Extraction and Physicochemical Properties of the Pectin in Citron Peel (유자로부터 펙틴의 추출 및 이화학적 특성)

  • A pectic substance from citron peel was extracted with different methods to establish the optimum extraction conditions. The extraction yields of pectin with HCl, citrate and tartrate (concentration : 0.1 N, extraction ratio : 1 : 20) were 17.9%, 15.6% and 11.4%, respectively. Six times of 65% ethanol washing step was followed after first ethanol precipitation of acid extract for pure pectin. The degree of esterification (DE) of pectins was in the range of 43.0~47.6% and intrinsic viscosity was in the range of 0.94~2.63 (η$_{sp}$ /C (dL/g)). The sugar compositions such as rhamnose, xylose, glucuronic acid, galacturonic acid, galactose and glucose were little different in three kinds of pectins except for the content of arabinose.

Role of Sugars in Early Stage of Spore Germination in Filamentous Fungi, Aspergillus nidulans (사상균인 Aspergillus nidulans의 무성포자 발아와 당의 역할)

  • Initiation of spore germination in filamentous fungi such as Aspergillus nidulans and Botrytis cinerea requires the presence of nutrients. In this study, involvement of sugar sensing machinery was suggested in the germination of A. nidulans spores. Germination did not occur when the spores of A. nidulans were incubated in distilled water, whereas they were successfully germinated in the presence of 5% glucose with a germination rate of over 98% after 6hr incubation. Similar results were obtained when the spores were incubated in the presence of various sugars such as fructose, sucrose, and starch. Interestingly, spore germination was not observed in the presence of D-arabinose, whereas L-arabinose could induce germination as determined by the formation of germ tubes, indicating the presence of sugar sensing machinery that distinguish between the enantiomers of sugars. This inference was further supported by a decrease in germination rate (less than 25%) upon treatment of spores with trypsin. Subsequent MALDI-TOF mass spectrometry analysis of the surface proteins of spores identified ten proteins among which eight were involved in sugar metabolism. Taken together, our results suggest that spore germination in A. nidulans is initiated by the interaction of sugars with sugar binding proteins on the surface of spores.

Structural Analysis of Anti-metastatic Polysaccharides Isolated from Opuntia humifusa (천년초에서 분리한 항전이 다당의 구조 분석)

  • To examine the new practical utilization of mucilages in Opuntia humifusa, polysaccharides were isolated from O. humifusa and their anti-metastatic activity and structural analysis were carried out. In experimental lung metastasis of B16BL6 melanoma cells, prophylactically intravenous (i.v.) administration of the crude polysaccharide (CNC-0) from O. humifusa significantly inhibited lung metastasis in a dose-dependant manner. The main polysaccharide, CNC-Ia was purified to homogeneity from CNC-0 by two successive column chromatographies using DEAE-Sepharose FF and Sephadex G-100 and its structure was characterized. Molecular mass of CNC-Ia was estimated to be 700 kDa and it mainly consisted of arabinose, galactose and xylose in addition to two minor sugars such as rhamnose and fucose. Methylation analysis indicated that CNC-Ia comprised at least 18 different glycosyl linkages such as terminal Araf, 5-linked Araf, 4-linked Galp and terminal Xylp in addition to three characteristic linkages such as full branched Araf, 3,4,6-branched Galp and full branched Galp. To analyze the fine structure of CNC-Ia, it was sequentially digested by exo-${\alpha}$-L-arabinofuranosidase and endo-${\beta}$-1,4-D-galactanase. These analyses suggested that CNC-Ia belongs to be a highly branched Type I arabinogalactan which has a ($1{\rightarrow}4$)-${\beta}$-galactan backbone with arabinosyl oligosaccharide side chains.

