Hydrodynamic Characteristics of Self-expandable Graft Stents in Steady Flow (정상유동에서 자가팽창성 그래프트 스텐트의 수력학적 특성)

  • This experimental study is aimed at evaluating the hydrodynamic performance of newly designed self-expandable graft stents under steady flow condition. Two graft stents with different coating materials and a bare TiNi metallic stent for comparison test were used in the experiment. Pressure variation and velocity distribution at the upstream and downstream of the stents were measured at flow rates of 5, 10, and 15 l/min, respectively. Pressure loss due to insertion of the stent increased with increasing flow rate exponentially as expected. At a flow rate of 15 l/min, pressure loss of Polyure-thane(PU)-coated graft stent was 6 times higher than that of TiNi metallic stent, while the pressure loss of a porous Polytetrafluoroethylene(PTFE)-coated graft stent was comparable to a bare TiNi metallic stent. Velocity profiles of the porous PTFE-coated graft stent were similar to those of a bare TiNi metallic stent regardless of flow rate. Furthermore, the velocity profile of PU-coated graft stent revealed an asymmetrical and relatively low central velocity at a higher flow rate than 10 1/min, expecially, where the effects resulted in increases of wall shear stress and normal stress. The worse hydrodynamic behavior of PU-coated graft stent than the other two stents might be attributed to formation of folds due to poor flexibility of coated material when inserting the graft stent into the pipe with a more smaller size, which later gave rise non-symmetry of flow area, increase of surface roughness and jet flow via the crevice between the stent and cylinder wall.

Bioequivalence of Kuhnil Propiverine Hydrochloride Tablet to BUP-4 Tablet (Propiverine Hydrochloride 20 mg) (비유피-4 정(염산프로피베린 20 mg)에 대한 건일염산프로피베린 정의 생물학적동등성)

  • The purpose of the present study was to evaluate the bioequivalence of two propiverine hydrochloride tablets, BUP-4 (Jeil Pharm. Co., Ltd.) and Kuhnil Propiverine Hydrochloride (Kuhnil Pharm. Co., Ltd.), according to the guidelines of the Korea Food and Drug Administration (KFDA). The propiverine release from the two propiverine hydrochloride formulations in vitro was tested using KP VIII Apparatus II method with a variety of dissolution media (pH 1.2, 4.0, 6.8 buffer solutions, water and blend of polysorbate 80 into pH 6.8). Twenty six healthy male subjects, $23.73{\pm}2.79$ years in age and $67.04{\pm}7.93\;kg$ in body weight, were divided into two groups and a randomized $2\;{\times}\;2$ cross-over study was employed. After one tablet containing 20 mg as propiverine hydrochloride was orally administered, blood was taken at predetermined time intervals and the concentrations of propiverine in serum were determined using HPLC method with UV detector. The dissolution profiles of two formulations were similar at all dissolution media. Besides, the pharmacokinetic parameters such as $AUC_t,\;C_{max}\;and\;T_{max}$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters using logarithmically transformed $AUC_t,\;C_{max}$ and untransformed $T_{max}$. The results showed that the differences between two formulations based on the BUP-4 were 0.17%, 7.98% and 4.55% for $AUC_t,\;C_{max}\;and\;T_{max}$. respectively. There were no sequence effects between two formulations in these parameters. The 90% confidence intervals using logarithmically transformed data were within the acceptance range of log(0.8) to log(1.25) $(e.g.,\;log(0.88){\sim}log(1.l2)\;and\;log(0.90){\sim}log(1.l5)\;for\;AUC_t\;and\;C_{max},\;respectively)$. Thus, the criteria of the KFDA bioequivalence guideline were satisfied, indicating Kuhnil Propiverine Hydrochloride tablet was bioequivalent to BUP-4 tablet.

