단기선 GPS측위 모델을 이용한 관측오차 분석 (Analysis of Measurement Errors Using Short-Baseline GPS Positioning Model)

  • GPS 관측값을 이용하여 측위를 하는 경우 조정계산이 수반되면 이때 관측오차에 대한 정확한 모델링이 필수적이다. 본 연구에서는 GPS 관측타입별 관측오차를 분석하기 위해 MINQUE(Minimum Norm Quadratic Unbiased Estimators) 방법을 사용하였으며 단기선 GPS측위 모델을 기반으로 하였다. C1, P2, L1, L2 관측타입에 대해 각각 단위분산을 계산하였으며 그 결과 관측오차는 각각 22.3cm, 27.6cm, 2.5cm, 2.2cm로 나타났다. 단위분산 계산에는 대용량의 컴퓨터 메모리와 복잡한 계산이 필요하다. 이러한 단점을 극복하기 위해 epoch별로 단위분산을 추정하였으며 결과를 분석함으로써 epoch별 계산방법의 유효성을 검증하였다. 분석 결과, 계산된 결과값에 차이가 있긴 했으나 그 차이가 비교적 작기 때문에 대부분의 GPS 응용분야에서의 활용에는 문제가 없을 것으로 판단된다.

Laminaria japonica와 Kjellmaniella crassifolia를 이용한 Pb의 생체흡착 : 흡착속도 및 흡착평형 모델링 (Biosorption of Lead ions onto Laminaria japonica and Kjellmaniella crassifolia : Equilibrium and Kinetic Modelling)

  • 본 연구에서는 해안가에서 자생하는 두 종의 해조류(Laminaria japonica와 Kjellmaniella crassifolia)를 생체흡착제로 사용하여 용액중의 중금속(납) 제거 실험을 통해 생체흡착제의 납의 흡착속도와 흡착평형 특성을 연구하였다. 두 종의 해조류을 사용한 납의 생체흡착은 흡착초기 2시간 이내에 평형에 도달하였다. 흡착속도는 유사 2차 흡착속도식에 의해 거의 정확한 모사가 가능하였으며, 여기에서 산출된 속도상수, $k_{2,ad.}$는 각각 $0.883{\times}10^{-3}$$0.628{\times}10^{-3}\;g/mg/min$이었다. 흡착평형은 Langmuir, Redlich-Peterson 및 Koble-Corrigan (Langmuir-Freundlich) 모델식에 의해 잘 모사되었다. 또한, L. japonica와 K. crassifolia의 4가지 중금속에 대한 선택성은 Pb>Cd>Cr>Cu와 Pb>Cu>Cd>Cr순으로 나타났다. 본 실험에 사용된 L. japonica는 pH의 증가에 따라 납의 흡착량도 증가되었다.

Comparison of response surface methods for the optimization of an upflow anaerobic sludge blanket for the treatment of slaughterhouse wastewater

  • This study was aimed at using the Central Composite Design (CCD) and Box-Behnken Design (BBD) to compare the efficiency and to elucidate the main interacting parameters in the upflow anaerobic sludge blanket (UASB) reactor, namely: Organic Loading Rate (OLR), Hydraulic Retention Times (HRT) and pH at a constant temperature of 35℃. Optimum HRT (15 h), OLR (3.5 kg.m-3.d-1) and pH (7) resulted in biogas production of 5,800 mL/d and COD removal of 80.8%. BBD produced a higher desirability efficiency of 94% as compared to the CCD which was 92%. The regression quadratic models developed with high R2 values of 0.961 and 0.978 for both CCD and BBD, respectively, demonstrated that the interaction models could be used to pilot the design space. BBD model developed was more reliable with a higher prediction of biogas production (5,955.4 ± 225.3 mL/d) and COD removal (81.5 ± 1.014%), much close to the experimental results at a 95% confidence level. CCD model predictions was greater in terms of COD removal (82.6 ± 1.06% > 80.8%) and biogas production (4,636.31 mL/d ± 439.81 < 5,800 mL/d) which was less than the experimental results. Therefore, RSM can be adapted for optimizing various wastewater treatment processes.

