Antioxidant and digestive enzyme inhibitory effects of Eisenia bicyclis extracted by different methods (추출방법에 따른 대황 추출물의 항산화 및 소화효소 저해 효과)

  • The antioxidant and digestive enzyme inhibitory effects of Eisenia bicyclis extracted by various extraction methods (RE, reflux extraction; SE, ultrasonification extraction; AE, autoclave extraction; LE, low-temperature high-pressure extraction) were investigated. The extraction yield (55.21%) and the laminarin (39.03%), fucoidan (24.75%), total polyphenol (115.68 mg GAE/g) and flavonoid (36.67 mg RHE/g) contents of AE were higher than those in other methods. The DPPH radical (86.60%, 500 mg%), ABTS radical (58.56%, 25 mg%), nitrite (86.38%, 100 mg%) scavenging activities of the Eisenia bicyclis extracted by AE were higher than those of Eisenia bicyclis extracted by other methods. The ABTS radical and nitrite scavenging activities were above 98% in all tested Eisenia bicyclis extracts and these activities were dependent on its concentration. The inhibitory effects of AE against amylase (50 mg%) and ${\alpha}$-glucosidase (5 mg%) were 64.76% and 86.71%, respectively. The AE showed the best inhibitory effect of Eisenia bicyclis extracts (50 mg%) against trypsin (24.37%) and ${\alpha}$-chymotrypsin (49.05%), respectively. These results suggest that Eisenia bicyclis extracted by AE can be used as a bioactive and functional material in the food industry.

Protective Effect of Mulberry and Lithospermum erythrorhizon Extracts on Anti-aging against Photodamage (오디 및 자초추출물의 피부 광노화 보호효과)

  • We investigated the protective effect of UVB inducing photodamage from mulberry extract (ME) and Lithospermum erythrorhizon extract (LE). The contents of total anthocyanin and shikonin as a color compound of ME and LE were 4.92 mg/g and 9.58 mg/g, respectively. The electron donating ability and superoxide radical scavenging activity of ME were 84.32% and 76.34%, respectively. The oxygen radical absorbance capacity of the ME ($545.37{\mu}moles$ TE/g) was higher than LE ($427.18{\mu}moles$ TE/g). MMP-1 production in the HS68 cells were exposed to UVB suppressed by treatment with $200{\mu}g/mL$ of ME (68.6%) and LE (32.7%). ME and LE were applied to a skin aging mouse model, which was induced by the irradiation of UVB to the backs of hairless mice. The value of skin erythema index, wrinkle depth and thickness, epidermis thickness, and collagenous fiber damage in the experiment groups (MEL: ME 3%, MEM: ME 5%, MEH: ME 7%, LEL: LE 3%, LEM: LE 5%, LEH: LE 7%) were remarkably reduced than in the control group (only UVB exposure group), while water capacity increased. The level of total wrinkles depth in the skin was decreased to be 30% of the control group by MEH and LEM. These results suggest that ME and LE are useful cosmetic materials for skin protection against UVB-inducing.

Study on Anti-oxidant Activity of Four Kinds of Korea Herb Medicine Materials (4가지 한방 소재(행인, 호장근, 자초, 강황)의 항산화활성에 관한 연구)

  • There was an increasing interest that herbal medicine and natural material extracts were proved processes of antioxidant, cosmeceutical activity and the other effects. The aim of this study was to assess the antioxidant of extraction of four kinds from Prunus armeniaca L., Reynoutria elliptica, Curcuma aromatica, Lithospermum erythrorhizon. RE (Reynoutria elliptica) and CA (Curcuma aromatica) have good electron donating ability. The water and ethanol extract of RE at a 100 ppm concentration showed over 70%, the water extract at 500 ppm concentration showed 83% and the ethanol extract at 100 ppm concentration showed 86% of CA. Xanthine oxidase inhibition activity of the water extract of LE (Lithospermum erythrorhizon) at a 1,000 ppm concentration showed over 44%, on the other hand, RE showed in all lowest effect and there was no inhibition activity of a couple more extracts. In the measurement of nitrite scavenging activity, all extracts showed highly scavenging activity. Especially the water and ethanol extract of RE showed over 99% at 500 ppm, also LE showed over 40% at 10 ppm concentration.

Analysis and Stability Test of the Water Extracts and Powders from Asiasari Radix et Rhizoma for Toxicity Study (세신의 독성평가를 위한 성분분석 및 안정성 시험)

  • For toxicity assessment, water extracts and powders from the Asiasari Radix et Rhizoma were manufactured and the component analysis were analyzed according to the Korean Pharmacopoeia method. Stability tests of water extracts and powders from the Asiasari Radix et Rhizoma were investigated at room temperature and refrigerated for one year. Two samples were found to have been stable for a year after analyzing them with HPLC. Therefore, the use of the water extracts and powders of Asiasari Radix et Rhizoma after preparation during the animal test turned out to be stable.

