생쥐의 근피로(筋疲勞) 및 혈액성분(血液成分)에 대한 단기(短期) 인삼투여(人蔘投與)의 효과(效果)에 관한 연구(硏究) (A study on the short-term effects of Ginseng Radix Alba extract on muscle fatigue and blood components of the rats.)

  • Purpose: This study was conducted to investigate the short-term effects of Ginseng Radix Alba extract solution on muscle fatigue and concentration of blood components especially in glucose and lactate dehydrogenase of the rats. Methods: The 4~6-week-old ICR rats are used in the study, and we administered the extract solution of Ginseng Radix Alba concentration of 1, 10, 100mg/0.3ml to each rats and water to another rats(control group) once a day for each 30 and 60 days. After 30 and 60 days. we measured the persistent time of swimming exercise test and the results of grip strength test. And we also examined the plasma concentration of glucose, and lactate dehydrogenase. Results: In swimming exercise test, the swimming time of the 1, 10, 100mg/0.3ml groups for 30 days and the 100mg/0.3ml group for 60 days significantly increased than control group. and we found that as the concentration increased, the swimming time was also increased for 30 and 60 days experiment. And in grip strength test, the grip strength of the 1, 10, 100mg/0.3ml groups for 30 days and the 10mg/0.3ml group for 60 days significantly increased than control group. and we found that as the concentration increased, the grip strength was also increased for 30 days experiment. Plasma concentration of lactate dehydrogenase was significantly increased in the 10mg/0.3ml group for 60 days than control group. And there was no significant differences between the control and the treatment group in the plasma concentration of glucose. Conclusion: This study show that Ginseng Radix Alba can increase the muscle strength and antifatigue effect.

Recovery of Lactic Acid from Fermentation Broth by the Two-Stage Process of Nanofiltration and Water-Splitting Electrodialysis

  • A two-stage process of nanofiltration and water-splitting electrodialysis was investigated for lactic acid recovery from fermentation broth. In this process, sodium lactate is isolated from fermentation broth in the first stage of nanofiltration by using an NTR-729HF membrane, and then is converted to lactic acid in the second stage by water-splitting electrodialysis. To determine the optimal operating conditions for nanofiltration, the effects of pressure, lactate concentration, pH, and known added impurities were studied. Lactate rejection was less than 5%, magnesium rejection approximated 45%, and calcium rejection was at 40%. In subsequent water-splitting electrodialysis, both the sodium lactate conversion to lactic acid and sodium hydroxide recovery, were about 95%, with a power requirement of $0.9{\sim}1.0\; kWh$ per kg of lactate.

생쥐 체외수정과 배아 발달에 미치는 에너지원의 영향 (Effects of Energy Substrates on In Vitro Fertilization of the Mouse Oocytes with Cumulus Mass and their Developments)

  • 난구세포는 lactate와 pyruvate를 쉽게 생성하고, 이로 인해 배양액내 에너지원의 농도를 변화시켜 난자의 수정과 배양에 영향을 주는 것으로 알려져 있다. Glucose, lactate 및 pyruvate의 농도가 상이한 M16, MTF 및 CZB배양액에서 난구세포를 포함한 생쥐난자의 체외 수정과 발달을 관찰하여, 이들 기질의 영향에 대하여 살펴보고 배양액의 유용성을 재검토하고자 하였다. Glucose를 제거한 배양액 (CZ2 배양액)에서 수정율과 배반포 형성율은 다른 배양액에 비해 유의하게 감소되었으나 (p<0.05), 생쥐 난관액과 동일한 기질 농도로 조성된 MTl (난간액이 난구세포를 포함하고 있을 때) 및 MT2배양액 (난관액이 난구세포를 포함하고 있지 않을 때)과 glucose를 포함한 modified CZB배양액에서는 영향이 없었다. 이와 같은 결과로 기질의 농도를 생리적 수준으로 조정한 배양액의 이용은 난구세포를 포함한 생쥐 난자의 체외수정과 그 발달을 향상시키지 못하고, glucose의 제거는 악영향을 나타내는 것으로 사료된다.

