Microscopic Study of Decomposition-Inhibition in Stabilized $ClO_2$ Gas in Kidney of Rat with Passage of Time (시간경과에 따른 안정화 이산화염소(Stabilized $ClO_2$)의 콩팥조직 부패억제에 대한 현미경적 연구)

  • The stabilized $ClO_2$ gas has been used for many years by the food industry as a strong oxidizing and sanitizing agent that has broad and high biocidal effectiveness. Therefore, "stabilized $ClO_2$" gas may be used in fields of disinfectant and sterilization. But, there have been few studies on the decomposition-inhibition effect of stabilized $ClO_2$ gas with passage of time. The main purpose of this study was to examine the decomposition-inhibition effect of stabilized $ClO_2$ gas and the morphological change of kidney by measuring of the light and electron microscope. Sprague-Dawley (SD) rats weighting from 230 gm to 250 gm were used as experimental animals. Under ether anesthesia, the right kidney of rat was obtained. Put each sample in $37^{\circ}C$ and humidity $80{\pm}5%$ incubator, we obtained each sample after 0 day, 1 day, 2 days, 3 days, 4 days and 5 days. We proceeded the observation of light and electron microscope. The results obtained in this study reveal that stabilized $ClO_2$ gas is an effective decomposition inhibitor until 2 days that was conducted at $37^{\circ}C$ and humidity $80{\pm}5%$ conditions.

Antimicrobial and Anti-halitosis Effects of Alnus firma Extracts (사방오리나무 추출물의 항우식 및 항구취 효과)

  • To investigate the antimicrobial and anti-halitosis effects of Alnus firma extracts and gallic acid (GA) isolated from A. firma, we measured their antimicrobial activities against oral pathogens and their inhibitory effects on the cell adhesiveness and acid production of oral pathogens. In addition, the levels of volatile sulfur compounds were determined by using oral chroma. The dichloromethane (DCM) fraction has broad antimicrobial activity, and the ethylacetate (EA) fraction showed a relatively high level of antimicrobial activity against Streptococcus mutans and Porphyromons gingivalis. Especially, the GA and DCM fractions had significant inhibitory effects on the attachment and acid production of S. mutans and Streptococcus salivarius, respectively. The 2% MeOH extract of A. firma showed a significant inhibitory effect on the production of volatile oral compounds, such as hydrogen sulfide, methyl mercaptan, and dimethyl sulfide, which can cause bad breath and halitosis. Two percent GA also had a significant inhibitory effect on the production of hydrogen sulfide. Our study showed that the active fractions and GA of A. firma could be suitable resources for development as a natural antibiotic agent for the treatment of infectious oral diseases.

Morphological Change of Men's Hair Shaft by Weathering (풍화작용에 의한 정상모발의 형태학적 변화)

  • The morphological changes in normal and weathering hair shafts of the human scalp were investigated by using the transmission and scanning electron microscopes. The hair shaft composed of cuticular layer, cortex and medula. The surface of normal hairs are smooth and covered by imbricated cuticular scales. The cuticular layer consists of five to seven cuticle cells. These cells, which are flat and thin, measuring about $100{\mu}m$ long and $0.4{\mu}m$ thick, appears intercellular membrane complex in diameter 25 nm. The cortex composed of melanin granules and cornified cells, which multicomponent concentric microfibrils in diameter about 8 nm give rise to macrofibrils in diameter $0.5{\mu}m$ to $0.8{\mu}m$ encased in limiting membrane. The melanin granules are spherical shaped about $0.5{\mu}m$ in size and scattered between macrofibrils. The medulla in the normal hairs are $16{\mu}m$ in diameter centrally region of cortex. Normal hair shafts undergo progressive degenerative changes due to a variety of environmental insults. In the initial weathering process of hair, the cuticular scales became irregularly raised and broken, and then cuticle cells formed cytoplasmic vacuolation, following dissociated intercellular membrane complex, ultimately entirely lost and nuded cortex. Occasionally, transverse fissures were seen at hair shafts indicating that the hairs were deteriorated. Complete removal of the cuticular layer in the heavily damaged cortex portions appeared splitting of the cortical cell into its macrofibrils and scattering of melanin granules.

