두툽상어 matrix metalloproteinase 유전자 cDNA의 클로닝 (Cloning of a matrix metalloproteinase cDNA from Scylliorhinus torazame)

  • Matrix metalloproteinases(MMP)는 배발생 및 재조직화 등의 정상적인 생체형성과 관절염, 암전이, 치근막염, 골조송증 등의 질병과정에서 collagen이나 proteoglycan과 같은 세포외기질의 구성성분을 분해하는 아연(zinc)효소군이다. 지금까지 다양한 종에서 mmp의 유전자가 클로닝되고 그 기능이 연구되어 왔지만 아직 어류에서는 연구결과가 보고된 바가 없다. 본 연구에서는 한국의 부산연안에 많은 연골어유 투툽상어(Scylliorhinus toraxzame)로부터 RT-PCR(reverse transcriptase dependent polymerase chain reaction)의방법으로 mmp cDNA의 일부를 클로닝하였다. 이것은 염기서열에서 인간, 쥐 및 닭의 membrane type matrix matalloproteinase-3(mt3-mmps)의 염기서열과 74% 동일성을 보이며, 아미노산서열에서는 90%이상의 동일성을 갖고 있다. 또한 MMP에 나타나는 cysteine switch domain, zinc binding domain(HExGH motif), propeptide cleavage site, and RRKR motif등을 가지고 있다. 이러한 결과로부터 본 연구에서 클로닝된 RT-PCR단편은 두툽상어의 mt3-mmp 또는 이와 유사한 유전자의 cDNA이라 믿어진다.

FPSO 다점 계류 시스템의 최적 설계 연구 (Study on Optimum Design of FPSO Spread Mooring System)

  • For a spread-moored FPSO (Floating Production, Storage, & Off-loading) subjected to environmental excitation from waves, current, and wind, a procedure to determine optimum length and stiffness of mooring lines is suggested using quasi-static frequency domain response analyses. Coupled relations between design parameters are closely examined. In consideration of this, optimized design parameters are proposed based on minimum weight condition. The initial design parameters for numerical analyses are calculated using the static catenary equation of mooring lines. It is demonstrated the line tension and vessel's offset are influenced by the mooring line length and stiffness. Accordingly it is suggested the optimum vessel's offset range should be determined considering line fatigue damage. The availability and limitation of the application of quasi-static analysis method for spread mooring system are explained by comparing the result of time domain analysis with one of frequency domain analysis.

  • 저가형 GNSS (global navigation satellite system) 수신 모듈에 DGNSS (differential GNSS) 서비스를 적용하기 위한 방안으로 거리 영역의 보정정보를 위치 영역으로 투영한 후, stand-alone으로 산출한 위치에 적용하는 DGNSS-CP 방식이 제안된 바 있다. DGNSS-CP 를 상용 수신기 또는 휴대폰에 적용하기 위해서는 항법 방정식의 관측행렬을 이용하여 위치영역 투영 방정식을 구성하므로, 각 위성의 시선벡터를 산출하여야 한다. GNSS 항법 메시지, 배치 정보 등이 시선벡터 산출을 위하여 사용되는데, 각 방법에 따라 정확도와 연산량 등의 성능에 차이가 발생한다. 본 연구에서는 제시된 두 가지 시선벡터 산출 방식에 따라 DGNSS-CP의 성능에 어떠한 영향을 끼치는지 확인하기 위하여, Septentrio PolaRx4 Pro 수신기에서 stand-alone mode 로 저장된 데이터에 해당 알고리즘을 적용하였고, 배치 정보를 사용하는 방법이 궤도정보를 사용하는 방법에 비해 정확도 면에서는 그 성능이 RMS (root mean square) 0.1 m 가량 저하되는 반면, 연산량은 약 1/15수준으로 줄일 수 있음을 확인하였다.

시간영역 변형형상을 이용한 철도교량의 손상평가 (Damage Evaluation of a Railroad Bridge Using Time-domain Deflection Shape)

  • 공용기간동안 철도교량의 안전성 및 사용성을 확보하기 위해서는 지속적인 감시를 통하여 교량의 구조적 성능을 유지하는 것이 필요하다. 구조물의 구조적 건전성을 감시하기 위하여 현재까지 개발된 대부분의 방법들은 모달 응답을 이용하고 있으나, 이러한 모달응답은 별도의 추출 과정이 필요하며 실제 구조물에서 얻을 수 있는 수가 제한된 다는 단점이 있다. 이 논문에서는 열차이동하중으로 인한 시간영역의 변형형상을 이용하여 자유진동응답에 기반한 손상평가방법의 적용성을 검토하였다. 검토된 방법은 이동하중으로 인한 시간영역의 변형응답을 이용하므로 별도의 모달 응답 추출과정이 필요 없어 실제 구조물에 적용이 용이하다. 제시된 방법의 적용성은 단순판형교 수치예제를 이용하여 검증하였다.

