  • In order to identify the bacteria living on the cocoons in Korea, the isolated bacterias' morphological. cultural and physiological characters has been determined through the detailed study. The second aim of this experiment was to protect against the bacteria which damage silk protein during storage. 1. The twelve strains of the bacteria were isolated and identified in the cocoons produced in Korea. The results of the identification are as the following. No 1, No 8; Bacillus subtilis variation No 2, ; Bacillus stearothermophilus No 3, ; Bacillus circulans No 5, No 6; Bacillus thuringiensis No 7, No 11; Bacillus brevis No 12, No l0; Bacillus cereus variation

Memory-improving Effects of Fermented Sea Tangle Saccharina japonica in Normal Mice (정상 동물모델에서 다시마(Saccharina japonica) 발효물의 기억력 개선 효과)

  • Marine organisms are sources of many bioactive compounds, such as essential fatty acids, essential amino acids, vitamins, and minerals, making them useful candidates for the production of safe bioactive substances. They also synthesize glutamic acid, which can be used to produce γ-aminobutyric acid (GABA), the major inhibitory neurotransmitter in the central nervous system (CNS), via fermentation with Lactobacillus brevis BJ-20. This study investigated the degree to which fermented sea tangle (FST) inhibits enzymes such as acetylcholine esterase (AChE) and prolyl endopeptidase (PEP) and affects memory of normal mice using the T-maze test. FST inhibited more than 90% of AChE at 1 mg/mL and 50% of PEP at 8 mg/mL. Oral FST (100 mg/kg) significantly improved performance of normal mice on the T-maze. Therefore, sea tangle fermented with L. brevis BJ20 effectively contributes to memory improvement and might be a useful functional food ingredient.

Characteristics of Fermented Omija (Schizandra chinensis Baillon) Sugar Treatment Extracts by Lactobacillus sp. (Lactobacillus sp. 균주를 이용한 오미자 당침액의 발효특성)

  • This study was carried out to investigate the characteristics of FOST (fermented omija sugar treatment extracts) using Lactobacillus brevis HLJ59. Antioxidant activities of FOST were evaluated through viable cell number of L. brevis HLJ59, DPPH radical scavenging activity, reducing power and SOD-like activity, compared to non-FOST(non-fermented omija sugar treatment extracts). Also it was to evaluate Angiotensin Converting Enzyme (ACE) and Urease inhibitory activity of FOST. The viable cell number of L. brevis was about $2.05{\pm}0.21{\times}10^8$, $6.31{\pm}0.56{\times}10^{11}$, and $8.14{\pm}0.14{\times}10^9$ at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. DPPH radical scavenging activity of FOST was about 60.3%, 71.8%, and 44.5% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. The reducing power of FOST was about 0.92, 1.19, and 0.73 (OD at 700 nm) at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, and SOD-like activity of FOST was about 50.4%, 53.7%, and 33.4% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. ACE and Urease inhibitory activity by FOST was about 47.4%, 78.2%, 56.4% and 58.1%, 83.4%, 63.2% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. The results indicated that the fermentation of omija sugar treatment extracts using Lactobacillus brevis HLJ59 increased the antioxidant activities campared to the non-fermented omija sugar treatment extracts.

Sleep Inducing Effect of Gastrodia elata Fermented with Lactic Acid Bacteria (유산균 발효 천마의 수면유도 효과)

  • Ethanol extract of Gastrodia elata fermented with Lactobacillus brevis was highly effective on the duration of pentobarbital hypnosis in mice. Pretreatment of mice with ethanol extract of the fermented Gastrodia elata (200 mg/kg, p.o.) prolonged markedly the duration of pentobarbital sleeping time and reduced the sleep latency. The mechanism of the extract of the fermented Gastrodia elata was investigated to inhibit the binding of $^3H$-Flumazenil, a selective benzodiazepine receptor antagonist, to benzodiazepine receptor of mice cortices. $IC_{50}$ value from displacement of $^3H$-Flumazenil binding was 62 ${\mu}g/mL$ at the treatment of the fermented Gastrodia elata. Therefore, these finding, such as increase of sleeping time and reduction of sleep latency, was examined by elevated concentration of GABA and parishin C, which were increased by Lactobacillus brevis.

