아가로오스의 효소가수분해산물로 부터 확보할 수 있는 3,6-무수갈락토오스(L-AHG), 네오아가로비오스(NA2), 네오아가로테트라오스(NA4) 및 네오아가로헥사오즈(NA6)의 항염증 활성을 규명하고자, 마우스 대식세포주 RAW264.7 세포를 대장균 유래 지질다당류(LPS)로 자극할 때 세포표면의 TLR4 수용체의 역할을 통해 유도되는 친염증성 M1 분극화 및 이에 따른 친염증성 반응에 미치는 영향을 조사하였다. 그 결과로서, 이들 아가로오스 분해산물들 중에서 단당류인 L-AHG만이 유일하게, LPS 자극에 의해 RAW264.7 세포에서 유도될 수 있는 M1 분극화의 대표적인 마커인 iNOS 효소의 발현과 이에 따른 NO 생성을 농도의존적으로 현저하게 저해하였고 염증 매개체인 프로스타글란딘 E2의 생성을 촉매하는 COX-2의 발현 수준도 LPS 자극후에는 증가하였지만, L-AHG의 존재에 의해서는 그 증가 수치가 다소 저해되는 것으로 나타났다. 이때 RAW264.7 세포에 대한 L-AHG의 세포독성은 확인되지 않았다. 또한 L-AHG는 LPS로 처리된 RAW264.7 세포내에서 유도되는 친염증 신호전달경로에 있어서 초기 신호전달 단계인 TAK1 활성화 단계까지는 별 영향을 나타내지 않았으나 TAK1의 촉매작용에 의한 JNK 및 p38 MAPK의 활성화 인산화(activating phosphorylation)는 현저하게 저해되었다. 특히, 대식세포의 친염증 신호전달경로에서 TAK1 활성화는, 그 하류 단계에서 NF-kB의 활성화가 성공적으로 일어날 수 있도록 해주며, 또한 TAK1에 의한 하류 신호분자인 p38 MAPK 활성화는 NADPH 활성화 및 이에 따른 친염증성 ROS 분자들의 생성을 유발하기 때문에, LPS에 의해 자극된 대식세포내의 친염증 신호전달경로에 있어서 TAK1-JNK/p38 MAPK 경로의 L-AHG에 의한 저해는 대식세포의 M1 분극화 및 친염증 반응을 억제하는 효과적인 기전이 되며, 그 활용성이 크게 기대된다. 아울러 L-AHG가 LPS와 RAW264.7 세포의 표면분자인 TLR4와 상호작용을 방해할 수 있는지에 대해서도 유세포분석기와 형광표지가 된 FITC-LPS를 이용하여 조사한 결과, L-AHG는 대식세포의 표면에서 이루어지는 LPS-TLR4 상호결합은 방해하지 않는 것으로 확인되었다. 이는 L-AHG의 항염증 작용의 표적이 LPS가 TLR4 수용체를 통해 유도하는 세포 내 신호전달경로에 있음을 지지해 준다. 이상의 연구결과들은 아가로오스 유래 희귀 단당류인 3,6-무수갈락토오스(L-AHG)가 LPS 자극에 의해 유발되는 RAW264.7 마우스 대식세포의 M1 분극화 및 염증 반응에 대해, TLR4의 친염증 신호전달경로의 TAK1-JNK/p38 MAPK 단계를 저해하는 항염증 활성을 효과적으로 발휘할 수 있음을 시사한다.
