Evaluation of Resistance to the Aphid (Aphis glycines Matsumura) in Soybean Cultivars and Germplasms

  • Native of soybean aphid (Aphis glycines Matsumura) is an Asia and aphid is one of the dangerous pests in soybean [Glycine max (L.) Merr.]. High density aphid populations can reduce crop production by causing severe damage. The objective of this study was evaluation of resistance to the soybean aphid in soybean cultivars and germplasms. A total of fifty five soybean cultivars or germplasms, including two susceptible and two resistant check varieties, were infested to evaluate their resistance in the field cage and greenhouse test by aphid colonies which derived from wild collected one soybean aphid biotype in Korea. The average number of reproduced soybean aphid was evaluated with 62.7 aphids in the resistant check variety PI 567598B and also estimated with 1,807 aphids for susceptible check variety Williams 82. In soybean varieties and germplasms, the average reproduced soybean aphid populations ranged from the lowest 497 aphids for Junjeori to the highest 3,862 aphids for Mansu. About seventy six percent of soybean cultivars and germplasms were shown high density soybean aphid populations when compared with another susceptible check variety PI 567543C in the field cage test. From the greenhouse test to evaluate aphid index, 87.3% of soybean cultivars or germplasms presented aphid index with 9.0. No soybean cultivars and germplasms were observed with soybean resistant phenotype when regarded a aphid resistant level as less than 10% aphid reproductions compared with susceptible check Williams 82. Although no Korean soybean cultivars were identified with resistant trait to the soybean aphid, we found one great resistant genetic resource PI 567598B in this study. This result will be helpful to further study for providing useful genetic information for soybean researchers.

Optimized Shoot Induction and Histological Study of in vitro Cultured Korean Soybean Cultivars

  • Soybean is the one of recalcitrant legume species for shoot induction. Shoot regeneration via direct organogenesis was investigated in five soybean cultivars, 'Dawon', 'Pungsan', 'Daewon', 'Taekwang' and 'Chongdoo 1' by using cotyledonary node explants. Out of 5 soybean cultivars, an efficient shoot regeneration condition was developed in the two soybean cultivars, 'Dawon' and 'Pungsan'. When various kinds of plant growth regulators with different concentration were estimated, the optimum medium condition for shoot induction in both soybean cultivars was MS + B5 vitamin supplemented with BA at concentration 2 mg/L. In addition, shoot formation efficiency was increased with 97.09% and 93.88% by the pretreatment of BA onto the explants before in vitro culture in both cultivars. Shoot induction in 'Dawon' cultivar was originated from epidermal tissue and sub-epidermal layers when histological changes were investigated under shoot regeneration after culturing cotyledonary node segments on shoot induction medium for 0 to 21 days. Especially, cell dedifferentiation was observed from parenchyma cells to meristematic cell in 3-day cultured segments.

Molecular Marker Analysis for Resistance of Soybean Cultivars to Soybean Cyst Nematode

  • Soybean cyst nematode (Heterodera glycines Ichinohe; SCN) is an important soybean pest and the use of resistant cultivars is the effective method to reduce or eliminate SCN damage. However, breeding for SCN resistance is difficult and expensive by the oligogenic nature of the resistance and genetic variability in the pathogen. Fortunately, SCN resistance loci, rhg1 and Rhg4 are generally accepted as a necessity for the development of resistant genotypes using any source of resistance. In this study, resistance of 44 Korean soybean cultivars to SCN was tested using two molecular markers. Seonheukkong and Pokwangkong were the homozygous to rhg1 locus. Seven cultivars were susceptible to SCN based on Satt309 marker linked rhg1 locus. All Korean cultivars estimated in this study were recessive homozygous to Rhg4 locus and were susceptible in the PCR reaction using primer 548/563 linked to the Rhg4 locus conferring resistance to SCN race 3. Among 44 cultivars estimated, seven cultivars were susceptible to SCN in both Satt309 and primer 548/563 markers. Based on both Satt309 and primer 548/563 markers, there is no resistant cultivar to SCN in Korea. Therefore, SCN resistant cultivars need to be developed in the future. These two markers can be used for improving SCN resistant cultivars.

Difference in Growth, Yield and Isoflavone Content among Soybean Cultivars under Drained Paddy Field Condition

  • Naturally occurring soybean isoflavones are known to be influenced by various genetic and environmental conditions. Growth, yield, and isoflavone content were determined in four different cultivars of soybean grown under drained paddy and upland fields. Most of growth characteristics and yield components of four different soybean cultivars harvested in drained paddy field were greater than those in upland field, regardless of cultivar. By means of high performance liquid chromatography, total daidzein and genistein contents of soybean in drained paddy field were increased up to 40 and 35%, respectively, compared with those in drained paddy field. Besides isoflavone contents, the growth and yield of soybean were significantly affected by cultivar and field conditions, indicating the necessity of genetic program for soybean cultivars appropriate to drained paddy field conditions. In conclusion, converting paddy field into upland may effectively improve soybean cropping system, especially in terms of isoflavone increment under paddy field conditions.

