${\beta}-galactosidase$ Activity of Lactobacillus spp. from Pickles (젓갈에서 분리한 Lactobacillus spp.의 ${\beta}-galactosidase$ 특성)

  • Two strains of Lactobacillus(L.) casfi and one strain of L. Pentosus, which were isolated from pickles, were used to investigate in studing their characteristics of ${\beta}-galactosidase$. The preferable carbon sources and pH of the MRS media for enzyme production from L. casei No.10 was found to be 1.0% lactose and pH 7.5, from L. Pentosus No.63 was 1.0% galactose and pH 7.5, and from L. casei No.36 was 1.0% lactose and pH 6.5, respectively. The maximum enzyme production from each strain was found after 48 hours culture at $30^{\circ}C$ in a medium with preferable carbon source. The optimum reaction temperature with substrate for ${\beta}-galactosidase$ activity was found at $60^{\circ}C$ for all three strains . The stability of enzyme from L. casei No.36 was found to be at $45^{\circ}C$, from L. Pentosus No.63 was found at $55^{\circ}C$. This stability from L. casei No.36 was found at $40^{\circ}C$, but it was reduced to 60% at $55^{\circ}C$. These stabilities of enzymes remained about 90% at $40^{\circ}C$ for all three strains. The optimal pH for enzyme activities was found to be pH 6.5 for all three strains. Enzyme activity remained over 90% for L. casei No.10 at $pH\;5.0{\sim}6.0$, for L. casei No.36 at $pH\;5.0{\sim}8.0$, and for L. pentosus No.63 at $pH\;6.0{\sim}7.0$.

Biosorption of Lead and Cobalt by Absidia coerulea and Thraustochitrium sp. (Absidia coerulea와 Thraustochitrium Sp. 에 의한 납과 코발트의 생물흡착)

  • Nonliving Absidia coerulea and Thraustochitrium sp. were used as biosorbents to remove lead and cobalt that are one of representative pollutant in wastewater and radioactive liquid waste. The optimum pH range for maximum lead and cobalt removal was increased 6.5~11.4 and 8.6~12.0 for Absidia coerulea and 4.2~10.5 and 8.9~11.6 for Thraustochitrium sp. to compared to biosorbent-free control, pH of 8.4~11.2 and 10.5~11.5, respectively. With 1 g biosorbent/L at initial solution pH 5.0. Absidia coerulea and Thraustochitrium sp. took up lead from aqueous solutions to the extent of 104 and 125 mg/g biomass, respectively, whereas Absidia coerulea and Thraustochitrium sp. at initial pH 6.0 took up only 2 and 20 mg/g biomass of cobalt, respectively. For initial 500 mg Pb/L at initial pH 5.0. optimum amount of biosorbent for maximum lead uptake was 0.2 g/L for Absidia coerulea and Thraustochitrium sp., whereas optimum 3.0 g biosorbent/L was needed for initial 200 mg Co/L at initial pH 6.0. Absidia coerulea and Thraustochitrium sp. had higher adsorption capacity for lead than that of cobalt.

Monitoring for the Resistance to Prochloraz of Fusarium Species Causing Bakanae Disease in Korea (국내 벼키다리병균의 Prochloraz 약제에 대한 저항성 변화)

  • To investigate the changes of the resistance to prochloraz of Fusarium species causing bakanae disease, Fusarium isolates were collected from various regions in Korea, and pathogenicity tests were performed using rice seeds. Minimum inhibitory concentration (MIC) and effective concentration of 50% ($EC_{50}$) values of isolates were determined using the agar dilution method. High frequency distribution of MIC values of prochloraz against isolates collected in 2006~2007 and 2013~2014 years were $3.125{\sim}6.25{\mu}g/mL$ and $6.25{\sim}12.5{\mu}g/mL$, respectively. The mean $EC_{50}$ value of isolates increased from $0.3142{\mu}g/mL$ in 2006~2007 to $0.8124{\mu}g/mL$ in 2013~2014. Based on the $EC_{50}$ value of isolates collected in 2006~2007, the resistant baseline of prochloraz was determined as $0.6{\mu}g/mL$. Compared with the ratio of resistant isolates in 2006~2007, the ratio of resistant isolates in 2013~2014 increased from 6.5% to 41.6%.