본 연구에서는 들깨 유래 기능성 물질인 rosmarinic acid (RA)와 luteolin이 RAW264.7 세포에서 항염증작용에 대한 상승 효과가 있는지 알아보고자 하였다. 그 결과 RAW264.7 세포에 RA($50{\mu}M$)와 luteolin ($1{\mu}M$)을 동시에 처리하였을 경우 염증 매개인자인 NO, iNOS, $PGE_2$, COX-2의 생성을 RA ($100{\mu}M$)와 luteolin ($2{\mu}M$)을 각각 처리하였을 때 보다 더 뛰어나게 억제하였다. 또한 RA ($50{\mu}M$)와 luteolin ($1{\mu}M$)을 동시에 처리하였을 경우 TNF-${\alpha}$, IL-6, IL-$1{\beta}$ 같은 염증성 사이토카인의 생성량을 RA ($100{\mu}M$)와 luteolin ($2{\mu}M$)을 각각 처리하였을 때 보다 더 뛰어나게 억제하는 것을 확인하였다. 그리고 RA ($50{\mu}M$)와 luteolin ($1{\mu}M$)을 동시에 처리하였을 경우 RA ($100{\mu}M$)와 luteolin ($2{\mu}M$)을 각각 처리하였을 때 보다 NF-${\kappa}B$의 subunit인 p65의 translocation과 $I{\kappa}B$-${\alpha}$의 degradation을 더 뛰어나게 억제하는 것을 볼 수 있어 두 화합물 간의 상승작용이 뚜렷함을 확인 할 수 있었고, RA와 luteolin 두 화합물을 동시에 처리할 경우 염증관련 질환 치료에 유용하게 활용될 수 있을 것으로 판단된다.
인삼은 가공처리방법에 따라 홍삼과 발효홍삼으로 구분된다. 또한, 이들의 가공처리방법에 따라 인삼의 효능을 나타내는 사포닌의 함량이 차이가 있다. 따라서, 본 연구에서 인삼, 홍삼 및 발효홍삼이 대식세포에서 LPS에 의한 염증에 대한 항염효과 및 그 기전을 규명하고자 하였다. 마우스의 대식세포인 RAW264.7 세포에서 LPS에 의해 유도되는 염증관련인자인 NO 및 COX-2의 발현 및 TNF-$\alpha$, INF-$\gamma$ 그리고 NF-${\kappa}B$의 활성을 인삼, 홍삼 그리고 발효홍삼에 의한 항염효과 차이를 비교 하였다. 그 결과, 인삼 및 홍삼 그리고 발효홍삼 모두에서 LPS에 의한 NO의 생성을 억제시키는 것을 확인하였으며, TNF-$\alpha$ 및 INF-$\gamma$의 생성 또한 억제시키는 것을 알 수 있었다. 또한, 인삼 및 홍삼 그리고 발효홍삼 모두 COX-2의 발현 및 LPS에 의한 $I{\kappa}B$의 인산화를 억제시킴으로써 NF-${\kappa}B$의 활성을 억제시키는 것임을 알 수 있었다. 홍삼이 인삼과 발효홍삼에 비하여 NO의 생성을 더 효과적으로 억제시키는 것은 각각의 제조과정에서 나타나는 인삼사포닌의 조성의 차이에 따른 것으로 추정된다. 본 연구는 단순한 동물세포 수준에서의 비교 차이이며 좀 더 정확한 기전의 규명을 위해서는 향후 동물실험을 통한 비교 실험이 수행되어야 할 것이다.
in vivo 및 in vitro에서 마황의 항염증효과를 검토하기 위하여 마황추출물을 급여한 흰쥐에게 LPS shock로 급성기 염증반응을 유발시킨 후, 혈액 및 간장의 전염증성 cytokines들의 농도를 경시적으로 조사하였으며, 한편으로는 Raw 264.7 cell에 LPS shock를 가한 후, 마황추출물이 각종 전염증성 cytokines들의 생산량에 미치는 영향을 조사했다. in vivo 실험에서, 각 처리군 별 plasma IL-$1{\beta}$, IL-6, TNF-$\alpha$ 및 IL-10 농도의 경시적 변동은 전 처리군 모두가 LPS 처리 후, 2h째 급격하게 증가하여, 5h째에 최고치를 나타내었다. Plasma IL-$1{\beta}$, IL-6 및 TNF-$\alpha$의 농도는 LPS처리 후 5h째에서 마황 첨가군 모두가 대조군보다 낮은 값을 나타내었다. Plasma IL-10의 농도는 LPS처리 후, 2h째 및 5h째 모두 에서 마황추출물 첨가군 들이 대조군보다 높은 값을 나타내었다. Raw 264.7 macrophages를 이용한 in vitro 실험에서, IL-$1{\beta}$, IL-6 및 TNF-$\alpha$의 농도들은 대조군보다 마황처리 군들 모두가 낮은 값을 나타내었다. IL-10의 농도는 대조군보다 마황처리군들이 다소 높은 경향을 보였으나, 유의한 차이는 아니었다. 이상의 결과들을 종합해보면 마황에 내재하는 기능성 물질들이 염증반응을 완화하는 효과를 가지고 있음을 시사해 준다.