Evaluation of Interfering Substances in Routine Chemistry Tests Using Toshiba TBA-C8000 Chemistry Analyzer

  • In clinical chemistry tests, the interfering substances such as hemoglobin, lipid, bilirubin, and drugs, etc. can cause the changes of test results performed by spectrophotometrical methods. We evaluated the effects of interfering substances on the test results by adding interfering substances on the samples in the 19 kinds of clinical chemistry tests such as aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, gamma-glutamyltransferase, total protein, albumin, glucose, total cholesterol, total bilirubin, triglyceride, uric acid, calcium, inorganic phosphours, high density lipoprotein cholesterol, low density lipoprotein cholesterol, creatinine, blood urea nitrogen, and C-reactive protein using newly implemented automatic chemical analyzer Toshiba TBA-C8000 under the direction of CLSI EP07-A guideline. Hemolytic samples show increased concentration of total protein, aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase and reduced concentration of total bilirubin, alkaline phosphatase by interfering effect. Hyperlipemic samples show increased concentration of total protein and alkaline phosphatase and reduced concentration of low density lipoprotein cholesterol. The samples with conjugated bilirubinemia show increased concentration of inorganic phosphours, otherwise the samples with unconjugated bilirubinemia show no interference or allowable range in the test result.

탈염전기투석에 의한 모사발효액으로부터의 암모늄 락테이트의 회수 (Recovery of ammonium lactates from simulated fermentation broth by desalting electrodialysis)

  • An experimental study was carried out on the recovery of ammonium lactate from simulated fermentation brothby desalting electrodialysis (DSED). All experiments, using AM-1 and CM-1 membranes, were operated in a constant current and a subsequent constant-voltage mode, and the switching point was determined based on the previous results. The effects of operating conditions such as operating current, initial feed concentration, and initial feed pH were investigated. Increased operating current resulted in a decreased operating time but an increased energy consumption per unit amount of ammonium lactate recovered. As the initial feed concentration was increased, the operating time increased while energy consumption decreased. The operating time and energy consumption slightly decreased as the initial feed pH was increased up to 6.0. However, no significant influences on the recovery of ammonium lactate were observed over 6.0. The rejections of acetate, glucose, and proteins were $10\%,\;93\%,\;and\;98\%$, respectively. Sulfate was not rejected at all.

2단(유산발효+광발효) 발효공정을 통한 음식물쓰레기로부터의 수소생산 (Biohydrogen Production from Food Waste by Two-Stage (Lactate+Photo)-Fermentation Process)

  • In the present work, it was attempted to produce $H_2$ from food waste by the two-stage fermentation system. Food waste was acidified to lactate by using indigenous lactic acid bacteria under mesophilic condition, and the lactate fermentation effluent (LFE) was subsequently converted to $H_2$ by photo-fermentation. $Rhodobacter$ $sphaeroides$ KD131 was used as the photo-fermenting bacteria. The optimal conditions for lactate fermentation were found to be pH of 5.5 and substrate concentration of 30 g Carbo. COD/L, under which yielded 1.6 mol lactate/mol glucose. By filtering the LFE and adding trace metal, $H_2$ production increased by more than three times compared to using raw LFE, and finally reached the $H_2$ yield of 3.6 mol $H_2$/mol lactate. Via the developed two-stage fermentation system $H_2$ yield of 5.8 mol $H_2$/mol glucose was achieved from food waste, whose value was the highest that ever recorded.

구자를 장기간(長期間) 투여(投與) 했을 때 생쥐의 근피로(筋疲勞)와 근력(筋力)에 미치는 영향(影響) (Long Term Effect of Allii tuberosi semen on Muscle Fatigue and Muscular Strength)

  • Purpose: This study was conducted to investigate the muscle anti-fatigue effects of Allii tuberosi semen after long term administration. Materials and Methods: 4-6 weeks old ICR mice were used in this study and we administered the water soluble extracts of Allii tuberosi semen in the concentration of 1, 10 and 100mg/0.3ml to each 5 mice (ATS group) and same volume of normal saline was administered to 5 mice (control group) once a day for 90 or 120 days. After the administration we performed the swimming exercise test and the grip strength test. And we measured the concentration of glucose and the activity of lactate dehydrogenase in plasma regarded as biochemical indicator related with the muscular fatigue. Results: In swimming exercise test, all ATS groups showed increased time compared with relative control group after 90 days administration. But after 120 days adminstration 10 and 100mg/0.3ml ATS groups showed increased results. In grip strength test, all ATS group increased grip strength after 90 days administration and 10mg/0.3ml ATS group showed increased result after 120 days administration. All the results of plasma glucose didn't showed statistically significant difference after 90 and 120 days administration. In the plasma concentration of activity level of lactate dehydrogenase, 1mg/0.3ml and 10mg/0.3ml ATS groups showed decreased concentration compared with relative control group after 90 days administration. But after 120 days adminstration 1mg/0.3ml ATS groups showed decreased results. Conclusion: From this study we could find the anti-fatigue effect of Allii tuberosi semen in the muscle strength related test.