  • 본 논문에서는, Marshall-Olkin의 이변량 지수모형을 따르는 두개의 부품으로 이루어진 시스템에서 두 부품의 수명 $(X_{1},\;X_{2}$들이 이변량 1종 중단된 자료로 관찰되는 경우, $P(X_{1}\;<\;X_{2})$에 대한 최우추정량을 구하고 근사적 정 규성을 밝힌다. 그리고 그 추정량을 기초로 $P(X_{1}\;<\;X_{2})$에 대한 근사적 검정법을 제안하고, 몬테칼로 모의실험을 통하여 여러가지 상황에서 제안된 추정량의 근사적 검정력을 계산하여 비교하였다.

Enhanced Production of Gamma-Aminobutyric Acid by Optimizing Culture Conditions of Lactobacillus brevis HYE1 Isolated from Kimchi, a Korean Fermented Food

  • This study evaluated the effects of culture conditions, including carbon and nitrogen sources, L-monosodium glutamate (MSG), and initial pH, on gamma-aminobutyric acid (GABA) production by Lactobacillus brevis HYE1 isolated from kimchi, a Korean traditional fermented food. L. brevis HYE1 was screened by the production analysis of GABA and genetic analysis of the glutamate decarboxylase gene, resulting in 14.64 mM GABA after 48 h of cultivation in MRS medium containing 1% (w/v) MSG. In order to increase GABA production by L. brevis HYE1, the effects of carbon and nitrogen sources on GABA production were preliminarily investigated via one-factor-at-a-time optimization strategy. As the results, 2% maltose and 3% tryptone were determined to produce 17.93 mM GABA in modified MRS medium with 1% (w/v) MSG. In addition, the optimal MSG concentration and initial pH were determined to be 1% and 5.0, respectively, resulting in production of 18.97 mM GABA. Thereafter, response surface methodology (RSM) was applied to determine the optimal conditions of the above four factors. The results indicate that pH was the most significant factor for GABA production. The optimal culture conditions for maximum GABA production were also determined to be 2.14% (w/v) maltose, 4.01% (w/v) tryptone, 2.38% (w/v) MSG, and an initial pH of 4.74. In these conditions, GABA production by L. brevis HYE1 was predicted to be 21.44 mM using the RSM model. The experiment was performed under these optimized conditions, resulting in GABA production of 18.76 mM. These results show that the predicted and experimental values of GABA production are in good agreement.

Responses of high-rise building resting on piled raft to adjacent tunnel at different depths relative to piles

  • In this study, 3D coupled-consolidation numerical parametric study was conducted to predict the deformation mechanism of a 20 storey building sitting on (4×4) piled raft (with length of piles, Lp=30 m) to adjacent 6 m diameter (D) tunnelling in stiff clay. The influences of different tunnel locations relative to piles (i.e., zt/Lp) were investigated in this parametric study. In first case, the tunnel was excavated near the pile shafts with depth of tunnel axis (zt) of 9 m (i.e., zt/Lp). In second and third cases, tunnels were driven at zt of 30 m and 42 m (i.e., zt/Lp = 1.0 and 1.4), respectively. An advanced hypoplastic clay model (which is capable of taking small-strain stiffness in account) was adopted to capture soil behaviour. The computed results revealed that tunnelling activity adjacent to a building resting on piled raft caused significant settlement, differential settlement, lateral deflection, angular distortion in the building. In addition, substantial bending moment, shear forces and changes in axial load distribution along pile length were induced. The findings from the parametric study revealed that the building and pile responses significantly influenced by tunnel location relative to pile.

