Taxonomic Review of the Family Triglidae (Pisces, Scorpaeniformes) from Korea (한국산 성대과 (횟대목) 어류의 분류학적 재검토)

  • The taxonomic review of the family Triglidae was made based on the specimens collected from the south-western coasts of Korea from 1986 to 1999. The family Triglidae from Korea was classified into 10 species belonging to 3 genera: Pterygotrigla hemisticta, Chelidonichthys kumu, Lepidotrigla, japonica. L. guentheri, L. alata, L. kishinouyei, L. kanagashira, L. microptera, L. hime and L. abyssalis. Of them, L. kanagashira was reported for the first time in South Sea of Korea. A key to species of the family Triglidae from Korea was provided, with description of their morphological characteristics and their distribution.

Selection of organic Nitrogen Source and Optimization of Culture Conditions for the Production of Arachidonic Acid from Mortierella alpina (Mortierella alpina를 이용한 아라키돈산의 생산에서 유기질소원의 선정과 배양 조건의 최적화)

  • Experiments were carried out to select an organic nitrogen source and optimize the culture conditions for the production of arachidonic acid by Mortierella alpina DSA-12. Corn steep powder(CSP) was selected as an organic nitrogen source based on arachidonic acid production and raw material price. The optimum C/N ratio was in the range of 15 to 17 with the medium containing glucose as carbon source and CSP as nitrogen source. The optimum culture conditions for arachidonic acid production showed 500 rpm agitation and 25$^{\circ}C$ culture temperature at 0.5 vvm aeration. Under the optimum conditions, the concentration of cell, total lipid and arachidonic acid were 21.8 g/L, 10.2 g/L and 3.70 g/L, respectively, from 50 g/L glucose and 18 g/L CSP. In the 500 L fermenter with 0.5 vvm aeration and 200 rpm agitation, the concentration of cell, total lipid and arachidonic acid were 19.8 g/L, 9.1 g/L and 3.67 g/L, respectively, from 50 g/L glucose and 18 g/L CSP. This result showed that an arachidonic acid production could be possible with a bench-scale fermenter using corn steep powder as a nitrogen source.

Taxonomical Studies on Korean Fungi of Ascomycetes for the Publication of Colored Illustrations (원색도감(原色圖鑑) 발간(發刊)을 위한 한국산(韓國産) 자낭균류(子囊菌類)의 분류학적(分類學的) 연구(硏究))

  • This study has been carried out for the publication of colored illustrations of Korean fungi. The fresh fungi of Ascomycetes were collected and photographed, for the most part, at Gwangneung, Kyonggi Province, Mt. Yongmun in Kyonggi Province, Mt. Sokri in Chungcheong Province, Mt. Jiyee in Kyeongsang Province, Mt. Mudeug in Jeonra Province, and Mt. Hanra in Jeuju Island from July 1, 1981 to June 31, 1982. These higher fungi were made into colored slides and dried specimens, then classified. According to the investigated result, colored slides and classification for the common and new to Korea 101 species were completed among 620 spp. which were known all of the fungi in Korea. They were included 10 oders, 19 families, 51 genera and 101 species. Their world distributions were investigated. Twenty six species of them, that is, Peziza vesiculosa, Rhizina inflata, Gelasinospora longispora, Sordaria fimicola, Cantharcmyces exiguus, Dichomyces biformis, D. furcifer, D. homalotae, D. hybridus, D. vulgatus, Dioicomyces anthici, Enathromyces indicus, Laboulbenia borealis, L. brachconychi, L. cristata, L. exigua, L. fusciculata, L. filifera, L. flagellata, L. rougetii, L. tachys, L. vulgaris, L. yoshidai, Rickia ancylopi, R. papuana and R. peyerimhoffii were found to be new in Korea. In addition their common names and descriptions prepared. According to study plan, this paper pressed only one page in colored plate among 101 species.

Effect of Bambusae Caulis in Liquamen on the Synthesis of Basement Membrane Proteins during Proliferation and Differentiation of 3T3-L 1 Cells (죽역이 3T3-L1 세포의 증식 및 분화시 기저영 단백질 합성에 미치는 영향)

  • The purpose of this research was to investigate effects of Bambusae Caulis in Liquamen (BCL) on the synthesis of basement membrane proteins during proliferation and differentiation of 3T3-L1 cells. BCL has been used to relieve the cough and asthma, and remove phlegm in traditional oriental medicines. In recent years. it was studied for its antiinflammatory, antiallergenic. immune-modulating and anticarcinogenic capabilities. We have previously observed that glycyrrhizin stimulates the adipose conversion of 3T3-L1 cells. To investigate effects of BCL on the basement membrane proteins during proliferation and differentiation of 3T3-L1 cells, we have analyzed synthetic amounts of basement membrane components such as type IV collagen and BM40. BCL stimulated the synthesis and secretion of type IV collagen from both 3T3-L1 preadipocytes and adipocytes. The synthesis and secretion of BM40 was not affected by BCL. The continuous addition of BCL markedly stimulated cell growth and increased cell density. These results suggest an important role for type IV collagen in adipocyte differentiation.

