  • A nuclear polyhedrosis virus (NPV) of the tobacco cutworm, Spodoptera litura would be a promisible agent for the control of the insect. To develop a viral insecticide using S. litura NPV, effect of spray on soybean leaves, temperature, storage, an sunlight on the pathogenicity of the virus were studies as follows: Median lethal concentration ($LC_{50}$) of the virus sprayed on the leaves against the third and the fifth instar larvae were $1.301\times10^{4 PIBS}/ml$ and $1.087\times10^{5 PIBS}/ml$, respectively. On the concentration of $1.0\times10^{5 PIBS}/ml$, median lethal times ($LT_{50}$) were 7.3 days for the 3rd and 8.9 days for the 5th instar larvae. Stability of S. litura NPV was quickly decreased at the higher temperate than $60^{\circ}C$ and at the longer exposure to the higher temperature. Storage of the virus at $-20^{\circ}C$ was kept higher pathogenicity than $4^{\circ}C$ and $25^{\circ}C$. Viral activity was maintained more than 10 days in the sprayed-under leaves, but decreased at 3 day after spray in th sprayed-on the leaf surface when exposed the virus to sunlight.

  • The aim of this study was to identify the effects of dithiopyr (DTP), a herbicide, on behavior in zebrafish. The toxicity of DTP has rarely been investigated in fish. In the present study, zebrafish were exposed to different concentrations of DTP in the range of 10-20 μM for 48 h in a test container, in order to measure the value of median lethal concentrations (LC50). Behavioral experiments were performed, including the novel tank test (NTT) and the open field test (OFT), to assess stress responses or locomotion. After exposure to the DTP solution at a sublethal concentration of 2.5–10 μM for 6 min, the behavior of the zebrafish was observed for 6 min. In the acute toxicity test, the LC50 value of DTP showed as 14.49 μM in the zebrafish. The NTT showed that the duration of immobility and the velocity were significantly increased by exposure at a concentration of 5 μM of DTP, compared with a control group (p<0.05). However, compared with the control group, DTP significantly decreased the distance moved and the frequency at the top of the tank, and significantly increased the turn angle and duration at the bottom, in a concentration-dependent manner (p<0.05). In addition, in the OFT, exposure to DTP significantly decreased the distance moved and velocity compared with the control group (p<0.05). Exposure to DTP also significantly increased the duration of immobility, the turn angle, and the meandering movement, in a concentration-dependent manner (p<0.05). Further, exposure to DTP at a low concentration elevated whole-body cortisol levels in the zebrafish. The results of this study thus suggest that DTP induces a toxic response and negative effects on behavior and the endocrine system in zebrafish.

  • Olive flounder (Paralichthys olivaceus) (Weight 110.9±17.1 g, length 22.3±1.2 cm) were exposed to waterborne nitrite at 0, 30, 60, 120, 240, 480 and 960 mg NO2-/L according to water temperature at 20℃ and 25℃ for 96 hours. The lethal concentration 50 (LC50) of olive flounder, P. olivaceus exposed to waterborne nitrite was 513.87 mg NO2-/L at 20℃ and 208.35 mg NO2-/L at 25℃, which means a significant difference in LC50 by the water temperature. Hemoglobin and hematocrit were significantly decreased by waterborne nitrite exposure. The inorganic component, plasma calcium, was significantly decreased, and the organic components such as plasma glucose and cholesterol were significantly decreased showing a similar tendency with calcium. In enzymatic components, the AST and ALP were also significantly decreased by nitrite exposure. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of P. olivaceus, and the effect of exposure to nitrite had a significant effect on nitrite toxicity depending on the water temperature.

  • Crucian carp, Carassius carassius (Weight 42.4 ± 9.0 g, Length 15.0 ± 1.0 cm) were exposed to waterborne zinc at 0, 5, 10, 20, 40 and 80 mg Zn2+/l for 96 hours. The lethal concentration 50 (LC50) at 96 hours of crucian carp, C. carassius exposed to waterborne zinc was 51.58 mg Zn2+/l. In hematological parameters, the RBC count was significantly decreased in the concentration of 40 mg Zn2+/l at 48 hours, whereas the hematocrit was significantly increased by zinc exposure. The MCV (mean corpuscular volume) (µl) and MCH (mean corpuscular hemoglobin) (pg) were significantly increased in the concentration of 40 mg Zn2+/l at 48 hours. The plasma components such as calcium, magnesium, glucose, cholesterol, total protein and ALT (Alanine aminotransminase) were significantly changed by zinc exposure. The results of this study suggest that the zinc exposure to C. carassius induced the significant physiological changes in the hematological parameters and plasma components as toxicity.

  • Surfactants are organic compounds that have both hydrophilic and non-polar parts in one molecule, classified as non-ion, anion, cation, and amphoteric surfactants according to the charge of hydrophilic parts in aqueous state. A trace amounts may remain when vegetables and fruits are washed using type1 detergent (Vegetable and fruit detergent), and there is a possibility of exposure to the human body through ingestion. This study developed the simultaneous analysis method for 5 surfactants with LC-MS/MS for analysis of detergent residues after washing vegetables and fruits with detergent. The mobile phase used distilled water and acetonitrile containing 50 mM ammonium formate and 0.1% formic acid and was analyzed using a gradient method using XBridge BEH C8 column. The accuracy of the established method was 83.9-112.1%, and the precision was less than 20%. The detection limit was 7.0 (SLS) to 29.0 (SLES-N3) ㎍/L, and the correlation coefficient (r2) of calibration line regression was greater than 0.99, it is considered suitable for the analysis of trace amounts of surfactant components remaining in vegetables and fruits.

