Effect of n-6/n-3 fatty acid ratio on lipid metabolism in obesity model rats (n-6/n-3 지방산 비율이 비만 랫드의 지질대사에 미치는 영향)

  • This study was determined the effects of dietary omega-6 and 3 fatty acid ratios (n-6/n-3, 0, 4:1, 15:1, 30:1) on lipid metabolism in obese model animal rats. Blood triacylglycerol, total cholesterol, LDL-C, glucose, ALT, AST, insulin, and leptin concentrations in n-6/n-3 group were decreased by 22.21, 20.60, 52.96, 15.71, 11.97, 9.13, 37.57, 45.98%, respectively, while HDL-C and phospholipid concentrations were increased by 28.38, 80.39% respectively, compared with control group, as especially in 4:1 group showed the greatest effect. SREPB-$1{\alpha}$ and SREPB-2 mRNA in liver tissues were down-regulated in n-6/n-3 group, but LPL-mRNA of PPARs in adipose tissue was up-regulated compared with control group. The adipocyte size in liver tissues was decreased in the order of n-6/n-3 ratio of 30:1, control, 15:1 groups, and the adipocyte size in adipose tissues was decreased in the order of n-6/n-3 ratio of control, 30:1, 15:1, 4:1 groups.

  • The purpose of this research was to study of the lower body obesity control with female university students, particularly has been done meridian message and aroma therapy, The data included in the remainder of this research was gathered from the experimental groups consisted of eighteen female students for tested group and compared group over 8 weeks. By the observation of tested group and compared group, we could see the measurement of thigh and calf sizing down overall, but they still show some differences depend on physical constitutions of each members who has been tested. This meant that the result could be changed by the will and persistent endurance of experimented ones Both of tested group and compared group's body mass index shrinked but still there were some exceptions, this result showed the difference of each participants' willingness as well as the age gap. We could find the reduction of Total Cholesterol, LDL, neutral fat and blood sugar in both tested group and compared group. However in case of HDL the measurement increased which means wrong method of controlling obesity can show some abnormal figures

Effects of natural raw meal (NRM) on high-fat diet and dextran sulfate sodium (DSS)-induced ulcerative colitis in C57BL/6J mice

  • BACKGROUND/OBJECTIVES: Colitis is a serious health problem, and chronic obesity is associated with the progression of colitis. The aim of this study was to determine the effects of natural raw meal (NRM) on high-fat diet (HFD, 45%) and dextran sulfate sodium (DSS, 2% w/v)-induced colitis in C57BL/6J mice. MATERIALS/METHODS: Body weight, colon length, and colon weight-to-length ratio, were measured directly. Serum levels of obesity-related biomarkers, triglyceride (TG), total cholesterol (TC), low density lipoprotein (LDL), high density lipoprotein (HDL), insulin, leptin, and adiponectin were determined using commercial kits. Serum levels of pro-inflammatory cytokines including tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin (IL)-$1{\beta}$, and IL-6 were detected using a commercial ELISA kit. Histological study was performed using a hematoxylin and eosin (H&E) staining assay. Colonic mRNA expressions of TNF-${\alpha}$, IL-$1{\beta}$, IL-6, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) were determined by RT-PCR assay. RESULTS: Body weight and obesity-related biomarkers (TG, TC, LDL, HDL, insulin, leptin, and adiponectin) were regulated and obesity was prevented in NRM treated mice. NRM significantly suppressed colon shortening and reduced colon weight-to-length ratio in HFD+DSS induced colitis in C57BL/6J mice (P < 0.05). Histological observations suggested that NRM reduced edema, mucosal damage, and the loss of crypts induced by HFD and DSS. In addition, NRM decreased the serum levels of pro-inflammatory cytokines, TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 and inhibited the mRNA expressions of these cytokines, and iNOS and COX-2 in colon mucosa (P < 0.05). CONCLUSION: The results suggest that NRM has an anti-inflammatory effect against HFD and DSS-induced colitis in mice, and that these effects are due to the amelioration of HFD and/or DSS-induced inflammatory reactions.

Effects of a Pueraria lobata-root based combination supplement containing Rehmannia glutinosa and aerobic exercise on improvement of metabolic dysfunctions in ovariectomized rats (갱년기 모델 유도 흰 쥐에서 갈근과 지황 복합물 및 유산소 운동이 대사이상 개선에 미치는 영향)