Soil CEC for Textural Classes in Korea (우리나라 토양(土壤)의 토성별(土性別) 양(陽)이온 치환용량(置換容量))

  • Mean values and the relative contribution of OM and clay to total CEC for soil textural classes were obtained from the analytical results of the typical profiles(sample size : 3,182) which were described by the detailed soil maps througthout Korea with an exception of Jeju island. The results are below. 1. Mean values of the soil CEC were 2.9 for S, 4.7 for LS, 6.7 for SL, 9.0 for L, 10.2 for SiL, 10.7 for CL, 8.6 for SCL, 12.2 for SiCL, 16.1 for SiC, and 17.4me/100gr for C, respectively. 2. The multiple regression equation and partial regression coefficient tended to show that OM and clay had the highly significant effect on CEC. 3. Clay content of the coarse, moderately coarse, and moderately fine soil was 1.10 to 1.89 times as important as OM content whereas OM of the medium and fine soil 1.09 to 2.94 times as important as clay in predicting CEC. 4. Mean values of CEC of the humus and clay in Korean soils were about 62.9 and 24.0me/100gr, respectively.

  • The pharmacokinetics of haloperidol were determined after single oral and intravenous doses in 13 male schizophrenic patients. Plasma concentrations of haloperidol(HP) and reduced haloperidol(RH) were measured by high performance liquid chromatography. Plasma concentration data obtained were analyzed by obth model dependent (one-or two exponential decay models using nonlinear regression) and model independent (AUC and first moment curve) approaches. The two methods were found to be in close results. After intravenous injections of HP in 8 patients (10 mg/man), the mean central and peripheral volume of distributions were $2.85\;{\pm}\;1.70$ and $8.09\;{\pm}\;2.10\;l/kg$, respectively, and mean steady state volume of distribution was $11.87\;{\pm}\;3.21\;l/kg$. Mean clearance, MRT and elimination half life were $12.39\;{\pm}\;3.25\;ml/min/kg$, $925.10\;{\pm}\;166.79\;min$ and $676.35\;{\pm}\;126.45\;min$, respectively. After oral administrations of HP in 5 patients, mean peak time and peak concentration were $217.63\;{\pm}\;61.60\;min$ and $9.77\;{\pm}\;2.92\;ng/ml$, respectively. Mean MRT and elimination half life were $1112.23\;{\pm}\;131.73\;min$ and $724.02\;{\pm}\;120.03\;min$, respectively, and these parameters were not significantly different from those of intravenous injection of HP. Absolute bioavailability of HP oral product was found to be about 44%. The profiles of plasma RH concentration-time curves after oral or intravenous doses of HP were similar. Also it was found that the elimination rate of RH was solwer than that of HP by comparing the slopes of plasma concentration-time curves of HP and RH.

Physicochemical characteristics and fatty acid profiles of muscle tissues from Hanwoo steers fed a total mixed ration supplied with medicinal plant by-products

  • Objective: Using medicinal plant by-products (MPBP) as feed additives may be an eco-friendly option as substitutes for feedstuffs and may assist in reducing the improper disposal of MPBP. Therefore, this study was conducted to evaluate the influences of MPBP on the meat quality of Hanwoo steers fed a total mixed ration (TMR). Methods: Twenty seven steers (body weight = $573{\pm}57kg$) were randomly divided into three treatments with a control group and two tested groups as follows: control, 1,000 g/kg TMR; treatment 1 (MPBP30), 970 g/kg TMR and 30 g/kg MPBP; treatment 2 (MPBP50), 950 g/kg TMR and 50 g/kg MPBP. Results: Average daily gain, feed conversion ratio and the Commission Internationale de l'Eclairage $L^*$ of muscle were improved (p<0.05, respectively) by MPBP30. Stearic acid ($C_{18:0}$) was decreased (linear effect, p = 0.012), while oleic acid ($C_{18:1}$) was increased (linear effect, p = 0.055) by MPBP level. Saturated fatty acid (SFA) and polyunsaturated fatty acid (PUFA) were decreased for MPBP50 while unsaturated fatty acid (USFA) and monounsaturated fatty acid (MUFA) were increased for MPBP 50. USFA and SFA ratio was increased for MPBP50 as well. Conclusion: These results indicated that MPBP supplementation in Hanwoo steers fed a TMR increased feed efficiency and meat color (lightness) with altering fatty acid proportions. Therefore, MPBP may be successfully used in ruminant feeding.

