Metabolic Adjustment of Lactate Dehydrogenase Isozymes to a Change in Dissolved Oxygen in Bluegill (Lepomis macrochirus) (파랑볼우럭(Lepomis macrochirus)에서 용존산소량의 변화에 대한 젖산탈수소효소 동위효소들의 대사조절)

  • The aim of this study was to examine the metabolic adjustment of lactate dehydrogenase (EC 1.1.1.27, LDH) isozymes to a change in dissolved oxygen (DO) in bluegill (Lepomis macrochirus). After bluegills were adapted to a constant environment in an aquarium, the DO was changed to investigate the activity of LDH isozyme and the relative ratio of subunits A, B, and C for each tissue. When the DO was decreased from 18 ppm to 6 ppm, LDH in skeletal muscle, heart, and brain tissues recovered to the level of control activity within 12, 12, and 6 hr, respectively. LDH activity changed in accordance with a change in DO. The compensation was performed rapidly and is thought to be an important function of LDH in enabling bluegills to adapt to their environment. In bluegill heart, eye, and brain tissues, the relative ratio of subunit A increased and showed a tendency to recover similarly to the subunit ratio of control groups up to 12 hr. It is thought that the anaerobic metabolism using subunit A was increased in the initial stage when DO was changed. In addition, the results revealed that subunit C was more similar to subunit A than subunit B. In bluegills, subunits A and C of LDH seem to be evolutionarily similar. LDH isozymes, mainly containing subunits A and C, are likely responsible for the function of pyruvate reductase, which plays a role in making the bluegill adapt to a hypoxic environment through anaerobic metabolism.

Distribution and Role of Mitochondrial Lactate Dehydrogenase Isozymes in Bird and Mammals (조류 및 포유류 내 미토콘드리아 젖산탈수소효소 동위효소들의 분포와 역할)

  • Mitochondria were isolated from bird and mammals. The activity of monoamine oxidase (EC 1.4.3.4) was then measured to identify mitochondrial isolation. Lactate dehydrogenase (EC 1.1.1.27, lactate dehydrogenase, LDH) isozymes in mitochondrial fractions were analyzed by biochemical and immunochemical methods. The activity of mitochondrial LDH was lower in mammals than in bird. Therefore, the role of mitochondrial LDH seems to be more important in bird than in mammals. The concentration of protein in all tissues of bird and mammals was less in the mitochondria than in the cytosol. In the cytosol of mice and golden hamsters, testis-specific LDH $C_4$ isozyme was expressed in testis in addition to the LDH $A_4$, $A_3B$, $A_2B_2$, $AB_3$, and $B_4$ isozymes. A single LDH AB hybrid isozyme was expressed in the chicken mitochondria. In mammals, mitochondrial LDH isozymes were differed according to tissues. LDH $A_4$ and testis-specific LDH $C_4$ isozymes were expressed in the mitochondria of mice. The mitochondrial testis-specific LDH $C_4$ isozyme was expressed only in the mice. In the golden hamster mitochondria, the LDH $B_4$ isozyme functioned as a lactate oxidase. As our results show, the mitochondrial LDH seemed to be playing the different role in the bird and mammals in relation with their metabolic conditions and habitats.

Effects of Guaruhaebaekbanha-tang Extract on Beating Rate and LDH Activity in Cultured Rat Myocardial Cells (과루해백반하탕 추출물이 배양 심근세포의 박동수와 LDH 활성도에 미치는 영향)

  • To certify the protective effect of herbal medicine against oxygen free radical-induced myocardiotoxicity, cytotoxicity was measured using MTT, LDH activity and Beating rate assay in the presence of Guaruhaebaekbanha-tang(GHBT) extracts or single constituents of this prescription. Myocardial toxicity was evaluated in neonatal rat myocardiocytes in cultures. In the present study, xanthine oxidase/hypoxanthine(XO/HX) resulted in a decrease in cell viability, increases in LDH activity in culture medium and decreases in beating rate in cultured myocardial cells. In the effect of GHBT extract, it showed the prevention from the XO/HX-induced cardiotoxicity by the increases of cell viability and beating rate as well as the decrease of LDH activity. In the protective effect of Fructus Trichosanthis(FT), Bulbus Allii Macrostemi(BAM) and Rhizoma Pinelliae(RP), all the extracts were significantly effective in the protection of XO/HX-induced cardiotoxocity in cultured myocardial cells by the increase of beating rate as well as th decrease of LDH activity. From these results, they show that XO/HX is cardiotoxic in cultured myocardial cells derived from neonatal rat, and it suggests that GHBT, FT, SAM, RP extracts are positively effective in the blocking in XO/HX-induced cardiotoxicity.