Changes of Serum Lipid Profiles after Eating Lycii Fructus in Rats Fed High Fat Diet (구기자 섭취에 의한 고지방식이를 하는 흰쥐의 혈중 지질상태 변화)

  • The purpose of this study was to investigate the effect of Lycii fructus on the serum lipid in rats fed high fat diet. We compared the effects of L. fructus and L.fructus water extract both adminstered with high fat diets on rats that had previously been on high fat or standard diets. Two separate experiments were conducted for 6 weeks. respectively. In experiment I, 4 groups of rats were fed experimental diets consisting of either \circled1 6 weeks of a standard diet(control), \circled2 6 weeks of a high-fat diet(HHC), \circled3 3 weeks of a high-fat diet followed by 3 weeks of a high-fat diet containing L. fructus(HHL) or \circled4 6 weeks of a high-fat diet with L. fructus extract in place of water for the last 3 weeks (HHT). In the second set of experiments, a high-fat diet (SHC), high-fat diet containing L.fructus(SHL) or high-fat diet with L. fructus extract in place of water (SHT) were fed for 3 weeks after 3 weeks of standard diet feeding. Rats fed L. fructus diet consumed more diets than high-fat diets. THe results of experiment I showed significant decreases(p<0.05) in serum triglyceride(TB) and low density lipoprotein-cholesterol (LDL-C) levels with L. fructus feedings, but did not show andy changes in total cholesterol (TC) level. High density lipoprotein-cholesterol (HDL-C) level was increased significantly(p<0.05) with L. fructus feedings. Therefore, the ration of LDL-C to HDL-C(LDL-C/HDL-C) which is used as an atherosclerosis index was significantly (p<0.05) low, while the HDL-c/TC ration was significantly(p<0.05) high with L.fructus intake. However, no significant were found in serum cholesterols and TG levels in experimentII. The results of these experiments indicate that , regardless of the feeding from, L. fructus can be beneficial in lowering serum TG and LDL-C levels for habitual high-fat diet intakers. L.fructus also seems to be effective in elevating serum HDL-C level, theregy having beneficial effects on atherosclerosis by influencing the serum lipoprotein profile.

Influence of Growth Regulators and Potassium Humate on in Vitro Multiplication of Apple Rootstock M.26 (생장조절제 및 Potassium Humate가 사과대목 M.26 기내 증식에 미치는 영향)

  • This experiment was designed to improve the in vitro production system of apple rootstock M.26 as being influenced by the growth regulators, TDZ, BA, IAA, IBA, zeatin, and GA$_3$. Different levels of potassium humate (KH), known as cytokinin and auxin-like substance, were also supplemented to the MS basal medium along with IBA 0.6 mg/L to find out it effect on root formation in apple rootstock M.26. ID initiate and establish the in vitro multiplication of shoots byway of meristem culture, MS medium added with zeatin 1.0 mg/L was found to be the most suitable, showing the 100% of survival rate of shoot tips. A combination of thidiazuron (TDZ) 0.2 mg/L and NAA 0.5 mg/L promoted the shoot proliferation when shoot tips were used as explants. MS basal medium plus IBA 0.6 mg/L was very effective for root induction, but an addition of potassium humate (250 mg/L) to the medium containing IBA 0.6 mg/L stimulated the induction and proliferation of the rook by far the better.

