Assessment of Soil Properties and Growth of Organically Cultivated Cucumber (Cucumis sativus L.) with Applications of Livestock Manure Compost and Fish Meal Liquid Fertilizer (가축분 퇴비와 어분 액비 시용이 유기농 오이 생육 및 토양환경에 미치는 영향)

  • This study was carried out to investigate the effects of livestock manure compost and fish-meal liquid fertilizer on the growth of cucumber and the soil properties for the stable production of organic cucumber. Cucumber was transplanted in greenhouse on the $6^{th}$ of April in 2017, and this experiment contained five treatments: livestock manure compost 100% (LC 100%), livestock manure compost 50% + fish-meal liquid fertilizer 50% (LC50 + LF50), livestock manure compost 50% (LC50), chemical fertilizer (NPK), and no fertilizer (NF). As a result, it was shown that soil chemical properties of LC50 + LF50 plot is not different from that of LC100 plot except for the EC content, but soil chemical properties of LC50 + LF50 plot is statistically significantly different from that of NPK plot except for pH. As a result of evaluating the functional diversity of soil microbial communities using Biolog system, the substrate richness (S) and the diversity index (H) were the highest in LC50 + LF50 plot. As a result of comparing the cucumber growth and yield, it was found that there was no statistically significant difference between the plant height and the fresh weight of LC100, LC50 + LF50, and NPK plot, but the plant height and the fresh weight of LC100, LC50 + LF50, and NPK plot were different from that of LC50 and NF plot. The yield of cucumber was the highest in NPK plot r(7,397 kg/10a), but there was no statistically significant difference in the yield of cucumber between NPK plot and LC100, LC50 + LF50 plot. The above-described results suggested that the livestock manure compost and fish meal liquid fertilizer can be used for organic cucumber production under greenhouse condition.

Study on Short-term Toxicological Evaluation of Treated Landfill Leachate Using Early Stage of River Puffer Fish, Takifugu obscurus (황복 Takifugu obscurus 자치어를 이용한 침출수 단기독성 연구)

  • Toxic effect of landfill leachate on the larvae and juvenile stage of the river puffer fish, Takifugu obscurus, were investigated in order to assess severity of environmental impact of leachate effluent on the general population of estuarine fishes. A short-term toxic experiment was designed to test both laboratory hatched larvae and juveniles (5, 10 and 15 mm in length) and in-situ juveniles (30 and 45 mm in length) using the leachate concentrations between 0 and 16%. Lethal concentrations of 50% mortality ($LC_{50}$) were observed using Spearman-karber Method. 24hr-$LC_{50}$ appeared at the leachate concentrations ranging from 3.03 to 8.57%, 48hr-$LC_{50}$ at 2.73 to 6.21 %, 72hr-$LC_{50}$o at 2.45 to 5.53%, and 96hr-$LC_{50}$ at 2.38 to 4.93%, respectively. Leachate concentrations between 0.69 and 1.51% induced 96hr-$LC_1$. Respiratory frequency was significantly affected even at low leachate concentrations between 0.5 and 1.0% (P < 0.05). These results suggest that the leachate effluents may cause a harmful impact on the physiology of river puffer fish, especially for younger cohorts.

Separation of Lentil Lectin Using Free-Flow Electrophoresis (자유유동 전기이동을 이용한 Lentil Lectin의 분리)

  • A Purification device with 30-channel free-flow electrophoresis was assembled to treat samples of 240m1 volume for purification of lentil lectin (LcH) from lentil seeds with no impurities in a silverstained PAGIEF gel. HEPES(50mM)-Ttis(50mM), Cycloserine(50mM)-urea(3M), Histidine(50mM)-urea(3M) were used as ampholytea among which Histidine(50mM)-urea(3M) (pI 7.65) was found best in resolution. LcH is known to be present in the form of LcH-A, LcH-B and the complex of the two. The complex, however, disappeared when urea was added in the ampholytes, which suggested that the complete purification of two isolectins is possible using the present multistep purificaton device.

