five 21-day experimental periods were adopted ($1^{\circ}$ to 14-day: diets adaptation period; $15^{\circ}$ to 21-day: data collection and sampling period). The diets consisted of four different levels of sugarcane bagasse (45%, 50%, 55%, and 60%) and a control diet, commonly adopted in the region, based on spineless cactus (25% sugarcane bagasse), formulated to meet 12 kg/d milk yield. Results: The dry matter (DM), organic matter (OM), and total digestible nutrients intakes and DM and OM digestibilities observed for 45% and 50% bagasse inclusion were similar to control diet, while that 55% and 60% bagasse inclusion were lower. Cows fed control diet, and bagasse diets of 45%, and 50% levels had the nutritional requirements attended, that guaranteed 12 kg/d of milk yield. The crude protein intake and digestibility of cows fed 45%, 50%, and 55% of bagasse inclusion were similar to control diet. The neutral detergent fiber (NDF) intake and digestibility differ for all bagasse diets related to control diet, while the non-fiber carbohydrates intake and digestibility for cows fed 45% of bagasse were similar for control diet. The intakes and digestibilities of nutrients decreased linearly in function of bagasse inclusion; NDF and indigestible NDF intakes did not vary. The ruminating time, feeding and rumination efficiency, microbial protein synthesis and milk yield decreased linearly with sugarcane bagasse inclusion. Conclusion: Sugarcane bagasse decreases milk production; however, its inclusion level in between 45% to 50% associated to concentrate could replace diets based on spineless cactus for crossbred dairy cow's producing 12 kg/d of milk.