대두품종의 렉틴 함량과 면역학적 특성 (Concentration and Immunological Characteristics of Lectin in Soybean (Glycine max L.) Cultivars)

  • 대두(Glycine max L.) 종자로부터 분리된 렉틴을 토끼에 면역화시켜 렉틴항혈청을 얻었다. 이를 이용한 면역화학적 방법을 사용하여 대두렉팅의 면역화학적 특성과 6개 대두 품종의 종자에서 렉틴의 존재를 조사하였다. Crossed immunoeiectrophoresis한 결과 종자는 4종의 침강선을 보임으로서 렉틴에 4종의 항원성분이 존재함을 보여 주었다. 1% agarose gel을 이용하여 Immunodouble diffusion test한 결과 6개 대두 공시품종 모두 렉틴항체에 대하여 침강선을 나타내었고 푸종간 침강선은 모두 유사한 양상을 보였다. ELISA 방법에 의하여 6개 대두 품종간의 렉틴함량을 조사, 비교한 결과 렉틴함량은 장엽콩>광교>황금콩>백운콩>장백콩=팔달콩 순으로 나타났다. 공시품종의 조자로부터 분리한 렉틴은 모두 토끼 적혈구는 응집시켰으나 Fusarium sp., Alternaria sp., Cuvalaria sp. 등의 곰팡이 포자는 응집시키지 못하였다.

Effects of Low Dose Gamma Radiation on the Root Growth of Soybean Cultivars

  • ${\gamma}-Radiation$ at very low doses frequently has a stimulating or hormetic effect on the growth of organism. Effects of low dose ${\gamma}-ray$ irradiation on the root growth of soybean cultivars were investigated and hormetic effects by environmental conditions were compared with the occurrence of increased economic yield, seeds of cultivars were irradiated with the dose of $0.5{\sim}20Gy$ and cultivated in growth chamber controling temperature, humidity, light, greenhouse and field respectively. To understand hormetic effect on root growth of cultivars and the difference of hormetic effect by cultivation environment, harvested root of soybean cultivars were scanned with image file, and root surface area, root length, root average diameter etc. were examined by WinRhizo program. Also, dry weight of cultivars was examined. Root growth and dry weight of soybean cultivars showed apparently hormetic effect at cultivation of growth chamber condition. In field experiment executed for whole life cycle, yields of pea were not different significantly in each ${\gamma}-ray$ irradiated cultivars but weight of one hundred peas increased in whole ${\gamma}-ray$ irradiated cultivars. Increment of yield was assumed to be induced through shortening of maturing stage caused by ${\gamma}-ray$ hormesis in early growth stage.

논 재배 시 한국 콩 품종의 수량성 관련요인분석에 의한 품종군 분류 (Cluster Analysis of Korean Domestic Soybeans based on Yields Characters in Paddly Field Cultivation)

  • In order to select the suitable soybean cultivars for paddly field cultivation, growth response and yields characters were investigated with 37 Korean domestic soybean cultivars, in Yeoncheon, Gyeonggi province. The results can be summarized as follows ; When the soybeans were cultivated in paddy field, significant differences were observed in growth responses at each growth stage and yields among the cultivars. Yields of six cultivars, such as Danbaekkong, Paldokong, Pungsannanulkong, Myeongjunamulkong, Saeolkong, and Namhaekong, were less than 200kg/10a, however, seven cultivars, such as Shinpaldalkong 2, Sohokong, Dormikong, keumkangkong, Bukangkong, Dajangkong, and Geomjeongkong 2, showed highest yields ranged from 301~385kg/10a. At V5 stage, highly positive correlation(r=091$^{{\ast}{\ast}}$) was observed between the top dry weight and leaf area while negative correlation was existed between T/R ratio and root dry weight. However, at R2 age, leaf area was correlated with both top and root dry weight while T/R ratio was correlated with leaf area and top dry weight. Thirty-seven soybean cultivars used in this experiments were classified into 3 different clusters based on 19 important agronomic characters, and genotypic properties of the cultivars were similar in each cluster. In results, high yields could be achieved by selected cultivation of suitable soybean cultivars such as Shinpaldalkong 2 which showed early maturation date and highly ripening ratio.