Determination of Bioconcentration Factor on Phosphamidon and Profenofos by Flow-through Fish Test (Flow-through fish test를 이용한 Phosphamidon과 Profenofos의 생물농축계수의 측정)

  • The present study was performed to investigate the bioconcentration of phosphamidon and profenofos. The BCFs(bioconcentration factors), depuration rate constants and LC$_{50}$ for two pesticides in zebrafish(Brachydanio rerio) were measured by the flow-through system(OECD guideline 305). The results obtained are summarized as follows: The 24-hrs LC$_{50}$, 48-hrs LC$_{50}$, 72-hrs LC.n and 96-hrs LC$_{50}$ were more than 100 mg/l for phosphamidon. The concentration of phosphamidon in zebrafish reached an equilibrium in 12 hrs at low and high concentrations(0.2 mg/l and 1 mg/1). The average BCF values of phosphamidon were less than 1 at low(0.96, n=7) and high concentrations (0.89, n=7) after 12~168 hrs. Depuration rate constants of phosphamidon were 0.18 hr-1 and 0.21 hr-1, half-life of phosphamidon were 3.85 and 3.30 at low and high concentrations(0.2 mg/l and 1 mg/l), respectively, The concentrations of phosphamidon in zebrafish at low and high concentrations were rapidly decreased after 8(0.04 $\mu\textrm{g}$/g) and 12 hrs(0.07 $\mu\textrm{g}$/g). The 24-hrs LC$_{50}$, 48-hrs LC$_{50}$, 72-hrs LC$_{50}$ and 96-hrs LC$_{50}$ were 2.9, 2.6, 2.2 and 2.0 mg/1 for profenofos. The concentration of profenofos in zebrafish reached an equilibrium in 12 hrs at five-hundredth and one-hundredth concentration of 96-hrs LC$_{50}$(0.004 mgA and 0.02 mg/1). The average BCF values of profenofos were 141.9(n=7) and 111.3(n=7) at five-hundredth and one-hundredth concentration of 96-hrs LC$_{50}$(0.004 mg/l and 0.02 mg/1) after 12~168 hrs. Depuration rate constants of profenofos were 0.09 hr$^{-1}$ and 0.10 hr$^{-1}$, half-life of profenofos were 7.70 and 6.93 at five-hundredth and one-hundredth concentration of 96-hrs LC50(0.004 mg/l and 0.02 mg/1), respectively. The concentrations of profenofos in zebrafish at five-hundredth and one-hundredth concentration of 96-hrs LC$_{50}$ decreased agter 8(0.18 $\mu\textrm{g}$/g) and 12 hrs (0.19 $\mu\textrm{g}$/g). The LC$_{50}$ value in zebrafish showed that acute toxicity of profenofos was higher than that of phosphamidon. The BCF values of profenofos were 100 times higher than those of phosphamidon, and depuration rate of phosphamidon was two times faster than that of profenofos.

Acute Toxicity of Cement on Mortality of Pond Smelt (Hypomesus olidus) (빙어(Hypomesus olidus)의 폐사에 미치는 시멘트의 급성독성)

  • The effect of cement on survival, LC/sub 50/, safe concentration (SC) and application factors of pollutant (AF) for pond smelt, Hypomesus olidus were investigated for 96 hours. Mean body length of pond smelt used in this experiment was 8.6±1.2 cm. Cement concentrations established in this experiment were 0 (control group), 10, 25, 50, 100, 250, 500, and 1,000 ppm, respectively. All fishes were dead in concentration more than 500 ppm cement within 4 hours. The LC/sub 50/ to cement toxicity was 123.03 ppm for 48h, 91.20 ppm for 72 h and 58.88 ppm for 96h. The value of SC and AF to cement toxicity for this species were 2.64∼9.14 ppm and 0.045∼0.155, respectively.

