Interaction between Invertebrate Grazers and Seaweeds in the East Coast of Korea (동해안 조식성 무척추동물과 해조류 간 상호작용)

  • We estimated the distribution of predator-prey interaction strengths for 12 species of herbivores (including amphipods, isopods, gastropods, and sea urchins) and made a regression model that may be applicable to other species. Laboratory experiments were used to determine per capita grazing rate (PCGR; g seaweeds/individual/day). Relationship between the biomass of individual grazers and fourth-root transformed PCGR was fitted to power curve ($y=0.2310x^{0.3290}$, r=0.8864). This finding supported that the grazing efficiency was not even as individual grazers increase in size (biomass). Therefore, the biomass-normalized PCGR was estimated and revealed that smaller size herbivores were more effective grazers. Grazing impact considering density of each taxon was calculated. The sea hare Aplysia kurodai had greatest grazing impact on the seaweed bed and the sea urchin Strongylocentrotus nudus and S. intermedius were ranked in descending order of the impact. The amount of seaweed grazed by the amphipod Elasmopus sp. (>4,000 $ind./m^2$) and Jassa falcata (>2,000 $ind./m^2$) were 3.435 and $1.697mg/m^2/day$ respectively. The combined grazing amount of herbivores was $5,045mg/m^2/day$ in the seaweed bed. Although sea hare and sea urchin had strong impacts on seaweeds, the effects of dense, smaller species could not be seen as negligible. Surprisingly, the calculated grazing potential of sea urchins with a mean density of 3 $ind./m^2$ exceeded the mean production of seaweed cultured in domestic coastal waters in Korea (ca., 5 ton/ha). Small crustaceans were also expected to consume up to 16% of the seaweed production if their densities were rising under weak predation conditions. Considering that the population density of herbivores are strongly controlled by fish, human interference like overfishing may have strong negative effects on persistence of seaweeds communities.

THE FOOD AND GROWTH OF THE LARVAE OF THE ARK SHELL ANADARA BROUGHTONI SCHRENCK (피조개의 먹이와 성장)

  • The larvae of the ark shell Anadare broughtoni(Schrenck) were grown at room temporature (approximately $20.4^{\circ}C$), and fed laboratory-cultured Cyclotella nana. The egg of the ark shell produced in the laboratory measured about $54.9\mu$ in diameter. The embryos gradually developed into larvae up to $110.8\mu$ shell length, $83.9\mu$ shell height and with shell breadth of $58.2\mu$ even in the absence of the algal food. Beyond this sire, however, the growth of the larvae was considerably retarded. The larvae showed better growth rate when they were fed the algal food two days after spawning, i. e., early straight-hinge stage. Daily rate of food consumption varies according to the larval sizes. But the rate increases considerably when the larvae begin to form umbos. In general the rate Is indicated by the following formula: $Y=0.0025161\;X^{2.76459}$. The growth experiments of the larvae indicate that the efficiency of food conversion was higher when fed centrifuged food. Regarding to the difference in the slopes of growth curve, centrifuged food showed better growth rate as compared to those grown with the non-centrifuged food. The smaller the larval size, the greater will be the difference in growth. The larvae began settling when they reathed 261.7 to $289.6\;{\mu}$ in shell length, 199.2 to $221.7\mu$ in shell height and 147.6 to $170.8\mu$ in shell breadth. The time which elapsed from spawning to the larval settlement was about 28 days. The mean growth of the larvae is indicated with regression line and exponential curve equations as follows. Comparison between phosphorus absorption coefficient and Langmuir adsorption maximum (전토양(田土壤) 인산(燐酸)의 흡수계수(吸收係數)와 Langmuir 최대흡착량(最大吸着量)과의 비교연구(比較硏究))