  • A specific analytical method able to identify and quantify traces of six glucocorticoids residues in eggs were developed. The extraction and clean-up parameters for simultaneous analysis were evaluated and HPLC and spectrometric conditions were also established. For determination of glucocorticoids, 5 g of egg was transferred into a test tube, adjusted pH 5.2 with acetate buffer and was $\beta$-glucuronidase/arylsulfatase from Helix pomatia added. The mixture was centrifuged and supernatant was extracted twice with 20 mL n-hexane. The extraction was performed with HLB cartridge using methanol, followed by clean-up with silica cartridge using methanol/ethyl acetate (4/6, v/v). The analytes were determined by HPLC/ESI-MS/MS operating in the negative ion mode. Validation studies with fortified egg samples for established method were performed. The result of method validation gave good efficiency, linearity, accuracy and precision. The correlation coefficients ($r^2$) of the calibration curves appeared to be higher than 0.99 in egg, indicating excellent linearity. LOD was ranged 0.09 to $0.17{\mu}g/kg$, and recoveries for most compounds were in the range of 55.7-69.8%. This method can be used to determine ${\mu}g/kg$ levels of glucocorticoids in eggs.

  • This study aimed to investigate the acrylamide content in frozen food products after cooking. Twenty samples of bread (Group 1) and 30 samples of processed tuberous and corn vegetable products (Group 2) were selected. Acrylamide levels were quantified using liquid chromatography tandem-mass spectrometry (LC-MS/MS). The frozen food samples were heated using the air fryer cooking method according to the product packaging and were compared to ready-to-eat French fries (Group 3). The results showed that the acrylamide content was the highest in group 3, followed by that in group 2 and group 1. The acrylamide content of all the samples was found to be within the domestic recommended standard of 1 mg/kg. However, when the samples that exceeded EU benchmark level (0.5 mg/kg) were selected and cooked using the deep-fat frying method according to the product packaging, one of them showed the acrylamide content of 1.83 mg/kg, which exceeded the domestic recommended standard. The present study highlights the need for continued evaluation and management to reduce acrylamide contents in frozen foods, as increasing domestic exposure to acrylamide is concerning.

  • To determine the toxic effects of the mirror carp, Cyprinus carpio nudus on waterborne nitric acid exposure, they were exposed to waterborne nitric acid at 0, 4, 20, 100, 500, and 2,500 mg NO3-/L for 96 h. The semi-lethal concentration (LC50) of the scented fish by 96 hours of acute exposure to nitric acid was 1,433.54 mg NO3-/L. The physiological changes of waterborne nitric acid exposure on mirror carp were evaluated through hematologic properties, and RBC count showed a significantly decreased (p < 0.05). The change in the ion control ability of the mirror carp according to exposure to waterborne nitric acid was evaluated through the plasma inorganic component, and calcium and magnesium were significantly decreased (p < 0.05) in the plasma inorganic component. The health and stress status of the mirror carp due to exposure to waterborne nitric acid were evaluated through plasma organic ingredients, and the plasma organic component glucose showed a significantly increase (p < 0.05). Changes in liver damage and enzyme activity due to waterborne nitric acid exposure were evaluated through plasma enzyme components, the enzymatic activities of plasma AST, ALT, and ALP were significantly decreased (p < 0.05) in high levels of waterborne nitric acid exposure. The results of the present study suggest that waterborne nitric acid exposure to C. carpio nudus can toxicly affect survival, hematologic properties, and plasma components.

  • Marine ecotoxicological standard method using fish larvae was established with the standard test species of Oryzias latipes(Japanese Medaka) and Paralichthys olivaceus(flounder) and with the 7 day $LC_{50}$ as endpoint. Test method referred to the USEPA(1994) with the replacement of test species found in the Korean water. Standard test species were selected in terms of the species supply and ecological importance in Korean waters. Japanese medaka can be reared with small tanks in the lab and has wide tolerance on salinity, and flounder eggs can be easily obtained from commercial fish hatcheries. General conditions for larval fish toxicity test are as follows. The possible salinity ranges for toxicity test were $0{\sim}35\;psu$ for medaka and >20 psu for flounder. Test type was designated as static non-renewal test if the dissolved oxygen in the test chamber does not fall below 4.0 mg/L. Ages of test species were selected as 7 days after hatched for medaka(about 5 mm TL) and 25 days for flounder(about 10 mm TL) because of the low natural mortality after these periods. Test can be accepted when the survival rates are over 80% in control. Also, species sensitivity on standard reference materials(copper, cadmium or zinc) must be provided with the toxicity test results.

  • To improve stability of recombinant DNA pLC1 encoding endoglucanase gene and pGL1 encoding $\beta $-glucosidase gene, DNA fragments of genes coding endoglucanase and $\beta $-glucosidase were cloned within the recA gene on a pDR1453, and the pDRE10 and pDRG20 of recombinant plasmids were integrated into the recA gene on the E. coli 1100 chromosomal DNAs. The stability of inheritance was completely maintained in E. coli 1100; Transformants E. coli 1100/pDREIO and pDRG20 were expressed well by recA promoter and increased endoglucanase and $\beta $-glucosidase activities. This method can be used as a model to improve the stability of recombinant plasmid in large scale culture.