The Inhibition Effects of Butylatedhydroxyanisole and Flavone on the Microsomal Activation of Cigarette Smoke Components in Rat (Butylatedhydroxyanisole과 Flavone에 의한 연기성분의 생체내 활성억제에 관한 연구)

  • The inhibition effect of flavone and butylatedhydroxyanisole (BHA) on the microsomal activation of Cigarette Smoke Condensate (CSC) or its Neutral Portion (NP) was investigated in Rat. The activities of Latic acid dehydrogenase (LDH) in serum was measured in the time intervals of 3, 6, 12, 18, 24 and 30 hr, respectively, after the injection (ip) of CSC (5mg/kg) or NP (10mg/kg) to Wistar male rat. Flavone (1mg/kg) and BHA (1mg/kg) were injected along with CSC or NP. The significant enhancement of the LDH activity in serum was observed in both cases of rats treated with CSC and NP. A drastic decrease of LDH activity from 1040 unit to 641 unit was observed after 12 hours of injection of CSC along with flavone. In contrast with the case of flavone, BHA reduced the enzyme activity from 825 unit to 652 unit at the same condition of flavone. Therefore, flavone can be considered to be a better inhibitor on action of CSC in vivo.

Effect of Ginseng Ethanol Extract on Lactate Dehydrogenate-5 in Rat Brain with Age (인삼 Ethanol Extract가 쥐의 뇌 Lactate Dehydrogenase-5에 미치는 영향)

  • Attempts were made to see if feeding of ginseng ethanol extract could affect proper- ties of rat brain lactate dehydrogenase such as specific activity, heat stability, Km for substrate, inactivation by 3-bromopyruvate and trypsin, and immune response. The following results were obtained. Specific activity of LDH was observed to reach maximum in 5 month after birth and then decrease steadily. However, that of LDH from rat fed with ginseng ethanol extract was found in rat fed with ginseng ethanol extract. 3-bromopyruvate was shown to inactivate LDH-5 from old rat fed. Inactivation of LDH-5 by trypsin was remarkable in old rat fed. Km value for pyruvate in old rat fed was remarkably decreased. Cumulative results suggest that ginseng ethanol extract could affect conformational change of LDH responsible for altered properties through unknown mechanism.

  • In this work, silver nitrate complexes of sulfanilamide-5-methyl-2-thiophene carboxaldehyde (SMTCA) ligand intercalated Zn/Al-layered double hydroxide [Ag-SMTCA-LDH] were synthesized for the potential application as an antimicrobial system. The SMTCA ligand was synthesized by reacting sulfanilamide and 5-methyl-2-thiophene carboxaldehyde in methanol and further complexation with silver nitrate metal ions [Ag-SMTCA]. The structural analyses of synthesized compounds confirmed an intercalation of Ag-SMTCA into Zn/Al-NO3-LDH by flake/restacking method. SMTCA, Ag-SMTCA and Ag-SMTCA-LDH were characterized by 1H nuclear magnetic resonance (1H NMR) spectroscopy, Fourier-transform infrared (FTIR), ultraviolet-visible (UV-Vis) spectrophotometer, scanning electron microscopy (SEM) and transmission electron microscopy (TEM), X-ray diffraction (XRD), and thermogravimetric analysis (TGA). It was found that Ag-SMTCA-LDH exhibited good antimicrobial activity against both gram-positive (Bacillus subtilis, [B. subtilis], Staphylococcus aures, [S. aureus]) and gram-negative (Escherichia coli, [E. coli], Pseudomonas aeruginosa [P. aeroginosa]) bacteria as well as excellent antioxidant activity.

Lactate Dehydrogenase and Monocarboxylate Transporters 1, 2, and 4 in Tissues of Micropterus salmoides (큰입우럭(Micropterus salmoides) 조직의 젖산탈수소효소 및 Monocarboxylate 수송체(MCT) 1, 2, 4)

  • The properties of lactate dehydrogenase (EC 1.1.1.27, LDH) and expression of monocarboxylate transporters (MCTs) 1, 2, and 4 were studied in tissues from Micropterus salmoides. Native-PAGE revealed that the LDH $A_4$ isozyme was predominantly located in skeletal muscle. The LDH $A_4$, $A_2B_2$, and $B_4$ isozymes were detected in heart, liver, eye, and brain tissues, while eye-specific $C_4$ isozyme was detected in eye tissue. In September, strong LDH $B_4$ isozyme activity was detected in heart tissue. High $A_4$ isozyme activity was noted in all other tissues except heart tissue. However, in November, strong $A_4$ isozyme activity was detected in heart tissue. The LDH/CS (Citrate synthase, EC 4.1.3.7) ratio in skeletal muscle and heart tissues indicated that anaerobic metabolism was high in those tissues. Native-PAGE after immunoprecipitation showed that eye-specific $C_4$ isozyme was more similar to the $A_4$ than the $B_4$ isozyme. The LDH $A_4$ isozyme was purified by affinity chromatography. The molecular weight of subunit A was 37,200. The LDH activity in tissues was consistently 11.05~28.32% due to inhibition by 10 mM pyruvate. The $K_m^{PYR}$ of LDH in eye tissue was very low. The optimum pH for LDH in tissues was pH 7.5~8.0. The LDH $A_4$ isozyme was detected in mitochondria of skeletal muscle, whereas the $B_4$ and $A_2B_2$ isozymes were detected in heart tissue mitochondria. Western blot analysis indicated that MCTs 1, 2, and 4 were located in the plasma membrane and mitochondria of skeletal muscle and heart tissues. The sizes of MCTs 1, 2, and 4 in skeletal muscle were 60, 54~38, and 63 kDa, while those in heart tissue were 57, 54~38, and 55.5 kDa, respectively. In conclusion, M. salmoides appears to use anaerobic metabolism predominantly when adapted to a hypoxic environment. In highly activated skeletal muscle and heart tissue, energy production is controlled by inward and outward flows of pyruvate and lactate through MCTs 1, 2, and 4 in the plasma membrane and mitochondria, with effective adjustment by LDH isozymes.

