In previous reports, the authors isolated the algicidal marine bacterium, Alteromonas sp. SR-14 and demonstrated its growth inhibition of diatom, Chaetoceros calcitrans (C. calcitrans). In this paper, we studied the effects of cell free culture filtrate of Alteromonas sp. SR-14 on the growth of C. calcitrans, and the characteristics of the algal growth inhibition substance. The culture filtrate of Alteromonas sp. SR-14 grown in peptone broth showed growth inhibition activity against C. calcitrans. The reasonable culture conditions of the bacterium for producing of algal growth inhibition substances were $15~20^{\circ}$ in temperature, 7.0-9.0 in pH and $23~30{\textperthousand}$ in salinity, respectively. The algal growth inhibition activity of culture filtrate was increased from stationary phase in growth curve of Alteromonas sp. SR-14. The molecular weights of algal growth inhibition substances produced by Alteromonas sp. SR-14 were ranged about from 3 KDa to 12 KDa. Among the substances, less than 10 KDa fraction were stable by heating at $100^{\circ}$ for 10 minutes, while more than 10 KDa fraction were heat labile. According to the experimental results, the algal growth inhibition substance produced by the bacterium was not a single compound.
In order to interpret the effect of IAA on the phosphate metabolism and biosynthesis of organic compounds, Saccharomyces uvarum were cultured in the media treated with various concnetration of IAA $(10^{-3}M,\;10^{-5}M,\;10^{-7}M)$. Sampling at the beginning and intervals of culture, yeast cells fractionated were traced the contents of inorganic phosphate and organic compounds of various fractions. 1. Growth of Saccharomyces uvarum were enhanced by IAA $(10^{-3}M,\;10^{-5}M)$ and phosphate contents in DNA and RNA fractions treated with IAA were accelerated 2.3 times and 2 times in comparison with those of control. 2. Amounts of poly-P"A" and poly-p"B" were increased but poly-P"C" decreased during the culture. Therefore, it is considered that poly-P"C" play on most important role as a phosphate pool. 3. It is suggested that because phosphate contents in DNA, protein and lipid fractions increased, inorganic phosphates required phosphates required RNA were transferred from phosphates in cytoplasm, because these increased slowly during the culture. 4. Alkali-labile protein were accelerated by IAA and alkali stable protein only were inhibiction were enhanced by IAA while, ethanol : ether soluble fraction was induced by $10^{-7}M$ IAA in comparison with those control.X> IAA in comparison with those control.
Understanding the distribution of heavy metals in sediment is necessary because labile heavy metals can partition into the water column and bioaccumulate in aquatic organisms. Here we investigated six heavy metals (Co, Cu, Mn, Ni, Pb, and Zn) in sediment cores using a five-step sequential leaching method to examine the occurrence of heavy metals in the sediment. The results showed that all elements, except Mn, are depleted in the exchangeable and carbonate fractions. However, heavy metal concentrations are much higher in the Fe-Mn oxide and organic matter fractions, especially for Cu, indicating enrichment in the organic matter fraction. Furthermore, contamination parameters (contamination factor and geoaccumulation index) indicate that Mn contamination is high, primarily derived from anthropogenic sources, presenting a potential risk to ecosystems in the Nakdong River.
Sorption and sequential desorption experiments were conducted for Zn using a natural soil (NS) in background status by aging (1, 30 and 100 days). The sorption isotherm showed that Zn had high sorption capacity but low sorption affinity in NS. Sequential desorption was biphasic with appreciable amount of sorbed Zn residing in the desorption-resistant fraction after several desorption steps. The biphasic desorption behavior of Zn was characterized by a biphasic desorption model that includes a linear term to represent labile or easily-desorbing fraction and a Langmuirian-type term to represent desorption-resistant fraction. The biphasic desorption model indicated that the size of the maximum capacity of desorption-resistant fraction ($q^{irr}_{max}$) increased with aging in NS. Desorption kinetics and desorption-resistance of Zn in the soils collected from mine tailings (MA, MB and MC collected from surface, subsurface soils and mine waste, respectively) were investigated and compared to the bioavailability to earthworm (Eisenia fetida). Desorption kinetic data of Zn were fitted to several desorption kinetic models. The ratio ($q_{e,d}/q_0$) of remaining Zn at desorption equilibrium ($q_{e,d}$) to initial sorbed concentration ($q_0$) was in the range of 0.53~0.90 in the mine tailings which was higher than that in NS, except MA. The sequential desorption from the mine tailings with 0.01M Na$NO_3$ and 0.01M $CaCl_2$ showed that appreciable amounts of Zn are resistant to desorption due to aging or sequestration. The SM&T (Standard Measurements and Testing Programme of European Union) analysis showed that the sum of oxidizable (Step III) and residual (Step IV) fractions of Zn was linearly related with its desorption-resistance ($q^{irr}_{max}$) determined by the sequential desorption with 0.01M Na$NO_3$ ($R^2$= 0.9998) and 0.01M $CaCl_2$ ($R^2$= 0.8580). The earthworm uptake of Zn and the desorbed amount of Zn ($q_{desorbed}$ = $q_0-q_{e,d}$) in MB soil were also linearly related ($R^2$ = 0.899). Our results implicate that the ecological risk assessment of heavy metals would be possible considering the relation between desorption behaviors and bioavailability to earthworm.
