Effect of Temperature and Salinity on Production of Resting Egg in Korean Rotifer, Brachionus plicatilis (L and S-type)

  • Production of resting egg from the Korean rotifer, Brachiunus plicatilis (L and S-type) was investigated at different temperatures (L-type : 20, 24, $28^{\circ}C$, S-type : 28 32, $36^{\circ}C$) and salinities (10, 20,30 ppt). The rotifer was cultured in 25 ml test tube and fed on Nannochloris oculata. With regard to mixis rate, L-type rotifer showed higher rate at lower temperature, and the highest rate was observed at 20 ppt of salinity at each temperature of the experiment. However, for S-type rotifer, the optimum temperature and salinity were $28\~32^{\circ}C$ and 20 ppt, respectively. The highest number of resting egg was 173 eggs/ml in 16 days at $24^{\circ}C$, 10 ppt for L-type rotifer and 410 eggs/ml in 14 days at $28^{\circ}C$, 10 ppt for S-type rotifer. The maximum number of resting egg produced per 10,000 rotifers was 8,122 eggs at $20^{\circ}C$, 20 ppt for L-type rotifer and 8,700 eggs at $28^{\circ}C$, 20 ppt for S-type rotifer. The maximum number of resting egg produced $10^8$ cells of N. oculata was 50.7 eggs for L-type rotifer ($24^{\circ}C$, 20 ppt) and 79.6 eggs for S-type rotifer ($32^{\circ}C$, 10 ppt). The number of resting egg produced per day was $1\~11$ eggs/ml for L-type rotifers and $21\~35$ eggs/ml for S-type rotifer in 9 combination experiments. In this study, S-type rotifer is better than L-type rotifer in resting egg production, and the optimum temperature and salinity for resting egg production were $20^{\circ}C$, 20 ppt for L-type rotifer and $28^{\circ}C$, 20 ppt for S-type rotifer. This result shows the difference of Korean rotifer in the optimum condition for resting egg production from other rotifers reported earlier.

Size and Resting Egg Formation of Korean Rotifer, Brachionus plicatilis and B. calyciflorus (한국산 Rotifer, Brachinus plicatilis와 B. calyciflorus의 크기 및 내구란 형성)

  • Sixteen strains of marine rotifer, Braohionus plicatilis were isolated from salt pond, estuary and lagoon. Among 16 strains, 2 strains were large (L)-type and the others were small (S) or ultra small (US)-type. Four strains of fresh water rotifer, B. calyciflorus were isolated from commercial fish ponds. The size of lorica and resting egg were measured. In B. plicatilis, the range of lorica length from S-type and S-type were $244.3{\~}255.3\;{\mu}m$ and $131.0{\~}165.8\;{\mu}m$, respectively. The major axis of resting egg in the marine rotifer were $93.7\~116.4\;{\mu}m$ for S-type and $142.4{\~}145.5\;{mu}m$ for L-type, respectively. In freshwater rotifer, B. calyciflorus, the size range of lorica and major axis of resting egg were $211.8\~229.9\;{\mu}m$ and $126.8\~140.2\;{mu}m$, respectively. The size of freshwater rotifer was larger than that of S-type marine rotifer, but smaller than that of L-type one. Growth and formation of resting egg of B. plicatilis were different among the strains. The maximum density of S-type and L-type rotifer was 753.3 inds./ml for H-S strain and 220 inds./ml for O-L strain, respectively. The largest production of resting egg of S-type and L-type rotifer were 86.7 inds./ml for YY-S strain and 45.8 inds./ml for O-L strain, respectively.

Effects of Density, Temperature, Size, Grain Angle of Wood Materials on Nondestructive Moisture Meters

  • The aim of this study was to investigate the effects of density, temperature, size, and grain direction on measurement of moisture contents (MC) of wood materials non-destructively. The MC of different sizes of solid wood, glulam, and CLT from larch (larix kaempferi, $560kg/m^3$) and pine (pinus koraiensis, $430kg/m^3$) were measured using the dielectric type and resistance type meters. The specimens were conditioned in the environmental chamber to be equilibrium moisture content (EMC) of 12 % and 19 %. When density setting in dielectric type meter was increased from $400kg/m^3$ to $600kg/m^3$, the MCs of specimen (S-L-100-E) were decreased from 13.4 % to 11.3 %. However, when wood group (WG) setting in resistance type meter was changed from WG1 to WG4, the measured MCs were increased from 9.2 % to 12.3 %. When temperature setting in resistance type meters was changed from 0 to $35^{\circ}C$, the MC was decreased from 17.0 % to 13.0 %. The MCs measured by dielectric type meter for larger specimens (S-L-100-E_11.3 %, G-L-240-E_11.7 % and C-L-120-E_12.8 %) were higher than those of small size specimens (S-L-30-E_8.7 %, G-L-150-E_10.3 %, and C-L-90-E_9.7 %). The MCs measured by resistance type meter for larger specimens (G-L-240-E_11.6 % and C-L-120-E_13.3 %) were also higher than those of small size specimens (G-L-150-E_10.4 %, and C-L-90-E_11.8 %). The resistance type meter was not affected by the grain direction but the dielectric type meter were affected by the grain direction. The MC measured by resistance type meter for G-L-120-E perpendicular to grain direction was 11.5 % and the measured MC parallel to grain direction was 11.3 %. The MC measured by dielectric type meter parallel to grain direction (12.1 %) was higher than that measured perpendicular to grain direction (10.7 %).