An Microscopic Study of the Irradiation Effect on the Submandibular Glands of the White Rat (방사선조사가 흰쥐의 악하선에 미치는 영향에 관한 현미경적 연구)

  • This study was undertaken to observe the histopathologic changes in submandibular glands of the white rats when exposed to megavoltage fractionated dose of CLINAC 2100 C-D 6 MV X-RAY irradiation and 42 female white rats, weighing approximately 100gm, were divided into control and 2 experimental groups. At sacrifice, submandibular glands were excised and examined microscopically and electromicroscopically. The results were as follows : 1. The acinar cells of submandibular gland showed damage varied with dose, 12 Gy resulted in very mild injury while 24 Gy caused extensive injury. 2. The acinar cells of sumandibular gland showed similar ultrastructual alterations, appeared as pleomorphic nucleus, decreased numbers and pleomorpgism of secretory granules, distention of rough endoplasmic reticulum, expansion and pallor appearance of mitochondria, and hypertrophy of Golgi complex. 3. A serous cells were the most sensitive components, displaying morphological alterations of radiation damage as early as 3 hours, followed by submandibular seromucinous cells and secretory tubular cells. 4. The mucous cells, as well as the whole ductal lining cells, displayed no significant alterations. 5. No evidence of microvascular injury through whole experimental groups indicated that microvascular impairment dose not contribute to early. salivary gland injury.

Isolation and Characteristics of a Bacterium Removing Chemical Softener, Organo-polysiloxane (화학유연제, Organopolysiloxane 분해세균의 분리 및 특성)

  • Thirty three strains of bacteria were isolated from the wastewater and soil contaminated a chemical softener, nrganopolysiloxane. Of these, five strains which showed higher activities for removal this chemical were finally selected for further study. By five strains the 2,500 mgll chemical softener was removed 65.2-67.9% at $37^{\circ}C$ for 5 days by shaking. The pH optimum for growth of W3721, S3712, and S3723 strain were at around pH 7.0-7.5, and W2811, and W2823 strain were at pH 6.5-7.0, respectively. The temperature optimum for growth of W3712 strain was at $37^{\circ}C$ and the other four strains were at TEX>$30^{\circ}C$. The optimal pH and temperature for removal by W3712 strain was initial pH 7.0 and $37^{\circ}C$ respechvely. The W3712 strain was identified and named as Corynebacterium pseudodiphtheriticum W3712 based on its morphological and physiological characteristics.

Microscopic Study of Decomposition-Inhibition Effect in Grapefruit-seed Extract in Rat Kidney (자몽추출물의 콩팥조직 부패억제 효과에 관한 현미경적 연구)

  • It has been well known that grapefruit seed extract (GSE) have effects on the deodorization and a wide range disinfectant. But anybody do not research about the decomposition-inhibition effect of GSE. The present study was performed to investigate the decomposition-inhibition effect of GSE by measuring the microscope and electroscope observation with passage of time. A total of 30 healthy Sprague-Dawley (SD) rats weighting from 230 gm to 250 gm were used as experimental animals. One group that do not treated by GSE named control group and the other group that treated by 55% GSE named Experimental group. Under ether anesthesia, right kidney was obtained. Put each sample in $37^{\circ}C$ and humidity $80{\pm}5%$ incubator, we obtained each sample after 0 day, 1 day, 2 days, 3 days, 4 days and 5 days. 4 ${\mu}m$ of paraffin sections were obtained, stained H-E stain and observed by use of a microscope. Morphological change in tissue was similar to control 1 day group and experimental 3 days group. Therefore, decomposition-inhibition effect of GSE continued about 2 days and it protected necrosis. According to above results, the author suggest that 55% GSE is an effective decomposition inhibitor until 2 days on $37^{\circ}C$ and humidity $80{\pm}5%$ conditions.

Effective Components on the Taste of Kanjang made with Barley Bran using Multiple Regression Analysis (중회귀 분석을 이용한 보리간장 맛에 영향을 미치는 성분 조사)

  • This study was conducted to find effective components on taste of kanjang made with barley bran. The result of stepwise multiple regression analysis with transformation of variables was that the 70.1% and 80.8% of taste of barley kanjang could be explained at step 6 and step 15, respectively, calculated from absolute value. And, the 83.0% and 84.3% of taste of barley kanjang was explained at step 6 and step 15, respectively, calculated from relative value transformed with logarithm. The contributing proportion of the ${\alpha}-ketoglutaric$ acid was the highest followed by citric acid, lavulinic acid and glutamic acid computed from absolute value, and the ${\alpha}-ketoglutaric$ acid was the highest followed by fructose, glutamic acid and valine calculated from relative value trans-formed with logarithm.

