  • Au debut de la dynastie de Choson, qui represente une periode de la regne des rois T'aejo et T'aejong, il fallait etablir et codifier la .nouvelle politique nationale, et c'est dans ce contexte que les caracteres kyemi, qui ont joue Ie role tres important dans l'histoire de la typographie mondiale, furent crees. Le sujet de ce memoire n'est qu'une enumeration des faits historiques de l'edition ou de l'imprimerie dans cet epoque, mais une recherche sur la vraie signification de la culture du livre imprime. Done, on peut etudier ici; d'abord, less activites de l'imprimerie a l'epoque du roi T'aejo et du roi T'aejong, et enfin, la signification et le bilan synthetique de ces activites, surtout des caracteres kyemi sur le point de l'histoire de la civilisation mondiale.

  • ADP-glucose pyrophosphorylase (AGPase) catalyzes the key regulatory step in starch biosynthesis. Two cDNA clones encoding AGPase subunits were isolated from the leaf cDNA library of Chinese cabbage (Brassica campestris L. spp. pekinensis). One was designated as BCAGPS for the small subunit and the other as BCAGPL for the large subunit. Both cDNAs have uninterrupted open reading frames deriving 57 kDa and 63 kDa polypeptides for BCAGPS and BCAGPL, respectively, which showed significant similarity to those of other dicot plants. Also, However, the deduced amino acid sequence of BCAGPL has a unique feature. That is, it contains two regions (Rl and R2) lacking in all other plant enzymes. This is the first report of BCAGPL containing Rl and R2 among plant large subunits as well as small subunits. From the genomic Southern analysis and BAC library screening, we inferred the genomic status of BCAGPS and BCAGPL gene.

  • A cDNA Fragment encoding iron storage protrin generated by polymerase chain reaction(PCR) using highly conserved regions of ferritin related genes were used to sereen a red pepper cDNA library. cDNA clone was designated as Fp1. Fp1 clone contatines a 5' nontranslated region of 51dp containing stop conds. Down stream from 5' UTP. an open reading frame of 750bp was observed. followed by a 3' UTR of 272bp. The deduces amino acid sequence of red pepper protein(Fp1) showed 84%, 48% and 36% identity with soybean(SolC). human(HuL H) and horse spleen(HoS-L) ferritin mRNA accumulation in response to iron. Ferritin mRNA accumulation was transient and particularly abundant in leaves. reaching a maxmum at 12h. The level of ferritin mRNA in roots was affected to a lesser extent than in leaves.

  • Heterogeneous nuclear ribonucleoprotein L (hnRNP L) is a major pre-mRNA binding protein and it is an abundant nuclear protein that shuttles between the nucleus and the cytoplasm. hnRNP L is known to be related to many cellular processes, including chromatin modification, pre-mRNA splicing, mRNA export of intronless genes, internal ribosomal entry site (IRES)-mediated translation, mRNA stability, and spermatogenesis. In order to identify the cellular proteins interacting with hnRNP L, this study performed a yeast two-hybrid screening, using a human liver cDNA library. The study identified insulin-like growth factor binding protein-4 (IGFBP-4) as a novel interaction partner of hnRNP L in the human liver. It then discovered, for the first time, that hnRNP L interacts specifically with IGFBP-4 in a yeast two-hybrid system. The authenticity of this two-hybrid interaction of hnRNP L and IGFBP-4 was confirmed by an in vitro pull-down assay.

Microbial Diversity in the Enrichment Cultures from the Fermented Beverage of Plant Extract Using Ribosomal RNA Sequence Analysis (라이보좀 RNA 염기서열 분석을 이용한 집식배양된 식물추출물발효음료의 미생물 다양성)

  • A beverage was produced by the fermentation of mixed extracts from the various fruits, vegetables, algae, and medical herbs. The physicochemical properties of the fermented beverage of plant extracts (FBPE) and microbial diversity were analyzed in cultures enriched from FBPE using 16S and 26S rRNA gene sequence analyses. The pH, acidity, $^{\circ}brix$, reducing sugar, and alcohol contents of the FBPE were determined to be the 3.48, 1.68%, 70.0, 1,026 g/L, and 3.5%, respectively. The most abundant free sugar and organic acid in the FBPE were glucose (567.83 g/L) and tartaric acid (93.68 mg/L), respectively. Lactobacillus homohiochii was the predominant species in all enriched culture samples: 100% of the species in 0B (0% sugar) and 40B (40% sugar) libraries and 95.6% of 20B library (20% sugar). Lactobacillus fructivorans was detected in the 20B library. The predominant species in the samples of enrichment cultures collected from FBPE with three different sugar concentrations were: Candida zeylanoides (45.2%) in the 0Y library (0% sugar), Candida lactis-condensi (35.7%) and C. zeylanoides (35.7%) in the 20Y library (20% sugar), and C. lactis-condensi (38.1%) in the 40Y library (40% sugar). This result may provide a useful frame of reference for further analyses of microbial population dynamics in FBPE.