대두 및 콩나물 부위별 Isoflavones 함량 비교 (Comparison in Isoflavone Contents between Soybean and Soybean Sprouts of Various Soybean Cultivas)

  • In this study, isoflavone (genistein, genistin, daidzein, daidzin) contents in various parts of twelve soybean cultivars and three soy sprouts were determined by high performance liquid chromatography with UV detector. Three cultivars of soybean were selected and cultured in the lab to produce sprout for five days. Total isoflavone (Total IF) varied greatly among differnt breeds of soybean in range of 99 - 649.9 $\mu\textrm{g}$/g and 522.3 - 1,277.7 $\mu\textrm{g}$/g respectively, domestic and foreign cultivars. There were greatly difference in total IF of various parts of the soybean sprouts. Sprout from the Myunjunamul-kong appeared to have 69% genistein and 22% genistin in head part, and 30% and 62% of daidzin and daidzein, respectively, in root. Meanwhile, the sprouts from Junjori contains most (84%) of daidzein in its root. Sprout from chinese black-soybean had the largest amount of genistein among the sprouts but, there were no differences in the average genistein content between three selected black and non-black soys. The glycosidic form of IF were dominant compared to aglycone forms both in soybean and sprouts by 24 times and 12 times, respectively, suggesting that during the sprouts cultivation glycosidic forms could change to aglycone forms. There are no difference in total content between genistein + genistin and daidzein + daidzin in soy and soy sprout. Therfore, considering the total IF contents, the intake of 1 soy sprout is similer to 1.5 times as soybean.

Comparison of Sudden Death Syndrome in Responses to Fusarium solani f. sp. glycines between Korea and U.S. Soybean Lines

  • In order to identify the responses of Korean soybean cultivars to sudden death syndrome (SDS), forty-two Korean cultivars and three check cultivars (Hartwig and PI 520733 are resistant; Hartz 6686 is susceptible) were tested with sorghum seed inoculum infested with Fusarium solani f. sp. glycines isolate 171 in the greenhouse. This isolate has blue pigment cultural shape on potato dextrose agar (PDA) medium. All Korean cultivars inoculated with F. solani isolate 171 showed the typical SDS symptoms and disease severity on soybean leaves in each cultivar varied at 4 weeks after inoculation. Nine cultivars were included in the most SDS susceptible group and six cultivars were included in the most susceptible group based on Duncan's multiple range tests (P$\leq$0.05). In results of the LSD analysis for SDS the resistant group, a total of twenty-five Korean cultivars were included in the same SDS resistant group as PI 520733 or Hartwig and fourteen Korean cultivars were included in the same SDS susceptible group as Hartz 6686. In the second experiment, ten Korean cultivars, ten U.S. cultivars, and one introduced line were compared in the same way as the first experiment Disease severity ranking of check cultivars, Hartwig, PI 520733, and Hartz 6686, were the same as in the first experiment. Within Korean cultivars, seven cultivars showed the consistent severity proportions of leaf symptoms. Disease rankings of these cultivars in this experiment were the same as those in the first experiment. Three US cultivars: Hartwig, Hartz 5454, and Forrest, three Korean cultivars: Keunolkong, Myeongjunamulkong, and Jinpumkong 2, and one introduced line, PI 520733, were included in the highest SDS resistant group. Shinphaldalkong 2, Milyang 87, and Samnamkong consistently showed the highest SDS susceptibility in both experiments. Average disease severity in the first and the second experiment were 49.56% and 45.39%, respectively.

Physiological Responses of Soybean Cultivars to Fusarium solani f. sp. glycines Causing Sudden Death Syndrome

  • Six soybean cultivars having different SDS susceptibility were planted with sorghum seedinoculum infested with F. solani isolate 171 in the greenhouse. First leaf symptoms appeared on unifoliar leaves at 9 days after inoculation and all cultivars showed the typical leaf symptoms at 13 days after inoculation, when trifoliar leaves emerged. Leaf symptoms development in susceptible cultivars was faster than in resistant cultivars. Leaf symptom severities during the period of 25 to 29 days after inoculation showed a significant difference between cultivars which had SDS resistance and sus ceptibility. In this period, area under the diseaseprogress curve (AUDPC) of Hartz 6686 was the highest and that of PI 520733 was the lowest. SDS caused serious damage to the growth of soybean in all cultivars. Average reductions of growth rate of root fresh weight and dry weight were greater than those of plant tops. Duyu-kong showed less severe leaf symptoms than that of SDS suscetible cultivars; however, average growth rate of plants top and roots of this cultivar was less but not significantly different than those of SDS susceptible cultivars. In all cultivars, as severity of leaf symptoms increased, plant top weight decreased. Root rot symptoms were observed in all cultivars before leaf symptoms appeared. Average proportions of tap root reddish-brown discoloration of all cultivars was up to 75 % at 15 days after inoculati on; however there was no significant differenc between cultivars at each rating date. Appearances of leaf symptoms on leaves varied in each cultivar. SDS resistant cultivars had a significantly higher level of crinkling than susceptible cultivars and SDS susceptible cultivars had a significantly higher level of necrosis than resistant cultivars. Further study will be needed to identify the relationships between the physiological growth rate and SDS severities in soybeans.