The Taste Compounds in Fermented Entrails of Trepang, Stichopus Japonicus (해삼내장(內臟)젓의 맛성분(成分))

  • Fermented trepang entrails, Stichopus Japonicus, is widely used and occupied an important position in foods of this country. But little study on its taste compounds has been reported. This study was attempted to establish the basic data for evaluating taste compounds of fermented trepang entrails. Changes of free amino acids, free sugars, nucleotides and their related compounds as taste compounds during the fermentation of trepang entrails were analyzed by amino acid autoanalyzer and high speed liquid chromatography. Glutamic acid, alanine, glycine and proline were dominant amino acid in the fresh extracts, having 32.3%, 16.4%, 12.0% and 10.5% of the total free amino acid content, respectively. The content of leucine, valine, phenylalanine, isoleucine, methionine and tyrosine were low. The free amino acid were not changed in composition but changed in amounts during the fermentation of trepang entrails. Glutamic acid, alanine, glycine, proline, lysine, arginine and leucine were abundant in both fresh sample and fermented products. Free sugars in fermented trepang entrails, the results showed that galactose(933.7-988.0 mg%) was dominant and the content of arabinose, xylose were 78.7, 55.2-771mg% on moisture and salt free base respectively but glucose was detected in trace amount. Nucleotides and their related compounds were increased during the fermentation and hypoxanthine(47.1-$62.5{\mu}mole/g$, on moisture and salt free base) were dominant, IMP was abundant in fermented trepang entrails. TMA was increased while TMAO was decreased during the fermentation. The content of TMAO nitrogen in fermented trepang entrails was 30.0mg% on moisture and salt free base. The content of betine was increased during the fermentation and was ranged from 734.2 to 934.2mg% on moisture and salt free base. It is believed that such amino acids as glutamic acid, alanine, glycine, lysine, proline, arginine, leucine, such free sugars as galactose, arabinose, xylose, glucose, such nucleotides and their related compounds as IMP, hypoxanthine play an important role as taste compounds in fermented trepang entrails.

