Mechanism of Intestinal Transport of an Organic Cation, Tributylmethylammonium in Caco-2 Cell Monolayers

  • Many quaternary ammonium salts are incompletely absorbed after their oral administration and may also be actively secreted into the intestine. However, the underlying mechanism(s) that control the transport of these cations across the intestinal epithelium is not well understood. In this study, the mechanism of absorption of quaternary ammonium salts was investigated using Caco-2 cell monolayers, a human colon carcinoma cell line. Tributylmethylammonium (TBuMA) was used as a model quaternary ammonium salts. When TBuMA was administrated at a dose of 13.3 imole/kg via iv and oral routes, the AUC values were $783.7{\pm}43.6\;and\;249.1{\pm}28.0{\mu}mole\;min/L$ for iv and oral administration, indicating a lower oral bioavailability of TBuMA $(35.6\%)$. The apparent permeability across Caco-2 monolayers from the basal to the apical side was 1.3 times (p<0.05) greater than that from the apical to the basal side, indicating a net secretion of TBuMA in the intestine. This secretion appeared to be responsible for the low oral bioavailability of the compound, probably mediated by p-gp (p-glycoprotein) located in the apical membrane. In addition, the uptake of TBuMA by the apical membrane showed a $Na^+$ dependency. Thus, TBuMA appears to absorbed via a $Na^+$ dependent carrier and is then secreted via p-gp related carriers.

과메기 통조림의 제조 및 특성(I) - 조미과메기 통조림의 제조 및 특성을 중심으로 - (Preparation and Characterization of Canned Kwamaegi(I) - Preparation and Characterization of Canned Seasoned Kwamaegi -)

  • This study was investigated to obtain basic data which can be applied to processing of canned seasoned kwamaegi. Commercial Kwamaegi was cut into $2{\times}3cm$ lengths, filled 90 g into can (301-3) and added with 60 g water before precooking for 10 min. at $100^{\circ}C$. The precooked Kwamaegi was packed into the can, and added with 60 g seasoning sauces, which was prepared by mixing soy sauce 23%, monosodium glutamate (MSG) 2%, sorbitol 2%, sesame oil 1%, vinegar 2%, starch syrup 17%, sake 5%, water 48%. The cans were sealed using a vacuum seamer and then sterilized for various Fo values (Fo 8~12 min.) in a steam system retort at $121^{\circ}C$. The factors such as pH, VBN, amino-N, total amino acid, free amino acid, color value (L, a, b), texture profile, TBA value, mineral, sensory evaluation and viable bacterial count of the canned seasoned Kwamaegi produced with various sterilization condition(Fo 8~12 min.) were measured. There was no remarkable difference between sterilization conditions and sensual characteristics. The results showed that the product sterilized at Fo 8 min. was the most desirable because this condition is most economical.

여러 자생식물의 내염성 정도 구명 (Salt Tolerance of Various Native Plants under Salt Stress)

  • 본 실험에서는 염토지대에 조경용 식재로 사용이 가능한지 알아보기 위해 여러 과에 속하는 자생식물의 염 스트레스에 대한 내염성 정도를 조사하였다. NaCl의 처리에 따른 식물의 생육반응과 이온흡수 특성도 파악하여, 여러 식물에 대한 내성 정도를 구명하고자 하였다. 실험재료로 노랑꽃창포, 밀사초, 여우꼬리사초, 줄사초, 홍노줄사초, 애기부들, 바위 및 땅채송화 등을 이용하였다. 염농도는 0, 100, 200, 300mM로 처리해 주었다. 처리 4주 후 지상부 및 뿌리의 생체중 건물중, 엽내 무기이온 함량 등을 조사하였다. 줄사초, 왕밀사초, 여우꼬리사초, 노랑꽃창포 등은 NaCl 200mM의 고농도에서도 잘 견디는 초종으로 판단되었다. 홍노줄사초, 애기부들 등은 지하부 생육이 NaCl 100mM의 농도에서 저하되어, 지상부다 뿌리가 민감하게 반응하는 것으로 나타났다. 홍노줄사초는 K의 흡수량이 감소되었고, Na/K의 비율은 300mM의 농도에서 3으로 다른 종보다 높은 경향을 보였다. 애기부들은 NaCl의 농도가 증가됨에 따라 K, Ca, Mg의 흡수량이 일시 증가하다가 200mM의 농도부터 감소하는 경향을 보였고, Na/K의 비율은 300mM의 농도에서 0.8로 조사되었다. 땅채송화 및 바위채송화 등도 NaCl 100~200mM의 범위에서 지상부 및 지하부의 생육이 영향을 받는 것으로 나타났다. 바위채송화는 NaCl의 농도가 증가됨에 따라 K, Ca, Mg의 흡수량이 일시 증가하다가 200mM의 농도부터 감소하는 경향을 보였고, 땅채송화는 염농도에 따라 흡수량이 영향을 받지 않는 것으로 나타났다. Na/K의 비율은 300mM의 농도에서 모두 1로 조사되었다. 그러므로, 여우꼬리사초, 왕밀사초, 줄사초, 노랑꽃창포 등은 식물의 생육 및 이온흡수 특성을 고려할 때 NaCl 200mM의 고농도에서도 잘 견디는 초종으로 판단되었고, 특히 사초과 식물들의 내염성이 검증되어 앞으로 많이 활용될 수 있으리라 사료되었다. 홍노줄사초, 애기부들, 바위채송화, 땅채송화 등은 NaCl 100mM의 농도에서도 뿌리의 생육이 저하되었으며, 염처리에 따른 잎의 이온흡수특성은 뚜렷한 경향을 보이지 않았다.