본 연구에서는 건조 고등어 추출물 및 분획물들에 의한 nitric oxide 생성에 미치는 영향을 살펴보았고 건조 고등어 85% aq. MeOH 분획물을 중심으로 면역과정의 생물학적 작용과 대사적 변화를 유도하는 IL-6, IFN-${\gamma}$ 및 TNF-${\alpha}$ 같은 pro-inflammatory 사이토카인의 생성을 측정하여 건조 고등어 추출물에 의한 항염증 효과에 대하여 검토하였다. 건조 고등어 추출물과 각 분획물은 control보다 낮은 nitric oxide 생성량을 나타내었으며, 특히 85% aq. MeOH 및 n-hexane 분획물에 의한 저해효과가 높았다. 건조 고등어 분획물은 Con-A 보다는 LPS에 의해 자극된 IL-6, TNF-${\alpha}$ 및 IFN-${\gamma}$의 생성을 감소시키는데 더 효과적이었다. IL-6 및 TNF-${\alpha}$의 생성은 배양시간 6시간 후 건조 고등어 85% aq. MeOH 분획물의 모든 첨가농도(1, 3 및 $10{\mu}g$)에서 감소되었다(p<0.05). IFN-${\gamma}$의 경우, 건조 고등어 85% aq. MeOH 분획물을 LPS와 함께 처리했을 때 특히 72시간 배양 시 IFN-${\gamma}$의 생성량이 농도 의존적으로 감소하였고 Con-A에 의해 자극된 IFN-${\gamma}$의 생성량은 6 및 24시간 배양 후 유의적으로 감소하였다(p<0.05). 이상의 결과로부터 건조 고등어 85% aq. MeOH 분획물은 nitric oxide 생성과 pro-inflammatory 사이토카인(IL-6, TNF-${\alpha}$, and IFN-${\gamma}$)을 감소시켜 염증반응을 예방할 것으로 기대된다.
등골나물(Eupatorium japonicum)은 식용으로 이용되어온 식물이지만 생리활성에 대한 연구가 없었다. 따라서 본 연구진은 등골나물 에탄올추출물이 쥐 대식세포인 Raw264.7 세포에 LPS로 유도된 염증 반응에 미치는 영향에 대하여 연구를 수행하였다. 등골나물 꽃 부분에 70% 에탄올을 가하여 얻은 등골나물 에탄올추출물(EJE)을 Raw264.7 세포에 LPS와 함께 0, 1, 2.5, 5, 10 mg/L로 처리하여 세포를 배양하였다. LPS에 의해 생성된 NO 및 $PGE_2$ 분비는 EJE를 처리함에 따라 감소하였고 이 결과는 EJE의 독성에 의한 것이 아님이 증명되었다. Raw264.7 세포에 LPS에 의해 생성된 iNOS, COX-2의 단백질과 mRNA의 발현이 EJE의 농도 의존적으로 억제되었으며, 염증 반응시 생성되는 IL-6, IL-${\beta}$, TNF-${\alpha}$의 mRNA 발현도 등골나물 추출물에 의해 현저히 억제되었다. 더욱이 EJE의 분획물 중 EA와 MC 분획물이 독성이 적으면서 효과적으로 NO의 생성을 억제하는 것으로 나타났다. NO 생성 억제효과가 뛰어난 이들 분획물 내의 생리활성 물질에 대한 연구가 추가적으로 수행되어야 한다고 사료된다. 등골나물 꽃을 추출물로 항염증효과를 연구한 논문이 없고 다른 식용 가능한 천연재료들의 항염증효과와 비교하여(data not shown) EJE 분획물의 항염증효과가 낮은 농도에서 탁월한 점을 미루어 볼 때, 등골나물 추출물은 상당히 낮은 농도에서 뛰어난 항염증 물질을 가진 단일물질을 포함할 가능성이 높다고 사료된다. 따라서 위의 결과는 등골나물 추출물이 독성과 부작용이 적은 염증 치료제로 활용될 수 있는 가능성이 높음을 제시한다.
