  • A simple GC method with a FID detector was developed in order to determine two main compounds (${\beta}$-sitosterol and lupenone) for Adenophorae Radix. ${\beta}$-Sitosterol and lupenone were analyzed by the gradient thermal ramping method. Nitrogen was used as the carrier gas at 108 kPa. The flow rate of gas was 2.0 mL/min; $2{\mu}L$ of filtered sample was injected at a split ratio of 1 : 80. This method was fully validated with respect to linearity, precision, accuracy and robustness. Further, this GC-FID method was applied successfully in order to quantify two compounds in an Adenophorae Radix extract. The GC analytical method for classification analysis was performed by repeated analysis of 59 reference samples in order to differentiate between Adenophora triphylla var. japonica Hara and 14 Codonopsis lanceolata. The results indicate that the GC-FID method is suitable and reliable for the quality evaluation of Adenophorae Radix.

  • Soy sauce was fermented with the addition of mountain edible herbs, Ligularia fischeri, Codonopsis lanceolata and Symphytum officinale. In general, the total nitrogen content of soy sauce was increased with the increment of the amount of added mountain herbs. The mineral contents of calcium and potassium in the soy sauce after four months of aging at 20% substitution of Codonopsis lanceolata were increased by 1.3 and 1.5 times, respectively. With 10% substitution of mountain herb mixtures, the contents of tyrosine and arginine were increased by about 2 times as compared to the control. In the Rec assay system, antimutagenic effect of soy sauce with 10 and 20% substitution of Codonopsis lanceolata was higher than other samples. The results of sensory evaluation revealed that overall acceptability of soy sauce with 7% substitution of Codonopsis landeolata and 5% of mountain herb mixture exceeded other groups of samples.

  • During the life cycle, plants have to suffer from various environmental stresses. A common element in response to many environmental stresses is cellular dehydration. Dehydrins are a family of proteins commonly induced by environmental stresses associated with low temperature or dehydration and during seed maturation drying. For the study in the defense mechanism against various stresses, a cDNA clone encoding a dehydrin gene was isolated from a cDNA library prepared from tab root mRNAs of Codonopsis lanceolata. The cDNA, designated ClDhn1, is 893 nucleotides long and has an open reading frame of 480 bp with a deduced amino acid sequence of 159 residues. The ClDhn1 amino acid sequence is highly hydrophilic and possesses two conserved repeats of characterized lysinerich Ksegment (KIKEKLPG), and a 7serine residue stretch prior to the first lysinerich repeat that is common to many dehydrins. The DEYGNP conserved motif is, however, modified in the sequence of ClDhn1 gene. The deduced amino acid sequence of ClDhn1 was compared with other plant dehydrinls and showed high homology with Solanum commersonii

  • The objective of this study was to determine whether Codonopsis lanceolata or Platycodon grandiflorum ethyl acetate fraction (CLEA or PGEA) protect cells against sodium nitroprusside (SNP)-induced oxidative stress via the expression of various antioxidant systems. The HepG2 cells exposed for 24 hr to 0.5 mM SNP showed a reduction in the cell viability by an MTT assay. Pretreatment with CLEA and PGEA resulted in an inhibition of SNP-induced cell death. In addition, the effects of CLEA and PGEA on the expression of antioxidant systems via RT-PCR analyses was assessed. The levels of catalase (CAT), glucose-6-phosphate dehydrogenase (G6PD) and metallothionein (MT)-1A mRNA were increased after 24 hr of CLEA exposure. The levels of Mn superoxide dismutase CAT, G6PD, MT-1A, and MT-2A mRNA were increased after PGEA treatment. In conclusion, CLEA and PGEA exert indirect antioxidant effects, perhaps via the induction of a variety of antioxidant systems which, may protect cells against oxidative stress.
