Production of Hydrogen and Volatile Fatty Acid by Enterobacter sp. T4384 Using Organic Waste Materials

  • In a study of hydrogen-producing bacteria, strain T4384 was isolated from rice field samples in the Republic of Korea. The isolate was identified as Enterobacter sp. T4384 by phylogenetic analysis of 16S rRNA and rpoB gene sequences. Enterobacter sp. T4384 grew at a temperature range of $10-45^{\circ}C$ and at an initial pH range of 4.5-9.5. Strain T4384 produced hydrogen at 0-6% NaCl by using glucose, fructose, and mannose. In serum bottle cultures using a complete medium, Enterobacter sp. T4384 produced 1,098 ml/l $H_2$, 4.0 g/l ethanol, and 1.0 g/l acetic acid. In a pH-regulated jar fermenter culture with the biogas removed, 2,202 ml/l $H_2$, 6.2 g/l ethanol, and 1.0 g/l acetic acid were produced, and the lag-phase time was 4.8 h. Strain T4384 metabolized the hydrolysate of organic waste for the production of hydrogen and volatile fatty acid. The strain T4384 produced 947 ml/l $H_2$, 3.2 g/l ethanol, and 0.2 g/l acetic acid from 6% (w/v) food waste hydrolysate; 738 ml/l $H_2$, 4.2 g/l ethanol, and 0.8 g/l acetic acid from Miscanthus sinensis hydrolysate; and 805 ml/l $H_2$, 5.0 g/l ethanol, and 0.7 g/l acetic acid from Sorghum bicolor hydrolysate.

  • A gram(+) bacteria that produced microbial flocculant was isolated from soil and classified as a Bacillus species and named as Bacillus sp. A56. The culture conditions of the strain for fluocculant production were studied in a shake flask. Optimum temperature and initial pH for flcculant production were 30C and 6.5, respectively. The optimized medium has gollowing composition: glucose 20g/l, NH4NO3 0.5g/l, K2HPO4 1.0g/l, KH2PO4 0.8g/l, MgSO4.7H2O 0.2g/l, MnSO4.4-6H2O 0.3g/l, CaCO3 0.5g/l, yeast extract 0.3g/l, tryptone 0.3g/l in tap water.

Rapid Acquisition of CM and CL Code for GPS L2C Software Receivers

  • The GPS modernization program offered a new civil signal on the L2 band, and the first modernized GPS Block IIR satellite was launched in September 2005. Currently, eight GPS Block IIRM satellites and two Block IIF satellites transmit L2C signal. The L2C signal contains two codes of CM and CL that are much longer than the L1 C/A code. Thus, the acquisition of the CM and CL codes takes more time compared with that of L1 C/A code. Under the assumption that the L2C signal is strong enough for detection, this paper suggests rapid acquisition methods for the GPS L2C signals for software receivers and compares its performance with that of other methods.

Effect of probiotics Lactobacillus paracasei GKS6, L. plantarum GKM3, and L. rhamnosus GKLC1 on alleviating alcohol-induced alcoholic liver disease in a mouse model

  • BACKGROUND/OBJECTIVES: Heavy alcohol consumption causes the development of alcoholic liver disease (ALD), a neglected but important public health problem. Many studies have pointed out that probiotics could improve gut health, which is also considered to be a cause of ALD. Therefore, this study screened the probiotics, Lactobacillus casei GKC1 (GKC1), L. fermentum GKF3 (GKF3), Bifidobacterium lactis GKK2 (GKK2), L. rhamnosus GKLC1 (GKLC1), L. paracasei GKS6 (GKS6), and L. plantarum GKM3 (GKM3), for their potential benefits in alleviating ALD for applications to disease prevention. SUBJECTS/METHODS: C57BL/6N mice were divided into 8 groups (n = 6 in each): normal control, positive control (alcohol-diet fed), and treatments of feeding probiotics GKC1, GKF3, GKK2, GKLC1, GKS6, and GKM3 under an oral dose 0.82 g/kg B.W. per day by oral gavage. The experiment was conducted for 8 weeks, and the concentrations of alanine aminotransferase (ALT), aspartate aminotransferase, triglyceride (TG), and total cholesterol (TC) in mice were measured. The glutathione (GSH), catalase (CAT), and histology were analyzed after sacrifice. RESULTS: The results showed a decrease in the serum ALT, liver TG, and liver TC levels in the GKS6, GKM3, and GKLC1 groups compared to the positive control. In addition, the decreasing GSH and CAT levels were inhibited in the GKS6 and GKM3 groups. The histopathological results showed that all probiotics could reduce the accumulation of liver fat. Furthermore, there was a significant difference in GKLC1 with lower stomach damage compared to the alcohol-fed mice without any addition of probiotics. CONCLUSIONS: GKLC1, GKS6, and GKM3 can be used as supplements for alleviating the development of ALD.