Efficient Plant Regeneration Using Mature Seed-Derived Callus in Zoysiagrass (Zoysia japonica Steud.) (성숙종자 유래 캘러스를 이용한 들잔디 (Zoysia japonica Steud.)의 효과적인 식물체 재분화)

  • Using mature seed-derived callus, optimal conditions for efficient callus growth and plant regeneration, and regeneration efficiency by callus type were investigated in zoysiagrass (Zoysia japonica steud.). Callus induction was highest when the seeds were cultured on MS medium containing 2 mg/L 2,4-D, 0.2 mg/L BAP, 4 mg/L thiamine-HCl and 100 mg/L $\alpha$-ketoglutaric acid. Callus growth was highest when callus were cultured on MS medium containing 0.5 mg/L 2,4-D, 0.05 mg/L BAP, 4 mg/L thiamine-HCl and 100 mg/L $\alpha$-ketoglutaric acid. Plant regeneration was highest when callus was transferred on MS medium containing 3% maltose and 1 mg/L BAP, or 1 mg/L thidiazuron (TDZ). The combinations and concentrations of 2,4-D and BAP were shown to be critical factors for both the frequency and the type of callus. And four morphologically distinct types of callus were induced from the 2,4-D and BAP treatment. Type I,II and III calli produced shoots upon subculture, while the watery callus, type IV produced roots without shoots. Of four types of callus, type I exhibited the maximum frequency (82%) of shoot regeneration and the minimum frequency (4%) of albinism.

A Study on the Mechanical Properties and Deformed Layer of STS 316L and 316LN Stainless Steels (STS 316L과 316LN 강의 고온 기계적 특성 및 가공 변질층에 관한 연구)

  • The deformed layers generated in face milling works were comparatively investigated to type 316L and nitrogen(N)-added type 316LN stainless steels. In order to characterize mechanical properties between type 316L and type 316LN, high-temperature tensile tests were conducted with different temperatures: R.T to $700^{\circ}C$. The cutting forces of three components, Fx, Fy and Fz were measured using a tool dynamometer through the face milling cutting tests. The deformed layers were measured by micro-hardness tests along deformed layers. The results of mechanical properties showed that type 316LN was superior to type 316L. The deformed layers of two steels were generated in the 1501m-3001m ranges, and type 316L was higher than type 316LN. The reason for this is due to the high strength properties by nitrogen effect. It was found that deformed structures were well observed for type 316L, but were minutely observed for type 316LN in this cutting conditions.

Shaking Table Tests for Evaluation of Seismic Performance of L-type Caisson Quay Walls (L형 케이슨 안벽 구조물의 내진성 평가를 위한 진동대 시험)

  • Shaking table tests and pseudo-static analysis were performed, in this study, on newly-designed aseismatic L-type caisson quay walls, which were constructed by extending the bottom plate of gravity quay walls into the backfill soil. The L-type quay walls are expected to give economical benefits by reducing the cross-sectional area of the wall while maintaining its aseismatic efficiency as much as the classical caisson gravity quay wall. To confirm the effectiveness of the L-type structure, the geometry of L-type quay walls were varied for shaking table tests. And, to verify the influence of backfill soils on the seismic behavior of quay walls, additional shaking table tests were performed on the L-type quay wall after the backfill soils were replaced by gravels and light materials. As a result, it was found that L-type caisson quay walls are good earthquake resistant structures but increasing the length of bottom plate did not proportionally increase the effectiveness of the structure in its aseismatic performance. Replacing the backfill soils by the gravels and light materials, contrary to our expectation, was not an effective measure in improving the seismic performance of L-type caisson quay wall.

  • It is well known that $L_2(m)$ type PBIB designs have the Property A, so they are BNAS PBIB designs. However, $L_3(m)$ type PBIB designs are not of type of Property A but do have the factorial structure (Cotter, John, and Smith(1973)). In this paper, the properties of the $L_3(m)$ type PBIB designs are investigated. Extended Property A and fractional BNAS are defined and a solution formula for the treatment effects in the $L_32(m)$ type designs is obtained.
