The Morphological Characteristics of Entomopathogenic Fungus, Isaria sinclairii (Berk.) Lloyd (곤충기생균 Isaria sinclairii(Berk.) Lloyd의 형태학적 특성)

  • NIAST has developed the artificial cultivation technique of entomopathogenic fungus, Isaria sinclairii by using the silkworm. NIAST is planning to provide the silkworm farmers the guidance of the technique. Screening of pharmacologically active principles of I. sinclairii is needed to register the fungal breeding, attain the approval of food materials, and ultimately position the fungus as a high value-added product. The result of investigating the characteristics of Isaria sinclairii was showed that conidia were irregularly long oval-shaped and measured 6.33$\times$3.33 $\mu\textrm{m}$ in average The optimal condition of cultivating hyphae was pH 5.0 in acidity and 21- $25^{\circ}C$ in temperature. And the best condition of producing conidia was pH 6.0 in acidity and 16-$20^{\circ}C$ in temperature.

Spatial Distribution Patterns and Population Structure of Doellingeria scabra at Mt. Maebong in Korea (한국 매봉산 참취의 공간적 분포 양상과 집단 구조)

  • Doellingeria scabra Thunb. (syn. Aster scaber Thunb.), a perennial herb in the family Asteraceae, is frequently found in the wild mountain regions of Korea. This aim of this work was to measure the characteristics of patchiness of D. scabra in a local population on Mt. Maebong in Taeback-ci, Gangwon-do. The spatial distribution pattern of this species was estimated by analyzing ecological data by methods including the index of dispersion, Lloyd's mean crowding, and Morisita's index. The mean population density of the D. scabra population was 2.94. The D. scabra individuals were uniformly or randomly distributed in small-scale plots and were aggregately distributed in two large-scale plots ($16{\times}32m^2$ and $32{\times}32m^2$). The mean crowding ($M^*$) was 0.916. The mean patchiness index (PAI) was 0.796. Morisita's coefficient tended to decrease the density of the population as the plot size increased. The expected value of Eberhardt's index ($I_E$) in the local population was 2.623. Moran's I of D. scabra significantly differed from the expected value in 6 of 8 cases (75.0%). The first five classes were positive, with four showing statistical significance, indicating similarity among individuals in the first four distance classes (I-IV, 8 m), The results presented here could provide a theoretical basis for the conservation of D. scabra (Korean: chamchwi) and for the rehabilitation and sustainable management of forest ecosystems on Mt. Maebong, as well as on other mountains.

Anticancer Effects of Leek Kimchi on Human Cancer Cells

  • The anticancer effects of leek (buchu in Korean) kimchi were evaluated in the human cancer cells: AGS gastric adenocarcinoma cells, HT-29 human colon adenocarcinoma cells and HL-60 leukemia cells. The leek kimchi (fermented for 6 days at 15$^{\circ}C$) was fractionated into 7 groups: methanol extract, hexane extract, methanol soluble extract MSE), dichloromethane (DCM) fraction (fr.), ethyl acetate fr., butanol fr. and aqueous fr. Most of the leek kimchi tractions inhibited the growth of AGS and HT-29 cancer cells in a dose dependent manner. In particular, the DCM fr. showed the highest inhibitory effect among the tractions. Treatment with the DCM fr. (0.1 mg/mL) reduced the survival rates of AGS and HT-29 cancer cells to 19% and 37% of the controls, respectively. Moreover the DCM fr. of the leek kimchi arrested G2/M phase in the cell cycle and induced apoptosis in HL-60 human promyelocytic leukemia cells. These results indicate that the leek kimchi exerted an anticancer effect on those human cancer cells, and that the DCM fr. arrested G2/M phase in the cell cycle and induced apoptosis in the leukemia cells.

An Analysis of Case on Frustration under Time Charter in the Sea Angel (정기용선계약상 이행불능에 관한 Sea Angel호 사건의 판례 분석)

  • A party to the charter will not be able to rely upon the doctrine of frustration if an event which makes further performance impossible has been caused by his breach of the charter. Strictly speaking, this is not a situation of frustration at all but rather a case of discharge of the contract by breach. In the Sea Angel case, the defendant entered into a Lloyd's Standard Form of Salvage Agreement with owners of the casualty on the LOF 2000 form, incorporating the SCOPIC clause. The time charter was on the terms of the Shelltime 4 form. This case the trial of the action brought by the claimants owners of the vessel Sea Angel claiming outstanding hire from the defendant charterers. This Case was issued whether charterparty frustrated by refusal of port authorities to issue "No Demand Certificate" allowing port clearance pending payment of oil spillage clean-up costs. The court concluded that no attempt had yet been made to invoke the assistance of the Pakistani court to obtain the release of the vessel. There was not so radical or fundamental a change in the obligation assumed by defendant as to establish frustration. The purpose of this study aims to analyse frustration and time charter in the Sea Angel case.

  • The purpose of this study is fatigue damage assessment for large sized casting parts (hub and mainframe) of the 3MW offshore wind turbine by computer simulation. Hub and mainframe durability assessment is necessary because wind turbine have to guarantee for 20 years. Fatigue life evaluation must be considered all of fatigue load conditions as the components are wind load transmission path. Palmgren-Miner linear damage accumulation hypothesis is applied for fatigue life estimation with stress-life approach. S-N curve for the spheroid graphite cast iron EN-GJS-400-18-LT is derived according to durability guidelines. Reduction factors were applied for survival probability, surface roughness, material quality and partial safety factor according to Germanischer Lloyd rules. To calculate fatigue damage, stress tensors, extracted from the unity load calculation from ANSYS is multiplied with time track of fatigue loads extracted from GH bladed. Damage accumulation is performed with all of fatigue load conditions at each finite element nodes. In this study maximum nodal damage value is under 1.0. casted parts are safe. Assessment of Ruminal and Post Ruminal Amino Acid Digestibility of Chinese and Canadian Rapeseed (Canola) Meals