뇌 기반 진화적 접근법을 적용한 초등학교 학생의 과학 자유탐구에서 인지적 영역 분석 (Analyses on Elementary Students' Cognitive Domain in Free Science Inquiry Activities Applying a Brain-Based Evolutionary Approach)

  • In National Curriculum of Science revised in 2007, the Free Inquiry was newly introduced to increase students' interest in science and to foster creativity by having students make their own question and find answer by themselves. The purpose of the study was to analyze characteristics, in cognitive domain, appeared in the processes of performing the Free Inquiry activities applying a brain-based evolutionary science teaching and learning principles. For this study, 106 fifth grade students participated, and they performed individually Free Inquiry activities. In order to characterize of the diversifying, estimating-evaluating-executing, and extending-applying activities in cognitive domain (C-DEF), the Free Inquiry diary constructed by the students, observations by a researcher, and interviews with the students were analyzed both quantitatively and qualitatively. The major results of this study were as follows: First, at C-D step, many students (71.5%) had difficulty in searching the meanings of their results and the contents of interpretations were at the level of simple description of their results. A few students (15.2%) derived interpretations based on causal relationships between specific variable and result. Also, the tendency that the numbers of interpretation about meaning of their results were increased as the scores of science attitude and achievement was appeared. Second, at C-E step, the students showed tendency of considering facts exactly explaining inquiry topic and being appliable to daily life rather than objectivity or accuracy of scientific knowledge. Third, at C-F step, there were three types of extension and application: simple repetition (8.2%), extension (64.0%), and upward application (17.6%) types. Based on these findings, implications for supporting appropriate interpretation, evaluation, and application of inquiry results are discussed.

Identification of another calmodulin-binding domain at the C-terminal region of AtCBP63

  • Calcium signals can be transduced by binding calmodulin (CaM), a $Ca^{2+}$ sensor in eukaryotes, is known to be involved in the regulation of diverse cellular functions. We isolated a CaM-binding protein 63 kD (AtCBP63) from the pathogen-treated Arabidopsis cDNA expression library. Recently, AtCBP63 was identified as a CaM bining protein. The CaM binding domain of AtCBP63 was reported to be located in its N-terminal region, In this study, however, we showed that ACaM2 could specifically bind to second CaM-binding domain (CaMBD) of AtCBP63 at the C-terminal region. The specific binding of CaM to CaM binding domain was confirmed by a gel mobility shift assay, a split ubiquitin assay, site-directed mutagenesis, and a competition assay using a $Ca^{2+}$/CaM-dependent enzyme. The gene expression of AtCBP63 was induced by pathogens and pathogens related second messengers. This result suggests that a CaM binding protein, AtCBP63, may play role in pathogen defense signaling pathway.

Co-expression of a novel ankyrin-containing protein, rSIAP, can modulate gating kinetics of large-conductance calcium-activated potassium channel from rat brain.

  • We isolated a novel ankyrin-repeat containing protein, rSIAP (rSlo Interacting Ankyrin-repeat Protein), as an interacting protein to the cytosolic domain of the alpha-subunit of rat large-conductance Ca$\^$2+/-activated K$\^$+/ channel (rSlo) by yeast two-hybrid screening. Affinity pull-down assay showed the direct and specific interaction between rSIAP and rSlo domain. The channel-binding proteins can be classified into several categories according to their functional effects on the channel proteins, i.e. signaling adaptors, scaffolding net, molecular tuners, molecular chaperones, etc. To obtain initial clues on its functional roles, we investigated the cellular localization of rSIAP using immunofluorescent staining. The results showed the possible co-localization of rSlo and rSIAP protein near the plasma membrane, when co-expressed in CHO cells. We then investigated the functional effects of rSIAP on the rSlo channel using electrophysiological means. The co-expression of rSIAP accelerated the activation of rSlo channel. These effects were initiated at the micromolar [Ca$\^$2+/]$\_$i/ and gradually increased as [Ca$\^$2+/]$\_$i/ raised. Interestingly, rSIAP decreased the inactivation kinetics of rSlo channel at micromolar [Ca$\^$2+/]$\_$i/, while the rate was accelerated at sub-micromolar [Ca$\^$2+/]$\_$i/. These results suggest that rSIAP may modulate the activity of native BK$\_$Ca/ channel by altering its gating kinetics depending on [Ca$\^$2+/]$\_$i/. To localize critical regions involved in protein-protein interaction between rSlo and rSIAP, a series of sub-domain constructs were generated. We are currently investigating sub-domain interaction using both of yeast two-hybrid method and in vitro binding assay.

