Characteristics of Microsatellites in the Transcript Sequences of the Laccaria bicolor Genome

  • In this paper, we analyzed the microsatellites in the transcript sequences of the whole Laccaria bicolor genome. Our results revealed that, apart from the triplet repeats, length diversification and richness of the detected microsatellites positively correlated with their repeat motif lengths, which were distinct from the variation trends observed for the transcriptional microsatellites in the genome of higher plants. We also compared the microsatellites detected in the genic regions and in the nongenic regions of the L. bicolor genome. Subsequently, SSR primers were designed for the transcriptional microsatellites in the L. bicolor genome. These SSR primers provide desirable genetic resources to the ectomycorrhizae community, and this study provides deep insight into the characteristics of the micro satellite sequences in the L. bicolor genome.

Antioxidative Activity and Irritation Response of Lespedeza bicolor

  • In the present study, we evaluated the free radical scavenging and xanthine oxidase inhibitory activities exhibited by extracts obtained from the dried stems (and leaves) of Lespedeza bicolor We also assessed its potential irritation activities with regard to cosmetic use. When the DPPH radical scavenging activities of L. bicolor were assessed at six different concentrations (0, 50, 100, 250, 500 and 1000 ${\mu}g/ml$), the concentration of L. bicolor required to inhibit DPPH radical formation by $50\%$ was found to be $164.90{\mu}g/ml$. The effects of L. bicolor on the inhibition of xanthine oxidase were determined at seven different concentrations. The $50\%$ effective concentration was found to be $282.75{\mu}g/ml$. In the skin irritation test, all animals survived for the duration of the study, and all exhibited normal gains in body weight. The control sites exhibited no response to the control procedures. No edema, erythema, or eschar formation was observed in any of the tested rabbits. In the ocular irritation study, all of the rabbit eyes remained normal. In summary, L. bicolor extracts were considered to be non-irritating to the skin and eye.

수수 추출물에 의한 마우스 비장세포 및 대식세포 활성의 항진 효과 (Enhancing Effect of Sorghum bicolor L. Moench (Sorghum, su-su) Extracts on Mouse Spleen and Macrophage Cell Activation)

  • Sorghum bicolor L. Moench(Sorghum, Su-Su) is a major cereal food crop used in many parts of the world. It is used as a human food resource and folk medicines in Asia and Africa. The stem of sorghum has been used as a digestive aid and an anti-diarrheal agent. Sorghum hybrids contain high levels of diverse phenolic compounds that may provide health benefits. High levels of polyflavanols, anthocyanins, phenolic acids, and other antioxidant compounds have been reported in sorghums, which have also been shown to possess various biological activities such as anti-mutagenic, anti-carcinogenic, and HMG-CoA reductase inhibitory activities. In an in vitro experiment, we examined mice splenocyte proliferation and production of three types of cytokine($IL-1{\beta},\;IL-6,\;TNF-{\alpha}$) by peritoneal macrophages cultured with ethanol and water extracts of Sorghum bicolor L. Moench. A single cell suspension of splenocytes was prepared and the cell proliferation of the splenocytes was examined by MTT assay. The splenocyte proliferation was increased when water extracts of Sorghum bicolor L. Moench were used as supplements in all concentrations investigated. The production of cytokine($IL-1{\beta},\;IL-6,\;TNF-{\alpha}$) by activated peritoneal macrophage was detected by ELISA using the cytokine kit. $IL-1{\beta},\;IL-6,\;and\;TNF-{\alpha}$ production by activated macrophages were increased by supplementation with Sorghum bicolor L. Moench water extracts. This study suggests that supplementation of with Sorghum bicolor L. Moench water extracts may enhance immune function by regulating the splenocyte proliferation and enhancing the cytokine production by activated macrophages in vitro.

  • This study was performed to evaluate the effect of Sorghum bicolor L. Moench(Sorghum, su-su) extracts on mouse immune cell activation. As ex vivo experiment, different concentrations(0, 50, 500mg/kg B.W.) of Sorghum bicolor L. Moench water extracts were orally administrated into mouse every other day for four weeks. The proliferation of mouse splenocytes, the number of plaque forming cells(PFC) and the cytokine IL-1β production by activated macrophage were used as indices for immunocompetence. Splenocyte proliferation was enhanced in mouse orally administrated with 50mg/kg B.W./day concentration compared to that of control group. Especially, the highest proliferation of spleoncyte was seen in the mouse orally administrated at the concentration of 50mg/kg B.W./day. The number of plaque forming cells(PFC) to SRBC were significantly enhanced when compared with control group. Also, the mouse of Sorghum bicolor L. Moench water extracts 50mg/kg B.W./day supplementation group with LPS stimulation enhanced level of IL-1$\beta$ cytokine production. This study suggest that supplementation of Sorghum bicolor L. Moench water extracts may enhance the immune function by regulating the splenocytes proliferation, increasing the number of PFC and enhancing the cytokine production by activated macrophage.