탄저병(Colletotrichum orbiculare)에 대한 박과작물의 저항성 (Resistance Degree of Cucurbits Cultivars to Colletotrichum orbiculare)

  • Colletotrichum orbiculare에 의한 탄저병은 오이, 수박 및 멜론 등의 박과작물에 전세계적으로 발생하여 많은 피해를 주고 있다. 본 연구에서는 시판 중인 박과작물 112개 품종의 탄저병에 대한 저항성 정도를 조사하였다. 각 품종의 2-3엽기 유묘에 C. orbiculare KACC 40809의 포자 농도가 4.0 C $40^5spores/mL$인 포자현탁액을 분무하여 접종하였다. 실험한 36개 오이 품종 중 '아시아스트라이크', '통일백다다기', '대선', '청록맛짱', '녹야청청' 및 '아시아노각' 등 6품종은 중도저항성을 나타내었으며, 나머지 품종들은 감수성을 보였다. 멜론 및 참외 33개 품종과 수박 4개 품종들은 C. orbiculare에 대하여 모두 감수성을 나타내었다. 이와 달리 호박 품종들은 실험한 다른 박과작물에 비하여 낮은 감수성을 보였다. 호박 17개 품종 중 '감미락단호박'과 '텃밭풋호박'은 저항성을, 12개 품종은 중도저항성을 나타냈다. 박과작물을 위한 대목 22개 품종 중 '쯔야까EX', '눈부셔', '유니온', 'RS111', '강근토좌', '황제토좌', 'NO.8', '신토좌', '불패토좌' 및 '뉴타입' 등 10개 품종은 저항성을 보였고, 4개 품종은 중도저항성을 나타냈다. 이상의 결과로부터 선발된 저항성 품종들은 박과작물 탄저병 저항성 육종을 위한 유전자원으로 이용될 수 있으리라 생각되었다.

Effect of Sphingosine-1-Phosphate on Intracellular Free Ca2+ in Cat Esophageal Smooth Muscle Cells

  • A comprehensive collection of proteins senses local changes in intracellular $Ca^{2+}$ concentrations ($[Ca^{2+}]_i$) and transduces these signals into responses to agonists. In the present study, we examined the effect of sphingosine-1-phosphate (S1P) on modulation of intracellular $Ca^{2+}$ concentrations in cat esophageal smooth muscle cells. To measure $[Ca^{2+}]_i$ levels in cat esophageal smooth muscle cells, we used a fluorescence microscopy with the Fura-2 loading method. S1P produced a concentration-dependent increase in $[Ca^{2+}]_i$ in the cells. Pretreatment with EGTA, an extracellular $Ca^{2+}$ chelator, decreased the S1P-induced increase in $[Ca^{2+}]_i$, and an L-type $Ca^{2+}$-channel blocker, nimodipine, decreased the effect of S1P. This indicates that $Ca^{2+}$ influx may be required for muscle contraction by S1P. When stimulated with thapsigargin, an intracellular calcium chelator, or 2-Aminoethoxydiphenyl borate (2-APB), an $InsP_3$ receptor blocker, the S1P-evoked increase in $[Ca^{2+}]_i$ was significantly decreased. Treatment with pertussis toxin (PTX), an inhibitor of $G_i$-protein, suppressed the increase in $[Ca^{2+}]_i$ evoked by S1P. These results suggest that the S1P-induced increase in $[Ca^{2+}]_i$ in cat esophageal smooth muscle cells occurs upon the activation of phospholipase C and subsequent release of $Ca^{2+}$ from the $InsP_3$-sensitive $Ca^{2+}$ pool in the sarcoplasmic reticulum. These results suggest that S1P utilized extracellular $Ca^{2+}$ via the L type $Ca^{2+}$ channel, which was dependent on activation of the $S1P_4$ receptor coupled to PTX-sensitive $G_i$ protein, via phospholipase C-mediated $Ca^{2+}$ release from the $InsP_3$-sensitive $Ca^{2+}$ pool in cat esophageal smooth muscle cells.