  • Purpose: There is a fair amount of evidence indicating that increased risk of obesity and insulin resistance is associated with postmenopausal state, but can be modulated by diet and exercise. In this study, we explored whether a Pueraria lobata root-based supplement containing Rehmannia glutinosa (PR) and/or aerobic treadmill exercise can modify the metabolic changes associated with estrogen deficiency. Methods: Seventy rats were randomly assigned to the following groups for 8 weeks (n=10 per group): SHAM, sham-operated; PR0, ovariectomized (OVX) control; PR200, OVX with PR200 mg/kg B.W; PR400, OVX with PR400 mg/kg B.W; EPR0, OVX with exercise; EPR200, OVX with exercise and PR200 mg/kg B.W; EPR400, OVX with exercise and PR400 mg/kg B.W. Results: OVX induced significant increases in body weight, food intake, fat mass, LDL-cholesterol, and fasting blood glucose, confirming induction of menopausal symptoms. PR supplementation or exercise significantly suppressed the above mentioned changes through different regulatory elements in adipose tissue: PR supplement upregulated adiponectin gene expression and aerobic exercise upregulated adiponectin and insulin receptor gene expression and a combination of PR supplement and aerobic exercise showed an additive effect on adiponectin gene expression. Conclusion: Taken together, the results of this study suggest that PR supplement has a potential to provide health benefits in OVX rats through leptin and adiponectin secretion. In addition, the data suggest that combination of exercise and PR would have additive effects on metabolic dysfunction associated with estrogen deficiency.

Effect of Dietary Cinnamon Powder on Savor and Quality of Chicken Meat in Broiler Chickens (닭고기의 품질 및 맛에 관한 계피 급여효과)

  • A 35-day trial was carried out to determine the influence of dietary cinnamon powder (CNP) on the sensory evaluation and quality of chicken meat, carcass characteristics, plasma lipid level and growth performance of broiler chickens. There were 5 treatment groups: control; CNP 2.0%; CNP 3.0%; CNP 4.0%; and CNP 5.0%. The body weight of the broilers fed the diets containing 3.0% CNP was higher than the broilers fed the control feeds (p<0.05). The concentration of triacylglyceride, HDL-C was higher in the plasma from broiler chickens fed diets with CNP (p<0.05) but the concentrations of total cholesterol and LDL-C were significantly lower (p<0.05) compared to the control group. The carcass percentage, chicken breast and thigh weight were not different between the CNP and control groups. The WHC was significantly higher in the chickens fed 4% CNP diet, while the TBARS was significantly lower (p<0.05) in the chickens fed 3% CNP diet compared to the control group. The color of the breast muscle from the chickens fed 3% CNP diet was lighter than those from the control groups (p<0.05). The sensory evaluation of the taste and savor related to CNP in fried or boiled chicken meat were significantly better from the broiler chicken fed diets containing CNP than the control group (p<0.05). These results suggest that dietary cinnamon powder may improve savor and quality of chicken meat in broiler chickens.

  • Soybean (SoyPep) was prepared and evaluated their action on blood lipid profiles, select biochemical indexes and histopathological changes in spontaneously hypertensive rat (SHR). Male SHRs were divided into three groups and fed the experimental diet containing 0% (control), 3%, 10% SoyPep for 8 weeks. Lipids, antioxidative vitamin levels, glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT) activities in plasma were measured, and angiotensin I converting enzyme (ACE) activities in kidney, vascular tissue and plasma were also measured. In addition, histopathological changes in various organs were investigated. Dietary SoyPep was effective for lowering plasma levels of triglyceride and LDL-cholesterol. ACE activities in aorta and kidney were decreased in the group fed SoyPep compared to control. There was no difference of plasma GOT and GPT activities, and no histopathological difference in various organs among the experimental groups. Our results suggest that SoyPep has beneficial effects on diseases such as hypertension and atherosclerosis. SoyPep could be useful to develop a novel functional safe food additive for preventing the chronic diseases.

Effects of Hot-Water Extract of Mulberry Leaf Tea Fermented by Monascus pilosus on Body Weight and Hepatic Antioxidant Enzyme Activities in Mouse Fed a Normal Diet (정상식이 마우스의 체중과 간 조직 항산화계 효소활성에 미치는 Monascus pilosus 발효 뽕잎차 열수추출물의 효과)

  • In this study, to evaluate the anti-obesity effects of mulberry leaf tea and its fermented product by Monascus pilosus, we investigated body and organ weight, blood and liver biomarkers in mice fed 1% tea infusions instead of water for 8 weeks. Mice were divided into three groups such as a normal control (NC), unfermented mulberry leaf tea infusion (UMI) and fermented mulberry leaf tea infusion (FMI). Although it is not significant, tea infusion groups showed reduction of body weight gains compared with NC group. Moreover, contents of LDL-cholesterol and lipid peroxide (LPO), altherogenic index, and xanthin oxidase (XO) activity were significantly decreased, and glutathione S-transferase (GST) activity was significantly elevated. The results from this study suggested that UMI and FMI may have an anti-obesity activity, upregulate antioxidant enzymes and reduce levels of oxidants related to liver damage.

