HaCaT Keratinocytes and Primary Epidermal Keratinocytes Have Different Transcriptional Profiles of Cornified Envelope-Associated Genes to T Helper Cell Cytokines

  • HaCaT cells are the immortalized human keratinocytes and have been extensively used to study the epidermal homeostasis and its pathophysiology. T helper cells play a role in various chronic dermatological conditions and they can affect skin barrier homeostasis. To evaluate whether HaCaT cells can be used as a model cell system to study abnormal skin barrier development in various dermatologic diseases, we analyzed the gene expression profile of epidermal differentiation markers of HaCaT cells in response to major T helper (Th) cell cytokines, such as $IFN{\gamma}$, IL-4, IL-17A and IL-22. The gene transcriptional profile of cornified envelope-associated proteins, such as filaggrin, loricrin, involucrin and keratin 10 (KRT10), in HaCaT cells was generally different from that in normal human keratinocytes (NHKs). This suggests that HaCaT cells have a limitation as a model system to study the pathophysiological mechanism associated with the Th cell cytokine-dependent changes in cornified envelope-associated proteins which are essential for normal skin barrier development. In contrast, the gene transcription profile change of human ${\beta}2$-defensin (HBD2) in response to $IFN{\gamma}$, IL-4 or IL-17A in HaCaT cells was consistent with the expression pattern of NHKs. $IFN{\gamma}$ also up-regulated transglutaminase 2 (TGM2) gene transcription in both HaCaT cells and NHKs. As an alternative cell culture system for NHKs, HaCaT cells can be used to study molecular mechanisms associated with abnormal HBD2 and TGM2 expression in response to $IFN{\gamma}$, IL-4 or IL-17A.

  • Rolled products often have residual stresses or strip waves that are beyond the customer’s tolerance. To resolve this problem, skin-pass rolling is widely used during post-processing of such products. Because a short contact length compared to the strip width is a characteristic of skin-pass rolling, several numerical analyses have been previously conducted based on a two-dimensional approach. In the current study, a series of simulations was conducted using numerical analysis of three-dimensional elastic-plastic finite element method.

  • We study the isothermal flow of a dusty viscous incompressible conducting fluid between two types of boundary motions- oscillatory and non-oscillatory, under the influence of gravitational force. Within the frame work of some physically realistic approximations and suitable boundary conditions, closed form solutions were obtained for the velocity profiles and the skin friction of the particulate flow. These results show that for a constant pressure gradient, only the velocity profile of the fluid and the skin friction are unaffected by gravity, while magnetic field is seen to affect both the fluid, particle velocities and the skin friction. Thus, our results are extension of previous results in literature, and graphical demonstration of some these solutions have been presented.

  • We investigate the transient temperature response in biological tissue whose surface is exposed to alternately varying sinusoidal oscillation. Based on the Pennes bio-heat equation, we apply numerical analysis using a finite element method to find the effects of the physical properties of the skin layers. Three layers of tissue-epidermis, dermis, and subcutaneous-are considered as the solution region. We investigate the effects of different properties of the skin layers on the temperature profile. We also investigate the effects of the perfusion rate for the dermis, which is the most sensitive layer. The results show that the temperature profile of tissue depth has a discontinuous point when different physical properties are used.

  • Background Scalp defects may be caused by various etiological factors, and they represent a significant surgical and aesthetic concern. Various surgical techniques can be applied for reconstructive work such as primary closure, skin grafting, pedicled or free flaps. In this article, the authors share their clinical experience with scalp operations using the technique of local flaps and discuss the application of this method from the perspective of not only the size of the defect, but also in relation to the anatomical area, quality of surrounding tissue, and patient's condition. Methods During the period from December 2007 to December 2012, 13 patients with various scalp defects, aged 11 to 86 years, underwent reconstruction with local pedicle flaps. The indications were based on the patients' condition (age, sex, quality of surrounding tissue, and comorbidities) and wound parameters. Depending on the size of the defects, they were classified into three groups as follows: large, 20 to 50 $cm^2$; very large, 50 to 100 $cm^2$; extremely large, 100 $cm^2$. The location was defined as peripheral (frontal, temporal, occipital), central, or combined (more than one area). We performed reconstruction with 11 single transposition flaps and 1 bipedicle with a skin graft on the donor area, and 2 advancement flaps in 1 patient. Results In all of the patients, complete tissue coverage was achieved. The recovery was relatively quick, without hematoma, seroma, or infections. The flaps survived entirely. Conclusions Local flaps are widely used in scalp reconstruction since they provide healthy, stable, hair-bearing tissue and require a short healing time for the patients.