  • 자연 친화적 하천관리는 현 치수위주의 하천정비 관행으로 인한 하천의 환경 기능의 훼손을 막고, 제도적으로 자연 친화적으로 하천 사업을 추진하기 위해서 수변조사, 계획, 설계, 시공, 모니터링, 그리고 유지관리 등 일련의 체계적인 절차에 따라 하천 사업을 시행하는 것이다. 특히 이러한 표준 절차 가운데 수변조사는 하천의 특성을 결정짓는 여러 가지 자연적, 인공적 형성과정들과 그 과정들을 지배하는 생태학적 원리들을 파악하여 하천사업의 정비주제와 방향을 설정하는데 도움을 준다. 모니터링은 계획과 설계, 시공을 통해 실시된 하천사업의 효과를 평가하고, 공법적용에 따른 하천의 변화과정을 파악하여 유지관리 및 적응관리를 위한 근거를 제시하는 과정이다. 수변조사 및 모니터링 매뉴얼은 건설교통부가 수행한 '자연친화적 하천정비기법 개발' 의 연구성과로서, 경기도의 탄천, 충청북도의 달천 그리고 경기도 오산천을 대상으로 각 관련분야의 전문가가 참여하여 직접 적용$\cdot$검증한 결과를 바탕으로 제작한 것이다. 이는 하천의 관리 및 하천관련 사업을 자연 친화적으로 수행하기 위하여 필요한 '수변조사'와 '모니터링'에 대한 일반적인 절차와 방법을 체계화 한 것이다. 특히 수변조사 매뉴얼의 경우는 $\ulcorner$하천설계기준$\lrcorner$의 '제12장 하천환경조사'를 보완하는 관계에 있다. 하천설계기준에서 제시한 하천환경조사는 본 수변조사 매뉴얼상과 동일한 절차와 양식을 따르고 있다. 하천설계기준에서 자세하게 기술하지 못한 조사방법, 정리, 분석, 평가에 대한 내용을 구체적으로 적용하여 이를 보완하고 있다.은 안양천 웹페이지(http://anyang.river.or.kr)에서 구현되고 있으며, 앞서 설명한 바와 같이 1단계 프론티어 사업으로 설치된 4개의 하천수위, 2개의 지하수위 관측시설과 함께, 2단계에 (주)웹솔루스에서 자체적으로 설치 운영하고 있는 2개소의 하수관거 모니터링 관측시설, 그리고 안양시에서 운영하고 있는 5개소의 강우관측소와 7개소의 수위관측소를 모두 통합하여 실시간 자료를 제공하고 있다. 수위자료는 10분단위의 텍스트정보와 그래프형태로 지원되며, 검색기간 설정을 통해 원하는 기간내의 자료를 선별, 검색할 수 있다.. 또한 이와 같은 기초적인 정보를 바탕으로 하류하천의 탁수 피해를 최소화할 수 있는 선택취수탑의 운영방안을 수립할 수 있다 본 연구에서는 이를 위해 선택취수탑 주위의 성층흐름을 기존의 실험자료와 수치해석을 통하여 분석하였고, 온도성층구조나 취수구의 위치변화에 따른 방류수 수질특성을 조사하였다.쇄파대(artifical reef)와 같은 완충지대를 갖는 호안을 축조함으로써 월파량을 감소시키는 대안으로 제시하고자 한다. 본 연구 수행을 통해 태풍 내습시 발생 가능한 자연재해에 대한 사전 방지를 목적으로 태풍피해의 원인을 제시하고 이를 해결하여 현재의 방재대책이 항구적인 방재대책으로 전환될 수 있는 방안 마련의 기초 자료로 활용되기를 기대한다., L-arabinose, 및 D-galactose; 제3차(第三次) 가수분해물(加水分解物)(C)에서 L-rhamnose, D-xylose, L-arabinose 및 D-galactose, 비가수분해물(非加水分解物)(C')에서 D-xylose와 D-galactose를 검출(檢出)하였다.   • 여름철 집중강우시 유입되는 고탁수층은 저수지의 밀도성층으로 인하여 표수층 하부에 위치하며, 이를 적기에 배제하지 않을 경우에는 수평방향의 확산현상과 연직방향의 전도현상으로 인해 저수지 전역에 분포하게 되어, 탁수현상의 장기화를 유발한다. 이와 같은 탁수장기화에 대한 저수지내 대책의 하나로서 홍수유입후 탁도가 높은 물을 단기간동안 방류하고, 갈수기에는 탁도가 낮은 물을 방류하는 선택취수 기법을 적용할 수 있다. 임하댐의 경우, 2004년 태풍 '디앤무'와 태풍 '메기'로 인해 발생한 탁수를 선택취수를 통하여 조기에 배제함으로써 호내탁도를 저감시킨 바 있다. 그러나 이와 같은 고탁수 우선배제 기법은 반드시 하류하천의 영향범위를 사전에 분석검토하고, 하류하천의 수질현황을 고려한 합리적인 운영방안이 제시되어야 한다. 2004년 태풍 '디앤무' 발생직후의 댐방수로에서 170 NTU의 고탁수를 방류한 경우, 유하거리 250 km인 지점까지 30 NTU이상 유지된 바 있으며, 태풍 '메기'에 의한 고탁수 유입시, 임하댐 방류수 157 NTU, 안동댐 방류수 37 NTU인 경우에 임하댐 하류 113 km인 구미 선산 취수장까지 63 NTU의 탁도가 유지된 바 있다. 