PURPOSE : Euonymi Lignum Suberalatum (EL) is the stem fin of Euonymi alatus. In traditional Korean medicine, EL is used for treatment of uterine bleeding, metritis and static blood. Recently, many studies have reported several pharmacological effects of EL including anticancer, antimicrobial, antidiabetic activity, and anti-oxidative stress. However, the mechanisms underlying anti-inflammatory effects by the EL is not established. METHODS : To investigate anti-inflammatory effects of Euonymi Lignum Suberalatum Water (ELWE), Raw 264.7 cells were pre-treated with $10-300{\mu}g/mL$ of ELWE, and then exposed to $1{\mu}g/mL$ of LPS. Levels of NO, IL-6, $IL-1{\beta}$ and $TNF-{\alpha}$ were detected by ELISA kit. Expression of pro-inflammatory proteins were determined by immunoblot analysis. To evaluate the anti-inflammatory effect in vivo, rat paw edema volume, and expressions of COX-2 and iNOS proteins in carrageenan (CA)-induced rat paw edema model. RESULTS : NO production activated by LPS, was decreased by $30-300{\mu}g/mL$ of ELWE. Production of inflammatory mediators such as $TNF-{\alpha}$, ILs, $PGE_2$ were decreased by ELWE 100 and $300{\mu}g/mL$. In addition, ELWE reduced LPS-mediated iNOS and COX-2 expression. Moreover, ELWE increased $I-{\kappa}B{\alpha}$ expression in cytoplasm and decreased $NF-{\kappa}B$ expression in nucleus. In vivo study, ELWE reduced the increases of paw swelling, and expression of iNOS and COX-2 proteins in paw edema induced by CA injection. CONCLUSION : The results indicate that ELWE could inhibit the acute inflammatory response, via modulation of $NF-{\kappa}B$ activation. Furthermore, inhibition of rat paw edema induced by CA is considered as clear evidence that ELWE may be a useful source to treat acute inflammation.
Objectives : $18{\beta}$-Glycyrrhetinic acid (18betaGA) is an metabolite of glycyrrhizin in Glycyrrhiza (licorice). The present study investigated anti-inflammatory and anti-apoptosis effect of 18betaGA on the brain tissue of lipopolysaccharide (LPS)-treated C57BL/6 mice. Methods : 18betaGA was administered orally with low (30 mg/kg) and high (100 mg/kg) doses for 3 days prior to LPS (3 mg/kg) injection. Pro-inflammatory cytokines mRNA including tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin (IL)-$1{\beta}$, IL-6, and inflammatory enzyme cyclooxygenase-2 (COX-2) mRNA were measured in the cerebral cortex, hippocampus, and hypothalamus tissue using real-time polymerase chain reaction at 24 h after the LPS injection. Histological changes of Cornu ammonis area 1 (CA1) neurons, Bax, Bcl-2, and caspase-3 expression in the hippocampus was also evaluated by immunohistochemistry and Western blotting method. Results : 18betaGA significantly attenuated the up-regulation of TNF-${\alpha}$, IL-$1{\beta}$, IL-6 mRNA, and COX-2 mRNA expression in the brain tissues induced by the LPS injection. 18betaGA also significantly attenuated the reductions of the thickness of CA1 and the number of CA1 neurons. The up-regulation of Bax protein expression in the hippocampal tissue by the LPS injection was significantly attenuated, while the ratio of Bcl-2/Bax expression was increased by 18betaGA treatment. 18betaGA also significantly attenuated the up-regulation of Bax and caspase-3 expression in the CA1 of the hippocampus. Conclusion : This results indicate that 18betaGA has anti-inflammatory and anti-apoptosis effect under neuroinflammation induced by the LPS injection and suggest that 18betaGA may be a beneficial drug for various brain diseases accompanied with the brain tissue inflammation.