Coal combustion fly ash, which has a high available Si content and alkaline pH, was selected as a potential source of soil amendment in this study. Two field experiments were carried out to evaluate the possibility of heavy metal accumulation in silt loam (Pyeongtaeg series) and loamy sand (Nagdong series) of rice (Oryza sativa) paddy soils to which 0, 40, 80, and $120Mg\;ha^{-1}$ of fly ash were added. Rice yields increased significantly with fly ash application and the highest rice yields were achieved following the addition of around $90Mg\;ha^{-1}$ fly ash. Fly ash increased the soil pH but did not increase heavy metal uptakes of rice and heavy metal concentration in soils, due to very low concentration of heavy metals in the selected fly ash. Labile fraction of heavy metals (exchangeable + acidic fraction) was scarcely contained and most of them were stable and unavailable form (oxidizable and residual fraction). In conclusion fly ash could be a good supplement to an inorganic soil amendments without heavy metal contamination in paddy soils.

The environmental changes related to hypoxic water mass were investigated at Gamak bay in summer times, June, July and August 2006. The hypoxic water mass was found, in first, at the northern area of Gamak bay on 27 June. This water mass has been sustained until the end of August and disappear on 13 September. In Gamak bay, the hypoxic water mass was closely related to geography. During the formation of oxygen deficiency, changes in dissolved nutrients was studied and found that on surface layer and lower layer, DIN were 0.80 ${\mu}M$~19.8 ${\mu}M$(6.03 ${\mu}M$) and 1.13 ${\mu}M$~60.83 ${\mu}M$(10.66 ${\mu}M$), and DIP were 0.01 ${\mu}M$~0.92 ${\mu}M$(0.24 ${\mu}M$), and 0.01 ${\mu}M$~3.57 ${\mu}M$(0.49 ${\mu}M$), respectively, far higher distribution on lower layer of the water where hypoxic water mass was occurred. The configuration of phosphorus was analyzed to figure out the possibility of release of phosphorus from sediments. It was found that the Labile-Phosphorus, which is capable of easy move to water layer by following environmental change was found more than 70%. Therefore, in Gamak bay, it was found that the possibility of large amount of release of soluble P into the water, while hypoxic water mass was occurred in deep layer was higher. It is suggested that DIP in the northern sea of Gamak bay mainly sourced from the soluble P from lower layer of the waters where hypoxic water mass was created more than that from basin. However, existence form of phosphorus in sediments during normal times, not during creation of hypoxic water mass, needs further study.
Fluorescence characteristic and amino acids composition of organic matter were determined from extracted seawater samples at eight stations in the East Sea of Korea. Organic compounds have been extracted onto C-18 Sep-Pak cartridges. Three dimensional excitation/emission fluorescence contouring of extracts showed two markedly distinct characterized fluoroscopies representing protein-like biomacromolecule and humic-like geomacromolecule. Protein-like biomacromolecule showing fluorescence maxima at 280 nm/330 nm (excitation/emission) were abundant in the surface mixed layer and then apparently decreased below the thermocline at most stations. It suggests that source of biomacromolecule is comely related with vigorous biological synthetic activity in the surface layer and bacteria decompose its biologically labile components near the thermocline and in the deeper layer. On the other hand, humiliate geomacromolecule showing fluorescence maxima at 330 nm/430 nm (excitation/emission) were low in the surface mixed layer implying photochemical oxidation and then increased below the thermocline at most stations. It suggests that geomacromolecule might be transformed by condensation of bio-refractoryorganic fraction after decomposition of biomacromolecule and particulate organic carbon derived from the surface mixed layer. HPLC measurements of amino acids showed similar composition between seawater and extracted organic macromolecule after hydrolysis. Glycine, serine and alanine were predominant, accounting for more than 50% of total amino acids. Dissolved free amino acids of seawater were more abundant in the surface layer(0.7∼1.8 uM) than the deeper layer (0.2∼0.4 uM). D/L racemic ratio of alanine of extracted organic matter showed lower value in the surface layer than the deeper layer. It suggests that biomacromolecule predominant in the surface layer is relatively young, rapidly recycling and biologically labile.