  • Laboratory experiments on the phosphorus adsorption by soil were conducted to evaluate the parameters for determination of phosphorus adsorption capacity of soil, which serve as a basis for establishing the amount of phosphorus required to improve newly reclaimed soil and volcanic ash soil. The calculated Langmuir adsorption maxima varied from 6.2-32.9, 74.7-90.4 and 720-915mg p/100g soil for cultivated soils, non-cultivated soils, and volcanic ash soils respectively. The phosphorus absorption coefficient ranged from 116-179, 161-259 and 1,098-1,205mg p/100g soil for cultivated soils, non-cultivated soils, and volcanic ash soils respectively. The ratio of the phosphorus absorption coefficient to Langmuir adsorption maximum was low in soils of high phosphorus adsorption capacity (1.3-1.5) and high in soils of low phosphorus adsorption capacity (2.2-18.7). Changes in the amount of phosphurus adsorption induced by liming and preaddition of phosphorus were hadly detected by the phosphorus absorption coefficient, which is measured using a test solution with a relatively high phosphorus concentration. The Langmuir adsorption maximum was a more sensitive index of the phosphorus adsorption capacity. The Langmuir adsorption maxima of the non-cultivated soils, which were treated with an amount of calcium hydroxide equivalent to the exchangeable Al and incubated ($25-30^{\circ}C$) for 40 days at field capacity, were lower than the original soils. The change in the adorption maximum on incubation following the liming of soils was insignificant for other soils. The secondary adsorption maximum of soils, which received phosphorus equivalent to the Langmuir adsorption maximum of the limed soils incubated ($25-30^{\circ}C$) for 50 days at held capacity, was 74.5, 5.6 and 23.8% of the primary adsorption maximum for volcanic ash soils, non-cultivated soils, and cultivated soils respectively. The amount of phosphorus adsorbed by soils increased quadratically with the concentration of phosphorus solution added to the soils. The amount of phosphorus adsorbed by 5-g soil samples from 100ml of 100- and 1,000mg p/l solution for the mineral soils and volcanic ash soils respectively was found to be close to the Langmuir adsorption maximum. The amount of the phosphorus adsorbed at these concentrations is defined as a saturation adsorption maximum and proposed as a new parameter for the phosphorus adsorption capacity of the soil. The evaluation of the phosphorus adsorption capacity by the saturation adsorption maximum is regarded as a more practical method in that it obviates the need for the various concentrations used for the determination of the Langmuir adsorption maximum.

Gene Expression Profiles in Cervical Cancer with Radiation Therapy Alone and Chemo-radiation Therapy (자궁경부암의 방사선치료 및 방사선항암화학 병용치료에 따른 유전자발현 조절양상)

  • Purpose : To analyze the gene expression Profiles of uterine ceulcal cancer, and its variation after radiation therapy, with or without concurrent chemotherapy, using a CDNA microarray. Materials and Methods :Sixteen patients, 8 with squamous ceil carcinomas of the uterine cervix, who were treated with radiation alone, and the other 8 treated w14h concurrent chemo-radiation, were Included in the study. Before the starling of the treatment, tumor biopsies were carried out, and the second time biopsies were peformed after a radiation dose of 16.2$\~$27 Gy. Three normal cervix tissues were used as a control group. The microarray experiments were peformed with 5 groups of the total RNAs extracted individually and then admixed as control, pre-radiation therapy alone, during-radiation therapy alone, pre-chemoradiation therapy, and during-chemoradlation therapy. The 33P-iabeled CDNAS were synthesized from the total RNAs of each group, by reverse transcription, and then they were hybridized to the CDNA microarray membrane. The gene expression of each microarrays was captured by the intensity of each spot produced by the radioactive isotopes. The pixels per spot were counted with an Arrayguage, and were exported to Microsoft Excel The data were normalized by the Z transformation, and the comparisons were peformed on the Z-ratio values calculated. Results : The expressions of 15 genes, including integrin linked kinase (ILK), CDC28 protein kinase 2, Spry 2, and ERK 3, were increased with the Z-ratio values of over 2.0 for the cervix cancer tissues compared to those for the normal controls. Those genes were involved In cell growth and proliferation, cell cycle control, or signal transduction. The expressions of the other 6 genes, Including G protein coupled receptor kinase 5, were decreased with the Z-ratio values of below -2.0. After the radiation thorapy, most of the genes, with a previously Increase expressions, represented the decreased expression profiles, and the genes, with the Z-ratio values of over 2.0, were cyclic nucleotlde gated channel and 3 Expressed sequence tags (EST). In the concurrent chemo-radiation group, the genes involved in cell growth and proliferation, cell cycle control, and signal transduction were shown to have increased expressions compared to the radiation therapy alone group. The expressions of genes involved in anglogenesis (angiopoietln-2), immune reactions (formyl peptide receptor-iike 1), and DNA repair (CAMP phosphodiesterase) were increased, however, the expression of gene involved In apoptosls (death associated protein kinase) was decreased. Conclusion : The different kinds of genes involved in the development and progression of cervical cancer were identified with the CDNA microarray, and the proposed theory is that the proliferation signal stalls with ILK, and is amplified with Spry 2 and MAPK signaling, and the cellular mitoses are Increased with the increased expression oi Cdc 2 and cell division kinases. After the radiation therapy, the expression profiles demonstrated 4he evidence of the decreased cancer cell proliferation. There was no sigificant difference in the morphological findings of cell death between the radiation therapy aione and the chemo-radiation groups In the second time biopsy specimen, however, the gene expression profiles were markedly different, and the mechanism at the molecular level needs further study.