토마토 추출액 복합체가 전립선 암 세포와 전립선 비대증에 미치는 영향 (The Effect of the Compound of Tomato Extract to the Prostatic Cancer Cell and the Prostate of the Rat Model of Benign Prostatic Hyperplasia)

  • Benign prostatic hyperplasia (BPH) is one of the common disease in elderly men. Recently old-age population is increased and we are growing more and more interested in clinical importance of BPH. In this study, the effect of PLX, which was the mixture of tomato extract (including 2% of lycopene) and chitooligosaccharide, on prostatic cancer cell and testosterone-induced BPH in adult rats of the Sprague Dawley strain was determined. The cell viability was evaluated by MTT method using L929 and LNCaP cell line, pretreated with various concentrations of PLX. The expression of prostatic specific antigen (PSA) and 5${\alpha$}$-reductase genes were evaluated by realtime PCR using LNCaP cell line and compared various concentrations of PLX with 50 ${\mu}$M of finasteride. An experimental prostatic hyperplasia was induced in male Sprague Dawley rats by giving testosterone for 8 weeks. After 2 weeks from start of giving testosterone, PLX and finasteride were administered orally once a day. The results were analyzed with prostate weight per body weight at 8 weeks. Cell viability of L929 cell line decreased specifically at the concentration of 2000 ${\mu}$g/mf of PLX. The cytotoxicity of PLX to the LNCaP cell line was shown at above 500 ${\mu}$g/ml of PLX. The inhibitory effect of PLX to the expression of PSA and 5${\alpha$}$-reductase genes in LNCaP cell line increased with the concentration of PLX. In vivo study, the results of PLX and finasteride administered group were 3.75${\pm}$0.60 and 3.49${\pm}$0.49 prostate weight ${\times}10^3$/body weight, which were lower than the result of BPH induced group (4.74${\pm}$0.58). These results suggested that PLX may be an effective material in BPH by having the role of the 5a-reductase inhibitor.

Seven Days of Consecutive Shade during the Kernel Filling Stages Caused Irreparable Yield Reduction in Corn (Zea mays L.)

  • In monsoon climates, persistent shade is a troublesome weather condition with an impact on the growth and yield of corn (Zea mays L.). We imposed 7, 14, 21, and 28 days of consecutive shade (CS) on Gwangpyeongok and P3394 corn hybrids at the beginning of the kernel filling stages. Shade had little impact on leaf area and dry matter accumulation in the stem and leaves. However, dry matter accumulation in the ear was severely reduced by approximately 28% and 53% after 14 and 28 days of CS, respectively. For the components of grain yield, 7 and 14 days of shade did irreparable damage to the number of filled kernels, the kernel number per ear row, and the percent of filled kernels, but did little damage or reversible damage after removal of the shade to the 100-grain weight and the row number per ear. Shade significantly reduced the relative growth rate (RGR) due to a decrease in the net assimilation rate (NAR). These results suggest that source activity limitation by shade during the kernel filling stages leads to the inhibition of sink activity and size. The yield of biomass, ear, and grain logistically declined as the length of CS increased. Probit analysis revealed that the number of days of CS needed to cause 25% and 50% reductions in grain yield were 3.7 and 23.1, respectively. These results suggest that the plant yield loss induced by shade at the beginning of the kernel filling stages is mainly achieved within the first 7 days of consecutive shade.

Effects of strong shading on growth and yield in sweet potato (Ipomoea batatas L. LAMK.)

