Development of Jelly Fortified with Lactic Acid Fermented Prickly Pear Extract (백년초 젖산 발효액을 이용한 젤리의 제조)

  • Rheological properties and sensory characteristics of prickly pear jelly made from κ-carrageenan and glucomannan were examined. Prickly pear extract (PPE) fermented by Lactobacillus casei LS (isolated from soy milk curd residue) exhibited pH 3.12, 1.15% acidity (lactic acid) and 2.7 × 10/sup 9/ CFU/mL lactic acid bacteria. In the effect of concentration of fermented PPE, the jelly containing 4% fermented PPE had higher springiness and slightly lower hardness than that without fermented PPE, and was superior to color and taste. The hardness and brittleness of jellies were enhanced by the addition of 0.25% citric acid. The addition of 0.25% ascorbic acid resulted in the production of soft jelly like pudding. In the effect of concentration of κ-carrageenan, the hardness and springiness of jellies were increased by adding 0.75% κ-carrageenan, but was decreased by adding at the 0.9% level. In the overall preference of jellies including sweet and sour taste, higher score was obtained by the fortification of 0.6% and 0.75% κ-carrageenan, 0.25% citric acid and 30% fructose syrup.

Antioxidant Effects of Solvent Fraction from Sanguisorbae officinalis L. with Acetone (오이풀 아세톤 추출물을 이용한 용매 분획물의 항산화 효과)

  • The solvent extracts of Sanguisorbae officinalis L. were investigated for the activities of antioxidants as a functional ingredient for cosmetic products. The electron donating effect of ethyl acetate layer and n-butyl alcohol layer was appeared similar activity with positive control butylated hydroxy anisole (BHA) at all concentrations. In addition, in 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging assay, ethyl acetate layer, n-butyl alcohol layer and water layer were over 99% effect at all concentrations and higher than that of BHA. Also in hydrogen peroxide scavenging assay, ethyl acetate layer and n-butyl alcohol layer were higher than that of positive control ascorbic acid. The measured superoxide dismutase (SOD)-like activity of n-butyl alcohol was more than 50% at concentration of 1,000 ${\mu}g/mL$ and superoxide anion radical scavenging ability showed more than 45% at 1,000 ${\mu}g/mL$ of n-butyl alcohol layer. All these findings suggested that ethyl acetate layer and n-butyl alcohol layer have a great potential as a cosmeceutical ingredient with an antioxidant effect.

Antioxidative and Physiological Activities of Fractions from Pleurospermum kamtschaticumin Extracts (누룩치(Pleurospermum kamtschaticumin) 추출물의 항산화 효과 및 생리활성)

  • This study investigated the antioxidant activities, whitening effects, and antimicrobial activities of fractions from Pleurospermum kamtschaticumin extract. Total polyphenolic contents of fractions from Pleurospermum kamtschaticumin extract were 116.44~382.73 mg/g GAE (gallic acid equivalent), with the highest value in the ethyl acetate fraction. Fractions of Pleurospermum kamtschaticumin showed the highest DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity ($IC_{50}$=0.04 mg/mL) and ABTS radical scavenging activity (98.86% at 0.5 mg/mL), which was similar to ascorbic acid. Further, reducing power and FRAP activity were significantly high in the ethyl acetate and n-butanol fractions. The ethyl acetate and n-butanol fractions showed significantly high SOD-like activities (1 mg/mL, 86.93%, and 78.23%, respectively) compared to other fractions. Tyrosinase inhibition activities of the n-butanol fraction and 80% ethanol extract were 43.52% and 41.64%, respectively. Antimicrobial activity against Escherichia coli was only observed in the chloroform fraction (16.7 mm in 0.5 mg/disc). These results suggest that fractions from Pleurospermum kamtschaticumin extract show comparatively high antioxidant activity and thus could be used as a food additive or cosmeceutical ingredient.

