Gallic acid가 Lipopolysaccharide로 활성화된 마우스 대식세포의 케모카인과 성장인자 생성에 미치는 영향 (Inhibitory Effect of Gallic acid on Production of Chemokine and Growth Factor in Mouse Macrophage Stimulated by Lipopolysaccharide)

  • Chemokine and Growth Factor are major mediumtors of immuno-inflammatory pathway. The purpose of this study is to investigate whether productions of Chemokine and Growth Factor in lipopolysaccharide (LPS)-induced mouse macrophage RAW 264.7 cells are modulated by Gallic acid (GA), which is easily founded in tannin-containing natural materials such as red wine, green tea, grape juice, and Corni Fructus. Productions of Chemokine and Growth Factor were analyzed by High-throughput Multiplex Bead based Assay with Bio-plex Suspension Array System based on $xMAP^{(R)}$ (multi-analyte profiling beads) technology. At first, cell culture supernatant was obtained after treatment with LPS and GA for 24 hour. Then, the antibody-conjugated beads were added and incubated for 30 minutes. After incubation, detection antibody was added and incubated for 30 minutes. And Strepavidin-conjugated Phycoerythrin (SAPE) was added. After incubation for 30 minutes, the level of SAPE fluorescence was analyzed on Bio-plex Suspension Array System. Based on fluorescence intensity, concentrations of Chemokine and Growth Factor were determined. The results of the experiment are as follows. GA significantly inhibited the production of interferon-inducible protein (IP)-10, keratinocyte-derived chemokine(KC), and vascular endothelial growth factor (VEGF) in LPS-induced RAW 264.7 cells at the concentration of 25, 50, 100, 200 uM (p<0.05). GA significantly inhibited the production of monocyte chemoattractant protein-1(MCP-1) and macrophage-colony stimulating factor(M-CSF) in LPS-induced RAW 264.7 cells at the concentration of 50, 100, 200 uM (p<0.05). GA diminished the production of granulocyte macrophage-colony stimulating factor (GM-CSF) in LPS-induced RAW 264.7 cells. But GA did not show the inhibitory effect on the production of leukemia inhibitory factor (LIP) and macrophage inflammatory protein (MIP)-2 in LPS-induced RAW 264.7 cells. These results suggest that GA has the immuno-modulating activity related with its inhibitory effects on the production of IP-10, KC, MCP-1, VEGF, and M-CSF in LPS-induced macrophages.

Rutin alleviated lipopolysaccharide-induced damage in goat rumen epithelial cells

  • Objective: Rutin, also called vitamin P, is a flavonoids from plants. Previous studies have indicated that rutin can alleviate the injury of tissues and cells by inhibiting oxidative stress and ameliorating inflammation. There is no report on the protective effects of rutin on goat rumen epithelial cells (GRECs) at present. Hence, we investigated whether rutin can alleviate lipopolysaccharide (LPS)-induced damage in GRECs. Methods: GRECs were cultured in basal medium or basal medium containing 1 ㎍/mL LPS, or 1 ㎍/mL LPS and 20 ㎍/mL rutin. Six replicates were performed for each group. After 3-h culture, the GRECs were harvested to detect the relevant parameters. Results: Rutin significantly enhanced the cell activity (p<0.05) and transepithelial electrical resistance (TEER) (p<0.01) and significantly reduced the apoptosis rate (p<0.05) of LPS-induced GRECs. Rutin significantly increased superoxide dismutase, glutathione peroxidase, and catalase activity (p<0.01) and significantly decreased lactate dehydrogenase activity and reactive oxygen species and malondialdehyde (MDA) levels in LPS-induced GRECs (p<0.01). The mRNA and protein levels of interleukin 6 (IL-6), IL-1β, and C-X-C motif chemokine ligand 8 (CXCL8) and the mRNA level of tumor necrosis factor-α (TNF-α) and chemokine C-C motif ligand 5 (CCL5) were significantly increased in LPS-induced GRECs (p<0.05 or p<0.01), while rutin supplementation significantly decreased the mRNA and protein levels of IL-6, TNF-α, and CXCL8 in LPS-induced GRECs (p<0.05 or p<0.01). The mRNA level of toll-like receptor 2 (TLR2), and the mRNA and protein levels of TLR4 and nuclear factor κB (NF-κB) was significantly improved in LPS-induced GRECs (p<0.05 or p<0.01), whereas rutin supplementation could significantly reduce the mRNA and protein levels of TLR4 (p<0.05 or p<0.01). 구판의 LPS로 유도된 염증 반응 억제 효과 (Effects of Chinemys reevesii on Lipopolysaccaride-indused Inflammatory Reactions)