현재 임하댐의 경우, 하류하천으로의 방류수 수질을 모니터링할 수 있는 자동수질측정시스템이 발전취수탑 전면 1개소, 조정지댐 1개소에 설치되어 있다. 본 연구에서는 자동수질측정시스템과 연계하여 수질모형을 통하여 임하댐 하류하천인 낙동강 본류의 탁도관리시스템을 구축하고자 하였다. 환경부의 수질측정망 자료를 통하여 최근 3년간의 부유사농도의 변화양상을 분석하였으며, 태풍 직후의 낙동강 주요지점별 탁도측정자료를 이용하여 임하댐 고탁도방류에 의한 영향범위를 분석하였다. 또한 하천수질모형을 이용하여 탁수예측방법에 대하여 검토하였고, 이를 이용한 탁도관리시스템 구축을 위한 기본 방향을 제시하고자 한다.함께, 2단계에 (주)웹솔루스에서 자체적으로 설치 운영하고 있는 2개소의 하수관거 모니터링 관측시설, 그리고 안양시에서 운영하고 있는 5개소의 강우관측소와 7개소의 수위관측소를 모두 통합하여 실시간 자료를 제공하고 있다. 수위자료는 10분단위의 텍스트정보와 그래프형태로 지원되며, 검색기간 설정을 통해 원하는 기간내의 자료를 선별, 검색할 수 있다.. 또한 이와 같은 기초적인 정보를 바탕으로 하류하천의 탁수 피해를 최소화할 수 있는 선택취수탑의 운영방안을 수립할 수 있다 본 연구에서는 이를 위해 선택취수탑 주위의 성층흐름을 기존의 실험자료와 수치해석을 통하여 분석하였고, 온도성층구조나 취수구의 위치변화에 따른 방류수 수질특성을 조사하였다.쇄파대(artifical reef)와 같은 완충지대를 갖는 호안을 축조함으로써 월파량을 감소시키는 대안으로 제시하고자 한다. 본 연구 수행을 통해 태풍 내습시 발생 가능한 자연재해에 대한 사전 방지를 목적으로 태풍피해의 원인을 제시하고 이를 해결하여 현재의 방재대책이 항구적인 방재대책으로 전환될 수 있는 방안 마련의 기초 자료로 활용되기를 기대한다., L-arabinose, 및 D-galactose; 제3차(第三次) 가수분해물(加水分解物)(C)에서 L-rhamnose, D-xylose, L-arabinose 및 D-galactose, 비가수분해물(非加水分解物)(C')에서 D-xylose와 D-galactose를 검출(檢出)하였다.   • Two vibrio sp. strains were isolated from disease catfish(silurus asotus). The present isolates were identified as Vibrio ordalii based on their biological and biochemical characteristics ; they were positive for acid production from glucose, Iactose, maltose, sucrose and salicine, while negative for arabinose, galactose, inocitol and xylose. They are named KL-1 and KL-2, KL-1 and KL-2 strains were similar to physiological characteristics ; growth was observed at pH 5 to 10 and in 0% to 6.0% NaCl. Two strains did not growth at a concentration above 7.0% NaCl and pH10. This bacterium was infected into health catfish hypodermically. Such injection was found to induce haemorrhagic ulcers very similar to those observed in naturally infected fish. At 24h post-infection, the red spot developed around the injection site and grew bigger to from a red sport area. At 120h post-infection, the muscle ncerosis was extended near the ventral fin. The seventy percent lethal dosage was appeared to water temperature at $25^{\circ}C$. Two strains were tested for drug senistiveity by plate method. KL-1 and KL-2 strains were sensitive to GM. K, N, S and SxT, but resistant to CF, L and VA.

