Testosterone Relaxes Rabbit Seminal Vesicle by Calcium Channel Inhibition

  • Recent studies have documented that testosterone relaxes several smooth muscles by modulating $K^+$ channel activities. Smooth muscles of seminal vesicles playa fundamental role in ejaculation, which might involve testosterone. This study was aimed to assess the role of testosterone in seminal vesicular motility by studying its effects on contractile agents and on the ion channels of single vesicular myocytes in a rabbit model. The contractile responses of circular smooth muscle strips of rabbit seminal vesicles to norepinephrine ($10{\mu}M$), a high concentration of KCI (70 mM), and testosterone ($10{\mu}M$) were observed. Single vesicular myocytes of rabbit were isolated using proteolytic enzymes including collagenase and papain. Inside-out, attached, and whole-cell configurations were examined using the patch clamp technique. The applications of $10{\mu}M$ norepinephrine or 70 mM KCl induced tonic contractions, and $10{\mu}M$ testosterone (pharmacological concentration) evoked dose-dependent relaxations of these precontracted strips. Various $K^+$ channel blockers, such as tetraethylammonium (TEA; $10{\mu}M$), iberiotoxin ($0.1{\mu}M$), 4-aminopyridine (4-AP, $10{\mu}M$), or glibenclamide ($10{\mu}M$) rarely affected these relaxations. Single channel data (of inside-out and attached configurations) of BK channel activity were also hardly affected by testosterone ($10{\mu}M$). On the other hand, however, testosterone reduced L-type $Ca^{2+}$ currents significantly, and found to induce acute relaxation of seminal vesicular smooth muscle and this was mediated, at least in part, by $Ca^{2+}$ current inhibition in rabbit.

  • 일본인 전문식물재집자인 T. Uchiyama는 1900년과 1902년 두 번에 걸쳐 한반도의 식물채집을 실시하였다. T. Uchiyama의 수집물은 일본인으로서는 최초의 한반도 식물에 대한 채집활동의 결과로 생각되며, 당시 채집된 식물표본은 T. Nakai에 의해 Flora Koreana와 H. $L{\acute{e}}veill{\acute{e}}$ 연구 재료로 사용되었다. 본 연구는 동경대학 식물표본관에 소장된 T. Uchiyama의 채집표본과 문헌을 중심으로 당시의 채집 경로와 일정을 확인하여 채집일과 장소를 날짜순으로 정리하였으며 표본에 채집자에 의해 직접 기록된 자료와 Nakai에 의해 표기된 지명을 제공하였다. 문헌을 근간으로 추정된 전체 채집 표본수는 약 1.674개로 확인되나 현재 동경대학 표본관에 소장된 표본은 200여점에 불과하여 상당한 차이를 보인다. 표본의 2/3는 두 번째 채집인 1902년에 수집되었으며, Nakai는 이중 41개의 표본을 새로 기재한 종의 기준표본으로 사용하였다.

시판 가공식품의 영양표시 실태 모니터링에 관한 연구 (Monitoring the Current Nutrition Labeling Practice on the Packaging of Processed Foods)

  • The purpose of this study was to monitor the current nutrition labeling practice on the packaging of the processed foods providing consumers with reliable nutrition information, which has been considered as a useful aid for food selection and a potent educational tool for nutrition in a daily life. The 2,214 processed foods purchased at the a wholesale market in July, 2003, were divided by food categories issued from the 2003 food codes and assessed in the terms of the nutrition composition labeling and nutrition claims. Nutrition composition labeling was found on 533 out of the 2,214 processed foods items. The findings of this study were as follows : Milk and dairy products had 66.4% of nutrition composition labeling, which was the largest percentage among the food categories. Tables format (71.3%) is the most widely used type of nutrition labeling. The title of nutrition composition labeling used the most widely was nutrition composition (71.3%). Nutrition composition included many different ways of expression, such as a table of nutrition composition, and indication of nutrition composition, etc. The expression unit of the nutrition composition labeling was use per 100g or 100mL (40.5%). The processed foods with nutrition claims were 18.0% (399 items). Nutrition claims were divided into two categories: nutrient content claims and nutrient comparative claims. The most frequently used nutrient content claims were contained (43.9%) and more or plus in the nutrient comparative claims (16.0%). Ca was the most popular item as a nutrition claim nutrient (33.6%).

시판 가공식품의 영양성분 및 영양강조 표시 실태에 대한 연구 (A Study on Nutrition Composition Labeling and Nutrition Claim Practices for the Processed Foods)

  • The purpose of this study was to investigate the status of current nutrition labeling on the packaging of the processed foods that provide consumers with a reliable and consistent source of information, which has been considered as a useful aid for food selection and a potent educational tool for nutrition in daily life. The 2,160 processed foods purchased at the supermarket on September, 2002, were divided by food category issued from the 2002 food codes and assessed in the terms of the nutrition composition labeling and nutrition claims. Nutrition composition labeling was found on 356 of the 2160 processed foods items. Milk and dairy products had 49.7% of nutrition composition labeling, which was the largest number among the food category. Tables were most frequently used as the type of nutrition composition labeling (79.8%). Nutrition composition including many different ways of expression, such as a table of nutrition composition, indication of nutrition composition, analysis table of nutrition composition and comparative table of nutrition composition, made frequent use of nutrition composition labeling titles (78.7%). The various unit of measures were use in the nutrition labeling of the processed foods, per l00g or 100$m\ell$ was the highest (44.6%) under the currently practiced nutrition labeling. The correct labeling standard with nutrient content and % RDA except energy, was used on 47.8% of labels, and those with only liability indication nutrient and liability indication nutrients plus discretion indication nutrients were 25.3 and 22.5% respectively. The processed foods with nutrition claims were 8.0% (172 items). Nutrition claims were divided in two ways: nutrient content claims and nutrient comparative claims. The most frequently used claims were contained in the former (44.4%) and more or plus in the latter case (16.3%). Ca was the most popular item as a nutrition claim nutrient (50.6%).

