유한 요소법에 의한 수치 해석을 통하여 자기 트랜지스터(MAGFET)의 동작 특성을 분석하고, 소자의 최대감도를 얻기 위한 설계의 최적 조건을 제시하였다. 제시된 최적 조건에 따라 자기트랜지스터를 CMOS 표준 공정에 의하여 제작하고 전자기적 특성을 측정하였다. 소자의 감도는 활성 영역의 크기보다는 길이(L)에 대한 폭(W)의 비 W/L에 의존하며, W/L = 1 일 때 최대 감도를 나타내었다. 제작된 소자의 상대 감도는 2.53 %/T 이었다.
새송이버섯의 부가가치 이용방안 연구의 일환으로 마이크로웨이브 추출을 이용하여 새송이버섯의 추출수율, 전자공여 작용, 총 폴리페놀 함량 및 아질산염 소거작용의 효율적인 추출조건을 설정하고자 하였다. 에너지 용량(60, 90, 120 W), 시료 대 용매비(2, 5, 10 g/100 mL) 및 추출시간(1, 5, 10, 15 min)에 따라 물, $50\%$ 에탄올과 $99\%$ 에탄올을 용매로 사용하여 추출하였다. 에너지 용량을 $60\~120$ W로 증가시킴에 따라 각 추출물의 항산화성 성분들은 60 W에서 가장 높은 값을 보였고 90 W에서 평형에 이르러 변화하지 않았다. 시료 대 용매비를 1:50에서 1:10으로 감소시킴에 따라 추출수율을 제외한 각 추출물의 유용성분들은 1:10에서 가장 높았다. 추출시간의 영향에서 전자공여작용을 제외하고는 1분에서 가장 높은 값을 보였다. 5분 이상의 추출시간에서는 거의 영향을 받지 않았다. 전자공여작용은 추출시간이 증가함에 따라 높은 값을 보였다 추출용매의 영향에서는 물$>50\%$ 에탄올$>99\%$ 에탄을 순서로 높은 값을 보였다. 따라서 에너지 용량, 시료 대 용매비, 추출시간을 각각 60 W, 1:10(g/mL), 1 min으로 설정하여 물을 용매로 사용하였을 경우 새송이버섯의 항산화 성분을 최대로 추출할 수 있는 적당한 추출조건이었다.
The feasibility study of UV-disinfection system was performed for disinfection of effluent from wastewater treatment plant. Three low-pressure UV lamps of 17, 25, and 41 W were examined with various flow rates. Low-pressure UV lamps of 17W were examined with various turbidity, DOM (dissolved organic matter), and SS (suspended solid). The pilot plant was a flow-through type UV-disinfection system, and the range of exposure time varied from 5 to 40 seconds, turbidity from 0 to 40 NTU, DOM from 0 to 30 mg/L, and SS from 10 to 40 mg/L. The 41W lamp demonstrated complete disinfection showing no survival ratio in all the experimental conditions, and generally 17W and 25W lamps also showed high removal ratio over 97%. For the same UV dose (UV intensity times exposure time), high intensity-short exposure conditions showed better disinfection efficiency than low intensity-long exposure conditions. While the effects of turbidity and DOM were not apparent, the effects of SS was significant on the disinfection efficiency which indicates that SS control before UV-disinfection appears to be necessary to increase removal efficiency. Considering characteristics of effluent from existing wastewater treatment plants, cost-effectiveness, stable performance, and minimum maintenance, the flow-through type UV-disinfection system with high intensity and low-pressure lamps was thought to be a competitive disinfection system for wastewater reclamation.
샤르도네 품종을 이용한 포도주 양조용 효모의 발효 특성은 다음과 같다. S6U 균주는 다른 균주에 발효속도는 느렸지만 발효 종료시점에 완전히 발효되는 특징을 보였다. W-3 균주는 발효중 산 생성량이 적었으며 (0.75%), UCD530과 AC- 균주로 발효된 포도주에는 산류 extract 함량이 각각 3.26 mg/L 및 3.22 mg/L로써 다른 균주에 비해 많았다. CEG와 CS2 균주로 발효된 포도주는 특히 황색도 값이 높았으며, EC1118 균주는 다른 와인 효모에 비해 아세트알데하이드(49.9 mg/L)을 많이 생성하였다. 효모의 알콜 내성 특성을 나타내는 methylene blue dyeing ratio에 있어서, EC1118 및 CY3079 균주는 약 15% 미만의 낮은 methylene blue dyeing ratio을 나타내어 이 두 균주가 다른균주에 비해 높은 알콜 내성이 있음을 보여주었다. 글리세롤 함량에 있어서 UCD530 균주는 다른 균주에 비해 극히 많은 생성능을 보였고 또한 succinic acid 및 lactic acid 생성량도 높아 전형적인 S. bayanus 특징을 나타내었다. 포도주의 Acetic acid 함량에 있어서 UCD530, EC1118, AC-, CY3079, W-3 균주로 발효된 것은 170 mg/L 이하로 비교적 낮았으며 S6U, CEG, CS2로 발효된 포도주는 350 mg/L 정도로서 높았다.