  • 본 연구에서는 홍천 자운지구 고랭지 농업지역을 대상으로 장기간의 유역조사와 하천 모니터링을 통해 관측된 축척 데이터를 이용하여 비점오염저감 효과를 정량화하고 추후 모니터링 자료로서 탁수와 비점오염원 저감시설의 저감효과와 비점오염원의 효율적인 관리를 위한 기초자료를 제공하고자 하였다. 모니터링 결과 소양호 유역의 오염부하(1차 강우사상) 중 홍천군 자운지구의 유역단위 비점오염 저감효과의 분석에는 강우량과 단위면적당 오염부하를 이용하였으며, 자운천은 SS 5,396,761 kg, COD 82,261 kg, BOD 57,329 kg, T-N 68,711 kg, T-P 3,091 kg이었으며, 오염부하(2차 강우사상)는 SS 320,293 kg, COD 34,588 kg, BOD 22,350 kg, T-N 48,954 kg, T-P 640 kg으로 나타났다. 또한 소양호 유역의 EMC(1차 강우사상) 중 자운천은 SS 829.9 mg/L, COD 12.7 mg/L, BOD 8.8 mg/L, T-N 10.567 mg/L, T-P 0.475 mg/L 이었으며, EMC(2차 강우사상)는 SS 68.6 mg/L, COD 7.4 mg/L, BOD 4.8 mg/L, T-N 10.487 mg/L, T-P 0.137 mg/L로 나타났다. 소양호 유역의 단위면적당 오염부하(1차 강우사상) 중 자운천은 SS 402.0 kg/ha/event, COD 6.1 kg/ha/event, BOD 4.3 kg/ha/event, T-N 5.118 kg/ha/event, T-P 0.230 kg/ha/event 이었으며, 오염부하(2차 강우사상)는 SS 23.9 kg/ha/event, COD 2.6 kg/ha/event, BOD 1.7 kg/ha/event, T-N 3.646 kg/ha/event, T-P 0.048 kg/ha/event로 나타났다. 오염부하에서는 1차 강우에 비해 2차 강우에서는 SS 5,076,468 mg/L, COD 47,673 mg/L, BOD 34,979 mg/L, T-N 19,757 mg/L, T-P 2451 mg/L로 1차 강우사상에 비해 오염부하가 낮은 것으로 나타났다. 또한 EMC에서는 1차 강우에 비해 2차 강우에서는 SS 761.3 mg/L, COD 5.3 mg/L, BOD 4.0 mg/L, T-N 0.080 mg/L, T-P 0.338 mg/L로 1차 강우사상에 비해 EMC가 저감되었다. 단위면적당 오염부하는 1차 강우에 비해 2차 강우에서는 SS 378.1 mg/L, COD 3.5 mg/L, BOD 2.6 mg/L, T-N 1.472 mg/L, T-P 0.182 mg/L로 1차 강우사상에 비해 EMC가 저감되었다. 강우량과 강우강도 그리고 영농활동의 시기에 따른 EMC와 단위면적당 오염부하는 큰 차이를 보였으며, 예년에 비해 적은 강수량으로 인해 탁수와 비점오염부하의 배출이 상대적으로 적었던 것으로 판단된다. 소양호 유역과 같은 넓은 유역에서 시행되는 비점오염원의 저감연구는 오랜 시간 동안 자료를 축적해야 유의미한 평가가 이루어질 수 있으므로 장기적이고 지속적인 모니터링을 통한 유량 데이터 자료구축과 수질분석뿐만 아니라, 발생되는 비점오염 물질의 관리를 위한 체계적인 연구가 지속되어야하며, 아울러 농민의 인식도 변화 등을 포함하는 많은 인자들을 정밀히 조사하고 다각도로 분석하여 저감효과에 대한 지속적인 연구가 필요하다.

Mass Production of Resting Egg of Korean Rotifer, Brachionus plicatilis (L and S-type) (한국산 Rotifer, Brachionus plicatilis (L과 S-type)의 내구란 대량생산)

  • Mass production of resting egg of the rotifer, Brachionus plicatilis was carried out in 1 $m^3$ tank for L-type rotifer and in 1 $m^3$ and 4 $m^3$ tank for S-type rotifer. L-type rotifer was fed on concentrated Chlorella + baker's yeast and only baker's yeast for 15 days. S-type rotifer was fed on concentrated Chlorella in 1 $m^3$ tank for 7 days and frozen Chlorella + baker's yeast in 4 $m^3$ for $6\~8$ days. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for L-type rotifer fed on Chlorella + baker's yeast were $54.5{\times}10^6$ eggs, $30.5{\times}10^6$ eggs and $100{\times}10^3$ eggs, respectively. These were higher than those of rotifer fed on baker's yeast only. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for S-type rotifer fed on concentrated Chlorella in 1 $m^3$ tank were $50\~104{\times}10^6$ eggs, $47\~82{\times}10^6$ eggs and $136\~260{\times}10^3$ eggs, respectively. In 4 $m^3$ tank with frozen Chlorella + baker's yeast, these were $149\~567{\times}10^6$ eggs, $36\~123{\times}10^6$ eggs and $131\~338{\times}10^3$ eggs, respectively. This result suggests that S-type rotifer are better than L-type rotifer for the mass production of resting egg of the Korean rotifer.