  • To measure precisely the blood velocity in the skin microcirculation, we have used time domain correlation (called Cross-Correlation) based on the processing of the backscattered RF signal obtained with a wideband echographic imaging transducer, although it is difficulties of adaptation of the pulsed wave system, because of the data processing in real time and the hardware problem. This dedicated technology based on a 20MHz echographic imaging system has been developed. We present how the experimental data, i.e. the backscattered RF signal, have to be analyzed. After RF lines realignment, stationary echo canceling procedure and correlation level control, a velocity profile has been obtained. In-vitro result show that velocity measurements as low as 0.1mm/sec attainable with a 80${\mu}m$ in axial resolution. We have also validated with in-vivo experimentation on the external ear of a rabbit using B-mode sector scanning image and M-mode image of a custom made 20MHz skin image system. The flow of the "auriculares caudales" vein, a microvessel of 600 m diameter, has been detected and studied. This technique will allow a more precise exploration of circulatory troubles in cutaneous pathologies.

A Numerical Study of Planar Laminar Impingement Jet with a Confinement Plate (제한면을 가지는 이차원 층류 충돌젯의 수치적 연구)

  • The planar laminar impingement jet with a confinement plate has been studied numerically. Discretzing the convection term with the QUICKER scheme, the full Navier-Stokes equations for fluid flow were solved using the well known SIMPLER algorithm. The flow characteristics with Reynolds number and jet exit velocity profile effects on it were considered for H=3, Re=200 - 2000. Results show that vortical flow forms in turn along the confinement and impingement plates as the Reynolds number increases and such a complicated flow pattern has never been reported prior. The jet exit velocity profile is shown to do an important role in determining the position of vortex flow and its size as well as in stagnation and wall jet flow region. Parabolic jet exit profile results in peak of skin friction 1.4-1.6 times greater than that of uniform profile. The channel height effects are also studied and shown to have an effect on flow pattern similar to that of Reynolds number. Also shown is that effects of the jet exit velocity profile becomes less significant over a certain channel height.

Quality Properties of Chicken Nugget with Various Levels of Chicken Skin (닭고기 너겟의 닭고기 스킨 첨가수준에 따른 품질 특성)

  • This study aimed to investigate the effect of chicken skin on the proximate composition, pH, color, texture profile analysis, and sensory properties of chicken nugget. Chicken nugget samples were prepared by adding the following amount chicken skin 0% (control), 5%, 10% and 15%. The moisture and protein contents of the samples decreased with an increase chicken skin levels. And frying yield, redness, yellowness, Hardness, gumminess and chewiness of samples decreased with increasing chicken skin levels. On the other hand, the fat contents and lighteness of sample increased with an increase chicken skin levels. Futhermore, juiciness and overall acceptability increased with an increasing chicken skin levels. Therefore, usages of chicken skin 10% can improve quality properties of chicken nugget.

Evaluation of Dose Variation according to Air Gap in Thermoplastic Immobilization Device in Carbon Ion (탄소입자 치료 시 열가소성 고정기구의 공기층에 따른 선량 변화 평가)

  • Purpose: The purpose of this study is to find out the dose variation according to thickness of the air gap between the patient's body surface and immobilization device in the treatment plan. Materials and Methods : Four conditions were created by adjusting the air gap thickness using 5 mm bolus, ranging from 0 mm to 3 mm bolus. Immobilization was placed on top in each case. And computed tomography was used to acquire images. The treatment plan that 430 cGy (Relative Biological Effectiveness,RBE) is irradiated 6 times and the dose of 2580 cGy (RBE) is delivered to 95% of Clinical Target Volume (CTV). The dose on CTV was evaluated by Full Width Half Maximum (FWHM) of the lateral dose profile and skin dose was evaluated by Dose Volume Histogram (DVH). Result: Results showed that the FWHM values of the lateral dose profile of CTV were 4.89, 4.86, 5.10, and 5.10 cm. The differences in average values at the on the four conditions were 3.25±1.7 cGy (RBE) among D95% and 1193.5±10.2 cGy (RBE) among D95% respectively. The average skin volume at 1% of the prescription dose was 83.22±4.8%, with no significant differences in both CTV and skin. Conclusion: When creating a solid-type immobilization device for carbon particle therapy, a slight air gap is recommended to ensure that it does not extend beyond the dose application range of the CTV.

Development of Definition of Parameters and Reference Scales for Texture Profiling of Frankfurter Sausages (소세지의 텍스처 프로필 수행을 위한 용어와 표준척도의 개발)

  • The texture profile of frankfurter sausages was carried out by comparing the domestic(A) and imported(B) products. Fourteen character notes were identified: elasticity, surface moisture, surface smoothness, center hardness, skin toughness, cohesiveness, denseness, chewiness, moisture release, cohesiveness of mass, lumpiness, graininess, skin separation, and oiliness. Reference scales and the $0{\sim}3$ point numerical scaling method were also established. Panelists evaluated sausage A harder than the sausage B. It had higher skin separation and elasticity scores requring more chewing. Sausage B had lower surface moisture and cohesiveness of mass, but higher moisture release and graininess. Panelists commented that the sausage B were oilier but had milder tastes.