  • Two rapeseed meal samples (Sample A, hybrid 5900 and sample B, double low rapeseed No.4) obtained from China and one Canola meal sample obtained from a local crushing plant in Canada were used to investigate the amino acid degradability of rapeseed/Canola meal in rumen and amino acid digestibility of ruminal incubation residues by precision-fed rooster bioassay. Results show that in ruminal incubation the degradation rate of non amino acid nitrogen in crude protein is higher than that for amino acid nitrogen in crude protein, the results also suggest that the degradation rate of amino acid nitrogen in Chinese rapeseed meal sample B was lower than that for Canadian Canola, but that in Chinese rapeseed meal sample A is much close to that for Canadian canola meal. For all amino acids the digestibility of the bypass or residual protein as measured by the precision-fed rooster bioassay tended to be lower for Chinese rapeseed meal sample A than for sample B or Canadian canola meal which had similar digestibility values. However following a calculation of total amino acid availability, involving the digestibility of amino acids in the rumen and rooster bioassay the results are less contradictory. Results indicated that in traditional roasting-expelling process, heat treatment, especially dry heat treatmeat could decrease amino acids degradability in rumen of rapeseed/canola meal, but also may decrease total availability of amino acids of rapeseed/canola meal.

Structural Design and Cost Evaluation of Double Hull Bulk Carrier (이중선체 벌크화물선의 선체구조설계 및 경제성 검토)

  • After many casualties of conventional bulk carriers in recent years, a double hull bulk carrier was proposed to enhance the structural safety of a side shell and a transverse bulkhead. In this paper, two alternative structural designs of a double hull bulk carrier were carried out based on the Lloyd's rule. One has the double sided hull with longitudinal stiffeners and the other has that with a girder. The final structural design was examined in comparison with an existing single hull bulk carrier from the viewpoints of cargo hold capacity and the increases of weight and construction cost. Generally, the construction cost of a ship consists of the costs of material, labor and overhead cost. But, in this study, the relative construction cost concept was introduced to compare the economical validity more precisely. In this concept, fixed overhead cost is excluded in the assessment of construction cost, and only the variable overhead cost is added up to labor cost. As the result of this study, a double hull bulk carrier can be constructed within 1% increase of weight and construction cost.

Fatigue Life Prediction of Crank-type Rotavator

  • Purpose: This study was performed to predict the fatigue life of a crank-type rotavator operated in domestic soil conditions using Recurdyn$^{(R)}$, a dynamic analysis program. Methods: Torque on the PTO shaft was measured using experiments conducted on the uplands and paddy fields in Korea. On the basis of the experimental and analytical results, the fatigue life of the crank-type rotavator was predicted by constructing an S-N curve according to the GL (Germanischer Lloyd Wind Energie GmbH) guideline. Results: The torques experienced by the PTO shaft in the paddy soil and the uplands were in the range of 472~797 N m and 313~430 N m, respectively, for every condition. In case of load condition, the peak torques (846 N m, 770 N m) were applied for severe conditions, resulting in a maximum (von Mises) stress of 75 MPa at the crank arm. The fatigue life of the crank-type rotavator was predicted to be 1,167 h that satisfies the target value of 1,110 h, by substituting the analysis results into an S-N curve of crank arm. Conclusions: The fatigue life of the crank-type rotavator was within the target life for the studied soil conditions; however, further field experiments for various soil conditions would be required to verify the prediction results.

Sequential analysis of cell differentials and $IFN-{\gamma}$ production of splenocytes from mice infected with Toxopluma gondii

  • To assess the relationship between the changes of cellular components and the production of Th 1 cytokine in the immune tissue, inbred C57BL/6 mice were orally infected with 40 cysts of 76K strain of Toxoplosma gondii. The sequential change of cell differentials and $IFN-{\gamma}$ production of splenocytes were analyzed by Diff-Quik stain and RT-PCR. There were no significant proportional changes of cellular components of splenocytes until day 4 postinfection (Pl) as compared to those of day 0, and the relative percentage of macrophages and neutrophils/eosinophils increased significantly (p<0.01) thereafter. The expression of $IFN-{\gamma}$ mRNA of $CD3^{-}$ cells was observed from day 1 Pl at a low level. However, $IFN-{\gamma}$ production of $CD3^{+}$ cells increased significantly from day 4 Pl (p<0.01) which progressively increased thereafter. These findings provide the relative percentages of granulocytes and macrophages were increased in conjunction with increase of total number of splenocytes after oral infection with T. gondii in the susceptible murine hosts, and lymphocytes were the major cellular components and the important source of $IFN-{\gamma}$.

Complex sternal and rib reconstruction with allogeneic material

  • Sternal malunion, or loss, developed after a median sternotomy cannot only be difficult to manage and treat, but also may diminish one's quality-of-life drastically. The technique presented here represents a multispecialty approach in one stage for the reconstruction of an unstable thoracic cage. The procedure utilized a donated sternum and ribs. The sternum with ribs harvested from a single donor included adipose derived stromal vascular fraction (ADSVF) cells with marrow also from the same donor. Autologous muscle flaps, stabilized with acellular dermal matrix were utilized to provide a robust blood supply to the ADSVF cells and bone grafts. Acellular dermal matrix was used to construct the ribs and stabilize the plugs of stem cells and bone. These procedures, in the hands of multispecialty physicians, have led to several successful reconstructions involving complex chest wall deformities. This surgical intervention was performed in a one stage operation. This represents the first successful complete sternal transplant in a patient with return to normal activities and increased quality-of-life.