Removal of Red Tide Organisms -2. Flocculation of Red Tide Organisms by Using Loess- (적조생물의 구제 -2. 황토에 의한 적조생물의 응집제거-)

  • The objective of this study was to examine the physicochemical characteristics of coagulation reaction between loess and red tide organisms (RTO) and its feasibility, in developing a technology for the removal of RTO bloom in coastal sea. The physicochemical characteristics of loess were examined for a particle size distribution, surface characteristics by scanning electron microscope, zeta potential, and alkalinity and pH variations in sea water. Two kinds of RTO that were used in this study, Cylindrothen closterium and Skeietonema costatum, were sampled in Masan bay and were cultured in laboratory. Coagulation experiments were conducted using various concentrations of loess, RTO, and a jar tester. The supernatant and RTO culture solution were analyzed for pH, alkalinity, RTO cell number. A negative zeta potential of loess increased with increasing pH at $10^(-3)M$ NaCl solution and had -71.3 mV at pH 9.36. Loess had a positive zeta potential of +1,8 mV at pH 1.98, which resulted in a characteristic of material having an amphoteric surface charge. In NaCl and $CaCl_2$, solutions, loess had a decreasing negative zeta potential with increasing $Na^+\;and\;Ca^(+2)$ ion concentration and then didn't result in a charge reversal due to not occurring specific adsorption for $Na^+$ ion while resulted in a charge reversal due to occurring specific adsorption for $Ca^(+2)$ ion. In sea water, loess and RTO showed the similar zeta potential values of -112,1 and -9.2 mV, respectively and sea sand powder showed the highest zeta potential value of -25.7 mV in the clays. EDLs (electrical double-layers) of loess and RTO were extremely compressed due to high concentration of salts included in sea water, As a result, there didn't almost exist EDL repulsive force between loess and RTO approaching each other and then LVDW (London-yan der Waals) attractive force was always larger than EDL repulsive force to easily form a floe. Removal rates of RTO exponentially increased with increasing a loess concentration. The removal rates steeply increased until $800 mg/l$ of loess, and reached $100{\%}$ at 6,400 mg/l of loess. Removal rates of RTO exponentially increased with increasing a G-value. This indicated that mixing (i.e., collision among particles) was very important for a coagulation reaction. Loess showed the highest RTO removal rates in the clays.

Tissue Culture Method as a Possible Tool to Study Herbicidal Behaviour and Herbicide Tolerance Screening (조직배양(組織培養) 방법(方法)을 이용(利用)한 제초제(除草劑) 작용성(作用性) 및 제초제(除草劑) 저항성(抵抗性) 검정방법(檢定方法) 연구(硏究))

