영지버섯 균사체를 이용한 고체 발효 쌍화탕의 급성독성에 관한 연구 (Acute Toxicity Study on Ssanghwa-tang Extract Fermented with Ganoderma lucidum in Mice)

  • This study was carried out to investigate the acute toxicity and safety of Ssanghwa-tang extract fermented with Ganoderma lucidum. To evaluate the acute toxity and safety, 0(control group), 1250, 2500 and 5000 mg/kg of Ssanghwa-tang and fermented Ssanghwa-tang extracts were orally administered to 20 male and 20 female ICR mice. After single administration, we observed survival rates, general toxicity, changes of body weight, and autopsy. Compared with the control group, we could not find any toxic alteration in all treated groups (1250, 2500 and 5000 mg/kg). $LD_{50}$ of Ssanghwa-tang and fermented Ssanghwa-tang extracts might be over 5000 mg/kg and it is very safe to ICR mice.

  • In this research, the acute toxicity of fermented Yukmijihwangtang extract was examined using male and female ICR mice, To evaluate the acute toxity and safety, 0(control group), 1250, 2500 and 5000 mg/kg of fermented Yukmijihwangtang extract were orally administered to male and female ICR mice. After single administration, we observed survival rates, general toxicity, changes of body weight for the 14 days and autopsy at 1 day following the administration according to the Regulation of Korean Food and Drug Administration. Compared with the control group, we could not find any toxic alteration in all treated groups (1250, 2500 and 5000 mg/kg). $LD_{50}$ of fermented Yukmijihwangtang extract might be over 5000 mg/kg and it is very safe to ICR mice.

Single Oral Dose Toxicity Test of Platycodin D, a Saponin from Platycodin Radix in Mice

  • The object of this study was to evaluate the single oral dose toxicity of platycodin D, a saponin from the root of Platycodon grandiflorum in male and female mice. Platycodin D was administered to female and male mice as an oral dose of 2000, 1000, 500, 250 and 125 mg/kg (body wt.). Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after treatment, upon necropsy, organ weight and histopathology of 14 principle organs were examined. As the results, no platycodin D treatment related mortalities, clinical signs, changes on the body and organ weights, gross and histopathological observations against 14 principle organs were detected up to 2000 mg/kg in both female and male mice. Therefore, $LD_{50}$ (50% lethal dose) and approximate LD of playtcodin D after single oral treatment in female and male mice were considered over 2000 mg/kg - the limited dosages recommended by KFDA Guidelines [2009-116, 2009], respectively.

신경작용제 사린 및 소만 중독에 대한 기니픽에서의 예방패치의 해독 및 뇌보호 효능 (Antidotal and Neuroprotective Efficacies of a Prophylactic Patch against Sarin and Soman Poisonings in Guinea Pigs)

  • This study was designed to evaluate the prophylactic efficacy of a combinational patch system containing physostigmine and procyclidine against sarin and soman using guinea pig. The median lethal dose values of two nerve agents were calculated by a probit analysis of deaths occurring within 24 h. In this study, the values of median lethal dose of sarin and soman were determined to be 33.0 and 26.7 ㎍/kg in guinea-pigs, respectively. The guinea pigs treated with a prophylactic patch(4×5 ㎠) for 24 h were 100 % protected against a challenge of 1.5 LD50. The combinational KMARK-1(atropine and 2-PAM) and prophylactic patch were more effective than a single KMARK-1, a combination of pyridostigmine and KMARK-1 significantly. Epileptiform seizures in the guinea pigs treated with the combinational antidotes led to neuropathological changes, in comparison with intact feature of brain of the animal treated with the patch.

Methods: In this study, we investigated the acute toxicity of water-extracted Ojeok-san(Wuji-san). 0(control group), 1250, 2500, and 5000 mg/kg of Ojeok-san(Wuji-san) were orally administered to 20 male and 20 female for 14 days.   • 수증기 증류, 헥산 및 초임계 추출법에 의하여 추출된 고수 및 계피 정유성분을 통해 복숭아혹진딧물, 점박이응애 및 배추좀나방에 대한 살충효과를 검정하였다. 고수의 수증기 증류, 헥산 및 초임계 추출 정유성분의 살충 및 살비활성 결과, 수증기 증류 추출물은 복숭아혹진딧물 성충에 대해 $30.59{\mu}g/cm^2$ 로 우수한 활성을 나타내었다. 계피의 수증기 증류, 헥산 및 초임계 추출법 정유성분의 살충 및 살비활성 결과, 수증기 증류 및 헥산 추출물은 점박이응애 성충에 대해 각각 5.96, $4.64{\mu}g/cm^2$ 의 우수한 활성을 나타내었다. 또한 초임계 추출에서 복숭아혹진딧물 성충에 대해 $6.50{\mu}g/cm^2$의 활성을 나타내었다. 본 연구결과는 고수와 계피의 수증기 증류, 헥산 및 초임계 추출 정유성분을 이용한 친환경 방제 가능성을 보여준다.

나리 생장, 자구 발달 및 화분과 종자발아에 미치는 방사선의 영향 (Effect of Irradiation on Growth, Bulblet Formation, and Germination of Pollen and Seed of Several Lily Cultivars)

  • 나리 돌연변이 육종을 위한 기초자료로서 구근, 종자, 화분의 방사선 감수성을 구명하고자 하였다. 'New Wave'외 6품종의 구근은 감마선에 매우 민감하였다. 무조사구는 개화하였으나 조사구에서는 생육이 크게 지연되었으며 화서는 전혀 형성되지 않았다. 'New Wave'와 'Tiny Dino'는 125Gy 조사구에서 생존은 하였으나 생육은 저조하였다. 'New Wave'와 'Tiny Dino'를 제외한 다른 품종들은 잎의 일부 또는 전부가 고사하였다. 품종별 인편의 수분함량은 72~78%이었다. 인편으로부터 자구 형성은 무조사구에서 100%, 조사된 구에서는 'Siberia' 50Gy 처리구에서만 8.5% 일어났다. 화분의 발아율은 감마선 조사에 의해 일정한 경향을 나타내지 않았다. $F_1$ 'Augusta' 종자는 선량이 높아질수록 생존율은 급격히 떨어졌다. 자구는 무조사구에서만 형성되었으며 $LD_{50}$는 100Gy 미만이었다. 방사선 조사된 기내 $F_1$ 'Augusta'의 자구는 배양방법에 관계없이 구고와 구폭이 선량 증가와 함께 감소하였다. 신초 형성은 무조사구에서 100% 되었으나 다른 처리구에서는 15~16%로 떨어졌다.

Hypotensive and Toxicological Study of Citric Acid and Other Constituents from Tagetes patula Roots

  • Study of the effects of the methanolic extract of Tagetes patula roots on blood pressure led to the isolation of well known citric (1) and malic acid (7) as hypotensive, and pyridine hydrochloride (4) as hypertensive constituents of the plant along with a new constituent, 2-hydroxy, 5- hydroxymethyl furan (9). Citric acid and malic acid caused 71% and 43% fall in Mean Arterial Blood Pressure (MABP) of rats at the doses of 15 mg/kg and 30mg/kg respectively while pyridine hydrochloride produced 34% rise in the MABP of rats at the dose of 30mg/kg. $LD_{50}\;and\;LD_{100}$ of citric acid in mice have been determined as 545 mg/kg and 1000 mg/kg, respectively.