Insecticidal Activity and Effect on Biological Characteristic of 16 Insecticides Against Phthorimaea Operculella (Lepidoptera: Gelechiidae) (감자뿔나방에 대한 16종 살충제의 살충활성과 생물적 특성에 미치는 영향)

  • Susceptibility of each developmental stage of Phthorimaea operculella (Lepidoptera: Gelechiidae) were investigated using 16 insecticides which are available in the market in Korea. For the eggs and pupae, only spinosad showed a 71.1% inhibition rate for egg hatchability and a 66.7% inhibition rate for emergence. For the 3rd instar larvae, the feeding toxicities were over 90% for fenitrothion ($LC_{50}$ 336.6 ppm), esfenvalerate ($LC_{50}$ 8.6 ppm), ethofenprox ($LC_{50}$ 35.7 ppm), and emamectin benzoate ($LC_{50}$ 0.05 ppm). Furthermore, the contact toxicities were over 90% for esfenvalerate ($LC_{50}$ 0.87 ppm), ethofenprox ($LC_{50}$ 16.5 ppm), emamectin benzoate ($LC_{50}$ 0.53 ppm), and spinosad ($LC_{50}$ 2.48 ppm) at the recommended concentrations. Deltamethrin and spinosad yielded 100% mortality for adult P. operculella 48 h after treatment. The adult female fecundity was inhibited by deltamethrin, esfenvalerate, emamectin benzoate, spinosad and dinotefuran, which were significantly different from the control. The adult longevities (7.3-8.3 days) were reduced by approximately 1-2 days compared with the control (9.3 day). The emamectin benzoate maintained 100% insecticidal activity 14 days after treatment and ethofenprox maintained over 90% activity 7 days after treatment.

  • On protein equivalence base, fishmeal (FM) was replaced by lysine cell mass (LCM) in selected different diets in Korean rockfish, Sebastes schlegeli (Hilgendorf) Eight experimental diets were formulated to contain 100% FM (LC $M_{0}$), 90% FM+10% LCM (LC $M_{10}$),80% FM+20% LCM (LC $M_{20}$), 70% FM+30% LCM (LC $M_{30}$), 60% FM+40% LCM (LC $M_{40}$ ), 70% FH+30% LCM+lysine (LC $M_{+Lys}$), 60% FM+40% LCM+lysine (LC $M_{40+Lys}$), and 50% FM+50% LCM+lysine (LC $M_{50+Lys}$). Experimental individuals of the fish (12.6 g) were randomly fed on one of the experimental diets. After 6 weeks of feeding trial, weight gain (WG) and feed efficiency (FE) of fish fed LC $M_{0}$ diet was significantly (P〈0.05) higher than those of fish fed LC $M_{20}$, LC $M_{30}$, LC $M_{40}$ , LC $M_{30+Lys}$, LC $M_{40+Lys}$, and LC $M_{50+Lys}$ diets, however, there was no significant difference in WG of fish fed LC $M_{0}$ and LC $M_{10}$ diets. Supplementation of lysine has no effect on WG. There was no significant difference in condition factor (CF) of fish fed LC $M_{0}$, LC $M_{10}$ and LC $M_{20}$ diets. Hemoglobin (Hb) of fish fed LC $M_{0}$, LC $M_{10}$, LC $M_{20}$, LC $M_{30}$, LC $M_{40}$ , LC $M_{30+Lys}$, and LC $M_{40+Lys}$, diets were not significantly different from each other. No significant differences were observed in hematocrit (PCV) and hepatosomatic index (HSI) among all dietary treatments. Apparent digestibility of dry matter (ADM) and protein (ADP) of diets significantly decreased with increase in dietary LCM level, though there was no difference in ADM and ADP between LC $M_{0}$ and LC $M_{10}$. Diamondback moth (Plutella xylostella L.) resistance to organophosphorus and carbamate insecticides in Kangwon alpine vegetable croplands (강원도 고랭지대 배추경작지 배추좀나방(Plutella xylostella L.)의 유기인계 및 카바메이트계 살충제에 대한 저항성 발달)