Korean Red Ginseng aqueous extract improves markers of mucociliary clearance by stimulating chloride secretion

  • Background: Abnormal chloride (Cl-) transport has a detrimental impact on mucociliary clearance in both cystic fibrosis (CF) and non-CF chronic rhinosinusitis. Ginseng is a medicinal plant noted to have anti-inflammatory and antimicrobial properties. The present study aims to assess the capability of red ginseng aqueous extract (RGAE) to promote transepithelial Cl- secretion in nasal epithelium. Methods: Primary murine nasal septal epithelial (MNSE) [wild-type (WT) and transgenic CFTR-/-], fisher-rat-thyroid (FRT) cells expressing human WT CFTR, and TMEM16A-expressing human embryonic kidney cultures were utilized for the present experiments. Ciliary beat frequency (CBF) and airway surface liquid (ASL) depth measurements were performed using micro-optical coherence tomography (μOCT). Mechanisms underlying transepithelial Cl- transport were determined using pharmacologic manipulation in Ussing chambers and whole-cell patch clamp analysis. Results: RGAE (at 30㎍/mL of ginsenosides) significantly increased Cl- transport [measured as change in short-circuit current (ΔISC = ㎂/㎠)] when compared with control in WT and CFTR-/- MNSE (WT vs control = 49.8±2.6 vs 0.1+/-0.2, CFTR-/- = 33.5±1.5 vs 0.2±0.3, p < 0.0001). In FRT cells, the CFTR-mediated ΔISC attributed to RGAE was small (6.8 ± 2.5 vs control, 0.03 ± 0.01, p < 0.05). In patch clamp, TMEM16A-mediated currents were markedly improved with co-administration of RGAE and uridine 5-triphosphate (8406.3 +/- 807.7 pA) over uridine 5-triphosphate (3524.1 +/- 292.4 pA) or RGAE alone (465.2 +/- 90.7 pA) (p < 0.0001). ASL and CBF were significantly greater with RGAE (6.2+/-0.3 ㎛ vs control, 3.9+/-0.09 ㎛; 10.4+/-0.3 Hz vs control, 7.3 ± 0.2 Hz; p < 0.0001) in MNSE. Conclusion: RGAE augments ASL depth and CBF by stimulating Cl- secretion through CaCC, which suggests therapeutic potential in both CF and non-CF chronic rhinosinusitis.

The Effects of Vitamin D Supplementation to Peak-producing Hens Fed Diets Differing in Fat Source and Level on Laying Performance, Metabolic Profile, and Egg Quality

  • This experiment was designed to examine the effects of supplemental vitamin D on laying performance, metabolic profile and egg quality of hens fed diets containing different fat sources and levels. Lohman strains (n = 480) were assigned to one of 10 diets: basal diet (BD), BD plus 2.5 and 5.0% sunflower oil (SO) or tallow (T) at vitamin D provided $1{\times}$ and $3{\times}$ of the current recommendation. The experiment lasted from week 30 to 44 of age. Each diet was tested in 12 replicate cages of 4 hens. Production, metabolism, and egg quality data were subjected to three-way ANOVA. Both fats decreased feed intake (FI) as compared to BD. Increasing SO and T levels linearly decreased and quadratically increased FI, respectively. The dietary factors did not affect egg production (EP) and egg weight. Vitamin D supplementation increased and decreased EP when diets contained SO and T, respectively. Feed conversion efficiency (FCE) for hens fed SO was lower than for hens fed T. However, increasing T level improved FCE, whereas increasing SO level worsened FCR. Vitamin D supplementation increased serum vitamin D and glucose concentrations. Vitamin D supplementation also caused a decrease and an increase in serum vitamin D concentration when diets contained SO and T, respectively. Serum glucose concentration for hens fed SO was lower than hens fed T. Increasing fat level linearly increased serum triglyceride and VLDL concentrations, regardless of the fat type. Increasing SO level linearly decreased serum cholesterol concentration. Vitamin D supplementation did not alter lipid metabolites. The dietary factors did not affect serum total protein, Ca, and P concentrations. As compared with BD, feeding SO decreased dry tibia and ash weights more than feeding T. Vitamin D supplementation tended to increase dry tibia weight and decrease tibia ash weight. Eggshell strength and thickness, yolk and albumen indexes, and Haugh unit were not responsive to the dietary factors. Eggshell strength quadratically increased with increasing T level. Yolk color for hens fed SO was lower than for hens fed T. The dietary factors did not affect most of yolk fatty acids. Increasing SO level quadratically decreased yolk $C_{18:2}$ concentration. Vitamin D supplementation increased and decreased yolk $C_{18:2}$ concentration when diets contained SO and T, respectively. In conclusion, increasing fat level improved laying performance without altering metabolic profile and egg quality. Vitamin D supplementation had minor alteration effects on laying performance, metabolic profile, and egg quality in response to fat feeding.