  • On admet Jusqu'ici que le XVe siecle est une $\acute{e}poque$ phare de l'histoire de l'imprimerie avec l'apparition de la typographie en Orient et en Occident, $c'est-\acute{a}-dire$ en $Cor\acute{e}e$ sur le Rhim. Cependant, $l'\acute{e}poque$ de l'invention typographique en $Cor\acute{e}e$ doit $\hat{e}tre$ $devanc\acute{e}e$ encore d'un ou deux siecle. Outre cette question, il est $\acute{a}$ noter ici que les caracteres mobiles $m\acute{e}talliques$ Kyemi, fondus en 1403, ont $jou\acute{e}$ un $r\hat{o}le$ $tr\acute{e}s$ important dans l'histoire de la typographie mondiale et que les $caract\acute{e}res$ kabin qui leur $succ\acute{e}derent$ en 1434 menerent la typographie $cor\acute{e}enne$ a son $apog\acute{e}e$. Or, les recherches $cor\acute{e}ennes$ se satifait de la primaute de l'invention en Coree des caracteres mobiles metalliques, mais n'adoptent pas une attitude suffisament critique. Aussi, l'auteur a-toil voulu s'interroger sur la veritable valeur de la typographie de la dynastie de Choson tant au point de vue de l'histoire de l'imprimerie mondiale. En premier lieu, on etudie, comme une chronique, les caracteres mobiles metalliques importants tout au long de la dynastie de Choson sans donner une interpretation nouvelle. C'est dans deux derniers, qu'on essaie de developper ses idees sur la typographie en Coree notamment la question technologique qui devrait etre le fond des recherches. Aussi, on y etudie, la technique de la typographie ancienne en Coree interpretee selon une methode scientifique et moderne et, enfin, le bilan synthetique en cherchant les raisons qui freinerent son developpement en Coree malgre la performance de ses debuts. A l'issue de ces recherches, on ne peut s'empecher d'eprouver une certaine insatisfaction a cause de l'ambiguite des explications que l'on trouve dans des documents fondamentaux. Avec ce difficulte on a voulu, au moins, exposer des points discutatables qui devraient etre les recherches a futur.

  • As an attempt to grasp the current situation of traditional Korean books scattered about in the world, this study has bibliographically analyzed first of all traditional Korean books as well as old Korean maps and atlases, and topography descriptions collected by L. C.. Traditional Korean books have been analyzed classified by subject, printing or manuscript, and the kind of edition. Historical development in the compilation of geographical materials during the Chosun Dynasty has been studied. Especially on old Korean maps and atlases bibliographical characteristics have been researched. Accordingly. the foundation has been laid not only for the study on the bibliographical characteristics and the current situation of traditional Korean books and old Korean maps and atlases but also for the comparative analysis on traditional Korean books in Korea with the ones collected by L.C.

  • To understand the molecular structure and pathogenesis mechanism of Korean garlic viruses (GV), virus particles were isolated from field-grown garlic leaves and RNA genome was isolated from them. It was used for constructing cDNA library for GV. Several cDNA clones for GV were isolated and classified into 4 different groups on the basis of cross Southern hybridization. Northern blot analysis of GV RNA with one of these cDNA clones shows that the clone is a cDNA for GV RNA.

Toxic dinoflagellate Gymnodinium catenatum Graham(Dinophyceae) from the southern coast of Korea: morphology, phylogeny and effects of temperature and salinity on growth (남해안에서 분리한 유독 와편모조류 Gymnodinium catenatum Graham (Dinophyceae): 형태, 분자계통학적 특성 및 온도와 염분에 따른 성장 특성)

  • The toxic dinoflagellate Gymnodinium catenatum isolated from the southern coast of Korea was described under light and scanning electron microscopy, and its large subunit (LSU) rDNA was sequenced. In addition, the effects of temperature and salinity on its growth were investigated. The cells of G. catenatum, as viewed under the electronic microscope, were green-brown color, $38.1-77.4{\mu}m$ in length and $26.1-40.8{\mu}m$ in width. The epicone was conical, while the hypocone was trapezoidal. The nucleus was located at the central part of the cell. The apical groove was horseshoe-shaped and small pores were irregularly distributed on the cell surface. Molecular phylogeny based on LSU rDNA gene sequences showed that the Korean G. catenatum and previously reported species formed a monophyletic clade within Gymnodinium sensu stricto clade. The maximum growth rate of $0.37day^{-1}$, was obtained at $25^{\circ}C$ and 35 psu, and the maximum cell density of $1,073cells\;mL^{-1}$, was observed at $20^{\circ}C$ and 25 psu. However, G. catenatum did not grow at temperature < $15^{\circ}C$ and < $30^{\circ}C$. These results suggest that environmental conditions of summer and autumn in the southern coast of Korea may be favorable for the growth of G. catenatum.