Antioxidant and Anti-Adipogenic Activities of Bread Containing Corn Silk, Job's Tears, Lentinus edodes, and Apple Peel in 3T3-L1 Preadipocytes (옥수수수염, 율무, 표고버섯 그리고 사과껍질을 함유한 빵의 항산화 및 3T3-L1 지방 전구세포 분화 억제 활성)

  • Corn silk, Job's tears, Lentinus edodes, and apple peel 70% ethanol extracts (CS, JT, LE, and AP) were studied for their antioxidant activities. CS among all extracts showed the highest antioxidant activities based on total polyphenol and flavonoid contents, 2,2-diphenyl-${\beta}$-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate) radical scavenging activity, and reducing power. Adipocyte differentiation was investigated by Oil Red O staining assay using CS, JT, LE, AP, and extract of developed bread containing corn silk, Job's tears, Lentinus edodes, and apple peel (DB) treated to 3T3-L1 adipocytes. DB1 and DB2 showed anti-adipogenic and antioxidant effects. Triglyceride (TG) accumulation in 3T3-L1 cells was measured, and among the samples tested (CS, JT, LE, and AP), CS was found to have the highest inhibitory activity against TG accumulation of differentiated 3T3-L1 adipocytes and regulated factors associated with adipogenesis. CS suppressed lipid droplet formation and adipocyte differentiation in 3T3-L1 cells in a dose-dependent manner. We examined the effects of CS on the levels of CCAAT-enhancer-binding protein ${\beta}(C/EBP{\beta})$, peroxisome proliferator activated receptor ${\gamma}(PPAR{\gamma})$, and adipocyte-specific lipid binding protein (aP2) mRNA as well as protein levels in 3T3-L1 cells treated with CS at various concentrations (0, 10, 50, and $100{\mu}g/mL$) during adipocyte differentiation and treatment with CS in 3T3-L1 adipocytes down-regulated expression of $PPAR{\gamma}$ and aP2 mRNA. CS also significantly inhibited up-regulation of $C/EBP{\beta}$, $PPAR{\gamma}$, and aP2 proteins during adipocyte differentiation. These data indicate that DBs have anti-adipogenic activity induced by CS in 3T3-L1 preadipocytes, and CS exerts anti-adipogenic activity by inhibiting expression of $C/EBP{\beta}$, $PPAR{\gamma}$, and aP2 signaling pathway in 3T3-L1 adipocytes. JT, LE, and AP had no inhibitory effects on differentiation of 3T3-L1 preadipocytes but displayed strong antioxidant effects. These results suggest that the developed bread may be a health beneficial food that can prevent or treat obesity and diseases induced by oxidative stress.

Effect of Water Extracts of Cuscuta Japonica Chois in RANKL-induced Osteoclast Differentiation (파골세포 분화에서 토사자 물 추출물의 효과)

  • Osteoclasts are bone-resorbing multinucleated cells derived from the monocyte/macrophage lineage. The differentiation of osteoclasts are regulated by osteoblastic cells expressed RANKL, which is the most critical molecule for osteoclast differentiation. In this study, we found that water extracts of cuscuta inhibited RANKL-mediated osteoclast differentiation by direct action on bone marrow macrophages (BMMs) without cytotoxicity. In BMMs, water extracts of cuscuta inhibited the mRNA expression of c-Fos, NFATc1, TRAP, and OSCAR. Also, the protein expression of c-Fos and NFATc1 was inhibited by water extracts of cuscuta treatement. Water extracts of cuscuta inhibited the phosphorylation of p38, ERK, and JNK in BMMs treated with RANKL. However, water extracts of cuscuta did not inhibit RANKL-induced I-${\kappa}B$ activation. Water extract of cuscuta failed to inhibit bone resorption by osteoclasts cultured on hydroxyapatite plates. These results suggest that cuscuta may be a promising drug for use against bone disorders such as osteoporosis and rheumatoid arthritis.

Comparison of different colorimetric assays and application of the optimized method for determining the liberated fluoride contents in various tea extracts

  • The appropriate intake of fluoride (F-) is beneficial to human health; however, the over-consumption can result in various potentially harmful effects. This study compared different colorimetric reagents, i.e., aluminium-xylenol orange (Al-XO), zirconium-xylenol orange (Zr-XO), and zirconium-alizarin red S (Zr-ARS), for fluoride measurements by the UV-Vis, in terms of reaction mechanisms, method sensitivity, and interferences from aluminium and ferric ions. The colorimetric procedures were optimized, and the analytical methods were evaluated. The goodness of linearity (R2 > 0.998) was obtained for all three assays within the concentration range of 1.0-20.0 mg/L fluoride in deionized water, in which the method sensitivity followed the descending order of Zr-XO > Al-XO > Zr-ARS. The Zr-XO was applied for determining the fluoride in different tea extracts in water (90 ℃ and 60-minute-brewing) and black tea demonstrated the highest fluoride content (3.0-3.6 mg/L). The effects of brewing time and temperature on the release of fluoride in the tea extracts were also investigated, indicating these are critical factors for the fluoride extraction. This study highlighted the application potentials of the UV-Vis measurement as a simple, convenient, and cheap analytical approach and discussed different colorimetric reagents used for fluoride determination in tea extracts in the context that the UV-Vis spectrophotometers are commonly equipped in most laboratories.