Wet Oxidation of Phenol with Homogeneous Catalysts (균일촉매를 이용한 페놀의 습식산화)

  • The wet oxidation of phenol has been investigated at temperatures from 150 to $250^{\circ}C$ and oxygen partial pressures from 25.8 to 75.0 bar with initial pH of 1.0 to 12.0 and initial phenol concentration of 10 g/l. Chemical Oxygen Demand COD has bee measured to estimate the oxidation rate. Reaction intermediates have been identified and their concentration profiles have been determined using liquid chromatography. The destruction rate of phenol have shown the first-order kinetics with respect to phenol and the changes in COD during wet oxidation have been described well with the lumped model. The impact of various homogeneous catalysts, such as $Cu^{2+}$, $Fe^{2+}$, $Zn^{2+}$, $Co^{2+}$ and $Ce^{3+}$ ions, on the destruction rate of phenol and COD has also been studied. The homogeneous catalyst of $CuSO_4$ has been found to be the most effective for the destruction of phenol and COD during wet oxidations. The destruction rate of formic acid formed during wet oxidations of phenol have increased as increasing temperature and $CuSO_4$ concentration. The final concentrations of acetic acid which has been formed during wet oxidations and difficult to oxidize have increased with reaction temperature and with decrease in the catalyst load.

Isolation of Probiotic Piliated Lactobacillus rhamnosus Strains from Human Fecal Microbiota Using SpaA Antiserum-Based Colony Immunoblotting

  • Piliated Lactobacillus rhamnosus (pLR) strains possess higher adherent capacity than non-piliated strains. The objective of this study was to isolate and characterize probiotic pLR strains in human fecal samples. To this end, mouse polyclonal antiserum (anti-SpaA) against the recombinant pilus protein (SpaA) of L. rhamnosus strain GG (LGG) was prepared and tested for its reactivity and specificity. With the anti-SpaA, a method combining the de Man, Rogosa, and Sharpe (MRS) agar plating separation and colony immunoblotting (CIB) was developed to isolate pLR from 124 human fecal samples. The genetic and phenotypic characteristics of the resultant pLR isolates were compared by randomly amplified polymorphic DNA (RAPD) fingerprinting, and examination of adhesion to Caco-2 cells, hydrophobicity, autoaggregation, and in vitro gastrointestinal tolerance. Anti-SpaA specifically reacted with three pLR strains of 25 test strains, as assessed by western blotting, immunofluorescence flow cytometry, and immunoelectron microscopy (IEM) assays. The optimized MRS agar separation plus anti-SpaA-based CIB procedure could quantitatively detect $2.5{\times}10^3CFU/ml$ of pLR colonies spiked in $10^6CFU/ml$ of background bacteria. Eight pLR strains were identified in 124 human fecal samples, and were confirmed by 16S RNA gene sequencing and IEM identification. RAPD fingerprinting of the pLR strains revealed seven different patterns, of which only two isolates from infants showed the same RAPD profiles with LGG. Strain PLR06 was obtained with high adhesion and autoaggregation activities, hydrophobicity, and gastrointestinal tolerance. Anti-SpaA-based CIB is a rapid and inexpensive method for the preliminary screening of novel adherent L. rhamnosus strains for commercial purposes.

Geomagnetic Field Properties and Magnetic Interpretation in the Southern Part of the Ulleung Basin (鬱陵盆地 남단해역의 地磁場 特性 및 磁氣異常 解析)

  • Marine total magnetic intensity over the southern part of the Ulleung Basin and geomagnetic data measured at a land base station are analyzed. Fourteen days observation of geomagnetic field at a fixed on-land base station showed how the geomagnetic field around the study area behaves. geomagnetic data at the base station can also be used as correction data for a diurnal variation. Magnetic anomalies in the study area do not reflect an effect of sea bottom topography but mainly subsurface basement. The southern part of the Ulleung Basin can be devided into two zones according to a different anomaly pattern; along the coastal shelves the isolated anomalies with a short wave and a strong amplitude are dominant, and toward the open sea the anomalies become much more subdued. The high anomaly zone adjoined to land is interpreted to be caused by granitic intrusives or volcanic rocks, and the weak anomaly zone to the outer sea to be arisen from an existence of deep basement. A spectrum analysis is applied to estimate magnetic basement depths from three anomaly profiles with a long period and a weak amplitude toward the outer sea. The calculated depths are 7.0km, 5.0km, and 2.6km respectively from outer profile. The basement might be correlated with the mixed layer of tuff, basalt, and sediment, which had been defined as L-2 layer in the Yamato basin and the Japan Basin.

