It has been reported that Scolopendra morsitans L.(SML) has beneficial effects on human health and diverse diseases. The purpose of this study was to investigate the anti-inflammatory effects of ether extract from Scolopendra morsitans L. on lipopolysaccharide(LPS)-induced inflammatory response. Thus, we examined the inhibitory effect of SML ether fraction on LPS-induced increase of inflammatory mediators(NO, iNOS, COX-2, and $I{\kappa}B{\alpha}$) and pro-inflammatory cytokines(TNF-${\alpha}$) in RAW 264.7 cells. In the present study, SML ether extract itself decreased cell viability in a dose dependent manner(> 100 ${\mu}g/ml$). In addition, LPS increased NO production, iNOS expression and phosphorylation of $I{\kappa}-B{\alpha}$, which were blocked by the treatment of SML ether fraction in a dose dependent manner. Furthermore, the treatment of LPS increased TNF-${\alpha}$ production. However, the pretreatment of SML ether fraction prevented the LPS-induced TNF-${\alpha}$ production in dose dependant manner. Taken together, our results suggest that SML may be a beneficial drug against inflammatory diseases such as sepsis.

Estragole is a naturally occurring phenylpropanoid obtained from essential oils found in a broad diversity of plants. Although the phenylpropanoids show many biological activities, clear regulation of the inflammatory signaling pathways has not yet been determined. Here, we scrutinized the anti-inflammatory effect of estragole. The anti-inflammatory effect of estragole was determined through the inhibitory mechanisms of inducible nitric oxide synthase (iNOS), cyclooxygenase (COX-2), nuclear factor kappa B ($NF-{\kappa}B$), and mitogen-activated protein kinases (MAPK) pathways and the activation of nuclear factor erythroid 2-related factor 2 (Nrf-2)/heme oxygenase (HO)-1 pathways in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Estragole significantly inhibited NO production, iNOS and COX-2 expression as well as LPS-induced $NF-{\kappa}B$ and MAPK activation. Furthermore, estragole suppressed LPS-induced intracellular ROS production but up-regulated the stress response gene HO-1 via the activation of transcription factor Nrf-2. These findings demonstrate that estragole inhibits the LPS-induced expression of inflammatory mediators via the down-regulation of iNOS, COX-2, $NF-{\kappa}B$, and MAPK pathways, as well as the up-regulation of the Nrf-2/HO-1 pathway, indicating that this phenylpropanoid has potential therapeutic and preventive applications in various inflammatory diseases.
In this study, we investigated the anti-inflammatory effects of salidroside (SAL) isolated from the MeOH extract of Acer tegmentosum Maxim heartwood in RAW 264.7 macrophage cells. SAL pretreatment significantly inhibited nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) productions in the lipopolysaccharide (LPS)-induced RAW 264.7 cells. Western blot and RT-PCR analyses revealed that SAL inhibited the LPS-induced expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the protein and mRNA levels in a concentration-dependent manner. In addition, SAL reduced the release and the mRNA expressions of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and interleukin-6 (IL-6). Furthermore, nuclear factorkappa B ($NF{-\kappa}B$) luciferase reporter assay was performed to know the involvement of SAL in the production of pro-inflammatory cytokines, we confirmed that LPS-induced transcription activity of $NF{-\kappa}B$ was inhibited by SAL. Taken together, our data indicate that anti-inflammatory property of salidroside might be the result from the inhibition of iNOS, COX-2, $TNF-{\alpha}$ and IL-6 expressions via the down-regulation of $NF{-\kappa}B$ activity.