Effect of Medicinal Herbs' Extracts on the Growth of Lactic acid bacteria isolated from Kimchi and Fermentation of Kimchi (한약재 추출물이 김치 관련 유산균의 성장과 김치의 숙성에 미치는 효과)

  • This studies were carried out to investigate antimicrobial activity of 35 kinds of medicinal herbs against lactic acid bacteria isolated from home-made Kimchi. The most isolated lactic acid bacteria were inhibited by ethanol extract of various medicinal herbs such as Schizandra chinensis (SC), Salvia miltiorrhiza (SM), Glycyrrhiza uralensis (GU), Lithospermum erythrorhizon (LE) and Sophrora flavescens AITON (SF). But Cnidium monnier, Pinus densiflora, Paeonia suffruticosa, Acanthopanax sessiliflorum and Rhus chinensis inhibited the growth of only a few isolated lactic acid bacteria. The pH of Kimchi containing 1% of medicinal herbs extracts such as SM, GU, LE and SF was higher than that of control during fermentation for 25 days at $10^{\circ}C$, respectively. Titratable acidity and viable cells of total bacteria and lactic acid bacteria of the Kimchi were lower than that of control during fermentation. The sensory quality (taste, flavor and overall acceptability) of SM, GU, and LE added Kimchi was similar to that of control at 10th day of fermentation. But Sophrora flavescens AITON added Kimchi decreased significantly its sensory quality compared with control(P<0.05).

Anti-inflammatory effect of a mixture of Astragalus membranaceus and Lithospermum erythrorhizon extracts by inhibition of MAPK and NF-κB signaling pathways in RAW264.7 cells (RAW264.7 대식세포에서 MAPK 및 NF-κB 신호전달 경로 억제를 통한 황기 및 지치 복합물의 항염증 효과)

  • This study investigated a mixture of Astragalus membranaceus (AM) and Lithospermum erythrorhizon (LE) extracts (ALM16), exerts anti-inflammatory effects in lipopolysaccharide (LPS)-induced RAW264.7 macrophage cells, and its underlying mechanism. ALM16 was prepared by mixing AM and LE extracts in a ratio of 7:3 (w/w). Cytotoxicity of ALM16 in RAW264.7 cells was not shown up to 200 ㎍/mL of ALM16. The results of this study showed that ALM16 does-dependently inhibits the production of nitric oxide, prostaglandin E2 and pro-inflammatory cytokines (interleukin-1β, interleukin-6, and tumor necrosis factor-α) in LPS-induced RAW264.7 cells. ALM16 not only markedly reduced the protein expression levels of inducible nitric oxide synthase and cyclooxygenase-2 (COX-2) in LPS-stimulated RAW264.7 cells, but also inhibited the nuclear translocation and DNA-binding activity of nuclear factor-kappa B (NF-κB). In addition, ALM16 specifically inhibited the phosphorylation of c-Jun N-terminal kinase and extracellular signal-regulated kinases in LPS-stimulated RAW264.7 cells. In conclusion, these results suggest that ALM16 may exert anti-inflammatory effect by modulating mitogen-activated protein kinase and NF-κB signaling pathways.

Study on DPPH Free Radical Scavenging and Lipid Peroxidation Inhibitory Activities of Vietnamese Medicinal Plants

  • Among 90 Vietnamese medicinal plant extracts investigated for their antioxidant activity by DPPH assay at various concentrations from $10-100{\mu}g/mL$, 67 showed an inhibition rate over 50% at $100{\mu}g/mL$; 47 had greater than 50% inhibition at $50{\mu}g/mL$; 17 showed over 50% inhibition at $25{\mu}g/mL$. 8 extracts which exhibited strong inhibitory activity more than 50% inhibition at $10{\mu}g/mL$ were further tested for lipid peroxidation inhibition by TBA assay. They displayed activity with $IC_{50}$ values from 30.6 to $158.9{\mu}g/mL$. Until now, this is the first report on antioxidant activity of the female flower of Borassus flabellifer, and the stem of Combretum latifolium, Embelia ribes, Spatholobus parviflorus, and Tetrastigma erubescens. Fractionations of the EtOAc extract prepared from S. parviflorus led to the isolation of protocatechuic acid (1), ferulic acid (2), epicatechin (3), and gallic acid (4). These compounds showed significant DPPH inhibitory activity with $IC_{50}$ values from 6.5 to $23.6{\mu}M$.