  • Micro/nano-sized L $a_{0.6}$S $r_{0.4}$Co $O_{3-}$$\delta$/ particles are considered to improve oxygen permeability in highly selective inorganic oxygen separation membrane. A L $a_{0.7}$S $r_{0.3}$G $a_{0.6}$F $e_{0.4}$ $O_{3-}$$\delta$/ membrane with perovskite structure is fabricated by a conventional solid-state reaction. As the oxygen permeation flux of the L $a_{0.7}$S $r_{0.3}$G $a_{0.6}$F $e_{0.4}$ $O_{3-}$$\delta$/ membrane was lower than commercial gas separation membranes, we coated the L $a_{0.6}$S $r_{0.4}$Co $O_{3-}$$\delta$/ particles to enhance the oxygen permeation flux. It has been demonstrated that the effective area of reactive free surface is an important factor in determining the effectiveness of the introduction of coating layer for oxygen permeation. The introduction of micro/nano L $a_{0.6}$S $r_{0.4}$Co $O_{3-}$$\delta$/ particles was very effective for increasing oxygen flux, as the flux was as much as 2 to 6 times higher than that of an uncoated L $a_{0.7}$S $r_{0.3}$G $a_{0.6}$F $e_{0.4}$ $O_{3-}$$\delta$/ membrane.\delta$/ membrane.>/ membrane.brane.

Changes in SARS-CoV-2 antibody titers 6 months after the booster dose of BNT162b2 COVID-19 vaccine among health care workers

  • Purpose: In Japan, the data on severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antibody titers after the booster dose of the coronavirus disease 2019 (COVID-19) vaccine are insufficient. The aim of this study is to evaluate changes in SARS-CoV-2 antibody titers before, 1, 3, and 6 months after the booster dose of the BNT162b2 COVID-19 vaccine among health care workers. Materials and Methods: A total of 268 participants who received the booster dose of the BNT162b2 vaccine were analyzed. SARS-CoV-2 antibody titers were measured before (baseline) and at 1, 3, and 6 months after the booster dose. Factors associated with changes in SARS-CoV-2 antibody titers at 1, 3, and 6 months were analyzed. Cutoff values at baseline were calculated to prevent infection of the omicron variant of COVID-19. Results: The SARS-CoV-2 antibody titers at baseline, and 1, 3, and 6 months were 1,018.3 AU/mL, 21,396.5 AU/mL, 13,704.6 AU/mL, and 8,155.6 AU/mL, respectively. Factors associated with changes in SARS-CoV-2 antibody titers at 1 month were age and SARS-CoV-2 antibody titers at baseline, whereas changes in SARS-CoV-2 antibody titers at 3 and 6 months were associated with the SARS-CoV-2 antibody titers at 1 month. The cutoff values of the SARS-CoV-2 antibody titers at baseline were 515.4 AU/mL and 13,602.7 AU/mL at baseline and 1 month after the booster dose, respectively. Conclusion: This study showed that SARS-CoV-2 antibody titers increase rapidly at 1 month after the booster dose of the BNT162b2 vaccine and begin to decrease from 1 to 6 months. Hence, another booster may be needed as soon as possible to prevent infection.

Quantification of kerosene and Diesel in Mixed Petroleum Fuels for Environmental Sample Characterization (다종유류 오염 환경매체에서의 유류 분리.정량에 관한 연구(I) - 등유, 경유 정량을 중심으로 -)

  • The objective of this study was to develop an effective separation and quantification method for kerosene and diesel in a mixed petroleum fuel (gasoline, kerosene, and diesel) contaminated environmental samples. This investigation was directed to prove the hypothesis that if the source of petroleum fuels were identical, the peak-area ratios of a reference n-alkane to other n-alkane peaks should be a constant even at the different concentrations. In addition, experimental recovery rates were determined to select the reference peaks of kerosene and diesel for peak area ratio measurements. The experimental results showed that the peak area ratios were constant among the samples having different concentrations when the ratios were calculated from areas of $C_{l3}$, $C_{l4}$, and $C_{15}$ peaks for kerosene and $C_{l6}$ and $C_{l7}$ peak for diesel as reference n-alkane peaks. The recovery rates were evaluated by comparing the relative peak area ratios of each reference peaks after making pairs of the kerosene and diesel reference peaks in the samples contained a known amount of gasoline, kerosene, and diesel. The recovery rates(%) Were 107.0$_{{\pm}20.6}$/86.6/ sub $\pm$15.9/ for kerosene- $C_{13}$/diesel- $C_{16}$, 99.6$\pm$$_{17.2}$/86.6$_{{\pm}15.9}$ for kerosene- $C_{14}$/diesel- $C_{16}$, 73.9/$\pm$14.4//86.6$_{{\pm}sub 15.9}$ for kerosene- $C_{15}$ /diesel- $C_{16}$, 109.4$_{{pm}0.8}$/75.9$_{{pm}4.7}$ for kerosene- $C_{13}$/diesel- $C_{17}$, 107.4$_{{pm}7.9}$/75.9$_{{pm}4.7}$ for kerosene- $C_{14}$/diesel- $C_{17}$, and 95.7$_{{pm}4.6}$ /75.9/$\pm$14.6//75.9$_{{pm$}4.7}$ for kerosene- $C_{15}$ /diesel- $C_{17.}$ The above experimental results confirm that all of the reference peak pairs of kerosene and diesel are applicable to the quantitative analysis for the mixed fuel contaminated samples, but the kerosene- $C_{15}$ /diesel- $C_{l7}$ peaks are recommended since the pair has a lower standard deviation than the other pairs.s..s.s.s..s..s.s.s.s.s.

