Antioxidant Activity and Anti-Adipogenic Effect of Ligularia stenocephala Extract (곤달비 추출물의 항산화 활성 및 지방세포 분화 억제 효과)

  • The purpose of this study was to evaluate the antioxidant and anti-adipogenic activities of Ligularia stenocephala (L. stenocephala) extract. The contents of the total polyphenol of the extract was 55.950 mg GAE/g residue. Antioxidant activities of L. stenocephala were evaluated by free radical scavenging ability and a reducing power test. 2,2'azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and ${\alpha}$-${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) free radical scavenging activities of the extract were approximately 90% and 70%, respectively. Reducing power of the extract was 258.833 mg TE/g residue. The anti-adipogenic activity of L. stenocephala extract was examined in 3T3-L1 cells. During adipocyte differentiation, the 3T3-L1 cells were treated both with and without the extract. L. stenocephala extract suppressed the lipid accumulation in a concentration-dependent manner in the 3T3-L1 cells. The L. stenocephala extract inhibited the expression of peroxisome proliferator activated receptor ${\gamma}$ ($PPAR{\gamma}$) and adipocyte protein 2 (aP2) proteins, compared with control adipocytes. These results indicate that L. stenocephala could be regarded as a potential source natural antioxidant and an anti-obesity agent.

Purification and Characterization of an $\alpha$ -L-Arabinofuranosidase from Bacillus sp. DSNC 101 (Bacillus sp. DSNC 101이 생산하는 $\alpha$-L-Arabinofuranosidase의 정제 및 특성)

  • ${\alpha}$-L-Arabinofuranosidase was purified from the culture supernatant of Bacillus sp. DSNC 101. The enzyme had a molecular weight of 56 kDa. Optimum temperature and pH for ${\alpha}$-L-arabinofuranosidase activity were 55$^{\circ}C$ and 7.0 respectively. The Michaelis constant(Km) and maximal reaction velo-city(Vmax) for p-nitrophenyl-${\alpha}$-L-arabinofuranoside were 1.0 mM and 113.6 U/mg protein, respe-ctively. ${\alpha}$-L-Arabinofuranosidase was completely inhibited by HgCl$_2$ and CuSO$_4$. The enzyme was spe-cific for the ${\alpha}$-linked arabinoside in the furanoside configuration. The enzyme was produced during growth on agricultural residue such as rice straw, but not during growth on spelt xylan, glucose or cellobiose.

Three Cyanide-Bridged One-Dimensional Single Chain CoIII-MnII Complexes: Rational Design, Synthesis, Crystal Structures and Magnetic Properties

  • Two pyridinecarboxamide dicyanidecobalt(III) building blocks and two mononuclear seven-coordinated macrocycle manganese(II) compounds have been rationally selected to assemble cyanide-bridged heterobimetallic complexes, resulting in three cyanide-bridged $Co^{III}-Mn^{II}$ complexes. Single X-ray diffraction analysis show that these complexes $\{[Mn(L^1)][Co(bpb)]\}ClO_4{\cdot}CH_3OH{\cdot}0.5H_2O$ ($\mathbf{1}$), $\{[Mn(L^2)][Co(bpb)]\}ClO_4{\cdot}0.5CH_3OH$ ($\mathbf{2}$) and ${[Mn(L^1)][Co(bpb)]\}ClO_4{\cdot}H_2O$ ($\mathbf{3}$) ($L^1$ = 3,6-diazaoctane-1,8-diamine, $L^2$ = 3,6-dioxaoctano-1,8-diamine; $bpb2^{2-}$ = 1,2-bis(pyridine-2-carboxamido)benzenate, $bpmb2^{2-}$ = 1,2-bis(pyridine-2-carboxamido)-4-methyl-benzenate) all present predictable one-dimensional single chain structures. The molecular structures of these one-dimensional complexes consists of alternating units of $[Mn(L)]^{2+}$ ($L=L^1$ or $L^2$) and $[Co(L^{\prime})(CN)2]^-$ ($L^{\prime}=bpb2^{2-}$, or $bpmb2^{2-}$), forming a cyanide-bridged cationic polymeric chain with free $ClO_4{^-}$ as the balance anion. The coordination geometry of manganese(II) ion in the three one-dimensional complexes is a slightly distorted pentagonal-bipyrimidal with two cyanide nitrogen atoms at the trans positions and $N_5$ or $N_3O_2$ coordinating mode at the equatorial plane from ligand $L^1$ or $L^2$. Investigation over magnetic properties of these complexes reveals that the very weak magnetic coupling between neighboring Mn(II) ions connected by the diamagnetic dicyanidecobalt(III) building block. A best-fit to the magnetic susceptibility of complex ${\mathbf}{1}$ leads to the magnetic coupling constants $J=-0.084(3)cm^{-1}$.