Biochemical and Scanning Electron Microscopic Study on the Enamel Organ of Fetal Rat following a Ingestion of Fluoride (불소투여에 따른 태내백서 치아의 생화학적 및 주사전자현미경적 연구)

  • The present study has been carried out to investigate the effect of fluoride toxicity on the morphology as well as inorganic chemical constituents of rat teeth. Rats were administered sodium fluoride at dose of 0 ppm, 100 ppm, 200 ppm and 300 ppm at the beginning of pregnancy. Animals were perfused intravascularly with glutaraldehyde and the incisors were removed. Changes in the protein composition of the secretory and maturation enamel were investigated using polyacrylamide gel electrophoresis (SDS PAGE). And the enamel surface of incisors was examined under scanning electron microscope (SEM). Changes of protein quantities were found significantly in high levels fluoride administration for experimental groups compared with control. The SDS PAGE analysis demonstrated as follows In control group, secretory phase enamel protein, amelogenins, was detected more quantities than experimental group. The enamelin, presence in maturation phase enamel , showed more quantifies than control enamel with an increasing fluoride concentration in the drinking water. Also, the scanning electron micrographic data showed hypoplastic, tough, uneven, pitted and cracked enamel surfaces covered with granular deposits as a result of excessive intake of fluoride. From these results we conclude that high dose of fluoride administration leads to severe structural alterations on the enamel surface and these structural changes could be through defective mineralization.

Autophagy and Oral Cancer (자가포식작용과 구강암)

  • Autophagy plays an important role in cellular homeostasis and survival for cell recycling and various stresses within the cell. Recent studies have shown that autophagy activity modulates the expression of oncogene and tumor suppressor genes, leading to the development or suppression of cancer. Induction of autophagy is involved in preventing cancer development in normal cells and plays an important role in prompting a specific cell death mechanism in cancer cells with damaged cell death function. It is also known that autophagy inhibition increases the therapeutic efficacy by sensitizing cancer cells that are resistant to chemotherapy. However, the role of autophagy has not yet been fully understood in cancer treatment. Oral squamous cell carcinoma accounts for more than 90% of oral cancer and is the sixth most common cancer in the world. The incidence of oral cancer has increased by 50% over the last 20 years and the mortality rate is over 40% within 5 years after the onset. In oral cancers, the role of autophagy are described to look for tumor inhibitory in the early stages of tumor formation, like other cancers, indicating the dual functions involved in tumor cell survival include tumor progression stages. This review summarizes the various roles of autophagy in cancer cells and suggests the possibility of autophagy as a promising target for effective oral cancer therapy.

Fine Structural Analysis of Principal and Secondary Eyes in Wandering Spider, Pardosa astrigera (배회성 거미 (Pardosa astrigera) 주안과 부안의 미세구조적 분석)

  • The wandering spider, Pardosa astrigera, had four pairs of ocelli that arranged in three rows on the cephalothorax. Along the anterior margin lay a pair of small anterior median (AM) eye flanked on each side by anterior lateral (AL) eye. Two large posterior median (PM) eye was situated on the clypeus behind the anterior row and still more posteriorly was a pair of posterior lateral (PL) eye. The visual cell of retina consisted of cell body, rhabdome, and intermediate segment. Bipolar neuron was found in anterior median eye (principal eye) and unipolar neuron in others (secondary eye). Rhabdome showed that arranged in PMeye and PLeye. But rhabdomes of AMeye and ALeye were irregular in retina. Except AMeye, incontinuous tapetum found in ALeye, PMeye, PLeye. Anterior median eye was similar to anterior lateral eye in length and posterior median eye similar to posterior lateral eye. Component size of eye were similar to 4 pairs eye in cornea. Size of lens, cell body, and rhabdome was similar not only anterior median eye and anterior lateral eye but also posterior median eye and posterior lateral eye. Vitreous body was large posterior median eye than others.