제주 자생 새우란, 금새우란, 왕새우란의 분포와 외형적 특성에 관한 연구 (Studies on Distributions and Morphological Characteristics of Calanthe discolor, C. sieboldii, and C. bicolor Native to Cheju Province)

  • 한라산을 중심으로 새우란(Calanthe discolor)은 해발 120~600m 범위에서, 금새우란(C. sieboldii)은 300~600m에서 분포하였고, 왕새우란(C. bicolor)은 해발 300~400m의 범위에서 새우란과 금새우란이 동시에 분포하는 지역에서만 분포하였다. 엽(葉)과 화기(花器)의 형태적인 면에서 새우란이 가장 작았고 금새우란이 가장컸으며, 왕새우란은 그 중간형태를 나타내었다. 화색(花色)에서는 새우란은 적갈색 계통의 꽃잎과 흰색의 설판과 같은 단순한 색상을 나타내었고, 금새우란은 노란색계통만을 나타내었다. 반면 왕새우란은 두 종의 색이 혼합된 다양한 화색을 나타내었다. 따라서 왕새우란은 새우란과 금새우란의 자연 교잡에 의해 출현된 새로운 종으로 사료된다.

  • 제주 자생 새우란, 금새우란, 왕새우란의 분류를 위한 동위효소 분석과 RAPD법을 이용한 유전적 근연관계 조사 결과, peroxidase와 esterase의 동위효소(isozyme)분석에서 왕새우란의 밴드양상은 새우란과 금새우란이 동시에 나타나는 밴드에서만 밴드가 나타났다. RAPD를 이용한 유전적인 근연관계에서 왕새우란은 새우란과 금새우란 모두와 비교적 가까웠지만 새우란과 금새우란의 상호간 근연관계는 가장 멀었다. 즉, 왕새우란은 유전적인 근연관계에서 새우란과 금새우란의 중간적인 위치를 나타내었다. 따라서, 왕새우란은 새우란과 금새우란의 자연적인 교잡으로 출현된 새로운 종으로 사료된다.

하형 청예 사료작물의 생산성과 사료가치 비교 (Comparative Tield and Quality of Summe Annual Grasses as Fresh-cut Forage)

  • 본(本) 시험(試驗)은 새로 도입(導入)된 13개(個) 품종(品種)의 수수-수단그라스잡종(雜種)(Sorghum bicolor (L.) Moench), T-E Goldmaker 수수잡종(雜種) (S.bicolor (L.) Moench), $NC^+$ 88SS 수단그라스 잡종(雜種) (S. bicolor (L.) Moench), Supermill permillet(Pennisetum americanum (L.) Leeke) 및 teosinte(Euchlaena mexicana Schrad.)를 Pioneer 988 수수-수단그라스 잡종(雜種)을 대조 품종으로 하여 건물생산성(乾物生産性), 조단백질함량(粗蛋白質含量) in vitro 건물소화율(乾物消化率)(IVDMD) 및 그밖의 생육특성(生育特性)을 비교하여 농가(農家)에 새로운 자료(資料)를 제시하기 위하여 수행되었다. 건물수량(乾物收量)에 있어서 14품종간(品種間)의 유의성은 인정되지 않았으나 teosinte 및 TE Goldmaker는 수량이 가장 낮았다. 각(各) 품종간(品種間)에 다소의 차이는 있었으나 대부분의 품종(品種)들이 엽병(葉病)(잎마른병)에 강하였으며 pearlmillet과 teosinte는 전혀 엽(葉)에 이병이 안되었고 $NC^+$ 88SS는 엽고병(葉枯病)에 매우 약하였다. 수수-수단그라스계(系) 잡종(雜種)의 평균(平均) 조단백질(粗蛋白質) 함량(含量)은 pearlmillet과 수수잡종(雜種) 보다는 낮았으나 수단그라스 잡종(雜種)보다는 높았다. G-83F, TE Goldmaker, Teosinte 및 HW 5111의 IVDMD는 대조 품종인 P-988보다 높았으며 고온기(高溫期) 청예용(靑刈用) 사초(飼草)의 조단백질(粗蛋白質) 함량(含量)과 IVDMD간에는 일정한 상호관계(相互關係)가 없었다.