Effect of Electrolyzed Water and Organic Acids on the Growth Inhibition of Listeria monocytogenes on Lettuce (전해수 및 유기산처리에 의한 양상치에 오염된 Listeria monocytogenes의 생육저해)

  • This study was conducted to determine the inactivation effect of electrolyzed water and organic acids either alone or in combination on L. monocytogenes or natural microflora on lettuce. Acidic electrolyzed water completely inactivated L. monocytogenes in broth system within 60 sec, but alkalin electrolyzed water caused approximate 1.7 log CFU/g reduction. However, acidic electrolyzed water reduced only 2.5 log CFU/g of L. monocytogenes on lettuce, and similar antimicrobial effect was observed with alkalin electrolyzed water. In the meantime, acidic and alkaline electrolyzed water caused approximately 2 log CFU/g reduction compared to control, whereas both electrolyzed water combined with $1\%$ organic acids ranged from 2.6 to 3.7 log CFU/g reduction. Among the organic acids, both electrolyzed water combined with $1\%$ citric acid showed the strongest synergistic antimicrobial effect to reduce L. monocytogenes on lettuce as well as total counts, yeast and molds. When antimicrobials, alone or in combination were treated into L. monocytogenes inoculated lettuce at $5^{\circ}C\;and\;15^{\circ}C$ for designed periods, the combined alkalin electrolyzed water with $1\%$ citric acid showed the greatest potential to inhibit growth of the bacteria. According to Scanning Electron Microscopy(SEM), the treatment of electrolyzed alkali water in combination with $1\%$ citric acid highly reduced the growth of the L. monocytogenes compared to single treatment and resulted in causing the destruction of cell membrane.

Determination of L-Dopa by Spectrofluorimetry Using Co-fluorescence Enhancer (보조 형광증가제를 이용한 L-Dopa의 형광분광법적 정량)

  • Methods to determine L-dopa(L-3,4-dihydroxyphenylalanine) in aqueous solution by spectrofluorimetry based upon the ligand sensitized luminescence of Tb(III) ion L-dopa complex have been studied. Tb(III) ion and Lu(III) ion were used as ligand sensitized fluorescencer and co-fluorescence enhancer, respectively. The effects of excitation wavelength, pH, concentration of Tb(III) ion, concentration of Lu(III) ion and emission wavelength on the fluorescence intensity were investigated. The fluorescence intensity of the Tb(III) ion L-dopa complex was further increased with addition of Lu(III) ion. The calibration curve for L-dopa was linear over the range from 5.0 ${\times}$ $10^{-7}$ M to 1.0${\times}$ $10^{-4}$ M and the detection limit was 4.0 ${\times}$ $10^{-8}$ M under the optimal experimental conditions of 300 nm, 8.0, 1.0 ${\times}$ $10^{-4}$ M and 545 nm for excitation wavelength, pH, concentration of Tb(III) ion and emission wavelength, respectively. When Lu(III) ion was added to Tb(Ⅲ) ion L-dopa complex, the concentration range of linear response and detection limit were 1.0${\times}$$10^{-8}$ M to 2.0 ${\times}$ $10^{-4}$ M and 1.0 ${\times}$ $10^{-9}$ M, respectively under the optimal experimental conditions of 300 nm, 8.5, 1.0 ${\times}$ $10^{-5}$ M, 1.0 ${\times}$ $10^{-5}$ M, 545 nm for excitation wavelength, pH, concentration of Tb(III) ion, concentration of Lu(III) ion and emission wavelength, respectively.

  • We define the spherical non-commutative torus $L_{\omega}$/ as the crossed product obtained by an iteration of l crossed products by actions of, the first action on C( $S^{2n+l}$). Assume the fibres are isomorphic to the tensor product of a completely irrational non-commutative torus $A_{p}$ with a matrix algebra $M_{m}$ ( ) (m > 1). We prove that $L_{\omega}$/ $M_{p}$ (C) is not isomorphic to C(Prim( $L_{\omega}$/)) $A_{p}$ $M_{mp}$ (C), and that the tensor product of $L_{\omega}$/ with a UHF-algebra $M_{p{\infty}}$ of type $p^{\infty}$ is isomorphic to C(Prim( $L_{\omega}$/)) $A_{p}$ $M_{m}$ (C) $M_{p{\infty}}$ if and only if the set of prime factors of m is a subset of the set of prime factors of p. New Records of Hippolytid Shrimps, Lebbeus speciosus and Lebbeus comanthi (Crustacea: Decapoda: Caridea) from the East Sea, Korea

  • Two species of the hippolytid shrimps, Lebbeus speciosus and Lebbeus comanthi, are described and illustrated for the first time in Korea. They were collected from the East Sea. Lebbeus speciosus was found in a scallop farm of Jumunjin, Gangneung, and L. comanthi associated with crinoids, was found in Gampo, Gyeongju. The difference between these two related species is in the number of dorsal teeth of the rostrum, five teeth in L. speciosus and a single tooth in L. comanthi. The Korean Lebbeus species has increased to 6 species by the present report. They are L. grandimana, L. polaris, L. unalaskensis, L. groenlandicus, L. speciosus, and L. comanthi.