  • Sweet potatoes (Ipomoea batatas (L.) LAMK.,) have been cultivated in Central and South America for about 2000 years and are now grown mainly in Asia and South America. Sweet potatoes are annual in the temperate region, but are classified as perennial in the tropical region. In 2000, the cultivation area of sweet potatoes decreased to about 16,000 ha in 2000, but the cultivation area increased slightly in recent 20,000 ha in Korea. Sweet potatoes do not show higher maximum dry matter production of 120 ~ 150g per plant, and the leaf area index (LAI), which maximizes dry matter production, is known as 3.0 ~ 4.0. As the leaf area increase, the penetration of light into the canopy becomes poor, and sufficient photosynthesis cannot be achieved in the lower leaves, on the other hand the respiration increase, which results in poor dry matter production. This study was conducted to know the responses of sweet potatoes to intensive shading treatment of 80% shading. This experiment was conducted for about 42 days from September 6, 2016 to October 18, 2016 at Gyeongsang National University Experimental Farm, Jinju, Korea. The plant canopy was shaded with black nylon 80% shade cloth suspended 1.2 m above the ground. The photosynthetic rate, stomatal conductance, chlorophyll fluorescence, SPAD and NDVI were measured in 3 replicates every 7 days after shading initiation. After the fresh weight was measured, the samples were dried at $80^{\circ}C$ in a dry oven and measured. By the 80% shading treatment, chlorophyll fluorescence of the treated plants was slightly higher than that of the control, the SPAD value was higher by 3.4 and NDVI value was higher by 0.01. However, photosynthetic rate and stomatal conductance were lower than those of the control. The stomatal conductance of the control were two times higher than those of the control and the photosynthetic rate of the control was four times higher than that of the control. In control, plant showed a tendency to steadily increase in fresh weight and dry weight. However, in the case of shading treatment, the tendency to increase in the fresh and dry weight of tuberous roots was not clear. The fresh weight of shoot showed a tendency to increase steadily while the difference between treatment and control was not large, but tended to decrease after frost.

금 나노 입자 응집 원리를 이용한 유기인계와 카바메이트계 비색-신속 농약검출법 개발 (Development of a Colorimetric Rapid Detection Method for Organophosphorus and Carbamate Pesticides using Gold Nanoparticle Aggregation Principle)

  • 본 연구에서는 시판되고 있는 혈청 내 AChE가 acetylthiocholine과 반응하여 GNP에 aggregation 일으키는 원리를 이용하여 신선채소 농산물 중에 저농도 농약을 신속하고 간편하게 분석할 수 있는 비색-신속 농약 검출법을 개발하는 연구를 수행하였다. 먼저 비색-신속 농약 검출법의 최적화를 위해 GNP 입자의 크기에 따른 응집정도를 확인하여 15~20 nm 직경의 GNP를 선정하였고, 혈청의 희석배수와 acetylthiocholine의 농도를 확인하여 GNP 응집 차이가 가장 큰 혈청 1000배 희석과 acetylthiocholine 1 mM을 최적화 조건으로 선정하였다. 비색-신속 농약 검출법의 평가를 위해 최적화된 비색농약분석법을 이용하여 유기인계 농약은 dimethyl amine으로 카바메이트계 농약은 carbofuran으로 민감도를 분석한 결과 모두 7.5 ng/mL까지 검출이 가능한 것으로 확인되었으며 이는 기존의 비색-신속 농약 검출법과 비교했을 때 높은 민감도와 특이성을 나타내었다. 농약 이외에 화학물질인 곰팡이독소 등에 대한 반응성은 확인되지 않아 높은 특이성을 나타내었다. 또한 상추, 깻잎, 양상추에 대한 시료 전처리법을 확립하고 임의로 오염시킨 3종(상추, 깻잎, 양상추)의 농산물에 대해서 회수율을 확인한 결과유기인계와 카바메이트계 농약을 83.85~133.16% 정도의 회수율이 확인되었다. 이상의 결과 볼 때 본 연구에서 개발한 비색-신속 농약 검출법을 이용한다면 시판 농산물의 잔류농약을 신속하고 민감도 높게 검출할 수 있을 것으로 판단된다.

동위원소희석 액체크로마토그래피 질량분석법에 의한 분유 내 콜레스테롤의 정량 (Quantitative analysis of cholesterol in infant formula by isotope dilution liquid chromatography-tandem mass spectrometry)

  • 분유 내 콜레스테롤을 정량분석하기 위한 일차분석법으로 동위원소희석 질량분석법을 개발하였다. 내부표준물질로 콜레스테롤-d4를 사용하여 분유에 첨가하였다. 분유에 지방산과 에스테르형태로 결합되어 있는 콜레스테롤을 자유 콜레스테롤로 바꾸어주기 위해 비누화과정을 수행하였다. 비누화과정은 반응온도, 반응시간, 사용한 KOH의 농도에 따라서 최적화하였으며, 그 결과로 분유시료 0.1 g에 대해 70 ℃에서 180 분 동안 8 M의 KOH 0.8 mL를 첨가하여 반응을 진행시키는 최적화조건을 확립하였다. 이와 같은 조건으로 실험을 진행하여 재현성은 0.23%, 확장불확도는 95%의 신뢰범위에서 1.9%로 추정되었다. 확립된 동위원소희석 질량분석법의 유효화를 위해 분유내에서 콜레스테롤의 인증값을 가지는 NIST SRM을 측정하였고 이 결과가 인증값과 불확도 범위내에서 일치하는 것을 확인하였다.