Antioxidant properties have been proposed as a mechanism for the putative anti-inflammatory effects of phenolic compounds. To reveal the relationship between antioxidant activity and anti-inflammatory effects of various antioxidants, we measured 1, 1-diphenyl-2-picryhydrazyl(DPPH)-reducing activity and examined the inhibitory effects on LPS-induced inflammation-related gene expression in the BV2 microglial cell line. Lipopolysaccharide(LPS)(0.2 ${\mu}g/ml$) was used with or without antioxidants to treat cells, and the regulation of iNOS and cytokine gene expression was monitored using an RNase protection assay(RPA). Although, all tested antioxidants had similar DPPH-reducing activity and inhibited nitrite production, but the curcuminoid antioxidants(ferulic acid, caffeic acid, and curcumin) inhibited LPS-induced gene expression(iNOS, $TNF-\alpha,\;IL-1{\beta}$, IL-6, and IL-1 Ra) in a concentration-dependent manner. Other tested antioxidants did not exhibit the same effects; N-acetylcysteine(NAC) only began to suppress $IL-1{\beta}$ gene expression just below the concentration at which cytotoxicity occurred. Moreover, the antioxidant potency of curcuminoids appeared to have no correlation with anti-inflammatory potency. Only curcumin could inhibit LPS-induced microglial activation at a micromolar level. These data suggest that curcumin may be a safe antioxidant possessing anti-inflammatory activity.
Objectives: The purpose of this study was to investigate the anti-inflammatory effects of aqueous extract from Cicadidae Periostracum(CP) on the RAW 264.7 cells. Method: We examined the cytokine productions including nitric oxide(NO), interleukin(IL)-1b, IL-6 and tumor necrosis factor-a(TNF-a) in lipopolysaccharide (LPS)-induced RAW 264.7 cells and also inhibitory mechanisms such as mitogen -activated protein kinases(MAPKs) and nuclear factor kappa BNF-kB) using Western blot. Results: CP inhibited LPS-induced production of NO, IL-1b and TNF-a but not of IL-6 in RAW 264.7 cells. CP respectively inhibited the activation of MAPKs such as extracelluar signal-regulated kinase(ERK 1/2), c-Jun $NH_2$-terminal kinase(JNK), p38 and NF-kB in the LPS-stimulated RAW 264.7 cells. Also oral administration of CP inhibited CLP - induced endotoxin shock. Conclusion: our results showed that CP down-regulated LPS-induced NO, IL-1b and TNF-a productions mainly through ERK, JNK, p38 MAPK and NF-kB pathway, which suggest the anti-inflammatory effects of CP.
Objectives : The purpose of this study was to investigate the toll-like receptor (TLR)-4 mediated anti-inflammatory effects of extract from Shigyungbanha-tang (SBT) on the peritoneal macrophage. Methods : To evaluate of TLR-4 mediated inflammatory of SBT. we examined NO and cytokine production in TRL-4 ligand (LPS : lipopolysaccharide) induced macrophages. Furthermore, we examined its molecular mechanism using western blot. Results : Extract from SBT itself does not have any cytotoxic effect in the peritoneal macrophages. Extract from SBT reduced LPS-induced nitric oxide (NO). tumor necrosis factor-alpha ($TNF-{\alpha}$), interleukin (IL)-6 and IL-12 production in peritoneal macrophages. SBT inhibited degradation of inhibitor kappa B-alpha ($I{\kappa}B-{\alpha}$) in the TLR-4 mediated peritoneal macrophages. Conclusions : These results suggest that SBT inhibits NO and cytokines production through inhibiting nuclear factor-kappaB (NF-${\kappa}$B) activation in peritoneal macrophage and that SBT may be beneficial oriental medicine for inflammation.
Background: Lung inflammation occurs in many lung diseases, but has limited effective therapeutics. Ginseng and its derivatives have anti-inflammatory effects, but their unstable physicochemical and metabolic properties hinder their application in the treatment. Panaxadiol (PD) is a stable saponin among ginsenosides. Inhalation administration may solve these issues, and the specific mechanism of action needs to be studied. Methods: A mouse model of lung inflammation induced by lipopolysaccharide (LPS), an in vitro macrophage inflammation model, and a coculture model of epithelial cells and macrophages were used to study the effects and mechanisms of inhalation delivery of PD. Pathology and molecular assessments were used to evaluate efficacy. Transcriptome sequencing was used to screen the mechanism and target. Finally, the efficacy and mechanism were verified in a human BALF cell model. Results: Inhaled PD reduced LPS-induced lung inflammation in mice in a dose-dependent manner, including inflammatory cell infiltration, lung tissue pathology, and inflammatory factor expression. Meanwhile, the dose of inhalation was much lower than that of intragastric administration under the same therapeutic effect, which may be related to its higher bioavailability and superior pharmacokinetic parameters. Using transcriptome analysis and verification by a coculture model of macrophage and epithelial cells, we found that PD may act by inhibiting TNFA/TNFAR and IL7/IL7R signaling to reduce macrophage inflammatory factor-induced epithelial apoptosis and promote proliferation. Conclusion: PD inhalation alleviates lung inflammation and pathology by inhibiting TNFA/TNFAR and IL7/IL7R signaling between macrophages and epithelial cells. PD may be a novel drug for the clinical treatment of lung inflammation.