  • Diamondback moth (Plutella xylostella L.) recently became a single worst insect which is not controlled effectively by organophosphorus and carbamate insecticides in Kangwon alpine croplands. The objective of this study was to determine if diamondback moth has developed a resistance to organophosphorus and carbamate insecticides. Resistance of diamondback moth, collected at Keichon, Jangpyong Taebaek, Chunchon, and Hongchon, was determined by the concentration required to kill fifty percent of population, $LC_{50}$. Their response of resistance varied to insecticides and locations: Taebaek populations were 35 and 70 times more resistant to chlorpyrifos and fenitrothion, respectively, than susceptible(S) population. Hongchon populations were 94 and 254 times more resistant to chlorpyrifos and fenitrothion, respectively, than S population. In addition, Chunchon populations were 37 and 19 times more resistant to profenofos and benfuracarb, respectively, than S population. However, the field populations did not differ in resistance to diazinon, phenthoate, flupyrazofos, carbofuran, and furathiocarb. This study show that field populations of diamondback moth found in Kangwon alpine vegetable croplands have developed a resistance and/or multiple resistance to some insecticides, implying that farmers are losing organophosphorus and carbamate insecticide options for selective control in vegetable crops.

  • The lethal concentration(LC50) of several micropollutants for three species of the fish Paralichthys olivaceus, Mugil cephalus and Sebastes schlegeli were determined by acute toxicity tests. For the determination of definitive test concentrations, the fish were exposed to three test material concentrations spaced at order-of-magnitude intervals based on a logarithmic ratio in range finding tests. LC50 was determined by five concentrations of test material in a geometric progression by means of range finding tests. The 96hr-LC50 values(mg/l) were estimated by the graphical interpolation of probability-logarithm transformations. These indicated that the order of sensitivities to three kind of micropollutants was Mugil cephalus > Paralichthys olivaceus > Sebastes schlegeli.

A Study on the Toxicity of Pb and Cu Compound in Carassius auratus(goldfish) (납 및 구리화합물이 Carassius auratus(goldfish)에 미치는 독성에 관한 연구)

  • In order to investigate acute toxicity and bioconcentration of heavy metals for a freshwater fish, the fish used in this experiment was goldfish, Carassius auratus. Each ten goldfish was accommonidated in a water and was treated with different concentration of Pb and Cu compound. The 24 $hr-LC_{50}$ was obtained by plotting on the log-normal distribution graph. Furthermore, the combined effect of Pb and Cu was also investigated the fish was treated with Pb or Cu compound only, and Pb and Cu compound together, respectively. These results were summarized as follows: 1. The 24 $hr-LC_{50}'s$ of Pb and Cu were 7.48 mg/l and 0.666 mg/l, respectively. 2. When single or/and combined treatment with Pb(7.0 mg/l) or/and Cu(0.6 mg/l) to Carassius auratus for 24 hours were performed, there was significant difference between the single or/and the combined treatment in their bioaccumulated Cu concentrations. Cu concentrations in goldfish were higher in the combined treatment than in the single treatment. 3. When Carassius auratus was exposed to 0.748 mg/l (1/10 of 24 $hr-LC_{50}$) and 1.496 mg/l of Pb (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 79.14 and 100.11 for Pb, respectively. The BCF of Pb was obtained as a linearity according to the concentration and exposure time as follows log BCF=1.014 log $P\cdot T$+1.011 ($r^2$=0.9041) where, P: pollutant concentration(mg/l) T: exposure time(day) 4. When Carassius auratus was pxposed to 0.0666 mg/l (1/10 of 24 $hr-LC_{50}$) and 0.1332 mg/l of Cu (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 55.42 and 63.24 for Cu respectively.