  • The extensor digitorum brevis (EDB) muscle island flap is a reliable, safe method for coverage of foot and ankle. There are many variation in approach such as curvilinear, zigzag, L-shaped or vertical longitudinal incision for exposure of the EDB muscle. These approaches use only single incision excluding the distal incision for exposure of the distal tendon. Since dorsalis pedis artery vascular bundle and sinus tarsi branch of the lateral tarsal artery both requires careful dissection, single incision alone may cause not only difficulty in exposure but also skin sloughing at donor site. So we tried to modify the approach into two parallel longitudinal incision, one for dorsalis pedis vascular bundle and the other for sinus tarsi branch exposure. The author treated 9 patient with EDB muscle flap. We used single incision in six patients, and two parallel incision in three patients. All the flap survived. In two parallel incision group, dissection was more easy and rapid. So we would like to suggest that two parallel longitudinal incision approach is better method than the single incision technique for exposure of the EDB muscle flap.

Optimization of Lactic Acid Fermentation of Prickly Pear Extract

  • Lactic acid fermentation of prickly pear extract (PPE) was performed by Lactobacillus rhamnosus LS, Lactobacillus bulgaricus, and Lactobacillus brevis. The PPE was pasteurized to eliminate indigenous microorganisms as well as to dissolve the partially insoluble pulp. The PPE fermented without yeast extract by L. rhamnosus LS exhibited 0.57% acidity and 3.5${\times}$10$^{8}$ CFU/mL bacteria count. With the addition of 0.2% edible yeast extract the PPE fermented by L. rhamnosus LS exhibited 1.15% acidity,2.7${\times}$10$^{9}$ CFU/mL bacteria count and 95.0% retention of red color. When 5% fructose syrup was added, the PPE fermented by L. rhamnosus LS had 1.09% acidity, 6.5${\times}$10$^{8}$ CFU/mL, and 97.7% retention of red color. With 1∼3% (w/v) concentrations of starter, the PPE fermented by L. bulgaricus and L. brevis showed 0.97% and 0.65% acidities, respectively. The viable cell counts from L. rhamnosus LS fermentation were higher compared with those of other LAB. During cold storage at 4$^{\circ}C$, the viable cell count was well maintained for 3 weeks, but then rapidly decreased. The red pigment was highly stable during cold storage for 4 weeks. The pasteurized PPE fortified with 5% fructose syrup, 0.2% yeast extract, and 0.05% CaCO$_3$ was successfully fermented by inoculating with 3% LAB and incubating at 3$0^{\circ}C$ for 2 days. Both viable cell counts and the red color of the fermented PPE were well maintained during cold storage for 3 weeks.

Resistant Activity to Hydrogen Peroxide of Lactobacillus spp., Bifidobacterium spp., Bacillus coagulans and Hydrogen Peroxide Generation Capability of Lactobacillus spp. (Lactobacillus spp., Bifidobacterium spp. 및 Bacillus coagulans의 과산화수소 저항성과 과산화수소 생성 능력)

  • Studies on the resistance of Lactobacillus ssp., Bifidobacterium spp. and Bacillus coagulans to hydrogen peroxide were conducted by determination of the viable cells after the test cells in 2mM hydrogen peroxide solution for a predetermined time; L. acidophilus CU4111 and L. casei CU4114 were most resistant to the hydrogen peroxide among the fifteen test lactobacilli strains, whereas L. brevis Cu4206 was the strain which was the most susceptible to hydrogen peroxide. Bifidobacterium longum Cu4131 was one of the resistant strains. A prominant tendency found out that Bacillus coagulans possessed a strong resistance to hydrogen peroxide. The results of level of hydrogen peroxide determination in the cell extracts showed all the test strains contained hydrogen peroxide in the cytoplasm, the amount varied depending on the strain and species of lactic acid bacteria. Bifidobacterium bifidum CU 4134 and L. casei CU 4114 were potent hydrogen peroxide producer strain.