  • A series of laboratory and greenhouse experiments were conducted to find out the possibility of tissue culture and cell culture methods as a tool to study herbicidal behaviour and herbicide tolerance screening from 1985 to 1986 at the Yeongnam Crop Experiment Station. For dehulled-rice culture, pure agar medium was the most appropriate in rice growth campared to other media used for plant tissue culture method. All the media but the pure agar medium resulted in growth retardance by approximately 50% and this effect was more pronounced to root growth than shoot growth. Herbicidal phytotoxicity was enhanced under light condition for butachlor, 2.4-D, and propanil while this effect was reversed for DPX F-5384 and CGA 142464, respectively. And also, herbicides of butachlor, chlornitrofen, oxadiazon, and BAS-514 resulted in more phytotoxic effect when shoot and root of rice were exposed to herbicide than root exposure only while other used herbicides exhibited no significant difference between two exposure regimes. Similar response was obtained from Echinochloa crusgalli even though the degree of growth retardance was much greater. Particularly, butachlor, 2.4-D, chlornitrofen, oxadiaxon, pyrazolate and BAS-514 totally inhibited chlorophyll biosynthesis even at the single contact of root. Apparent cultivar differences to herbicide were observed at the young seedling culture method and dehulled rice cultivars were more tolerant in DPX F-5384, NC-311, pyrazolate and pyrazoxyfen, respectively. For derant than other types or rice cultivar in butachlor, pretilachlor, perfluidone and oxadiazon while Tongil-type rice cultivars were more tolerant in DPXF-5384, NC-311, Pyrazolate and Pyrazoxyfen, respectively. For dehulled rice culture, on the other hand, Japonica-type rice cultivar was less tolerant to herbicides of butachlor, propanil, chlornitrofen and oxadiazon that was reversed trend to young seedling culture test. Cultivar differences were also exhibited within same cultivar type. In general, relatively higher tolerant cultivars were Milyang 42, Cheongcheongbyeo, Samgangbyeo, Chilseoungbyeo for Tongil-type, Somjinbyeo for Japonica-type and IR50 for Indica-type, respectively. The response of callus growth showed similar to dehulled rice culture method in all herbicides regardless of property variables. However, concentration response was much sensitive in callus response. The concentration ranges of $10^{-9}M-10^(-8)M$ were appropriate to distinguish the difference between herbicides for E. crusgalli callus growth. Among used herbicides, BAS-514 was the most effective to E. crusgalli callus growth. Based on the above results, tissue culture method could be successfully used as a tool for studying herbicidal behaviour and tolerance screening to herbicide.

Arsenic Removal Mechanism of the Residual Slag Generated after the Mineral Carbonation Process in Aqueous System (광물탄산화 공정 이후 발생하는 잔사슬래그의 수계 내 비소 제거 기작)

  • Laboratory-scale experiments were performed to identify the As removal mechanism of the residual slag generated after the mineral carbonation process. The residual slags were manufactured from the steelmaking slag (blast oxygen furnace slag: BOF) through direct and indirect carbonation process. RDBOF (residual BOF after the direct carbonation) and RIBOF (residual BOF after the indirect carbonation) showed different physicochemical-structural characteristics compared with raw BOF such as chemical-mineralogical properties, the pH level of leachate and forming micropores on the surface of the slag. In batch experiment, 0.1 g of residual slag was added to 10 mL of As-solution (initial concentration: 203.6 mg/L) titrated at various pH levels. The RDBOF showed 99.3% of As removal efficiency at initial pH 1, while it sharply decreased with the increase of initial pH. As the initial pH of solution decreased, the dissolution of carbonate minerals covering the surface was accelerated, increasing the exposed area of Fe-oxide and promoting the adsorption of As-oxyanions on the RDBOF surface. Whereas, the As removal efficiency of RIBOF increased with the increase of initial pH levels, and it reached up to 70% at initial pH 10. Considering the PZC (point of zero charge) of the RIBOF (pH 4.5), it was hardly expected that the electrical adsorption of As-oxyanion on surface of the RIBOF at initial pH of 4-10. Nevertheless it was observed that As-oxyanion was linked to the Fe-oxide on the RIBOF surface by the cation bridge effect of divalent cations such as Ca2+, Mn2+, and Fe2+. The surface of RIBOF became stronger negatively charged, the cation bridge effect was more strictly enforced, and more As can be fixed on the RIBOF surface. However, the Ca-products start to precipitate on the surface at pH 10-11 or higher and they even prevent the surface adsorption of As-oxyanion by Fe-oxide. The TCLP test was performed to evaluate the stability of As fixed on the surface of the residual slag after the batch experiment. Results supported that RDBOF and RIBOF firmly fixed As over the wide pH levels, by considering their As desorption rate of less than 2%. From the results of this study, it was proved that both residual slags can be used as an eco-friendly and low-cost As remover with high As removal efficiency and high stability and they also overcome the pH increase in solution, which is the disadvantage of existing steelmaking slag as an As remover.