  • The purpose of this study was to evaluate anti-obesity activity of Cirsium setidens Nakai test material (CNTM) in 3T3-L1 adipocytes and obese C57BL/6J mice fed with a high-fat diet using various obesity-related in vitro experiments. During adipocyte differentiation, CNTM significantly inhibited lipid accumulation and ROS production compared to controls. To evaluate whether CNTM could exert glycerol release effects on mature 3T3-L1 adipocytes, we treated cells with various concentrations of CNTM for 1 h. Treatment of mature adipocytes with $160-320{\mu}g/mL$ of CNTM increased the release of glycerol, but not in a significant dose-dependent manner. Anti-adipogenic and anti-lipogenic effects of CNTM seemed to be mediated by the inhibition of $PPAR{\gamma}$ and $C/EBP{\alpha}$. Moreover, CNTM stimulated fatty acid oxidation in an AMPK-dependent manner. CNTM-treated groups of C57BL/6J mice showed reduced body weights and adipose tissue weight with improving serum lipid profiles and adiponectin protein expression in obese C57BL/6J mice fed with a high-fat diet. These results suggest that CNTM might have anti-obesity effect on adipogenesis and lipid metabolism in vitro and in vivo. This presents the possibility of developing a treatment for obesity using nontoxic natural resources.

Bifidogenic Effects of the Oral Administration of Fly Maggot Extract on Organic Acid, Cecal Microorganisms, Thymus and Spleen Weights, and Blood Lipids in Rats (파리유충 추출물이 랫드의 혈액지질, 면역세포 중량, 맹장 미생물 및 유기산 변화에 미치는 비피더스 활성효과)

  • Housefly (Musca domestica L.) maggots are used as biomedical material. Ethanolic extracts of fly maggot (EM) were orally administered to male rats at levels of 0 (control group), 4.0, 6.0, and 8.0 mg per 100 g live weight for 40 days. Serum triglycerides, total cholesterol, and LDL-C decreased by 17.90, 17.60, and 16.37%, respectively, whereas HDL-C increased by 20.48% in the EM group compared with these parameters in a control group (p<0.05). Thymus and spleen weights dose-dependently increased by 21.42% and 21.42%, respectively, but abdominal fat decreased by 39.66% after EM administration compared with that in the control group (p<0.05). IgG, IgA, and IgM increased 35.14, 68.65, and 190.16%, respectively, in the EM groups compared to the control group (p<0.05). Bifidobacterium and Lactobacillus increased by 41.68% and 35.55%, respectively, in the EM groups compared with the control group, and Bacteroides, Clostridium, Escherichia, and Streptococcus decreased by 24.96, 46.37, 25.00, and 34.05%, respectively, in the EM groups compared with the control group (p<0.05). Compared with the control group, total organic acids, acetic acid, and propionic acid increased by 31.11, 49.34, and 24.88%, whereas butyric acid, isobutyric acid, valeric acid, and isovaleric acid decreased by 30.79, 72.64, 32.90, and 63.16% respectively, in the EM groups (p<0.05). These results suggest that EM has a bifidogenic effect on immune cell development, blood lipid levels, and abdominal fat reduction by increasing the production of organic acid and numbers of cecal microorganisms in animals.

Effect of Crude saponin from Red-ginseng efflux on Blood biochemical parameters in Rats Acutely Exposed to 2,3,7,8-tetrachlorodibenzo-ρ-dioxin (TCDD) (홍삼유출액으로부터 분리한 조사포닌이 TCDD (2,3,7,8-Tetrachlorodibenzo-ρ-dioxin)로 급성독성을 유도한 흰쥐의 혈액 생화학지수에 미치는 영향)

  • This study was carried out to investigate the protective effect of crude saponin from red ginseng efflux (RGE-CS) on biochemical parameters in male rats acutely exposed to 2,3,7,8-tetrachlorodibenzo-$\rho$-dioxin (TCDD). Forty male rats ($200{\pm}20g$) were divided into 4 groups. Normal control group (NC) received vehicle and saline; only TCDD-treated group (TT) received TCDD ($5{\mu}g/kg$, single dose) intrperitoneally; RGE-CS 20 received 20 mg/kg of crude saponin i.p. for 4 weeks from 1 week before TCDD-exposure; RGE-CS 40 also received 40 mg/kg of crude saponin i.p. for 4 weeks from 1 week before TCDD-exposure. Body weight of TT group was significantly decreased after TCDD-exposure. However, body weight of crude saponin groups increased throughout the experimental period, although the increasing rate was slower than that of NC group. Decrease in body weight was not observed during the experimental period in RGE-CS 40. Increases in triglyceride (TG), total cholesterol (TC), low-density lipoprotein (LDL), AST, ALT and $Fe^{2+}$ levels by TCDD intoxication were significantly attenuated by the RGE-CS treatment. Decrease in glucose, amylase, lactate dehydrogenase (LDH) and creatinine kinase (CK) by TCDD also were inhibited by the RGE-CS. These results suggest that saponin from red-ginseng efflux might be a useful protective agent against TCDD, an endocrine disrupter.