혼합곡 식이의 지구력 향상 효과 (Improving Effects of Multigrain Feed on Endurance)

  • 본 연구에서는 4주간 혼합곡 식이를 공급하여 treadmill을 통한 운동시간과 glycogen 저장능력 변화를 관찰함으로써 지구력 개선에 대한 평가를 하였다. 7주령 SD rats은 정상군 (Nor, 정상식이, n=6), 대조군(Con, 20% 정상식이+80% milled rice, n=6), GI(혼합곡 식이 I, n=6), GII(혼합곡 식이 II, n=6), GIII(혼합곡 III, n=6)와 GIV(혼합곡 식이 IV, n=6)으로 임의 군배정을 하였다. 지구력 시험은 24일간의 적응훈련과 혼합곡 식이 공급을 한 후 실시하였다. 운동 지속시간과 glycogen 저장능력은 혼합곡 식이 공급에 의해서 증가하였다. 더욱이 25 min 운동 그리고/또는 탈진 시까지의 운동 후에서 혈액 내 무기질 인, CPK와 lactate의 농도는 혼합곡 투여에 의하여 감소하였으나, GOP, GTP, lactate LDH의 농도는 군간 차이는 보이지 않았다. 본 연구에서는 4주간의 혼합곡 식이가 운동 지속 시간과 glycogen 장 능력이 증가되었으며, 피로물질인 무기질 인, CPK와 lactate의 농도가 감소시킴으로써 지구력 개선에 도움을 줄 것으로 생각된다.

탄소나노튜브로 개질된 탄소종이 기반 젖산산화효소 - 카탈레이즈 전극 제작 및 특성 분석 (Fabrication and Characterization of Carbon Nanotube-modified Carbon Paper-based Lactate Oxidase-catalase Electrode)

  • 본 연구에서는 전극의 전기 전도도 증대와 젖산 산화반응의 부산물인 과산화수소 생성 완화가 젖산 산화효소 전극 성능에 미치는 영향을 조사하였다. 유연성 있는 탄소종이 표면을 단일벽 탄소나노튜브로 개질하여 전극의 전기 전도도를 향상시켰다. 카탈라아제를 도입하여 젖산 산화반응에서 발생하는 과산화수소를 제거하였다. 젖산 산화효소와 카탈라아제가 동시에 고정화된 탄소종이 전극은 젖산 산화효소만 고정화된 탄소종이 전극보다 1.7배 많은 전류를 생성하였다. 단일벽 탄소나노튜브로 개질된 탄소종이 표면에 젖산 산화효소와 카탈라아제가 동시에 고정화된 전극은 171 µA의 전류를 생산하였는데, 이는 탄소종이 표면에 젖산 산화효소만 고정화된 전극이 생산하는 전류보다 2배 높은 값이다. 최적화된 전극은 젖산 농도가 20 mM까지 선형반응을 보여 센서용 전극으로 사용 가능함을 확인하였다.

흐름주입분석 기술을 이용한 젖산의 온라인 모니터링

  • On-line monitoring technique for the concentration of lactate by a FIA(Flow Injection Analysis) system was studied. The lactate oxidase(LOD) was immboilized on VA-Epoxy carrier and integrated into the FIA system. The pH, buffer flow rate and temperature for the LOD-FIA were optimized, and the effects of salts and metabolites dissolved in the sample on the activity of immobilized enzyme were investigated. The LOD-FIA has been applied to monitor the concentrations of lactate in a simulated bioprocess. The on-line monitoring data by the LOD-FIA agreed with the off-line data measured by a fluorescence spectroscopy well.