염증 반응과 산화적 스트레스는 다양한 질환의 발생과 진행에 중요한 역할을 담당한다. 따라서 염증과 산화적 스트레스를 동시에 억제할 수 있는 소재의 발굴은 인체 질환 제어에 매우 유용하게 적용될 수 있다. 본 연구의 목적은 RAW 264.7 대식세포에서 검은콩 품종인 소청자와 소청2호 추출물이 염증성 및 산화적 스트레스에 미치는 영향을 알아보기 위함이다. 본 연구의 결과에 의하면 소청자 및 소청2호 추출물은 LPS에 의한 iNOS와 COX-2의 발현을 억제하여 NO와 $PGE_2$의 생성을 억제하였으며, $TNF-{\alpha}$, $IL-1{\beta}$와 같은 염증성 cytokine의 분비와 발현을 감소시켰다. 또한 소청자 및 소청2호 추출물은 LPS로 자극된 세포 내 ROS 축적을 유의적으로 차단시켰고, Nrf2와 HO-1 발현을 증가시켰다. 또한 LPS에 의해 유도된 MAPKs의 활성화도 소청자 및 소청2호 추출물에 의하여 억제되었다. 결론적으로 소청자 및 소청2호 추출물은 RAW 264.7 대식세포에서 MAPKs 경로를 차단시킴으로써 LPS-유도 염증 및 산화 반응에 대한 보호 역할을 할 수 있으며, 이러한 효과에는 최소한 Nrf2/HO-1 경로 활성화가 관련되어 있었다. 이러한 결과를 고려해 볼 때 소청자 및 소청2호 추출물은 대식세포의 과다 활성화로 인한 염증성 및 산화적 반응 차단에 잠재적인 효과가 있음을 일 수 있었다.
본 연구의 목적은 LPS로 자극한 RAW264.7 세포를 이용하여 크랜베리에서 분리한 폴리페놀 분획물의 항산화능을 분석하는 것이었다. 크랜베리 분말의 에틸 아세테이트 분획(EF)과 메탄올 분획(MF)은 C18-Sep-Pak 카트리지를 사용하여 분리했다. 20시간 동안 LPS로 자극한 RAW264.7 세포는 활성산소종(ROS)과 DNA 손상이 유의하게 증가하였으며, EF, MF 및 TF(EF+MF)로 전처리하고 LPS로 자극한 RAW264.7 세포에서는 ROS와 DNA 손상이 유의적으로 감소했다. 그러나 superoxide dismutase의 활성에는 차이가 없었다.
Objective : This study was performed to evaluate the effect of Forsythiae Fructus (FF) ethanol extract on inflammatory cytokine production and its underlying mechanisms in mouse macrophages. Methods : Peritoneal macrophages from thioglycollate medium-injected mice were cultured and stimulated with lipopolysaccharide(LPS) or LPS/interferon(IFN)-${\gamma}$ for cytokine measurement and cellular signaling molecule analysis. Results : FF ethanol extract decreased the levels of secreted tumor necrosis factor(TNF)-${\alpha}$ and interleukin(IL)-6 in IFN-${\gamma}$/LPS-stimulated cells in a concentration-dependent manner. FF extract reduced IFN-${\gamma}$/LPS-induced STAT1 phosphorylation and LPS-induced p38 and JNK activation, but not ERK1/2 activity. The extract also inhibited LPS-induced $I{\kappa}B{\alpha}$ degradation through suppression of $I{\kappa}B{\alpha}$ kinase. Conclusions : These results suggest that FF ethanol extract affects the production of TNF-${\alpha}$ and IL-6 through inhibition of activation of STAT-1, $I{\kappa}B{\alpha}$, p38, and JNK.