비만 소아의 혈중 아디포넥틴과 인슐린저항성에 관한 연구 (Serum Adiponectin Levels and Insulin Resistance in Obese Children)

  • 목 적 : 식생활과 생활양식의 서구화로 인하여 우리나라도 비만의 유병률이 급격히 증가하는 추세이며 특히 소아비만은 이미 소아기에 합병증 발생의 위험과 더불어 성인비만으로 이행될 가능성이 크기에 이에 대한 관심이 날로 높아지고 있는 추세이다. 아디포넥틴은 최근 발견된 지방세포에서 분비되는 adipocytokine으로 비만에서 아디포넥틴의 감소가 인슐린저항성과 동맥경화증의 발병과 관련이 있는 것으로 알려져 있다. 본 연구는 비만아와 정상 체중아의 혈중 아디포넥틴 농도의 차이와 체질량지 수(BMI) 및 인슐린저항성과의 상관관계를 알아보고자 하였다. 방 법 : 2003년 1월부터 2003년 6월까지 BMI가 95 백분위 이상인 8-15세 사이의 비만아 82명(남아 54명, 여아 28명)과 대조군으로 정상 체중아 31명(남아 21, 여아10), 총 113명을 대상으로 신체계측을 하고 공복 후 채혈하여 혈중 아디포넥틴, 인슐린, 혈당 농도를 측정하였다. 혈중 아디포넥틴 농도를 비만여부와 사춘기 시기별로 비교하였으며 아디포넥틴과 체질량지수, 인슐린, HOMA 등과의 상관관계를 알아보았다. 결 과 : 1) 비만아의 평균 혈중 아디포넥틴 농도는 19.7 mg/mL, 정상 체중아는 27.6 mg/mL로 비만군이 유의하게 낮았으며(P<0.01), 전체 대상아 중 남아와 여아의 성별에 따른 차이는 없었다. 2) 사춘기 전보다 사춘기 시기의 소아에서 아디포넥틴의 농도가 의미있게 낮았다(P<0.01). 3) 비만아 중 27명을 대상으로 시행한 경구 당부하 검사에서 공복시에 비해 식후 2시간 후 인슐린 농도는 유의하게 높았으며($17.0{\mu}IU/mL$ vs $35.8{\mu}IU/mL$, P<0.01), 아디포넥틴 농도는 반대로 유의하게 낮았다(25.8 mg/mL vs 19.8 mg/mL, P<0.01). 4) 혈중 아디포넥틴과 BMI(r=-0.39, P<0.01), 인슐린(r=-0.28, P<0.01)은 각각 음의 상관관계를 보였으며, 혈중 지질치중 저밀도 지질단백(LDL) 콜레스테롤(r=-0.20, P<0.05)과도 음의 상관관계를 보였으나 총 콜레스테롤과 고밀도 지질단백(HDL) 콜레스테롤과, 중성지방과는 유의한 상관관계가 없었다. 결 론 : 혈중 아디포넥틴 농도는 비만아에서 정상 체중아보다 유의하게 낮았으며, 아디포넥틴은 BMI 및 혈중 인슐린과 음의 상관관계가 있었다. 혈중 아디포넥틴이 소아 비만아에서 인슐린 저항성의 예측인자로 유용하게 사용될 수 있을 것으로 생각된다.

The Evaluation of the Effect of Herbal Extract on Osteoarthritis: In Vitro and In Vivo Study

  • In this study, the anti-osteoarthritis effects of Cynanchum wilfordii, Phlomis umbrosa, and Angelica gigas extract (CPAE), observed and confirmed in previously clinical studies were further investigated by in vitro and in vivo studies. Anabolic biomarkers related to healthy cartilage maintenance, such as aggrecan, type II collagen ${\alpha}$-1 (Col2a1), sex determining region Y-box-9 (Sox-9), and catabolic biomarkers related to osteoarthritis, such as cyclooxygenase-2 (Cox-2), matrix metalloproteinase-13 (Mmp13), and nuclear factor kappa-light-chain-enhancer of activated B cells ($Nf{\kappa}b$), were evaluated by quantitative reverse transcriptase polymerase chain reaction and reporter gene assay. In vitro study results showed significant changes in both anabolic and catabolic biomarkers. For anabolic factors, significant changes in the level of aggrecan (P<0.05), Col2a1 (P<0.05), and Sox-9 (P<0.01) activation were shown after treatment of cartilage cells with CPAE (50 ng/mL) with similar efficacy compared to insulin growth factor, the positive control (100 ng/mL). For catabolic factors, significant changes in the inhibition activity of Cox-2 (P<0.05), Mmp13 (P<0.01), and $Nf{\kappa}b$ (P<0.05) were shown for CPAE (50 ng/mL) with similar efficacy compared to Celecoxib, the positive control ($10{\mu}M$). In the in vivo carrageenan-induced paw edema model study results showed that CPAE-treated groups (100 mg/kg) and Celecoxib-treated groups (60 mg/kg) showed comparably significant efficacy of inhibition by 37.1% and 52.1%, respectively. Furthermore, CPAE (200 mg/kg) showed similar effect to Celecoxib (60 mg/kg) with an inhibition rate of 54.3%. This result confirms that CPAE effectively inhibited the inflammation-induced osteoarthritis symptoms.