  • This study is to investigate the effects of Chinemys reevesii (CR) on allergic inflammation mechanism related chronic dermatitis. To investigate the effects of CR, we study inhibitory effect of CR on the levels of pro-inflammatory cytokines released from RAW 264.7 cell stimulated with lipopolysaccaride (LPS), and EoL-1, THP-1, Jutkat cell stimulated with Dermatophagoides pteronyssinus (DP), and LPS induced acute inflammatory BALB/c mouse model. CR reduced the levels of IL-$1{\beta}$ released from RAW 264.7 cell stimulated with LPS at 20 ug/ml, 10 ug/ml concentration. CR significantly reduced the levels of MCP-1 released from EoL-1 cell, IL-6 from THP-1 cell, and IL-4, IL-5, TNF-${\alpha}$ from Jutkat cell stimulated with DP at all the concentration. CR significantly reduced the levels of TNF-${\alpha}$, IL-6, IL-$1{\beta}$, in LPS induced inflammatory BALB/c mouse model, in a dose-dependent manner. These results suggested that CR has suppressive effects on pro-inflammatory cytokines in various inflammation related cell lines through the regulation of immune system. CR could be a therapeutic agent for treatment of chronic inflammatory dermatitis in the future.

LPS에 의해 유도된 3T3-L1 지방세포의 염증반응에 대한 Triacsin C의 효과 (Effect of Triacsin C on LPS-induced Inflammation in 3T3-L1 Adipocytes)

  • 본 연구에서는 long fatty acyl CoA synthetase의 저해제이며, 죽상동맥경화 억제 및 혈관확장 효과가 있는 것으로 알려진 triacsin C의 LPS로 유도된 3T3-L1 지방세포의 염증반응에 대한 효과를 알아보고자 하였다. 10 ${\mu}M$의 triacsin C를 2시간 동안 처리한 결과, LPS로 유도된 배양액내 IL-6 분비량과 mRNA IL-6 발현이 유의적으로 감소하였고 이는 선행연구를 통해 항염증 효능이 입증된 resveratrol(50 ${\mu}M$, 24시간 처리)에 비해서도 배양액으로의 분비된 IL-6의 억제효과가 더 높게 나타났다. 그러나 triacsin C과 resveratrol의 동반상승 효과는 없는 것으로 나타났다. Triacsin C나 resveratrol은 LPS로 유도된 지방세포의 TLR2, MCP-1, iNOS 유전자 발현에 대한 억제효과는 없는 것으로 나타났다. 그러나 triacsin C 처리 후에 resveratrol을 혼합 처리한 경우 iNOS의 발현은 LPS 대조군에 비해 유의적으로 감소하였다. 향후 triacsin C가 LPS로 유도된 염증반응을 어떤 기전으로 억제하는 지에 대해서는 추후 연구가 필요한 것으로 사료된다. 결론적으로 이 연구에서는 지방산 합성 저해제인 triacsin C는 3T3-L1 지방세포에서 IL-6 특이적 항염증 효과가 있으며, triacsin C와 resveratrol을 동시에 처리했을 때 iNOS의 발현을 유의적으로 감소시킴을 확인하였다. 이는 내독소에 의한 지방세포의 염증반응에 대한 triacsin C의 항염증 효과를 입증하기 위한 기초자료로 활용이 가능할 것으로 기대된다.

생쥐에서 급성 폐렴에 대한 여정자 열매 추출물의 억제효과 (Ligustrum lucidum Fruits Extract Inhibits Acute Pulmonary Inflammation in Mice)

  • Traditionally, Ligustrum lucidum fruits (LL) is one of the well-known oriental herb used in the treatment of skin and lung inflammation. This study investigated anti-inflammatory effects of LL in the pathogenesis of acute pulmonary inflammation in mice. Acute pulmonary inflammation was induced by intratracheal instillation of cigarette smoke condensate (CSC) and lipopolysaccharide (LPS) 5 times within 12 days in mice. LL extract was administered orally at a dose of 50 or 200 mg/kg. Administration of LPS and CSC significantly elevated airway hyperresponsiveness (AHR) to mice, and increased in the levels of inflammatory cells and mediators in mice. However, the LL extract significantly reduced the elevated AHR, and the increase of neutrophils, $CD4^+/CD3^+$ cells and $CD8^+/CD3^+$ cells, along with reducing the expression of TNF-${\alpha}$, IL-6, and MIP-2. Moreover, the LL extract alleviated the infiltration of inflammatory cells in expanded airway walls histologically. These results indicate that the LL extract has an inhibitory effects on acute pulmonary inflammation and AHR in murine model, and plays a crucial role as a immunomodulator which possess anti-inflammatory property.

