Induction of Differentiation on the Human Histocytic Lymphoma Cell Line U-937 by Costunolide (Costunolide의 백혈병 세포주 U-937에 대한 분화 유도 작용)

  • The present work was carried out to examine the effect of costunolide on the growth of several cells and characteristics of U-937 human leukemia-derived cell line. Costunolide produced a potent antitumor activity in vitro dependent on concentration against several tumor cells such as P-388, L-1210 leukemia and SNU-5 stomach cancer cells. However, it showed less cytotoxicity on normal cells such as Maccaccus rheus monkey kidney cells (MA-104) up to 200 ${\mu}M$ concentration. An effect of cell differentiation by costunolide was assessed by its ability to reduce nitroblue tetrazolium (NBT), and to induce phagocytosis of latex particles. In order to establish whether costunolide induces U-937 cells to differentiate toward macrophage or granulocyte, esterase activities was measured. Based on these results, we found that costunolide having cytotoxicity on U-937 human leukemia cells was explained through differentiation inducing activity.

Study on 2-thioDMNQ-S160, A Derivative of Shikonin, Antitumor Constituent of Lithospermum Erythrorhizon (자초의 항암성분 수도체 2-thioDMNQ-S160에 관한 연구)

  • The purpose of this study was to investigate the effect of 2-thioDMNQ-S160, shikonin analogue isolated from Uthospermum Erythrorhizon, on the antitumor activity. In the present study, 2-thioDMNQ-S160 exhibited a significant cytotoxicity against L1210, A549, U937, and 816-BL6 cell lines with IC/sub 50/s of 2.4ug/ml, 2,0ugjml. 4ug/ml and 20 ug/ml, respectively. 2-ThioDMNQ-S160 strongly inhibited adhesion of B16-BL6 cells to gelatin and matrigel coated matrices. Also, 2-ThioDMNQ-S160 exhibited anti-invasive effect of B16-BL6 cells. The T/C% was 123 % in 2-ThioDMNQ-S160 treated group in S-180 bearing ICA mice. These results suggested that 2-thioDMNQ-S160 might be a potent candidate of cancer drug.

  • The rate of the GSH conjugate formation, the inhibition of DNA topoisomerase-I and the cytotoxic activity against L1210 cells of the naphthoquinones showed the same order; 5,8-dimethoxy-1,4-naphthoquinone (DMNQ)>6-(1-hydroxyethyl)-DMNQ>2-(1-hydroxyethyl)-DMNQ; the steric hindrance of the substituents, particularly 2-substutuent, in reacting with cellular nucleophiles must be the main cause for lowering the bioactivities. Acetylation of 2-(1-hydroxyethyl)-DMNQ producing 2-(acetyloxyethyl)-DMNQ potentiated the bioactivities; 2-(-hydroxyethyl)-DMNQ did not react with GSH and the enzyme, and showed $ED_{50}$ of 0.146 mg/ml for the cytotoxcity. Furthermore, the acetylation 2-(1-hydroxyethyl)-DMNQ(T/C, 119%) enhanced the T/C values for the mice bearing S-180 tumor {T/C of 2-(1-acetyloxyethyl)-DMNQ, 276%]. It was assumed that the difference in bioactivities ensued by acetylation was based on the mechanism of the so-called bioreductive alkylation.

Antitumor and Anti-metastatic Effect of Mineral Powder (광물석분의 항암 및 항전이 효과에 관한 연구)

  • Antitumor and anti-metastatic effects of mineral powder(MP) were studied. In the present study, MP did not exhibit the any cytotoxic activity against leukemic cells such as L1210 and U937 tumor cell lines in vitro. Also, MP did not exhibit the any cytotoxic activity against solid cells such as A549 and B16-BL6 tumor cell lines in vitro. However, in vivo, MP exhibit a significant antitumor activity in BDF1 mice bearing Lewis lung carcinoma cells(LLC) with inhibition rates of 46 and $23\%$ at 200 and 100 mg/kg/day, respectively. Furthermore, in pulmonary colonization assay, MP exhibit the inhibitory effect of tumor metastasis. From these results, it was concluded that MP had antitumor and anti-metastatic activity suggesting its application for the prevention and treatment of cancer.

