Distributions of Nicotine and Toluene in Teenagers' Urine (청소년의 뇨 중에서 nicotine과 톨루엔의 분포에 관한 연구)

  • We identified nicotine, cotinine and toluene in high school volunteer's urine by using GC/NPD, GC/FID and GC/MS. To analyze of nicotine and cotinine, urine samples were extracted with diethylether and centrifuged on a benchtop centrifuge for 5 min. The upper organic layer was injected into a GC. The distributions of nicotine and cotinine were $4{\sim}630{\mu}g/L$ and $63{\sim}1,602{\mu}g/L$ in smoking-group, respectively. To analyze of toluene, head space vial was filled with 2mL sodium citrate solution and 1mL of urine. The vial was warmed in a water bath at $55^{\circ}C$ for 20min, and then $250{\mu}L$ of head space air was injected into a GC. The result show that toluene was not detected in all of the volunteers' samples. However, the range of toluene was 0.1~28.0mg/L in glue sniffer's urine samples(NISI data).

Antioxidative Activities and Tyrosinase Inhibition of Water Extracts from Ailanthus altissima (가죽나무(Ailanthus altissima) 물 추출물의 항산화 활성과 Tyrosinase 저해)

  • Water extracts from root, stem and leaf of Ailanthus altissima were utilized to determine antioxidant properties such as electron donating ability (EDA), nitrite scavenging ability, superoxide dismutase (SOD)-like activity, and the inhibitory activities of xanthine oxidase (XO) and tyrosinase. The EDA of root extract was the highest as 77.33% at 0.5 mg/mL concentration and that of stem extract was 70.01% at 1.0 mg/mL. The nitrite scavenging ability of leaf extract revealed the highest effect as 95.18% at pH 1.2, 1.0 mg/mL while those of stem and root extracts were 55.17% and 33.33%, respectively. The leaf extract showed the highest SOD like activity as 26.77% at 1.0 mg/mL, the measurement of root extract was 3.82% and that of stem extract was not effective. All kinds of extracts had strong inhibitory activities on XO of over 92% at 1.0 mg/mL. The highest activity on tyrosinase inhibition was obtained from leaf extract of 16.33% at 2.0 mg/mL. The results indicated that among the three extracts, the leaf extract has a strong and extensive antioxidant activity.

Bacterial Contamination in Disposable Wet Wipes from General Restaurants (대중음식점 일회용 물티슈의 세균 오염도 조사)

  • This study, funded by Jeju Special Self-Governing Province, evaluated the bacterial contamination in disposable wet wipes used in restaurants, coffee shops, and bakery shops. The materials used are 55 disposable wet wipes collected from general restaurants. The number of average bacteria found was $4.21{\times}10^3CFU/mL$ in 50 disposable wet wipes; in the other five disposable wet wipes, no bacterial contamination was found. The highest number of bacteria was measured as $16.67{\times}10^3CFU/mL$. Total bacterial count of separated are 71 strains. Among the 71 identified strains, 15 were Staphylococcus aureus and 3 were Pseudomonas aeruginosa. This can be an important problem. This study measured the level of bacterial contamination and identified the strains isolated in wet wipes. This study may be important as it provides the basis for establishing sanitation standars for wet wipes.

Serum anti-M$\ddot{u}$llerian hormone levels as a predictor of the ovarian response and IVF outcomes

  • Objective: The aim of this study was to investigate whether anti-M$\ddot{u}$llerian hormone (AMH) levels could be predict ovarian poor/hyper response and IVF cycle outcome. Methods: Between May 2010 and January 2011, serum AMH levels were evaluated with retrospective analysis. Three hundred seventy infertile women undergoing 461 IVF cycles between the ages of 20 and 42 were studied. We defined the poor response as the number of oocytes retrieved was equal or less than 3, and the hyper response as more than 25 oocytes retrieved. Serum AMH was measured by commercial enzymelinked immunoassay. Results: The number of oocytes retrieved was more correlated with the serum AMH level (r=0.781, $p$ <0.01) than serum FSH (r=-0.412, $p$ <0.01). The cut-off value of serum AMH levels for poor response was 1.05 ng/mL (receiver operating characteristic [ROC] curves/area under the curve [AUC], $ROC_{AUC}$=0.85, sensitivity 74%, specificity 87%). Hyper response cut-off value was 3.55 ng/mL $ROC_{AUC}$=0.91, sensitivity 94%, specificity 81%). When the study group was divided according to the serum AMH levels (low: <1.05 ng/mL, middle: 1.05 ng/mL - 3.55 ng/mL, high: >3.55 ng/mL), the groups showed no statistical differences in mature oocyte rates (71.6% vs. 76.5% vs. 74.8%) or fertilization rates (76.9% vs. 76.6% vs. 73.8%), but showed significant differences in clinical pregnancy rates (21.7% vs. 24.1% vs. 40.8%, $p$=0.017). Conclusion: The serum AMH level can be used to predict the number of oocytes retrieved in patients, distinguishing poor and high responders.