Comparison of serum isoenzyme levels of CPK and LDH in patients ungergoing thoracic operations (흉부수술 환자에서의 CPK 와 LDH I soenzyme 의 변화)

  • A analysis of CPK & LDH Isoenzyme was done on the consecutive patients undergoing thoracic operations from July 1982 to October 1982 in the Department of Thoracic and Cardiovascular Surgery, Capital Armed Forces General Hospital. Eighteen patients were analysed by three groups, such as open heart surgery [group A], major thoracic operation [group B] , minor thoracic operation group [group C]. In all patients serial determination of total level and Isoenzyme of CPK, LDH wad done on preoperative operative and up to 8th post-operative day, The results obtained are as follows. 1. The average value of serum CPK before the operation was 61 IU/L. The value of serum CPK was increased following the operation mainly MM portion and reached to the maximal level of 536107 IU/L in A group 1200191 IU/L in B group, 306150 IU/L in C group on the first postoperative day. The enzyme activity was gradually decreased thereafter and returned to the normal range on the 3rd or 4th day after the operation. 2. The average value of serum LDH before the operation was found to be 83 IU/L. The value was increased during the operation and reached to the maximal level of 481108 IU/L in group A, 14827 I U/L in group B, 10035 IU/L in group C on the second day after the operation. The enzyme activity was gradually decreased thereafter and returned to the normal range on the seventh day after the operation. The enzyme activity was dependent to the duration of operation, severity of muscle damage, type of thoracotomy, effect of extracorporeal circulation, state of disease.

  • In order to study the difference in metabolic efficiency of the cotyledon and seedlings of soybean during germination, the authors assayed lactic dehydrogenase activities in the two tissues for the purpose of comparison, with the following conclusions. 1. The LDH activity in the cotyledon of soybean increases during the 3 days after germination, followed by abrupt decrease during the later 3 days of germination. 2. The LDH activity in the seedlings of soybean increases during the 3 days after germination under fairly good correlation with the variation observed in the cotyledon; but, unlike the cotyledon LDH, the activity remains high through the later 3days of germination. 3. It is, therefore, concluded that the metabolic efficiency in the cotyledon seems to begin to stop at the 3rd day following germination, while the efficiency in the seedlings remains high.

Metabolic Adjustments of Lactate Dehydrogenase Isozymes to the Environmental Temperature in Bluegill (Lepomis macrochirus) (환경온도에 대한 파랑볼우럭(Lepomis macrochirus) 젖산탈수소효소 동위효소들의 대사조절)

  • The aim of this study was to examine the metabolic adjustment of lactate dehydrogenase (EC 1.1.1.27, LDH) isozymes to the environmental temperature in bluegill (Lepomis macrochirus). This study included three groups of bluegill collected in April (group Ⅰ), May (group Ⅱ), and September (group Ⅲ). The LDH activities of skeletal muscle, heart, and brain tissues were higher in group Ⅲ than in groups Ⅰ and Ⅱ. The citrate synthase (EC 4.1.3.7, CS) activity was higher in skeletal muscle but lower in heart and brain tissues of group Ⅱ as compared to group Ⅰ. In contrast, the CS activity was lower in skeletal muscle and higher in heart and brain tissues in group Ⅲ than in group Ⅱ. Furthermore, the LDH/CS activity ratio was higher in the skeletal muscle and brain in group Ⅲ than in groups Ⅰ and Ⅱ. Accordingly, anaerobic metabolism was increased in group Ⅲ. LDH A4, A2B2, and B4 isozymes were expressed in skeletal muscle, heart, liver, and brain tissues. The LDH C hybrid was detected in brain tissue. The LDH A4 isozyme was successfully purified by affinity chromatography. The molecular weight of the purified LDH A4 isozyme was 136 kDa and its optimal pH for enzymatic activity was 8.0. The KmPYR values of LDH in skeletal muscle were 0.161-0.227 mM using pyruvate as a substrate. These kinetic properties of LDH in skeletal muscle are consistent with the fact that bluegill is a cold-adapted species. These results may be useful for predicting the habitat use of this fish.