한국산 나리속 7종의 화분 관찰에 의한 나리속의 종간 유연관계를 구명하고자 본 실험을 수행하였다. 화분 표면을 주사전자현미경으로 관찰한 결과 모두 단립(monad)으로 발아구가 한 개인 장형(prolate)을 띠었다. 화분 길이와 폭의 비율은 종별로 차이를 나타내어서 말나리가 2.59로 제일 큰 장타원형을 나타냈고, 하늘말나리는 1.82로서 원형에 가까운 타원형을 나타냈다. 화분의 길이와 폭은 참나리가 각각 115.90와 $46.54{\mu}m$로 가장 컸으며 하늘말나리는 길이와 폭이 각각 $70.33{\mu}m$와 $38.72{\mu}m$로 가장 작았다. 외막의 표면 무늬는 망상(reticulate)으로서 망강(lumen)은 형태가 일정하지 않은 다각형으로 7종 모두 유사한 것으로 나타났다. 나리속의 종간 분류는 말나리와 중나리가 오차범위 내에서 유사한 화분 크기를 지니고 있었다. 한편, 나머지 종들의 화분 길이는 뚜렷한 차이를 나타내어 이러한 결과를 바탕으로 화분의 길이를 통한 나리속의 분류는 외막 모양이나 화분의 단면적보다는 화분 크기가 유용할 것으로 생각된다.
본 연구에서는 소목(Caesalpinia sappan L.) 분말을 활용한 모발 염색의 최적 조건을 결정하였고, 키토산 처리가 소목 분말의 모발 염색성에 미치는 영향을 조사하였다. 소목 분말의 혼합비율, 염색시간 및 온도에 따른 염색성을 확인한 결과 소목 분말과 물의 비율을 1:3(w/w)로 하여 40℃에서 60분 동안 처리하는 것이 모발 염색에 가장 효과적인 것으로 나타났다. 또한 키토산의 분자량, 처리 농도와 처리 순서가 소목 분말의 염색성에 미치는 영향을 확인한 결과 저분자량(~30 kDa)의 키토산을 1%(w/w)의 농도로 소목 분말 처리에 앞서 전처리를 하는 것이 염색성 향상 및 유지에 가장 효과적이었다. 결론적으로 모발 염색에 있어서 저분자량의 키토산은 매염제로서 소목 분말의 염색 효율과 유지력 향상에 크게 도움이 되는 것으로 판단된다.
A microbial cryoprotectant formulation using a W/O/W multiple emulsion system was developed. The psychrotolerant microorganism, B4, isolated from soil in South Korea, was observed by the drop freezing method, in which the microorganism sample inhibited ice nucleation activity. The antifreeze activity was eliminated when the microorganism sample was treated with protease, indicating that the antifreeze activity was due to the presence of antifreeze protein. The result of the l6S rDNA sequencing indicated the B4 strain was most closely related to a species of the genus Bacillus. Culture broth of B4 strain (Bacillus sp.) and rapeseed oil containing 1 % polyglycerine polyricinolate (PGPR) were used as core and wall material, respectively. The most stable W/O emulsion was prepared at a core/oil ratio of 1:2. The highest W/O/W emulsion stability was achieved when the primary emulsion to external aqueous phase containing 0.5% caster oil polyoxyethylene ether $(COG25^{TM})$ ratio was 1:1. Microcrystalline cellulose showed better W/O/W emulsion stability than other polymer types. The viability of cells in a W/O/W emulsion was higher than free cells during storage at $37^\circ{C}$. An acidic pH and UV exposure decreased the viability of free cells, but cells in W/O/W emulsion were more stable under these conditions.
This study presents the characteristics of growth and morphology of Anabaena sp., a representative filamentous cyanobacterium, depending on temperature variation from 10 to 30 ℃. Both the filament density (or number) and its length of Anabaena were highly affected by temperature, as well as growth stage. Rapid growth at a higher temperature led to an increase in Anabaena filament density, as well as optical density at 680 nm (OD680). However, the number of vegetative cells within a single filament of Anabaena grown at 30 ℃ was smaller than those grown at lower temperatures, due to the intercalary division of the filament. Of the three different cells comprising a single Anabaena filament, the vegetative cell marginally affects the growth of Anabaena. The main dimensions of the vegetative cell, i.e., length and width, depend on the temperature and growth stage. The length-to-width (L/W) ratios of vegetative cells and akinetes were relatively consistent regardless of the temperature. However, in vegetative cells with dichotomous growth, the L/W ratio shows clear differences depending on their growth stage. It has been demonstrated that the L/W ratio could be used as an indicator to indirectly predict the growth stage of on-sit Anabaena samples.
The availability of enokitake cultural waste for Lentinus edodes cultivation was investigated, although hardwood sawdust has traditionally been used as a substrate for this fungus. Firstly, physiochemical characteristics of cultural waste were analysed. Secondly, mycelial growth characteristics and fruiting yields of L. edodes on waste treated in some methods were determined. Physiochemical characteristics of enokitake cultural waste showed that the millwaste complex was a little degraded by enokitake fungus and suggested the probability that most component lost by enokitake could be rice bran. Mycelia of L. edodes grew and fruited well on waste supplemented by fresh rice bran and Quercus sawdust although didn't on waste only. Mycelial growths of these fungi on waste were accelerated when supplemented by rice bran to the percent of 40(w/w) but decreased or suppressed at above ratios(30, 40%, w/w). Supplementations of oak sawdust at above 40%(w/w) of the waste and rice bran at 20%(w/w) of the sawdust allowed such a good mycelial growth as to be selected as a pertinent mixing ratio for fruiting medium. A fruiting yield on enokitake cultural waste supplemented by oak sawdust (at 40% of the waste, w/w) and rice bran (at 20% of the sawdust, w/w) was not inferior to that on oak sawdust supplemented by rice bran only (at 20% of the sawdust, w/w). These results indicated strongly the potentiality of enokitake cultural waste as raw materials for shiitake cultivating substrates.