Studies on the Effects of Several Factors on Soil Erosion (토양침식(土壤侵蝕)에 작용(作用)하는 몇가지 요인(要因)의 영향(影響)에 관(關)한 연구(硏究))

  • This study was conducted on the major factors affecting soil erosion and surface run-off. In order to investigate the processes and mechanisms of soil erosion on denuded forest-land in Korea, and to systematize the magnitudes of influences and interactions between individual factors, the five major factors adopted in these experiments are soil textures (coarse sand and clay loam), slope steepness ($10^{\circ}$, $20^{\circ}$, $30^{\circ}$ and $40^{\circ}$), rainfall intensities (50, 75 and 100mm/hr), slope mulching methods (bare, coarse straw-mat mulching, grass mulching and anti-erosion liquid mulching) and vegetation densities (sparse, moderate and dense). The processes and mechanisms of soil erosion, and the effects of mulchings on soil erosion as well as surface run-off rates were studied algebraically with four parts of laboratory experiments under the simulated rainfall and another part of field experiment under the natural rainfall. The results in this study are summarized as follows: 1. Experiment factors and surface run-off rates The surface run-off rates under the natural rainfall were resulted about 24.7~28.7% from the bare slopes, about 14.0~16.4% from the straw-mat mulched slopes, about 7.9~9.1% from the liquid mulched slopes, and about 5.6~7.2% from the grass mulched slopes respectively. The surface run-off rates under the simulated rainfall differed greatly according to the rainfall intensity and the mulching method. 2. Magnitudes of influences and interactions of the individual factor on the surface run-off rates. The experimental analyses on the major factors(soils, slopes, rainfalls, mulchings and vegetations) affecting the rates of surface run-off, show that the mean differences of surface run-off rate are significant at 5% level between the soil texture factors, among the slope steepness factors, among the rainfall intensity factors, among the mulching method factors, and among the vegetation density factors respectively. The interactions among the individual factor have a great influence(significant at 1% level) upon the rate of surface run-off, except for the interactions of the factors between soils and slopes; between slopes and vegetations; among soils, slopes and rainfalls; and among soils, slopes and mulchings respectively. On the bare slopes under the simulated rainfall, the magnitude of influences of three factors(soils, slopes and rainfalls) affecting the rate of surface run-off is in the order of the factor of rainfalls, soils and slopes. The magnitude of influences of three factors (soils, rainfalls and mulchings) affecting the rate of surface run-off, on the mulched slopes under the simulated rainfall is in the order of the factor of mulchings, rainfalls and soils and that of influences of the factor of soils, slopes and mulchings is in the order of the factor of mulchings, soils and slopes. On the vegetation growing slopes under the simulated rainfall, the magnitude of influences of three factors (soils, slopes and vegetations) affecting the rate of surface run-off is in the order of the factor of vegetations, soils and slopes. In the same condition of treatments on the field experiment under the natural rainfall, the order of magnitude of influences affecting the rate of surface run-off is the factor of mulchings, soils and slopes. 3. Experiment factors and soil losses The soil losses of the experiment plots differed according to the factors of soil texture, slope steepness, rainfall intensity and mulching method. The soil losses from the coarse soil were increased about 1.1~1.3 times as compared with that of fine soil under the natural rainfall, while the soil losses from the fine soil were increased about 1.2~1.3 times compared with that of coarse soil under the simulated rainfall. The equation of $E=aS^b$ (a, b are constant) between the slope steepness (log S) and soil losses (log E) under the simulated rainfall were developed. The equation of $E=aI^b$ (a, b are constant) between the rainfall intensity (log I) and soil losses (log E) were developed, and b values have a decreasing tendency according to the increase of the slope steepness and rainfall intensity. The soil losses under the natural rainfall were appeared about 38~41% from the coarse straw-mat mulched slopes, about 20~22% from the liquid mulched slopes, about 14~15% from the grass mulched slopes as compared with that of the bare slopes respectively. The soil loss from the vegetation plots showed about 7.1~16.4 times from the sparse plot, about 10.0~17.9 times from the moderate plot and about 11.1~28.1 times from the dense plot as compared with that of the bare slopes. 4. Magnitudes of influences and interactions of the individual factor on the soil erosion. The experimental analyses on the major factors(soils, slopes, rainfalls, mulchings and vegetations) affecting the soil erosion, show that the mean differences of soil losses are highly significant between the soil texture factors, among the slope steepness factors, among the rainfall intensity factors, among the mulching method factors and among the vegetation density factors respectively. The interactions among the individual factor have mostly great influences upon the soil erosion. The magnitude of influences of three factors (soils, slopes and rainfalls) affecting the soil erosion on the bare slopes under the simulated rainfall is in order of the factor of rainfalls, soils and slopes. On the mulched slopes under the simulated rainfall, the magnitude order of influences of three factors(soils, rainfalls and mulchings) affecting the soil erosion is the factor of mulchings, rainfalls and soils, and the order of influences of factor of soils, slopes and mulchings is the factor of mulchings, soils and slopes.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers.