Purification and Characterization of Carboxypeptidase B from Wild Salmon (Salmo Salar) Eggs (연어알중의 Carboxypeptidase B 분리정제 및 그 특성에 관한 연구)

  • Carboxypeptidase B from Salmo Salar eggs was purified by CM-cellulose, 0.5 ammonium sulfate saturation, DEAE-cellulose, and Sephadex G-75 gel filtration and its enzymatic properties were investigated. Optimum temperature was 55$^{\circ}C$, pH optima were 4.0 and 7.0 at 37$^{\circ}C$, and the enzyme was stable at pH 2.0∼3.0 and 5.5∼7.0 for 1.5h. This enzyme showed substrate specificity hydrolyzing the peptide bond of glycyl-L-arginine. Its K$_m$ values was 0.21mM, and the enzyme activity was stimulated by Cu$^{2+}$ and Fe$^{3+}$, while inhibited by Zn$^{2+}$. The lysine was found to be competitive inhibitor and its K$_i$ value was determined to be 4.3mM. Molecular weight of this enzyme was determined to be 36,400 daltons by SDS-PAGE and the enzyme was monomeric protein composed of 19 kinds of amino acid residues.

Anti-Oxidative and Anti-Proliferative Effect of 70% Ethanol Extracts from Green Pepper (Capsicum annuum L. cv. DangZo) (당조고추 70% 에탄올추출물의 항산화 및 항암활성측정)

  • To evaluate the anti-oxidant and anti-tumor potential of the green pepper (Capsicum annuum L. cv. DangZo), total polyphenol content, radical scavenging activities and anti-tumor properties were measured. The total polyphenol content of the 70% ethanol extracts from green pepper (Capsicum annuum L. cv. DangZo) was 30.29 mg gallic acid equivalent/g extract. The DPPH radical and hydroxyl radical scavenging activities of 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 2.87 and 10.55, respectively. For ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity, 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 35.67% and 58.41% respectively. The green pepper (Capsicum annuum L. cv. DangZo) demonstrated greater capability in terms of anti-neoplastic activity vis-a-vis colon cancer cell lines when compared to other cancer cell lines.s. er (Capsicum annuum L. cv. DangZo) higher activities of anticancer activities on colon cancer cell lines compared to other cancer cell lines.

Optimal Dimension of PE Film Bag According to Fruit Size in MAP Storage of 'Fuyu' Persimmon Fruit ('부유' 단감 MAP 저장에서 과실 크기에 따른 PE 필름 봉지의 적정 규격)

  • Model for optimally dimensioned PE film bag was developed according to fruit size to maintain best quality of 'Fuyu' persimmon in modified atmosphere packaging (MAP) storage based on relationship between quality and oxygen and carbon dioxide in PE film bag. Harvested persimmons were graded into five sizes, and average weights were LL:261, L:217, M:188, S:168, and SS:154 g. Five fruit units of each grade were optimized in five PE film bag sizes of $150{\times}376,\;140{\times}357,\;130{\times}344,\;130{\times}333,\;and\;120{\times}3l8\;mm$. To minimize quality deterioration such as softening and discoloration, optimal oxygen and carbon dioxide concentrations in PE film bag were 0.5-1.0 and 6.0-8.0%, respectively, and optimal thickness of PE film bag according to fruit size were LL:45, L:50, M:55, S:60, and $SS:65\;{\mu}m$. For all fruit sizes, model for PE film bag area $(mm^2)$ was good quadratic simple equation by fruit weight (g): $Y=-4055.707+627.993X_1-0.701{X_1}^2$. Model far optimal oxygen and carbon dioxide (Y) concentration in PE film bag was suited to linear multiple equation by fruit weight $(X_1,\;g)$ and PE film thickness $(X_2,\;{\mu}m)$. Equations for oxygen and carbon dioxide concentrations (%) were $Y=5.798-0.0109X_l-0.0491X_2\;and\;Y=-2.427+0.01927X_l +0.09646X_2$, respectively.

Manufacturing and Characteristics of Fruit Wine from Acanthapanax sessiliflorus (오가피 열매 발효주의 제조 및 특성)

  • Fruit wine from Acanthopanax sessiliflorus(A. sessiliflorus) including many pharmacological components was manufactured and its functional properties were investigated. The fruit part of A. sessiliflorus contained 75.74${\pm}$0.49%(w/w) moisture, 12.51${\pm}$1.23%(w/w) crude protein, 4.20${\pm}$0.51.%(w/w) crude fat and 5.21${\pm}$1.64%(w/w) crude ash. Minerals of fruit were potassium(12.94${\pm}$0.08 mg/g), calcium(1.53${\pm}$0.06 mg/g) and magnesium(1.12${\pm}$0.05 mg/g). Initial soluble solid and fermentation temperature were 24-30$^{\circ}$Brix and 20${^{\circ}C}$ for manufacturing fruit wine from A. sessiliflorus. When initial soluble solid of a must was adjusted to more than 30$^{\circ}$Brix, ethanol production was suppressed slightly. The polyphenol content of the fruit wine fermented at 20${^{\circ}C}$(125.24${\pm}$1.86 mg/mL) was higher than those at 25${^{\circ}C}$(99.69${\pm}$2.11 mg/mL) and 30${^{\circ}C}$(95.55${\pm}$1.54 mg/mL). Electron donating activities of wines fermented at 20, 25 and 30${^{\circ}C}$ were 85.9${\pm}$2.3, 55.7${\pm}$2.5 and 55.2${\pm}$3.4%, respectively. The content of eleutheroside B increased up to 146.58${\pm}$4.10 $\mu$g/mL during fermentation. There was no significant effect of fermentation temperature on eleutheroside B content. The fruit part of A. sessiliflorus can be used as a valuable resource for development of nutraceutical foods.