Dietary Effect of Hemicellulose from Soy Fiber on Blood Glucose and Cholesterol Content in Streptozotocin-Induced Diabetic Rats (콩 식이섬유로부터 추출한 헤미셀룰로즈의 식이가 Streptozotocin유도 당뇨 흰쥐의 혈당과 혈청 콜레스테롤 함량에 미치는 영향)

  • The effect of hemicellulose extracted from soy fiber on the level of blood glucose and serum cholesterol in the streptozotocin-induced diabetic rat were investigated. The experimental plots were divided to cellulose group (control, 0.5% hemicellulose group (H-l) and 1% hemicellulose group (H-2) group. The groups were fed for 6 weeks, then fed for 1 week more after streptozotocin injection. Food intakes, weight gain and food efficiency ratio of H-2 group were higher, while water intakes and liver weight were lower than those of control and H-l group. The content of blood glucose and urine glucose were 212.8 mg/dL, 0.97 mg/dL in the control group, 160.5 mg/dL, 0.53 mg/dL in the H-l group, 141.0 mg/dL, 0.35 mg/dL in the H-2 group, respectively. There was no significantly difference in the content of neutral lipid, while the content of total serum cholesterol was 101.6 mg/dL in the control group, 73.8-78.4 mg/dL in the hemicellulose groups. The content of serum HDL-cholesterol in the all experimental groups showed no significantly difference showing 39.8-44.7 mg/dL. HTR and atherogenic index were 0.44 and 1.27 in the control group, but 0.54 and 0.46-0.85 in the hemicellulose groups, respectively.

Change in Growth of Chrysanthemum zawadskii var. coreanum as Effected by Different Green Roof System under Rainfed Conditions (빗물활용 옥상녹화 식재지반에 따른 한라구절초의 생육 변화)

  • This study aims to suggest a suitable soil thickness and soil mixture ratio of a green roof system by verifying the growth of Chrysanthemum zawadskii var. coreanum as affected by different green roof systems using rainwater. The experimental planting grounds were made with different soil thicknesses(15cm, 25cm) and soil mixing ratios (SL, $P_7P_1L_2$, $P_6P_2L_2$, $P_5P_3L_2$, $P_4P_4L_2$) and with excellent drought tolerance. Ornamental value Chrysanthemum zawadskii var. coreanum was planted. The change in plant height, green coverage ratio, chlorophyll content, fresh weight, dry weight, and dry T/R ratio of Chrysanthemum zawadskii var. coreanum were investigated from April to October 2009. For 15cm soil thickness, the plant height of Chrysanthemum zawadskii var. coreanum was not significantly different as affected by the soil mixing ratio. However, it was found to be higher in the amended soil mixture, $P_7P_1L_2$, $P_6P_2L_2$, $P_5P_3L_2$ and $P_4P_4L_2$ than in the sandy loam soil, as it was SL overall. For 25cm soil the plant height differences were in order to SL < $P_7P_1L_2$, $P_6P_2L_2$, $P_5P_3L_2$ < $P_4P_4L_2$. The green coverage ratio was observed not to be different by soil mixing ratio with soil thickness of 15cm, but, the lowest green coverage ratio in the SL. In the 25cm soil thickness, the green coverage ratio was 86-89% with a good coverage rate overall. The change in chlorophyll contents with 15cm soil thickness was found to be the highest in the SL treatment and the lowest in the $P_5P_3L_2$ treatment. For 25cm thickness, the highest value was in the $P_4P_4L_2$ and SL, and the lowest in the$P_7P_1L_2$. Fresh weight and dry weight were larger in soil with 25cm thickness. Therefore, the growth of Chrysanthemum zawadskii var. coreanum as affected by a different green roof system for using rainwater was higher in soil with 25cm thickness than 15cm, and in PPL amended soil than in sandy loam.