이동 애드혹 네트워크에서 효율적인 네임 서비스 제공을 위한 도메인 네임 시스템 (Domain name system for the efficient name service in mobile ad hoc networks)

  • 지금까지의 이동 네트워크 환경에서의 연구는 주로 경로 설정 방법에 대해 이루어겼으나, 실제 사용자에게 서비스를 제공하기 위해서는 DNS(Domain Name System)가 우선적으로 지원되어야 한다 이동 애드혹 네트워크의 고유한 특성으로 인하여 유선 네트워크에서 사용되고 있는 DNS를 그대로 도입하는 것은 문제가 발생한다. 분산 방식 DNS를 사용하는 경우에는 노드의 이동에 대해 효율적으로 대처할 수 있다는 장점을 가지나, 도메인 네임 충돌 문제를 해결하기 위한 오버헤드가 발생하게 되며, 중앙 방식 DNS를 사용하는 경우에는 네임 정보 획득을 위한 통신이 유니캐스트 기반으로 동작하고 네임의 할당 및 관리에 따른 오버헤드 문제를 해결할 수는 있으나 DNS서버의 이동에 따른 DNS네임 서비스의 연속성 있는 지원 문제가 해결되어야 한다. 따라서 본 논문에서는 이동 애드혹 네트워크 환경에서 중앙 방식 DNS를 기반으로 연속성 있는 서비스를 지원하면서도 네임 할당 및 관리에 있어서 효율적으로 동작하는 Manet DNS를 제안하며 서버의 이동성으로 인해 발생할 수 있는 네트워크 병합 및 분할 문제에 대한 해결책을 제시한다.

초등과학 실험 수업에서 탐구 요구 수준에 따른 초등학교 학생의 정의적 영역 학습의 특성 (Elementary School Students' Science Learning Characteristics in the Affective Domain and It's Relationship to the Levels of Inquiry Requirement in Science Experiment Instruction)

  • In this study, elementary school students' science learning characteristics together with the levels of inquiry requirement in elementary school science experiment instruction were investigated in terms of the affective domain. We divided seventy-three students of the fifth ade into two groups according to the levels of inquiry requirement. After each instruction had been implemented, the characteristics of the students' learning in terms of the affective domain were compared and analyzed within the context of the levels of inquiry requirement. There were no significant differences between Level 1(LL) and Level 2(HL) groups in terms of the affective characteristics. These results may be caused by the burden imposed by the difficulty in thinking through and setting up the experimental methods. However, the HL group students' levels of interest in science experiments was higher than that of the LL group. In conclusion, the HL instruction contributed little to the improvement of affective characteristics. Some possible reasons might be that students felt difficulties with the somewhat unfamiliar instructions which required them to design and perform experiments. Higher levels of interest in science experiments amongst HL students' implies that well-organized and continuous exercises involving the design and performance of experiments could improve students' scientific affective characteristics.

The purpose of the study was to analyze characteristics deployed in the processes of elementary school students' free inquiry activities applying a brain-based evolutionary science teaching and learning principles. For this study, 106 the fifth grade students participated, and they performed individually free inquiry activities according to a brain-based evolutionary approach. In order to characterize the diversifying, estimating-evaluating-executing, and extending-applying activities in behavioral domain, the free inquiry diary constructed by the students, observations by the researcher, and interviews with the students were analyzed both quantitatively and qualitatively. The major results of this study were as follows: First, the students preferred basic inquiry process skills and the majority of the students selected observation as a major approach of their inquiry. The reason was found to be that they were accustomed to only typical basic inquiry skills which is frequently presented at textbooks and regular instruction and didn't have appropriate experience for using relevant integrative inquiry skills. Second, most of the methods diversified and selected by the students were confined to descriptive explanation rather than causal one. Third, both of the science attitude and academic achievement were associated with the number of diversified methods and the selection of appropriate method. Based on these findings, implications for supporting domain novices in inquiry learning environments are advanced.