Effects of Endocrine Disruptors, Nonylphenol in Daphnia magna (물벼룩 (Daphnia magna)에서 내분비계장애물질인 노닐페놀의 영향)

  • Nonylphenol is one of endocrine disruptors, as structurally stable, hydrophobic compounds exhibit high condensability and long-lasting in the natural environment. The purpose of this study was to determine the toxic effects of nonylphenol on Daphnia magna. In acute toxicity test, D. magna was exposed for 48 h at concentrations of 0, 10, 18, 32, 56 and $100{\mu}g\;L^{-1}$ nonylphenol. In chronic toxicity test, D. magna were exposed through water for 21 days at concentrations of 0, 1.0, 1.8, 3.2, 5.6 and $10{\mu}g\;L^{-1}$ nonylphenol. Acute toxicity was assessed on the basis of immobility, while chronic toxicity was assessed on the basis of fecundity. The acute toxicity test on nonylphenol was showed that the values of 24 h and 48 h $EC_{50}$ were $25.0{\mu}g\;L^{-1}$ and $13.7{\mu}g\;L^{-1}$, respectively. In chronic test, fecundity was reduced significantly at $5.6{\mu}g\;L^{-1}$ of nonylphenol. These results indicated that nonylphenol have some hazard for acute or chronic toxicity to freshwater invertebrate organism.

Development of Oil Flowmeter and Preliminary Evaluation for Establishing National Leak Standard (국가 리크표준기용 오일유량계 개발 및 예비 평가)

  • We preliminarily evaluated the performance of a flowmeter by the measurement of the pressure of a ionization gauge and the produced throughput (Q) using a vacuum system and pistons with the radius of 10 mm and 20 mm. The throughput range with the later piston was from $1{\times}10^{-1}Pa{\cdot}L/s$ to $1.2Pa{\cdot}L/s$ and that with the former piston was from $5.3{\times}10^{-1}Pa{\cdot}L/s$ to $1.05Pa{\cdot}L/s$. In these ranges, both pistons showed good repeatability and linearity. The developed oil flowmeter as the leak standard will be used in the calibration of standard leak.

Plant Regeneration and Somatic Embryogenesis from Zygotic Embryo-derived Callus of Native Prunus yedoensis in Mt. Halla (한라산 자생 왕벚나무 접합자배 유래의 캘러스로부터 체세포배 형성과 식물체 재분화)

  • Somatic embryos were induced through embryogenic callus derived from immature zygotic embryo culture of native Prunus yedoensis in Mt. Halla and regenerated into plantlets successfully. Embryogenic callus was induced most effectively on MS medium with 1.0 mg/L 2, 4-D and 0.1 mg/L BAP at an efficiency of approximately 60% using 45 day-old zygotic embryos after full blooming. Globular somatic embryos were induced from embryogenic callus on MS medium with 1.0 mg/L 2, 4-D and 0.1 mg/L BAP and these globular embryos developed to heart-shaped and cotyledonary embryos on hormone-free MS medium. Normal somatic embryos germinated 49% on 1/2 MS medium and the plants regenerated from the somatic embryos were morphologically normal.

  • Near-ir spectra have been obtained for the $ν_3$+ Amide II combination thioacetamide(TA) band in CC$l_4$ and TA-DMP in CC$l_4$ in the range of 5 to 55${\circ}C$. Absorbance of the weak bands of the DMP and solvent has been compensated. The spectra are analyzed by the computer resolution into two Lorentzian-Gaussian product bands which have been identified with monomeric TA and 1 : 1 TA-DMP complex. Equilibrium constants and thermodynamic parameters for the hydrogen bonding between TA and DMP have been evaluated by the analysis of the concentration and temperature dependent spectra for the very dilute CC$l_4$ solutions. The $ΔH{\circ} and ΔS{\circ}$ of TA and DMT have been found to be -14.6 kJmo$l^{-1}$ and -16.2 Jmo$l^{-1}$ de$g^{-1}$.

Varietal Variation of Pigmentation and Some Nutritive Characteristics in Colored Rices (유색미 색도 및 영양특성의 품종간 변이)

  • Colored rices have been used for specific purposes by rice consumers due to the color and nutritive values empirically recognized. In this study, varietal variations of pigmentation and nutritive values were investigated in brown, red, purple and black rices. Pigments were localized in seed coat to pericarp region in all varieties tested. Pigments were slightly residued on the surface of milled rice. Anthocyanin content per g brown rice was 1.63~17.62 $\mu\textrm{g}$ in brown and reddish-brown rices, 3.56~11.10 $\mu\textrm{g}$ in red rices, 28.11~401.22 $\mu\textrm{g}$ in purple rices, and 3, 665.98 $\mu\textrm{g}$ in a black rice. A vatiety DZ 78 showed the highest protein content out of colored rices analyzed for protein. Normal and colored rices were found to have the similiar composition of amino acids, and so was in between brown rice without embryo and milled rice. Colored rices, L $K_1$B-4-12-1-1 and DK 1, showed higher content of vitamin $B_1$ compared with Hwacheongbyeo, a check variety of no specific color, and L $K_1$B-2-1-1 and L $K_1$B-4-12-1-1 showed much higher content of vitamin B2 in brown rice without embryo. Cation contents such as $K^{+}$, $Mg^{2+}$, $Ca^{2+}$ and F $e^{2+}$ were significantly increased in most of the colored rices tested implying that the increase might be associated with color pigmentation.ation.

  • Dendrobium candidum Wallich ex Lindley is a traditional Chinese medicine plant and has been widely used for medicinal and ornamental purposes. In this study, several different factors affecting micro propagation of protocorm-like bodies (PLBs) such as basal media, plant growth regulators, and carbon sources. The proliferation PLB derived from seeds was the best in $H_3P_4$ basal medium containing $0.1mg{\cdot}L^{-1}$ NAA and $0.1mg{\cdot}L^{-1}$ Kinetin. PLB growth was the best when $10g{\cdot}L^{-1}$ sucrose was added to the carbon atoms in the medium. The rate of shoot formation from the propagated PLB was the highest in 1/4 MS or $H_1P_2$ medium containing $10g{\cdot}L^{-1}$ sucrose, and the shoot length was longer than the others.