영지(Ganoderma lucidum) 균사체의 액체배양에 의한 세포외 수용성 다당류의 분획 및 항암활성

  • Exo-polysaccharide (BWS) obtained from submerged cultivation of Ganoderma lucidum mycelium was fractionated. Antitumor activity of their fractions was investigated in comparison with the mycelial polysaccharide fractions. Eight kinds (BWS-DN, BWS-DA, BWS-DN-GI, BWS- DA-GI, MWS-DN, MWS-DA, MWS-DN-GI and MWS-DA-GI) of polysaccharide fractions were obtained by DEAE-cellulose ion exchange chromatography and Sepharose CL-4B gel chromatography from BWS and MWS, which were isolated from culture fiuid and mycelial cell, respectively. The anticomplementary activities (ITCH$_{50}$%) of the exo-polysaccharide fractions showing 15% to 30% were lower than those of mycelial polysaccharide fractions showing 15% to 70%. The acidic fractions of BWS-DA and BWS-DA-GI fractionated from BWS, showed the highest activity of 30%. In the MTT assay, BWS-DN and MWS against mouse leukemia L1210 exhibited high inhibition ratio of 86 and 89%, respectively at the concentration of 600 $\mu$g/ml. High inhibition ratio of 50% (IC$_{50}$) was achieved for BWS, BWS-DA and MWS-DA fractions against human colon adenocarcinoma COLO-205 and for BWS-DA, BWS-DN and MWS-DN fractions against human leukemia HL-60 at the concentra- tion of 300 $\mu$g/ml among the six polysaccharide fractions, respectively.

Antitumor Activity of Pedunculagin, one of the Ellagitannin

  • As a part of trials to develop the antitumor agent from tannins isolated from plants, the antitumor activity of peduculagin, an ellagitannin, isolated from Alnus hirsuta var. microphylla was examined in vitro and in vivo. In vitro, the cytotoxicity was determined by 0.4% typanblue dye exclusion method. peduculagin showed the dose-dependent cytotoxicity against human chronic myelogenous leukemia (K-562), human promyelocytic leukemia (HL-60), mouse lymphoid neoplasm (P388), mouse lymphocytic leukemia (L1210) and mouse sarcoma 180(S180) cell lines. $ED_{50}\; values\; (ED_{50})$ of each cell line were 5.30, 0.92, 2.78, 9.35 and $1.38 \mug/ml$ respectively. The most sensitive cell line was HL-60. In vivo, pedunculagin was administered to ICR mouse with the doses of 50 and $100{\;}{\mu}g/ml$intraperitoneally once at 20 days before S180 inoculation. peduculagin showed the antitumor activity and its T/C ratio (%) was 120.82% in the group of both concentrations.

Anti-cell Adhesive Effect of Phenylacetylshikonin Analogues Related to their Cytotoxicity in A549 Cells

  • An attempt to estabilish the relationship between anti-cell adhesive action of phenylacetylshikonin analogues and their cytotoxicity against A549 cells was done. In the one hour incubation with A549 cells,${\alpha}$-methoxyphenylacetyl-(9), ${\alpha}$-acetoxyphenylacetyl-(13), 3,4-methylenedioxyphenylacetyl-(15) and 4-(N,N-dimethylamino)-phenylacetylshikonin (17) analogues showed a high anti-cell adhesive activity $(IC_100; value, 4-8{\mu}g/ml)$, while halophenylacetyl- and dimethoxy- or trimethoxyphenylacetyl analogues expressed no activity at $40{\mu}g/ml$, indicating that the presence of a bulky group at $ C^I-{\alpha}$ and a polar group at C-4 of phenylacetyl moiety may be important. A similar structure activity relationship exists for the 48 hr cytotoxocity $(ED_{50})$ of phenylacetylshikonin analogues in A 549 cells, but not in either K562 or L1210 cells. Furthermore, the difference between $IC_{100}$ values for anti-cell adhesive activity and$ED_{50}$ values for cytotoxicity of potent compound in A549 cells was not so great (1.5 to 3 times). Based on these observations, it is proposed that the anti-cell adhesive action of phenylacetylshikonins might be responsible for their cytotoxicity in A549 cells.