Genome-wide Association Study to Identify Quantitative Trait Loci for Meat and Carcass Quality Traits in Berkshire

  • Meat and carcass quality attributes are of crucial importance influencing consumer preference and profitability in the pork industry. A set of 400 Berkshire pigs were collected from Dasan breeding farm, Namwon, Chonbuk province, Korea that were born between 2012 and 2013. To perform genome wide association studies (GWAS), eleven meat and carcass quality traits were considered, including carcass weight, backfat thickness, pH value after 24 hours (pH24), Commission Internationale de l'Eclairage lightness in meat color (CIE L), redness in meat color (CIE a), yellowness in meat color (CIE b), filtering, drip loss, heat loss, shear force and marbling score. All of the 400 animals were genotyped with the Porcine 62K SNP BeadChips (Illumina Inc., USA). A SAS general linear model procedure (SAS version 9.2) was used to pre-adjust the animal phenotypes before GWAS with sire and sex effects as fixed effects and slaughter age as a covariate. After fitting the fixed and covariate factors in the model, the residuals of the phenotype regressed on additive effects of each single nucleotide polymorphism (SNP) under a linear regression model (PLINK version 1.07). The significant SNPs after permutation testing at a chromosome-wise level were subjected to stepwise regression analysis to determine the best set of SNP markers. A total of 55 significant (p<0.05) SNPs or quantitative trait loci (QTL) were detected on various chromosomes. The QTLs explained from 5.06% to 8.28% of the total phenotypic variation of the traits. Some QTLs with pleiotropic effect were also identified. A pair of significant QTL for pH24 was also found to affect both CIE L and drip loss percentage. The significant QTL after characterization of the functional candidate genes on the QTL or around the QTL region may be effectively and efficiently used in marker assisted selection to achieve enhanced genetic improvement of the trait considered.

Adsorption isotherm and kinetics analysis of hexavalent chromium and mercury on mustard oil cake

  • Adsorption equilibrium and kinetic behavior of two toxic heavy metals hexavalent chromium [Cr(VI)] and mercury [Hg(II)] on mustard oil cake (MOC) was studied. Isotherm of total chromium was of concave type (S1 type) suggesting cooperative adsorption. Total chromium adsorption followed BET isotherm model. Isotherm of Hg(II) was of L3 type with monolayer followed by multilayer formation due to blockage of pores of MOC at lower concentration of Hg(II). Combined BET-Langmuir and BET-Freundlich models were appropriate to predict Hg(II) adsorption data on MOC. Boyd's model confirmed that external mass transfer was rate limiting step for both total chromium and Hg(II) adsorptions with average diffusivity of $1.09{\times}10^{-16}$ and $0.97m^2/sec$, respectively. Desorption was more than 60% with Hg(II), but poor with chromium. The optimum pH for adsorptions of total chromium and Hg(II) were 2-3 and 5, respectively. At strong acidic pH, Cr(VI) was adsorbed by ion exchange mechanism and after adsorption reduced to Cr(III) and remained on MOC surface. Hg(II) removal was achieved by complexation of $HgCl_2$ with deprotonated amine ($-NH_2$) and carboxyl (COO-) groups of MOC.