Pharmacokinetics of oxytetracycline in rabbits after intravenous and intramuscular administrations (토끼에서 oxytetracycline의 정맥 및 근육 투여시의 약물동태학)

  • The study was carried out to determine the pharmacokinetic parameters after intravenous(iv) and intramuscular(im) administration (10mg/kr) in healthy rabbits. The results obtained through the experiments were summarized as follows: 1. Bioassay (Bacillus cereus 11778) was evaluated very useful for the determination of oxytetracycline(OTC) in the rabbit serum and tissues, with the detection limit of $0.125{\mu}g/ml$. 2. The pharmacokinetic profiles of OTC (10mg/kg, iv) in rabbits were best described with a two compartment open model $(C=29.5e^{-4,3t}{\pm}3.6^{-0.2t})$, whereas that of OTC (10mg/kg, im) showed a one compartment curve fitting. 3. Following iv administration, a rapid distribution phase was predominant [$t_{\frac{1}{2}}({\alpha}):1.43{\pm}0.98hr$ (♂), $0.5{\pm}0.1hr$(♀)], and then more slow elimination phase ensued [$t_{\frac{1}{2}}({\beta}):4.52{\pm}0.76hr$(♂), $7.32{\pm}2.52hr$(♀)]. Other computer generated pharmacokinetic values were as follows:C1 [$67.76{\pm}18.59ml/kg/h$(♂), $76.03{\pm}22.98ml/kg/h$ (♀)] Vd [$257.74{\pm}180.47ml/kg$ (♂), $92.33{\pm}23.62$ (♀)] AUC [$25.6{\pm}4.44mgh/L$ (♂), $39.6{\pm}12.13mgh/l$ (♀)]. There were no statistical significance between both sexes for all the parameters at the confidence level of 95%. 4. After im administaration, the absorption from the injection sites was very rapid [ Ka:$0.18{\pm}0.03h^{-1}$ (♂), $0.24{\pm}0.02h^{-1}$ (♀)] followed by a monoexponential elimination fashion. The time to peak blood level (Tmax) were calculated $1.64{\pm}0.15hr$ and $1.34{\pm}0.24hr$, in the male and female, respectively. The peak levels (Cmax) at the corresponding time were $1.69{\pm}0.23{\mu}g/ml$ (♂) and $2.08{\pm}0.16{\mu}g/ml$ (♀), with no statistical differences (p>0.05).

Effect of Gamma Irradiation on the Conformational Changes of Garlic (Allium sativum) Protein, Allivin (감마선 조사가 마늘(Allium sativum) 단백질 Allivin의 구조적 변화에 미치는 영향)

  • Conformational changes of allivin, a major allergen protein in garlic, by gamma irradiation were observed. Allium solution (3.0 mg/mL in 0.01 M PBS, pH 7.4) and raw garlic were irradiated in a gamma irradiation facility to be absorbed with doses of 2.5, 5.0, 7.5 and 10.0 kGy. After irradiation, a half of irradiated allivin solution was filtered through 0.22 ${\mu}{\textrm}{m}$ filtration kit. UV spectrum and SDS-PAGE were used for observing the conformational changes of allivin by gamma irradiation. Turbidity of allivin solution was increased by optical observation, and the result was validated by the increase of absorbance at 660 m. Turbidity of irradiated and filtered allivin solution significantly (p<0.05) decreased, and the concentration of soluble protein in the solution decreased by increase of the dose. Aggregation among allivin molecules was observed by SDS-PAGE, but irradiated and filtered samples showed decrease of allivin with reducing aggregation trend. SDS-PAGE profiles of irradiated raw garlic were not different. Results indicate that gamma irradiation can decrease the allivin in solution state which may impact on allergenic reaction of garlic.