Study on the Antitumor Substance Isolated from Oldenlandiae diffusae Herba (백화사활초(白花蛇活草)로부터 분리(分離)된 항암성물질(抗癌性物質)에 관(關)한 연구(硏究))

  • Hexane fraction of Oldenlandiae diffusae Herba(ODH) which was being used for the treatment of caner in oriental medicine showed the best cytotoxicity against L1210 and A549 in the solvent fractions. Antitumor substance isolated from hexane fraction of ODH was identified as ursolic acid(UA) by photometric analysis. IC50 of UA against cancer cells as SNU-1, HCT15, XF498, SK-MEL2 and A549 was $13{\mu}g/m{\ell}$, $15{\mu}g/m{\ell}$, $12{\mu}g/m{\ell}$, $9{\mu}g/m{\ell}$ and $11{\mu}g/m{\ell}$ respectively. It significantly inhibited the metastasis to lungs and kidneys from pulmonary colonization assay and study on histological changes of organs and showed the enhancing effect on B cell dosage-dependently by FACS analysis. T/C % of UA against S-180 cells was 171 % and its cytotoxicity against SNU-1 iant was confirmed from the morphological changes by elctronic microscopes such as SEM and TEM that it induced undulated membrane 4 hr after UA treatment, and the breakdown of cell membrane and nucleus 24 hr after UA treatment.

Study on Antitumor Activity of Kamisagoonjatang, Kamijihwangtang and Kamigoonjajihwangtang (가미지황탕(加味地黃湯), 가미사군자탕(加味四君子湯) 및 가미군자지황탕(加味君子地黃湯)의 항종양활성(抗腫瘍活性))

  • To evaluate the antitumor activity of Kamisagoonjatang(KST), Kami-jihwangtang (KJT) and Kamigoonjajihwangtang(KKJT), studies were done experimentally. The results were obtained as follows: 1. In cytotoxicity against B16-F10, HT1080, SNU, and L1210, con-centration inhibiting cell growth up to below 50% of control was recognized at $10^{-3}g/ml$ of KKJT. 2. In cytotoxicity against A549, SK-OV-3, XF498 and HCT15, concentration inhibiting cell growth up to below 30% of control was over $400{\mu}g/ml$ of KKJT only and also over $200{\mu}g/ml$ against SK-MEL-2. 3. In Inhibitory effect on activity of DNA topoisomerase I, the $IC_{50}$ was shown $200-400{\mu}g/m{\ell}$, of KST, over $400{\mu}g/m{\ell}$ of KJT and $100-200{\mu}g/m{\ell}$ of KKJT. 4. The T/C% was 122.8 in KJT, 127.4 in KST and 158.4 in KKJ-Ttreated group in S-180 bearing ICR mice. 5. In hematological changes in S-180 bearing ICR mice, numbers of WBC were decreased significantly in KJT and KKJT treated groups as compared with control, whereas those of platelet were increased with no significance in all groups as compared with control. From above results it was concluded that KKJT could be usefully applied for the prevention and treatment of cancer.

Synthesis, Structure, and Antitumor Activity of Novel Platinum(II) Complexes Involving Asymmetric Chiral Diamines as Carrier Ligands

  • New platinum(II) complexes with asymmetrically substituted chiral diamine ligands A₂PtX₂, (A₂ = NH₂CH(CH₃)CH₂NH($c-C_5H_9)$ (apcpa), NH₂CH(CH₃)CH₂NH($c-C_6H_11)$ (apcha); X₂ = 2Cl, isopropylidenmalonate (IPM), 1,1'-cyclobutandicarboxylate (CBDCA)) have been synthesized and characterized by means of elemental analyses, infrared and NMR spectroscopies, and X-ray crystallography. The crystal structures of (S-apcha)Pt[CBDCA] ·3H₂O (orthorhombic, P2₁2₁2(No. 18), a = 6.926(3), b = 15.243(3), c = 19.319(4)Å, V = 2039.5(10) ų, Z = 4, R = 0.072) and (S-apcha)Pt[IPM] ·2.5 H₂O (monoclinic, P2/C(No. 13), a = 9.882(1), b =18.502(1), c = 22.056(1)Å, V = 4032.8(5)ų, Z = 8, R=0.093) exhibit that the platinum atoms achieve a typical square planar arrangement with two nitrogen atoms in cis position and with the chiral center retained. The spectroscopic data disclose that these platinum complexes are stable and their molecular structures are retained in aqueous solution. Among these platinum complexes, the asymmetric diamine-Pt(II) complexes with chloride leaving group exhibit high in vivo activity comparable to cisplatin against leukemia L1210 cell line.