  • In the screening of Korean traditional tea sources for the cellular lysosomal enzyme activity of peritoneal macrophage from mice, CT-0, a cold-water extract from Carthamus tinctorius L., showed the highest macro-phage-stimulating activity. CT-1-IIa-2-1, a purified macrophage-stimulation polysaccharide was obtained by a series of purification steps such as anion exchage chromatography with DEAE-Toyopearl 650M, gel permeation chromatography with Sepharose CL-6B, Sephacryl S-200, and HPLC with Superdex G-75. The molecular weight of homogeneous purified polysaccharide was estimated about 68 kDa. CT-1-IIa-2-1 consisted of xylose 27.44%, arabinose 16.14%, mannose 15.92% and glucose 14.47%. To measure acute toxicity, dose of 50, 100, 500, and 1000 mg/kg were intraperitoneally injected to ICR mice. The LD$\_$50/ was about 397 mg/kg.

Changes of Fruit Characteristics and Cell Wall Component during Maturation and Ripening in Asian Pear 'Hanareum', 'Manpungbae', and 'Niitaka' (Pyrus pyrifolia Nakai) ('한아름', '만풍배' 및 '신고' 배의 성숙 기간 중 과실특성 및 세포벽 관련물질의 변화)

  • We investigated the changes of fruit quality parameters, polysaccharide contents and cell wall components during maturation and ripening of two Korean pear cultivar 'Hanareum' and 'Manpungbae' compared with 'Niitaka' pear (Pyrus pyrifolia Nakai) which showed different physiological maturity based on days after full bloom (DAFB). Flesh firmness decreased continuously with fruit development and maturation, reaching a final level of 29.4, 33.5, and 27.4N at maturity in 'Hanareum' (127 DAFB), 'Manpungbae' (163 DAFB), and 'Niitaka' (170 DAFB), respectively. The level of ethylene production was very low in early season 'Hanareum' pear which showed at most 0.39 ${\mu}L{\cdot}L^{-1}$ at maturity and no ethylene was detected in 'Manpungbae' and 'Niitaka' at maturity. Fructose was the most abundant soluble sugar during fruit maturation in the pears tested and an increase of sucrose was observed during fruit ripening in the Asian pears commonly. Ethanol insoluble solids (EIS) content decreased gradually with different levels among the pear cultivars as fruit ripens consisted of 10.79, 12.72, and 12.75 $mg{\cdot}g^{-1}$ FW. The amount of total soluble polyuronides was higher in early season cultivars 'Hanareum' than those of mid-season cultivar 'Manpungbae' and 'Niitaka'. In 'Niitaka' which harvested most late season, the level of 4% KOH soluble hemicelluloses was lower than 'Hanareum' and 'Manpungbae' and maintained constantly during fruit ripening period. Cellulosic residues were determined high level in 'Niitaka' which showed 612.33 ${\mu}g{\cdot}mg^{-1}$ EIS at maturity when compared with 'Hanareum' (408.0 ${\mu}g{\cdot}mg^{-1}$ EIS) and 'Manpungbae' (538.67 ${\mu}g{\cdot}mg^{-1}$ EIS). The main constituents of cell wall neutral sugars which consisted of arabinose, xylose, galactose, and glucose were decreased gradually with onset of fruit ripening regardless of cultivar. Arabinose which was predominant in 'Hanareum' pear decreased at the last stage of ripening, but the changes of cell wall neutral sugar during ripening were not occurred in 'Niitaka' pear. The change of molecular mass distribution in water soluble pectin observed dominantly at the early stage of fruit development. Depolymerization of 4% KOH-soluble hemicelluloses and degradation of xyloglucan showed in early-season cultivar 'Hanareum' during fruit maturation, and degradation of those fractions were detected only at the early stage fruit development in mid-season cultivar 'Manpungbae' and 'Niitaka'. The molecular mass profile of CDTA soluble pectin, $Na_2CO_3$-SP and 24% KOH soluble hemicelluloses showed no significant change during fruit maturation regardless of cultivar.

Characterization of a New Acidophilic Acetobacter sp. Strain HA Isolated from Korean Traditional Fermented Vinegar

  • A new strain of acidophilic, acetogenic bacterium, Acetobacter sp. strain HA was isolated by selective enrichment from the traditionally fermented rice wine vinegar in Korea. It was a gram-negative, non-motile short rod and oxidized acetate and lactate. The optimal temperature and pH for growth were $28^{\circ}C$ and 4.0, respectively. The strain HA differed from other Acetobacter species by growing well on methanol, xylitol, inositol, dulcitol, D-xylose, L-arabinose, and D-mannose as sole sources of carbon and energy. The isolated strain HA did not produce $\gamma$-pyrones from glucose and did not produce ketone bodies from glycerol. The quinone system used in this study was an ubiquinone-9 isoprene unit. The guanine-plus-cytosine content of the DNA was 50.7 mol%, and the major cellular fatty acids were $C_{18:1} and C_{16:0}$.