By the successive enrichment culture, more than 250 methanol-utilizing bacteria were isolated from various samples such as soil, waste water and sewage. Two strains of which were selected and tentatively identified as Acinetobacter sp. and Pseudomonas sp. experiments were carried out to determine the growth conditions for the higher biomass yield and to demonstrate the difference to protein composition dependent upon carbon sources of these two species. the results were as follows ; 1. the optimum pH was determined as 8 in the both species. The optimum temperature in Acinetobacter sp. was $25^{\circ}C{\sim}30^{\circ}C$ and pseudomonas sp. was $30^{\circ}C-35^{\circ}C$. The optimum initial concentration of mthanol was determined as 1-2% in Acinetobacter sp. and 2-3% in pseudomonas sp. 2. The optimum concnetrations of nitrogen source, micro-elements, and vitamins such as biotin and thiamine-HCl in Acnetobactar sp. were 1g $(NH_4)_3SO4,\;1{\sim}3mg\;Mn^{++},\;4mg\;Fe^{++},\;10{\mu}g\;biotin,\;and\;100{\mu}g$ thiamine-HCl per liter medium. In the Pseudomonas sp., 2g $(NH_4)_3SO4,\;1mg\;Mn^{++},\;trace\;amounts\;of\;Fe^{++},\;5{\mu}g\;biotin,\;and\;100{\mu}g$ thiamine HCl per liter were effective. Maximum biomass yield was 2.5g/l in Acinetobacter sp. and 4.8g/l in Pseudomonas sp. 3. Protein composition of the two strains exhibited that alkai-labile protein was higher than alkali-stable protein. In Pseudomonas sp., the contents of acid soluble fraction and alkali-stable protein of the cells grown in the methanol medium were higher than in sucrose medium. On the other hand, in Acinetobacter sp., alkalilabile protein of the cells grown in sucrose medium was higher than in methanol medium.

The purpose of this study was to investigate the release of p-hydroxybenzoic acid and other compounds from cell wall materials(CWM) and their cellulose fraction from carrot with chemical and enzymatic hydrolysis. To investigate this effect on cell wall chemistry of carrot, alcohol insoluble residue(AIR) of CWM were prepared and were extracted sequentially with water, imidazole, CDTA(-1, -2), Na$_2$CO$_3$(-1, -2), KOH(0.5, 1.0 and 4M), to leave a residue. These were analysed for their carbohydrate and phenolic acids composition. Arabinose and galactose were the main noncellulosic sugars. Phenolics esterified to cell walls in carrot were found to consist primarily of p-hydroxybenzoic acid with minor contribution from vanillin, ferulic acid and p-hydroxybenzaldehyde. p-Hydroxybenzoic acid was quite strongly bound to the cell wall. The contents of p-hydroxybenzoic acid in 0.5M KOH, Na$_2$CO$_3$-2, IM KOH, and ${\alpha}$-cellulose were 2,097, 1,360, 1,140, and 717 $\mu\textrm{g}$/g AIR from CWM, respectively. Alkali labile unknown aromatic compound(C$\sub$7/H$\sub$10/O$_2$) was found in ${\alpha}$ -cellulose hydrolyzate digested with driselase and cellulase. This compound was also found in hydrolyzate of 2 M trifluoroacetic acid at 120$^{\circ}C$ for 2 hours. Driselase treatment solubilized only 46.6 $\mu\textrm{g}$/g of the p-hydroxybenzoic acid from carrot AIR. These results indicate that p-hydroxybenzoic acid was associated with neutral polysaccharides, long chain galactose and branched arabinan from graded alcohol precipitation.

Total acid digestion and 1 M HCl extraction methods were used to investigate the pollution status and the degree of ecological risk of trace metals in surface sediments from the Ulsan-Onsan coast. Total concentrations of trace metals (Cu, Cd, Pb, Zn, and Hg) were two-fold higher in surface sediments from Onsan coast than in those from Ulsan coast. The mean labile fractions of the total concentrations of Cd and Pb were 72% and 78%, respectively, indicating a high contribution from anthropogenic sources, whereas Cr, Li, Ni, and As in the residual fraction exceeded 80%, indicating a high contribution from natural sources. According to the results of assessment of trace metal pollution using the sediment quality guidelines in Korea, the concentrations of Cu, Pb, Zn, and Hg were higher than the values of the probable effects level (PEL) at some stations of Onsan coast, and the concentrations of Cr and Ni were lower than the values of the threshold effects level (TEL). The pollution level and ecological risk of the trace metals were analyzed using various indexes (EF, $I_{geo}$, m-PEL-Q, and ERI). Our results showed that the degree of pollution by trace metals (Cu, Cd, Pb, Zn, and Hg) on the Ulsan-Onsan coast was high, and Hg and Cd were the major potential ecological risk factors.