Microbial Dynamics of Commercial Makgeolli Depending on the Storage Temperature

  • Market fresh makgeolli was stored at different temperatures of $4^{\circ}C$ and $25^{\circ}C$ to assess the change of the microbial diversity according to the storage temperature and period. Yeast counts increased until day 3 of storage and decreased thereafter. General and lactic acid bacterial counts continuously increased during storage. The data indicated that the control of growth of microorganisms, particularly general bacteria and lactic acid bacteria (LAB), is essential. Total acid levels started to decrease in the makgeolli stored at $4^{\circ}C$, and increased from day 6 of storage in the makgeolli stored at $25^{\circ}C$. The increase of total acid in the non-refrigerated condition greatly affected the quality of makgeolli. In both the fresh makgeolli samples stored at $4^{\circ}C$ and $25^{\circ}C$, yeast (Saccharomyces cerevisiae) and molds (Aspergillus tubingensis, Candida glaebosa, and Aspergillus niger) were noted. Denaturing gradient gel electrophoresis (DGGE) band patterns were almost constant regardless of the storage period. As for bacteria, Lactobacillus crustorum, L. brevis, and Microlaena stipoides were found in the makgeolli stored at $4^{\circ}C$, and L. crustorum, Lactobacillus sp., L. plantarum, L. brevis, L. rhamnosus, and L. similis were found in the makgeolli stored at $25^{\circ}C$. In particular, in the makgeolli stored at $25^{\circ}C$, L. crustorum and L. plantarum presented dark bands and were identified as the primary microorganisms that affected spoilage of fresh makgeolli.

Electromyographic Study of Lumbosacral Radiculopathy by Lumbar Disc (요추 추간원판에 의한 요천추 선경근 병변의 근전도 연구)

  • The aim of this study is to examine the sensitivity and characteristics of electromyography abnormalities detected by using various paramenters in patients with lumbosacral radiculopathies. EMG is widely used for disgnosing and localizing the level of radiculopathy. The results of the study were as follow : 1. In electromyography, L5 radiculopathy usa 95 cases(51.690). S1 radiculopathy was $45m(24.5\%)$ L4 radiculopathy was 18cases $(9.8\%)$, and L2, 3 radiculopathy was 8cases$(4.3\%)$. Remains 18cases$(9.8\%)$ had no definite radiculopathy. 2. Peroneal and tibial motor nerve conduction velocity studies were not significant as compared to the side to side. 3. Latency of H-reflex in L5 radiculopathy was $30.55\pm2.47$ in affected side, $29.47\pm2.29$ in unaffected side, in S1 radiculopathy was $33.00\pm2.03$ in affected side, R30.18\pm2.21$ in unaffected side. It was statistically significant(p<0.01). H-reflex mean difference of S1 radiculopathy group was significantly prolonged as compared to the L5 and S1 radiculopathies(p<0.001). 4. In L2, 3 radiculopathy, abnormal spontaneous activities and motor unit action potentials were showed high sensitivity in upper lumber paraspinal, hip adductors, quadriceps and iliopsoas muscles. 5. In L4 radiculopathy, lower lumbar paraspinal, tibialis anterior, quadriceps muscles were showed high sensitivity. 6. In L5 radiculopathy, lower lumbar paraspinal, extensor hallucis longus, extensor digitorum longus, peroneus longus, extensor digitorum brevis, gluteus maximus, tensor fasciae latae muscles were showed high sensitivity. 7. In S1 radiculopathy, lower lumbar paraspinal, gluteus maximus, peroneus longus, soleus, abductor hallucis, hamstrings, extensor digitorum brevis, extensor hallucis lognus, gastrocnemius muscles were showed high sensitivity.

Enzyme-assisted extraction of Ecklonia cava fermented with Lactobacillus brevis and isolation of an anti-inflammatory polysaccharide

  • Fermentation and enzyme-assisted extraction (EAE) improve nutritional and functional properties of foods by increasing the extraction of active compounds, ingestion rates, and body absorption. In this study, we investigated whether applying the EAE process improves the extraction and isolation efficiency of a polysaccharide from fermented Ecklonia cava (FE), which inhibited NO production in lipopolysaccharide (LPS)-activated RAW 264.7 cells. The results showed that the FE using the fungi Candida utilis and two different bacteria, namely Lactobacillus brevis and Saccharomyces cerevisiae increased protein and carbohydrate contents in comparison with those in non-fermented E. cava (NE). Aqueous extracts of fermented E. cava increased extraction yields and carbohydrate content, compared with the aqueous extract of NE. In addition, treating LPS-stimulated RAW 264.7 cells with aqueous extracts resulted in reduced NO production compared to that in LPS-treated cells. Ten EAEs of L. brevis-fermented E. cava (LFE) improved NO inhibitory effects in LPS-activated RAW 264.7 cells and the Viscozyme extract (VLFE) from the resulting extracts showed the highest NO inhibitory effect. We found that the >30 kDa fraction of VLFE led to markedly high inhibition of LPS-induced NO production as compared to that in the <30 kDa fraction. The crude polysaccharide isolated from >30 kDa fraction (VLFEP) consisted of fucose and markedly decreased NO production induced by LPS stimulation. VLFEP could be useful as an anti-inflammatory agent to suppress macrophage activation.