A Study on the Marine Traffic Congestion by Analysis of Ship's Domain (선박 점용영역 분석을 통한 해상교통혼잡도 고찰)

  • This research concluded that the area is most sensitively affected by the maritime traffic congestion rather than other factors including the ship's speed and fairway width. We performed a literature review for the area used in Japan, Denmark, and Shanghai ports, and analyzed by comparing with the area in Jinhae, Korea through the observation survey of marine traffic. The result of sensitivity analysis concluded that the congestion is affected by amount of 10% by changing 1 knot of the ship's speed, 18% of 100m of the fairway width, and 34%~43% of 1L of ship's length. By applying the result into the current audit system using $8L{\times}3.2L$, $6L{\times}1.6L$, we found that there is big difference with $3.5L{\times}1.5L$ in Japan, $5.9L{\times}2.2L$ in Shanghai, $4L{\times}5B$ in Denmark, and $3L{\times}2L$ in Jinhae, Korea. For this reason, further studies should be carried out in near future to the area mostly affected by congestion.

Inhibitory effects of Capsicum annuum L. water extracts on lipoprotein lipase activity in 3T3-L1 cells

  • Obesity, an intractable metabolic disease, currently has no medical treatment without side effects, so studies have been actively carried out to find natural compounds that have anti-obesity activity with minimum side effects. In this study, the anti-obesity effects of water extracts of seven Capsicum annuum L. varieties being Putgochu (Pca), Oyee gochu (Oca), Kwari putgochu (Kca), Green pepper (Gca), Yellow paprika (Yca), Red paprika (Rca) and Cheongyang gochu (Cca), were examined through the evaluation of lipoprotein lipase (LPL) mRNA expression level in 3T3-L1 cells (mouse pre-adipocytes). After capsaicin elimination by chloroform defatting, freeze-dried powder of Cca was treated to 3T3-L1 cells and anti-obesity effects were examined by determining the LPL mRNA level using the RT-PCR method. Of the primary fractions, only proven fractions underwent secondary and tertiary refractionating to determine anti-obesity effects. From seven different Capsicum annuum L., there was a significant decrease of the LPL mRNA expression level of 50.9% in Cca treatment compared to the control group. A significant decrease of the LPL mRNA expression level was shown in primary fractions (Fr) 5 (36.2% decrease) and 6 (30.5% decrease) of the Cca water extracts. Due to the impurities checked by UPLC chromatography, Fr 5 and 6 were refractionated to determine the LPL mRNA expression level. Treatment of Fr 6-6 (35.8% decrease) and Fr 5-6 (35.3% decrease) showed a significant decrease in the LPL mRNA expression level. When analyzed using UPLC, major compounds of Fr 6-6 and Fr 5-6 were very similar. Subsequently, we refractionated Fr 6-6 and Fr 5-6 to isolate the major peak for structure elucidation. Treatment of Fr 5-6-1 (26.6% decrease) and Fr 6-6-1 (29.7% decrease) showed a significant decrease in the LPL mRNA expression level. Consequently, the fractions may have a possibility to ameliorate obesity through the decrease of the LPL mRNA expression level.

  • It is necessary to provide a fast and secure handover for seamless real-time multimedia services based on IEEE 802.11. In this paper, we propose FMIPv6 handoff protocol integrating L2/L3 layer based on IEEE 802.11 WLAN environment. In that, we propose a hierarchical key management scheme and authentication mechanism for protecting the handover signaling messages. The number of connections with AAA server is minimized for the fast handover. It is also compared and analyzed the handover cost with previous method.