Uncaria sinensis(OIi.) Havil Suppressendotoxin-induced Proinflammatory Responses through Blocking ERK Signaling Pathways

  • Uncaria sinensis(OIi.) Havil (USH) is used in traditional Korean medicine to treat inflammation such as amebic dysentery. In this study, we investigated the anti-inflammatory effect of USH. The water extract of USH significantly inhibits lipopolysaccharide (LPS)-induced nitrite oxide (NO), tumor necrosis factor $(TNF)-{\alpha}$, interleukin (IL)-6and IL-12 productions in murine peritoneal macrophages. Furthermore, USH selectively inhibited activation of the extracellular signal-regulated kinase (ERK) but not of p38 MAPK, c-Jun N-terminal kinase (JNK) and nuclear $factor-{\kappa}B$ $(NF-{\kappa}B)$. In murine model, we found that administration of USH reduced serum levels of $TNF-{\alpha}$, IL-6 and IL-12 productions in LPS-treated mice. Our results suggest that USH exerts ant-inflammatory effects in macrophages via inhibition of ERK activation and may be a useful therapeutic approach to inflammatory diseases.

  • Objectives : Danguieumja is a traditional medicinal prescription to treat skin disease. It was commonly used for the treatment of itching, chronic urticaria and atopic dermatitis in Korea by the addition or omission of several herbs. This study investigated the anti-inflammatory potential of Gagam-Danguieumja (GDE) water extract. Methods : We examined the effects of GDE on the lipopolysaccharide (LPS)-induced production of nitric oxide (NO) in a murine macrophage cell line, RAW 264.7 cells. Results : GDE inhibited production of NO in a dose dependent manner and also decreased the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2). As a possible molecular mechanism of anti-inflammatory effect increased phosphorylation of mitogen-activating protein kinases (MAPK) by LPS were blocked by GDE treatment. Conclusions : These results suggest that GDE has an anti-inflammatory therapeutic potential through the inhibition of MAPK phosphorylation, thereby decreasing the expression of pro-inflammatory genes.

Inhibition of Inducible Nitric Oxide Synthase by Agaricus bisporus Extract in RAW 264.7 Macrophages

  • Agaricus bisporus, also known as white button mushroom, is one of the most popular mushrooms consumed in Korea. This mushroom contains high concentrations of flavanoids and exhibits antioxidant activity. In this study, we examined the effects of Agaricus bisporus ethanol extract (ABE) on lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 cells. Nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) protein levels were assessed in cells treated with $100\;{\mu}M$ LPS in the presence or absence of ABE. 0.5 mg/mL of ABE suppressed NO production significantly. Moreover, ABE inhibited levels of iNOS protein. Taken together, these results suggest that ABE exerts anti-inflammatory activity in LPS-induced inflammation in RAW 264.7 cells.

뽕잎 n-부탄올 추출물의 치주인대세포에서 LPS로 유도된 염증성 사이토카인의 억제와 골 형성 분화 조절 (n-Butanol Extract of Mulberry Leaves Suppresses LPS-induced Inflammatory Cytokines and Modulates Osteogenic Differentiation in Periodontal Ligament Cells)

  • Periodontitis is a chronic inflammatory disease that is known to have the characteristics of destructed periodontal tissue. Anti-oxidant and anti-inflammatory effects of mulberry leaves in periodontal tissue is not well known until now. We investigated the effects of n-butanol extract of mulberry leaves on the lipopolysaccharide (LPS)-induced proinflammatory cytokines, such as $IL-1{\beta}$, IL-6, IL-8 and modulates osteogenic differentiation in periodontal ligament cells. The expression levels of Runx2, ALP and mRNA were increased by n-butanol extract of mulberry leaves at the concentration of $100{\mu}g/ml$ in periodontal ligament cells. n-Butanol extract of mulberry leaves extract reduced the range of pathophysiological processes, such as inflammation and increase in the level of osteogenic-related genes. These findings suggest that n-butanol extract of mulberry leaves has therapeutic effects on periodontitis and periodontal tissue regeneartion.

Anti-inflammatory Effects of Scrophularia Koraiensis Nakai via NF-κB and MAPK Signaling Pathways in LPS-induced Macrophages

  • Scrophularia koraiensis Nakai is widely used to remedy fever, edema, and neuritis. S. koraiensis has harpagoside and angoroside C, these compounds have been reported to alleviate inflammation, rheumatic diseases, and analgesic stimulation. We evaluated the anti-inflammatory effects of the ethanol extract of S. koraiensis (SKE) in lipopolysaccharides (LPS)-induced macrophages. At cellular levels, SKE decreased the production of nitric oxide (NO), the expression of inducible nitric oxide synthase (iNOS), and cytokines (IL-1b, TNF-a, and IL-6) under the LPS stimulation. SKE inhibited the phosphorylation of nuclear transcription factor-kappa B (NF-κB) p65 and its inhibitor (IκB-α). In addition, SKE suppressed the phosphorylation of extracellular signal-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 in the mitogen-activated protein kinase (MAPK) pathway. In conclusion, SKE could be considered a potential resource for attenuating inflammation response and it may be utilized in the material for cosmetics, food additives, and tea.