Rutin 동효물질의 국산천연약품자원에 관한 연구(I) -Lespedeza속 식물의 풀라보노이드 검색- (Studies on the Natural Resources of Rutin-like Constituents(I) -Screening for Flavonoids in the Genus Lespedeza Plants-)

  • In order to detect constituents with similar actions to rutin in the genus Lespedeza plants growing in Korea, twelve species were collected in Chung Buk and Kyung Ki Provinces and subjected to systematic extraction. When the extract was examined for flavonoid by adding flavonoid reagents, all the species gave positive cofirmatory tests. The examination of the flavonoid consitituents by T.L.C. gave the following results: as rutin-like constituents, quercetin was detected in L. cyrtobotrya, L. maritima and L. bicolor var. melanantha and eriodictin-like constituents were detected in seven species. Of those twelve species, five species, L. cuneata, L. cyrtobotrya, L. maritima, L. bicolor var. melanantha and L. patentibicolor were found to contain kaempferol. All of them appear to contain lespedin and eriodictiol.

조록싸리 종내분류군과 잡종의 분류학적 연구 (Taxonomic study on infraspecific taxa of Lespedeza maximowiczii and hybrids with related species)

  • 조록싸리(Lespedeza maximowiczii) 내에 많은 종내분류군들 및 근연분류군과의 잡종들이 기록되었지만, 이들의 분류학적 실체에 대해서는 학자들간에 이견이 있다. 본 연구는 조록싸리와 근연분류군간 잡종인 지리산싸리(L. chiisanensis), 진도싸리(L. patentibicolor) 및 조록싸리의 종내분류군인 털조록싸리(var. tomentella), 늦싸리(var. elongata), 삼색싸리(var. tricolor)의 실체를 규명하기 위해, 외부형태를 관찰하고 microsatellite를 이용하여 유전적 구조를 분석하였다. 연구 결과 털조록싸리와 늦싸리의 꽃과 잎의 형태는 조록싸리의 변이폭에 속했고, 유전적으로도 두 분류군이 조록싸리와 같이 묶였다. 삼색싸리는 꽃의 구조와 색에서 근연종인 L. buergeri와 유사한 반면 소엽 정단부와 소포엽의 형태는 조록싸리와 가까웠다. 유전적 구조에서도 삼색싸리의 개체들은 조록싸리와 L. buergeri의 계통이 혼합된 형태를 보이거나(K = 3), 독립되었다(K = 5). 잡종들의 경우, 지리산싸리의 꽃과 잎의 형태는 조록싸리와 구별되지 않는 반면, 진도싸리의 꽃 형질은 싸리(L. bicolor)의 변이폭에 속했는데, 유전적 계통도 각각 조록싸리와 싸리로 지정되었다. 본 연구결과를 종합하여 저자들은 털조록싸리와 늦싸리를 조록싸리의 품종, f. friebeana (Schindl.) D. P. Jin, J. W. Park & B. H. Choi로 처리하였는데, 이는 L. friebeana가 var. tomentella 보다 먼저 발표되었기 때문이다. 또한 삼색싸리는 독립종, L. tricolor (Nakai) D. P. Jin, J. W. Park & B. H. Choi 로 처리하였고, 지리산싸리는 조록싸리의 이명으로 처리하였다. 한편 진도싸리는 싸리의 이명으로 판단된다.

싸리꽃 에탄올 추출물의 피부(皮膚) 생리활성(生理活性) 효과 (Dermal Bioactive Properties of the Ethanol Extract from Flowers of Lespedeza bicolor)

  • Objective : In this study, the ethanol extract from flowers of Lespedeza bicolor was investigated for their dermal bioactive properties related to cosmeceuticals such as depigmentation and radical scavenging effect. Results : The ethanol extract from flowers of Lespedeza bicolor showed considerable radical scavenging activity ($SC_{50}:\;10\;{\mu}g/ml$) and inhibited the production of nitric oxide (NO) in Raw 264.7 macrophages activated with the endotoxin lipopolysaccharide (LPS). Although the proliferation of B16/F10 cells was slightly decreased by the ethanol extract from flowers of Lespedeza bicolor at the concentration of $100\;{\mu}g/ml$, it did not appear necrosis. The ethanol extract from flowers of Lespedeza bicolor down-regulated melanin formation effectively. Methods : The free radical scavenging activity of Lespedeza bicolor was assayed in cell free systems using a stable free radical, 1,l-diphenyl-2-picrylhydrazyl (DPPH). Nitrite accumulated in culture medium was measured as an indicator of NO production using a Griess reaction. Cell viability was measured by MTT assay and melanin content was assessed using the method of Hosei with some modifications. Conclusions : These results suggest that the ethanol extract from flowers of Lespedeza bicolor is a potent depigmetation agent and it may be a candidate for antioxidant and anti-inflammatory agent.