BACKGROUND/OBJECTIVES: Seaweeds have been reported to have various health beneficial effects. In this study, we investigated the potential anti-obesity and anti-inflammatory effects of four types of domestic brown seaweeds in a high-fat diet-induced obese mouse model and bone marrow-derived macrophages (BMDM). MATERIALS/METHODS: Male C57BL/6N mice were fed low-fat diet (LFD), high-fat diet (HFD) or HFD containing Undaria Pinnatifida, HFD containing Laminaria Japonica (LJ), HFD containing Sargassum Fulvellum, or HFD containing Hizikia Fusiforme (HF) for 16 weeks. RESULTS: Brown seaweed supplementation did not affect long-term HFD-associated changes in body weight or adiposity, although mice fed HFD + LJ or HFD + HF gained slightly less body weight compared with those fed HFD at the beginning of feeding. Despite being obese, mice fed HFD + LJ appeared to show improved insulin sensitivity compared to mice fed HFD. Consistently, we observed significantly reduced blood glucose concentrations in mice fed HFD + LJ compared with those of mice fed HFD. Although no significant differences in adipocyte size were detected among the HFD-fed groups, consumption of seaweeds decreased formation of HFD-induced crown-like structures in gonadal adipose tissue as well as plasma inflammatory cytokines. BMDM from mice fed HFDs with seaweeds showed differential regulation of pro-inflammatory cytokines such as IL-$1{\beta}$ and IL-6 compared with BMDM from mice fed HFD by LPS stimulation. CONCLUSION: Although seaweed consumption did not prevent long-term HFD-induced obesity in C57BL/6N mice, it reduced insulin resistance (IR) and circulation of pro-inflammatory cytokines. Therefore, seaweeds may ameliorate systemic inflammation and IR in obesity partially due to inhibition of inflammatory signaling in adipose tissue cells as well as bone marrow-derived immune cells.
The present study was conducted to investigate anti-nociceptive and anti-inflammatory effects of the leaves of Ilex latifolia Thunb (I. latifolia) in in vivo and in vitro. Writhing responses induced by acetic acid and formalin- and thermal stimuli (tail flick and hot plate tests)-induced pain responses for nociception were evaluated in mice. I. latifolia (50 - 200 mg/kg, p.o.) and ibuprofen (100 mg/kg, p.o.), a positive non-steroidal anti-inflammatory drug (NSAID), inhibited the acetic acid-induced writhing response and the second phase response (peripheral inflammatory response) in the formalin test, but did not protect against thermal nociception and the first phase response (central response) in the formalin test. These results show that I. latifolia has a significant anti-nociceptive effect that appears to be peripheral, but not central. Additionally, I. latifolia (50 and $100{\mu}g/mL$) and 3,5-di-caffeoyl quinic acid methyl ester ($5{\mu}M$) isolated from I. latifolia as an active compound significantly inhibited LPS-induced NO production and mRNA expression of the pro-inflammatory mediators, iNOS and COX-2, and the pro-inflammatory cytokines, IL-6 and $IL-1{\beta}$, in RAW 264.7 macrophages. These results suggest that I. latifolia can produce antinociceptive effects peripherally, but not centrally, via anti-inflammatory activity and supports a possible use of I. latifolia to treat pain and inflammation.

① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을
원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