Antioxidant and anti-aging effects of Alpinia galanga L. rhizome extracts and preservation of antioxidant effects in W/O type emulsion (갈랑가 뿌리 추출물의 항산화, 항노화 효과 및 W/O형 에멀젼에서 항산화 효과의 보존성)

  • As a result of this study, DPPH radical scavenging activity was 81.8% at a concentration of 100 ㎍/mL of Alpinia galanga L. rhizome 70% ethanol extract (AG.E), and ABTS+ radical scavenging activity was confirmed to be 99.8%, similar to L-Ascorbic acid (AA), at a low concentration of 50 ㎍/mL AG.E. To measure anti-aging activity, collagenase and elastase inhibitory activities were measured and AG.E showed higher inhibitory effects than epigallocatechin gallate (EGCG) starting at a low concentration of 50 ㎍/mL. In particular, AG.E showed inhibitory effects more than three times that of EGCG at a concentration of 500 ㎍/mL. In order to verify anti-aging effect of AG.E in CCD-986sk cell, good anti-aging effect was obtained in various experiments stimulated with UVB. In a gene expression analysis experiment using RT-PCR, the COL1A mRNA expression level was found to increase 2.90 times compared to no addition at a low concentration of 20 ㎍/mL AG.E, confirming the possibility of developing it as a good functional material related to anti-aging. As a basic study on temporal biological activity preservation ratio of material when applied to formulations, AG.E, and AA were added to a stable W/O type emulsion and stored in a thermostat at 25 ℃ for 60 days. As a result of measuring DPPH and ABTS+ radical scavenging activities on the 1st, 30th, and 60th days, it was confirmed that antioxidant effects are maintained at a high level over time in formulations.

  • Polyphenol oxidase was purified from an extract of tea leaf by ammonium sulfate fractionation followed by Sephadex G-150 column chromatography, which resulted in a 67-fold increase in specific activity. The enzyme had optimum pH 6.5 and was relatively heat stable. The substrate specificity of the tea leaf PPO showed high affinity toward pyrogallol and catechol. Potassium cyanide, sodium diethyldithiocarbamate, L-cysteine, 2-mercaptoethanol and ascorbic acid were potent inhibitors.

  • It has been known that quercetin is one of bioflavonid compounds and has anti-tumor effect by suppressing tumor growth in vitro and in vivo, including multiple biological effects by antioxidant and effective anti-inflammatory agent. The present study investigated whether quercetin can enhance antioxidant enzyme activity (glutathione proxidase: GPX, superoxide dismutase: SOD, catalase: CAT) and regulate the intracellular reactive oxygen intermediate levels on the B16F10 murine melanoma cells in the presensece of vitamin E, L-ascorbic acid (vitamin C) and reduced glutathione (GSH). Effect of Caffeic Acid on the Production of Reactive Oxygen Species in Raw 264.7 Cells (Raw 264.7 세포에서 유해산소 생성에 미치는 Caffeic Acid의 영향)

  • To investigate effect of caffeic acid on the intracellular reactive oxygen species production, we used DHE for intracellular superoxide anion production, DCF for intracellular ${H_2}{O_2}$ production and DHR for intracellular hydroperoxide production in Raw 264.7 cells. DPPH assay showed that antioxidant activity of caffeic acid with 39.5 ${\mu}M$ of ${IC}_{50}$ values was similar to that of ascorbic acid with 41.3 ${\mu}M$ of ${IC}_{50}$ values. Caffeic acid dose-dependently inhibited silica-induced ${H_2}{O_2}$ and hydroperoxide production but did not affect superoxide anion production in Raw 264.7 cells, which suggest that antioxidant effect of caffeic acid acts on the post-step of superoxide anion. On the other hand, caffeic acid showed a potent antioxidant effect in $lCuSO_4$-induced lipid peroxidation. Furthermore, plasma superoxide dismutase activity (3.43${\pm}$0.23 U/ml) in 10 mg/kg caffeic acid-fed mice was significantly higher than that (2.32${\pm}$0.24 U/ml) of control. From the above results, it is referred that caffeic acid appears to have potent anti-oxidant activity in both cell system and in vivo system.
